Comparison of Bifidogenic Growth Stimulation Activities of Fermented Whey Prototypes

  • Fermented whey solution presenting bifidogenic growth stimulation (BGS) activity was processed as prototypes such as sterilized fermented whey (SFW), spray-dried fermented whey (SDFW), and freeze-dried fermented whey (FDFW) and their BGS activities were compared. In optical density ($OD_{600}$) test, the BGS activity of three prototypes, which showed similar activities, were significantly different with non-fermented whey solution adjusted to pH 4.5 as a control (P<0.05). In viable cell count test, SDFW had the most positive influence than other prototypes on the BGS activity even though the difference was not significant. However, the activities of all prototypes were significantly different than the negative control (no addition). These results indicate that the processed prototypes of fermented whey solution show BGS activities and might be commercialized, with further evidences, in animal or human studies.

Anaerobic Biodegradation of PCP in Japanese Paddy Soils

  • Seven soil samples were collected from paddy fields located nearby Nagoya city in Japan. All the soils were subjected to flooded condition and incubated with PCP at $30^{\circ}C$ for two months, and their anaerobic PCP degradation have been monitored by checking the PCP concentration of the soils at regular intervals. The degradation of PCP did not occur in the soils autoclaved two times before pre-incubation. On the other hand, all the soils showed significant PCP degradation in non-sterilized condition after 30 days of incubation, except far one soil sample (Yatomi), in which PCP was rarely degraded until 30 days of incubation. This result showed PCP disappearance in the pad(rf soils was mainly caused by microbiological activity, and depended upon the physicochemical characteristics of the soils.

  • For the purpose of evaluating the cleansing efficiency against Candida albicans detected frequently in patients with denture stomatitis, two denture cleansers with or without enzymes were studied under the same conditions. The results were as fellows: 1. Enzyme-contain denture cleanser was showed more Candida albicans lytic ability than non-enzyme-contained denture cleanser. 2. It was observed that Candida albicans lytic activity in further diluted manufacturerers' recommended concentration was decreased. 3. In fungicidal test, the enzyme-contained denture cleanser sterilized Candida albicans, and the non-enzyme-contained denture cleanser did not sterilize Candida albicans. 4. Sterilizing time of Candid albicans was needed for at least 60 minutes in enzyme-contained denture cleanser solution which was diluted with manufacturerers' recommended concentrations., and was needed for more times with further diluted manufacturerers' recommended concentrations. 5. In vitro growth test of Candida albicans on acrylic resin surface, the only enzyme-contained denture cleanser inhibited growth of Candida albicans, and it was observed that inhibiton ability of growth of Candida albicans on arrylic resin surface was decreased in further diluted manufacturerers' recommended concentrations.

Sterilization of Freeze Dried Manila Clam (Ruditapes philippinarum) Porridge for Immuno-Compromised Patients

  • This study was conducted to evaluate the combined effect of gamma irradiation and different conditions (vacuum packaging, antioxidant and freezing) on the microbiological and sensory characteristics of freeze dried Manila clam porridge (MCP) for immuno-compromised patient food. MCP can be sterilized at 1 kGy to 10 kGy. The initial counts of total aerobic bacteria and yeast molds in the non-irradiated MCP were $2.4{\pm}0.5$ and $1.2{\pm}0.3{\log}\;CFU\;g^{-1}$, respectively, but gamma irradiation significantly decreased the total aerobic bacteria to below the detection limit ($1{\log}\;CFU\;g^{-1}$) (5 kGy). Moreover, gamma irradiation effectively eliminated yeasts/molds at dose below than 1 kGy. However, gamma irradiation accelerated the increase of lipid oxidation and therefore, decreased the sensory characteristics of MCP as irradiation dose increased. To improve the sensory qualities of gamma irradiated MCP, combination treatment (vacuum packaging, 0.1% vitamin C) were applied. There was no significant difference in the overall acceptance scores between the combined-treatment sample (5.6 points) and the non-irradiated samples (6.0). The results indicate that combination treatment (vacuum packaging, 0.1% vitamin C) may help to maintain the quality of MCP. Therefore, it considered that irradiation of MCP with combined treatment and this is an effective method for the consumption as a special purpose food such as for space travel or immuno-compromised patients.

