컴퓨팅 사고력 평가를 위한 3가지 상호보완적 접근 방안 (A Complementary Approach of Three Methods for Computational Thinking Assessment)

  • 컴퓨팅 사고력(CT: Computational Thinking)은 21세기 핵심 역량으로 주목받으며 소프트웨어교육을 통한 CT 역량 증진에 많은 관심이 모아지고 있으나, 효과적인 CT역량 평가 방법의 설계 및 구현에 대한 더 많은 연구가 필요하다. 본 연구는 한 학기 동안 CT역량 증진을 위한 비전공자 프로그래밍 수업을 수강한 52명의 예비교원을 대상으로 세 가지 CT 평가 방법을 활용하여 수업의 효과를 살펴보았다. CT 평가 방법은 (1) 사전-사후 베브라스 CT 검사, (2) 그룹 스크래치 프로젝트에 대한 Dr. Scratch 자동 평가, 그리고 (3) 예비교원의 스크래치 프로그래밍을 통한 CT역량 평가 지필 시험을 포함한다. 본 연구의 결과 CT 역량을 위한 평가 방법의 통합이 긍정적인 효과를 보이는 것으로 나타났다. 본 논문은 이러한 CT 평가 방법의 통합의 장점 및 시사점을 논의하고 있다.

배관부 감육 손상의 비파괴 평가를 위한 레이저 초음파 기술 적용 (Application of Laser Ultrasonic Technique for Nondestructive Evaluation of Wall Thinning in Pipe)

  • 원자력 발전소의 많은 배관들은 고온, 고압 환경에서 사용되고 있으며, 부식에 의한 감육 손상이 빈번하게 발생하고 있다. 이러한 배관의 감육 손상은 원전 설비가 노후화됨에 따라 점차 증가할 것으로 예상된다. 따라서, 배관 부식에 의한 원전 설비의 사고를 예방하기 위해 배관부의 감육 손상 및 부식 결함을 비파괴적인 방법으로 평가할 필요가 있다. 특히, 원자력 발전소의 배관에서 발생하는 감육 손상을 실시간으로 평가하기 위한 레이저 초음파 기술은 기존의 접촉식 방법이 가지는 한계를 넘어 접근이 어려운 부위에서도 측정이 가능하다. 본 연구에서는 비파괴, 비접촉식 평가를 위해 레이저를 이용한 광학적 방법을 적용하였다. 펄스 레이저로 초음파를 발생시키고 레이저 간섭계를 이용하여 초음파 신호를 획득하였으며, 먼저 감육 손상이 없는 배관에서의 초음파 신호를 검출하여 배관 내에서의 종파 속도를 측정하였다. 그리고, 배관의 두께대비 20, 30, 40, 50%의 인위적인 감육 배관을 제작하여 종파 속도를 측정하였다. 측정된 종파 속도를 이용하여 감육 배관의 두께를 계산할 수 있고, 감육 손상 부분(내부결함 깊이)의 정량적 평가도 가능하였다.

Analysis of CEA Expression and EGFR Mutation Status in Non-small Cell Lung Cancers