무균 로티퍼 Brachionus rotundiformis의 증식 (Growth of Axenic Rotifer Brachionus rotundiformis)

  • 이 연구에서는 해산어의 종묘 생산에 필요한 대량 배양용 로티퍼, Brachionus rotundiformis의 종 보존 방법으로서 무균 로티퍼의 이용을 검토했다. 로티퍼는 개량형 항생제 혼합액 AM9을 사용하여 무균 처리한후, 무균 Nannochloropsis oculata를 먹이로 공급하여 연속 유지 배양했다. 각각 다른 시기의 3회 (trial 1, 2 and 3)에 걸친 로티퍼 배양결과, 무균 배양 (axenic culture)에서는 비교적 안정적인 증식을 보인 반면, 유균 배양 (non-axenic culture)에서 로티퍼의 증식은 불안정적으로 각 시기에 따라서 심한 증식의 차이가 관찰되었다. 이 결과는 유균 배양에서 로티퍼의 증식에 직접적으로 악영향을 미치는 박테리아가 무균 처리과정을 통해서 제거되었음을 증명한다. 로티퍼 무균 배양 방법의 일반화 또는 대량 배양용 로티퍼의 씨앗으로서의 무균 로티퍼의 이용은 로티퍼의 안정 배양은 물론 유용 수산 생물의 대량 폐사를 일으키는 원인 생물을 사전에 제거할 수 있는 유효한 방법이 될 수 있다고 생각된다.

Sterilization effect of atmospheric pressure non-thermal air plasma on dental instruments

  • PURPOSE. Autoclaves and UV sterilizers have been commonly used to prevent cross-infections between dental patients and dental instruments or materials contaminated by saliva and blood. To develop a dental sterilizer which can sterilize most materials, such as metals, rubbers, and plastics, the sterilization effect of an atmospheric pressure non-thermal air plasma device was evaluated. MATERIALS AND METHODS. After inoculating E. coli and B. subtilis the diamond burs and polyvinyl siloxane materials were sterilized by exposing them to the plasma for different lengths of time (30, 60, 90, 120, 180 and, 240 seconds). The diamond burs and polyvinyl siloxane materials were immersed in PBS solutions, cultured on agar plates and quantified by counting the colony forming units. The data were analyzed using one-way ANOVA and significance was assessed by the LSD post hoc test (${\alpha}$=0.05). RESULTS. The device was effective in killing E. coli contained in the plasma device compared with the UV sterilizer. The atmospheric pressure non-thermal air plasma device contributed greatly to the sterilization of diamond burs and polyvinyl siloxane materials inoculated with E. coli and B. subtilis. Diamond burs and polyvinyl siloxane materials inoculated with E. coli was effective after 60 and 90 seconds. The diamond burs and polyvinyl siloxane materials inoculated with B. subtilis was effective after 120 and 180 seconds. CONCLUSION. The atmospheric pressure non-thermal air plasma device was effective in killing both E. coli and B. subtilis, and was more effective in killing E. coli than the UV sterilizer.

전기분해 격막 및 전해액에 따른 전해수의 특성 비교 (Characteristics of Electrolyzed Water Manufactured from Various Electrolytic Diaphragm and Electrolyte)

  • 본 연구에서는 전기분해 격막 및 전해액에 따른 전해산화 수의 물리적 특성 및 미생물의 표면살균 효과를 검토하였다. 격막 방식의 전기분해수 제조 시스템의 최적조건은 간극이 1.0 mm 20% NaCl 첨가량이 6 mL/mm 일 때 제조된 전해산화수의 물성치가 ORP 1,170 mV 수준, HClO 함량 100 ppm 수준, pH 2.5 수준으로 가장 우수하게 나타났으며, 1단 방식보다는 2단의 전기분해 방식이 물성 측면에서 우수함을 알 수 있었다. 무격막 방식의 전해수는 간극이 1.0 mm, 20% NaCl 첨가량이 4 mL/mm 일 때 격막 방식의 최적 조건과 가장 유사하게 나타났으나 차아염소산 함량은 132∼266 ppm 수준, pH는 9 수준으로 격막 방식과는 크게 달라지는 것을 알 수 있었다. 전해산화수는 실온에서 밀폐 용기에 보관한 경우에 있어 격막 및 무격막 방식에서 각각 1,150 및 750 mV 수준, 100 ppm 수준을 1개월 이상 유지할 수 있었다. 격막 및 무격막 방식으로 제조한 전해산화수는 Salmonella typhimurium 등 12 균주가 초기 $10^{5}$$10^{6}$CFU/ml에서 30초 이내에 모두 사멸되는 것으로 나타났다. 전해액으로 NaCl 및 CaCl$_2$, KCl을 사용하여 제조한 전해산화수의 ORP 및 HCIO 함량은 NaCl, KCl, CaCl$_2$순으로 높게 나타났으나 큰 차이는 없었으며, 미생물 사멸효과는 동일하게 나타났다.