  • Background: The serum carcinoembryonic antigen (CEA) level can reflect tumor growth, recurrence and metastasis. It has been reported that epidermal growth factor receptor (EGFR) mutations in exons 19 and 21may have an important relationship with tumor cell sensitivity to EGFR-TKI therapy. In this study, we investigated the clinical value of EGFR mutations and serum CEA in patients with non-small cell lung cancer (NSCLC). Materials and Methods: The presence of mutations in EGFR exons 19 and 21 in the tissue samples of 315 patients with NSCLC was detected with real-time fluorescent PCR technology, while the serum CEA level in cases who had not yet undergone surgery, radiotherapy, chemotherapy and targeted therapy were assessed by electrochemical luminescence. Results: The mutation rates in EGFR exons 19 and 21 were 23.2% and 14.9%, respectively, with the two combined in 3.81%. Measured prior to the start of surgery, radiotherapy, chemotherapy and targeted treatment, serum CEA levels were abnormally high in 54.3% of the patients. In those with a serum CEA level <5 ng/mL, the EGFR mutation rate was 18.8%, while with 5~19 ng/mL and ${\geq}20ng/mL$, the rates were 36.4% and 62.5%. In addition, in the cohort of patients with the CEA level being 20~49 ng/mL, the EGFR mutation rate was 85.7%, while in those with the CEA level ${\geq}50ng/mL$, the EGFR mutation rate was only 20.0%, approximately the same as in cases with the CEA level<5 ng/mL. Conclusions: There is a positive correlation between serum CEA expression level and EGFR mutation status in NSCLC patients, namely the EGFR mutation-positive rate increases as the serum CEA expression level rises within a certain range (${\geq}20ng/mL$, especially 20~49 ng/mL). If patient samples are not suitable for EGFR mutation testing, or cannot be obtained at all, testing serum CEA levels might be a simple and easy screening method. Hence, for the NSCLC patients with high serum CEA level (${\geq}20ng/mL$, especially 20~49 ng/mL), it is worthy of attempting EGFR-TKI treatment, which may achieve better clinical efficacy and quality of life.

영광 원자력발전소 6호기 가동중검사 수형 경험 (The Experience of Inservice Inspection for Yonggwang Nuclear Power Plant Unit 6)

  • 원자력발전소 운전에 따른 경년열화 등에 의하여 원자력발전소 주요 기기 및 재료 등에 손상 발생 가능성이 있어 원자력법 및 관련 기술기준에서는 비파괴검사 방법을 이용하여 원자력발전소 주요 기기 및 배관의 용접부 등 취약 부위에 대한 건전성을 주기적으로 평가토록하고 있다. 이에 따라, 영광 6호기 가동중검사는 기기, 배관 및 구조물 비파괴검사, 압력용기 자동 초음파탐상검사, 원자로 내부 구조물 육안검사 및 증기발생기 전열관 와전류탐상검사로 구분하여 수행하였다. 원자력발전소 계통의 주요기기에 대한 비파괴검사 결과, 기기, 배관 및 구조물과 원자로 압력용기 용접부에 대해서는 특이 사항 발생 없이 적용 규격에 만족되고 건전한 것으로 최종 평가되었다. 특히, 배관 용접부에 대한 초음파탐상검사는 영광 5호기에서와 마찬가지로 ASME Code Sec. XI 1995년도 판에 따라 기량검증(Performance Demonstration : PD) 방법을 적용함으로써 검사 신뢰도를 확보하였다는데 큰 의미가 있다.

Non-Contrast Cine Cardiac Magnetic Resonance Derived-Radiomics for the Prediction of Left Ventricular Adverse Remodeling in Patients With ST-Segment Elevation Myocardial Infarction

  • Objective: To investigate the predictive value of radiomics features based on cardiac magnetic resonance (CMR) cine images for left ventricular adverse remodeling (LVAR) after acute ST-segment elevation myocardial infarction (STEMI). Materials and Methods: We conducted a retrospective, single-center, cohort study involving 244 patients (random-split into 170 and 74 for training and testing, respectively) having an acute STEMI (88.5% males, 57.0 ± 10.3 years of age) who underwent CMR examination at one week and six months after percutaneous coronary intervention. LVAR was defined as a 20% increase in left ventricular end-diastolic volume 6 months after acute STEMI. Radiomics features were extracted from the oneweek CMR cine images using the least absolute shrinkage and selection operator regression (LASSO) analysis. The predictive performance of the selected features was evaluated using receiver operating characteristic curve analysis and the area under the curve (AUC). Results: Nine radiomics features with non-zero coefficients were included in the LASSO regression of the radiomics score (RAD score). Infarct size (odds ratio [OR]: 1.04 (1.00-1.07); P = 0.031) and RAD score (OR: 3.43 (2.34-5.28); P < 0.001) were independent predictors of LVAR. The RAD score predicted LVAR, with an AUC (95% confidence interval [CI]) of 0.82 (0.75-0.89) in the training set and 0.75 (0.62-0.89) in the testing set. Combining the RAD score with infarct size yielded favorable performance in predicting LVAR, with an AUC of 0.84 (0.72-0.95). Moreover, the addition of the RAD score to the left ventricular ejection fraction (LVEF) significantly increased the AUC from 0.68 (0.52-0.84) to 0.82 (0.70-0.93) (P = 0.018), which was also comparable to the prediction provided by the combined microvascular obstruction, infarct size, and LVEF with an AUC of 0.79 (0.65-0.94) (P = 0.727). Conclusion: Radiomics analysis using non-contrast cine CMR can predict LVAR after STEMI independently and incrementally to LVEF and may provide an alternative to traditional CMR parameters.

  • 혼합분포를 가정한 신용평가연구에서 부도차주를 정상으로 예측하거나 정상차주를 부도로 예측하는 오류를 최소화하는 분류점을 추정하는 방법을 토론한다. 확률변수 스코어와 정상과 부도상태의 모수공간으로 정의된 확률밀도함수들에 대하여 강력검정과 일반화가능도비검정을 이용하여 최적분류점의 추정방법을 제안하고, ROC와 CAP 곡선에서 분류정확도를 측정하는 정확도(accuarcy)와 진실율(true rate)을 이용하여 이 측도를 최대로 하는 최적분류점을 확률밀도함수의 관계식으로 추정하는 방법을 제안한다. 다양한 정규분포에서 가설검정, 정확도 그러고 진실율을 이용하는 세가지 방법의 최적분류점을 구하고 각최적분류점에 대응하는 제 I 종과 제 II 종 오류합의 크기를 비교하여 효율성을 토론한다.

국내 허가사항에 반영된 약물 유전정보 분석 (Analysis of Pharmacogenetic Information in Korea Drug Labels)

  • Background: Pharmacogenomics is the study of how genetic mutations in patients affect their response to drugs. Pharmacogenomic studies aim to maximize drug effects and minimize adverse drug events. The Food and Drug Administration and the European Medicine Agency published guidelines for pharmacogenetics in 2005 and 2006, respectively; the Korean Ministry of Food and Drug Safety followed suit in 2015. Methods: This study analyzed pharmacogenomic information in the Korean Ministry of Food and Drug Safety's integrated drug information system to evaluate whether domestic pharmaceutical products reflect the current research on pharmacogenomic differences. Results: In June 2020, the Korean pharmacogenomic database contained genomic data on 90 compounds. Of these, 45 compounds were classified as "Antineoplastic and immunomodulating agents." The other 45 non-antineoplastic agents were in the following categories: Anti-infectives, Mental & behavior disorder, Hormone & metabolism related diseases, Cardiovascular system, Skin & subcutaneous tissue disease, Genito-urinary system and sex hormones, Blood and blood forming organs, Nervous system, Alimentary tract and metabolism, Musculo-skeletal system, and Other conditions including the respiratory system. In addition, 30 additives unrelated to the main ingredient were associated with genetic precautions. Conclusion: This study showed that antineoplastic and immunomodulating agents accounted for half the drugs associated with pharmacogenetic information. For antitumor and immunomodulatory drugs, genomic tests were recommended depending on the indication; this was in contrast to genomic testing recommendations for non-antineoplastic medications. Genomic tests were rarely requested or recommended for non-antineoplastic medications because the relationships between genotype and efficacy among those drugs were relatively weak.

Clinical Significance of Upregulation of mir-196a-5p in Gastric Cancer and Enriched KEGG Pathway Analysis of Target Genes