무살균 배지를 이용한 느타리버섯 재배 (Cultivation of Oyster Mushroom Using Non Sterilized Media)

  • NSM으로 무살균 재배를 여러 가지 배지를 혼합하여 균사생장과 밀도, 오염율을 실험한 결과는 다음과 같다. 첫째, 방울솜을 주재료로 하여 옥수수강과 비트, 미강을 각각 10, 20, 30, 40%를 혼합하였을 때 옥수수강, 미강, 비트 순으로 균사생장과 밀도가 양호한 결과를 보였으며 방울솜:옥수수강을 80 : 20(V/V)으로 혼합한 경우가 가장 양호하다. 특히 방울솜에 비트를 10% 이상 혼합하면 균사생장과 밀도가 현저히 불량한 특징이 있다. 둘째, 면자각을 주재료로 하여 옥수수강과 비트, 미강을 각각 10, 20, 30,40%를 혼합하였을 때 비트, 미강, 옥수수강 순으로 양호한 것으로 나타남에 따라 방울솜과 반대의 경향을 나타내었다. 특히 면자각에 미강을 20%이상 혼합하면 잡균오염도 높아질뿐만 아니라 균사생장과 밀도도 현저히 낮아지는 특징이 있다. 셋째, 방울솜을 주재료로 하여 옥수수강과 비트를 각각 혼합한 경 우 방울솜:옥수수강:비트를 70:20:10(V/V)로 혼합한 배지가 가장 양호 하였다. 넷째, 면자각을 주재료로 하여 옥수수강과 비트를 각각 혼합한 경우 면자각:옥수수강:비트를 70:20: 10(V/V)로 혼합한 배지가 가장 양호하였다. NSM의 적정 약제 농도별로는 500배가 가장 좋았으나 2,000배액에서도 균사생장은 비교적 좋은 결과를 나타내었으나 잡균오염율은 희석을 많이 하면 오염율이 높았다. NSM 배지의 적정수분함량은 77%가 적당하였으며, $25^{\circ}C$에서 배양하였을 때 균사밀도와균사속도가 양호하였다. NSM에 배지를 침지한 시간별로 조사한 결과 침지한 즉시 꺼내어 접종한 것이나 32시간 동안 침지한 것이나 동일한 효과가 있었으며 48시간 이상 침지하면 균사속도와 균사밀도가 약간 나빠지는 경향이 있었다. 따라서 NSM에 침지한 즉시 꺼내어 접종하면 균사생장과 균사밀도가 양호하였다. 즉 N.S.M을 이용하여 느타리버섯 폐면재배에 있어서 방울솜을 주재료로 하여 옥수수강, 비트, 미강을 혼합하여 균사생장, 균사밀도, 잡균오염도를 조사한 결과, 방울솜에 옥수수강을 20~30% 혼합하였을 때 균사생장이 76mm/10일 정도로 가장 양호하였고 균사밀도도 양호하였다.

Increased Salinity Tolerance of Cowpea Plants by Dual Inoculation of an Arbuscular Mycorrhizal Fungus Glomus clarum and a Nitrogen-fixer Azospirillum brasilense

  • Pot greenhouse experiments were carried out to attempt to increase the salinity tolerance of one of the most popular legume of the world; cowpea; by using dual inoculation of an Am fungus Glomus clarum and a nitrogen-fixer Azospirillum brasilense. The effect of these beneficial microbes, as single- or dual inoculation-treatments, was assessed in sterilized loamy sand soil at five NaCl levels ($0.0{\sim}7.\;2ds/m$) in irrigating water. The results of this study revealed that percentage of mycorrhizal infection, plant height, dry weight, nodule number, protein content, nitrogenase and phosphatase activities, as well as nutrient elements N, P, K, Ca, Mg were significantly decreased by increasing salinity level in non-mycorrhized plants in absence of NFB. Plants inoculated with NFB showed higher nodule numbers, protein content, nitrogen concentration and nitrogenase activities than those of non-inoculated at all salinity levels. Mycorrhized plants exhibited better improvement in all measurements than that of non-mycorrhized ones at all salinity levels, especially, in the presence of NFB. The concentration of $Na^+$ was significantly accumulated in cowpea plants by rising salinity except in shoots of mycorrhizal plants which had $K^+/Na^+$ ratios higher than other treatments. This study indicated that dual inoculation with Am fungi and N-fixer Azospirillum can support both needs for N and P, excess of NaCl and will be useful in terms of soil recovery in saline area.

Electron Beam 조사가 인삼분말의 색도 및 관능적 품질에 미치는 영향 (Effects of Electron-Beam Irradiation on Color and Organoleptic Qualities of Ginseng Powders)

  • Electron beam, electrically produced from an electron accelerator, was compared with gamma energy in terms of its influence on color and organoleptic qualities of ginseng powders when exposed to the energy used for their microbial decontamination. Hunter color L and b values were suitable for measuring color characteristics of ginseng powders, which were not significantly changed by the exposure to 5 to 7.5 kGy electron beam and gamma energy. Fifty percent ethanol extracts of irradiated ginseng powders at 10 key showed negligible differences from the non-irradiated control in the pattern of absorption spectra at 280∼800 am, but showed increased values in overall color difference (AE) as compared with powdered samples. Irradiation more than 10 kGy and storage at ambient temperature for 4 months caused browning of powdered samples. Irradiation at more than 10 kGy of electron beam was found a critical level to bring about appreciable changes (p<0.05) in or-ganoleptic qualities such as color and odor of sterilized samples, and red ginseng powder was more susceptible than white one to organoleptic changes by irradiation.