  • Background: miRNAs are relatively recently discovered cancer biomarkers which have important implications for cancer early diagnosis, treatment and estimation of prognosis. Here we focussed on expression of mir-196a-5p in gastric cancer tissues and cell lines so as to analyse its significance for clinicopathologic characteristics and generate enriched KEGG pathways clustered by target genes for exploring its potential roles as a biomarker in gastric cancer. Materials and Methods: The expression of mir-196a-5p in poorly, moderate and well differentiated gastric cancer cell lines compared with GES-1 was detected by RT-qPCR, and the expression of mir-196a-5p in gastric cancer tissues comparing with adjacent non cancer tissues of 58 cases were also assessed by RT-qPCR. Subsequently, an analysis of clinical significance of mir-196a-5p in gastric cancer and enriched KEGG pathways was executed based on the miRWalk prediction database combined with bioinformatics tools DAVID 6.7 and Mirfocus 3.0. Results: RT-qPCR showed that mir-196a-5p was up-regulated in 6 poorly and moderate differentiated gastric cancer cell lines SGC-7901, MKN-45, MKN-28, MGC-803, BGC-823, HGC-27 compared with GES-1, but down-regulated in the highly differentiated gastric cancer cell line AGS. Clinical data indicated mir-196a-5p to beup-regulated in gastric cancer tissues (47/58). Overexpression of mir-196a-5p was associated with more extensive degree of lymph node metastasis and clinical stage (P < 0.05; x2 test). Enriched KEGG pathway analyses of predicted and validated targets in miRWalk combined with DAVID 6.7 and Mirfocus 3.0 showed that the targeted genes regulated by mir-196a-5p were involved in malignancy associated biology. Conclusions: Overexpression of mir-196a-5p is associated with lymph node metastasis and clinical stage, and enriched KEGG pathway analyses showed that targeted genes regulated by mir-196a-5p may contribute to tumorgenesis, suggesting roles as an oncogenic miRNA biomarker in gastric cancer.

식품 내 벤조피렌 분석법의 기존 연구동향 및 라만분광법 기반기술 전망 (Conventional Analytical and New Raman Spectroscopy-Based Methods for Detecting Benzo[a]pyrene in Food: Review and Perspective)

  • 국내 식품공전에 기술되어 시행중인 식품 내 벤조피렌 분석법은 회수율, 검출 및 정량 한계, 효용성과 정확성 등이 검증되어 적용되고 있다. 그러나, 기존의 시험법에는 시료의 추출 및 분리, 정제 등의 복잡한 전처리 과정이 포함되어 있다. 환경 및 식품 내 위해물질 검출법의 연구 및 개발의 현 추세는 시험비용과 시간소모를 최소화하고 시험과정을 단순화할 수 있는 비표지, 비파괴 분석법을 통해 관리 현장에서의 신속대응을 지향하고 있다. 본 총설에서는 현행의 벤조피렌 분석법에 대해 정리하고, 그 한계점을 극복하기 위한 새로운 시도로 라만 분광법을 이용한 벤조피렌 검출법에 대해 소개하고자 한다.

Assessment of quantitative structure-activity relationship of toxicity prediction models for Korean chemical substance control legislation

  • Objectives For successful adoption of legislation controlling registration and assessment of chemical substances, it is important to obtain sufficient toxicological experimental evidence and other related information. It is also essential to obtain a sufficient number of predicted risk and toxicity results. Particularly, methods used in predicting toxicities of chemical substances during acquisition of required data, ultimately become an economic method for future dealings with new substances. Although the need for such methods is gradually increasing, the-required information about reliability and applicability range has not been systematically provided. Methods There are various representative environmental and human toxicity models based on quantitative structure-activity relationships (QSAR). Here, we secured the 10 representative QSAR-based prediction models and its information that can make predictions about substances that are expected to be regulated. We used models that predict and confirm usability of the information expected to be collected and submitted according to the legislation. After collecting and evaluating each predictive model and relevant data, we prepared methods quantifying the scientific validity and reliability, which are essential conditions for using predictive models. Results We calculated predicted values for the models. Furthermore, we deduced and compared adequacies of the models using the Alternative non-testing method assessed for Registration, Evaluation, Authorization, and Restriction of Chemicals Substances scoring system, and deduced the applicability domains for each model. Additionally, we calculated and compared inclusion rates of substances expected to be regulated, to confirm the applicability. Conclusions We evaluated and compared the data, adequacy, and applicability of our selected QSAR-based toxicity prediction models, and included them in a database. Based on this data, we aimed to construct a system that can be used with predicted toxicity results. Furthermore, by presenting the suitability of individual predicted results, we aimed to provide a foundation that could be used in actual assessments and regulations.
