The Application of Thera-band on Non-weight Bearing Leg Influence Hip Abductor Activities During Pelvic Drop Exercise in Patients With Gluteus Medius Weakness

  • Background: The weakness of the gluteus medius (GM) is associated with various musculoskeletal disorders. The increasing GM activity without synergistic dominance should be considered when prescribing pelvic drop exercise (PD). Isometric hip extension or flexion of the non-weight bearing leg using thera-band at the ankle during PD may influence hip abductor activities. Objects: To determine how isometric hip extension or flexion of the non-weight bearing leg using thera-band at the ankle during PD influences the activities of three subdivisions of GM (anterior, GMa; middle, GMm; posterior, GMp), tensor fasciae latae (TFL), contralateral quadratus lumborum (QL), and GMp/TFL, GMm/QL activity ratios in patients with GM weakness. Methods: Twenty-three patients with GM weakness were recruited. Three types of PD were performed: PD, PD with an isometric hip extension of the non-weight bearing leg (PDE), and PD with an isometric hip flexion of the non-weight bearing leg (PDF). Surface electromyography (SEMG) was used to measure hip abductor activities. One-way repeated-measures analysis of variance was used to assess the statistical significance of muscle activities and muscle activity ratios. Results: GMa, GMm, and GMp activities were significantly greater during PDF than during PD and PDE (p < 0.001, p = 0.001; p = 0.001, p = 0.005; p = 0.004, p = 0.004; respectively). TFL activity was significantly greater during PDE than during PD and PDF (p < 0.001, p < 0.001, respectively). QL activity was significantly greater during PDF than during PD (p = 0.003). GMp/TFL activity ratio was significantly lower during PDE than during PD and PDF (p = 0.001, p = 0.001, respectively). There were no significant differences in the GMm/QL activity ratio. Conclusion: PDF may be an effective exercise to increase the activities of all three GM subdivisions while minimizing the TFL activity in patients with GM weakness.

Effects of Field-Grown Genetically Modified Zoysia Grass on Bacterial Community Structure

  • Herbicide-tolerant Zoysia grass has been previously developed through Agrobacterium-mediated transformation. We investigated the effects of genetically modified (GM) Zoysia grass and the associated herbicide application on bacterial community structure by using culture-independent approaches. To assess the possible horizontal gene transfer (HGT) of transgenic DNA to soil microorganisms, total soil DNAs were amplified by PCR with two primer sets for the bar and hpt genes, which were introduced into the GM Zoysia grass by a callus-type transformation. The transgenic genes were not detected from the total genomic DNAs extracted from 1.5 g of each rhizosphere soils of GM and non-GM Zoysia grasses. The structures and diversities of the bacterial communities in rhizosphere soils of GM and non-GM Zoysia grasses were investigated by constructing 16S rDNA clone libraries. Classifier, provided in the RDP II, assigned 100 clones in the 16S rRNA gene sequences library into 11 bacterial phyla. The most abundant phyla in both clone libraries were Acidobacteria and Proteobacteria. The bacterial diversity of the GM clone library was lower than that of the non- GM library. The former contained four phyla, whereas the latter had seven phyla. Phylogenetic trees were constructed to confirm these results. Phylogenetic analyses of the two clone libraries revealed considerable difference from each other. The significance of difference between clone libraries was examined with LIBSHUFF statistics. LIBSHUFF analysis revealed that the two clone libraries differed significantly (P<0.025), suggesting alterations in the composition of the microbial community associated with GM Zoysia grass.

  • 유전자변형 작물이 절지동물에 미칠 수 있는 잠재적인 부정적 영향은 유전자변형 작물의 주요한 환경위해성의 하나로 여겨지고 있다. 본 연구에서는 PPO (protoporphyrinogen oxidase) 저해 제초제 내성 유전자변형 벼가 절지동물에 미칠 수 있는 영향을 평가하기 위하여 절지동물의 다양성과 군집구조를 조사하였다. 절지동물은 야외포장에서 벼의 생육기간 동안 황색점착트랩을 이용하여 채집하였다. 유전자변형 벼는 채집된 절지동물군집의 다양도 지수에 유의한 영향을 주지 않았다. 또한 다변량분석(PerMANOVA, NMDS) 결과에서도 절지동물군집 구조는 채집시기에 따라 달랐지만 벼의 유전형(유전자변형 또는 비변형)에 의해 영향을 받지 않았다.

논문 및 특허의 분석을 통한 국내 GM식물의 개발 현황 (Current status on the development of GM plants based on the published articles and patents in Korea)

  • 국내의 GM식물개발과 관련된 R&D사업 중 대표적이라고 할 수 있는 "바이오그린21사업"과 작물유전체기능연구사업단에서 지난 2007년부터 2009년 말 현재까지의 사업성과 중 논문과 동록 된 특허를 분석하여 본 결과 두개의 사업단을 통하여 총 1,212건의 SCI논문과 451건의 비 SCI논문, 그리고 348건의 특허를 등록한 것으로 조사되었다. 이중에서 바이오그린21사업단에서는 6.0%의 SCI 논문, 10.2%의 비SCI논문, 12.6%의 등록된 특허가 그리고 작물유전체기능연사업단의 경우에는 15.7%의 SCI논문, 21.1%의 비SCI 논문, 81.6%의 등록된 특허가 GM식물의 개발과 직접적으로 관련된 것으로 조사되었다. 개발하고자 하는 GM식물의 농업적 특성은 생물학적 스트레스 또는 무생물적 스트레스에 저항성을 갖는 작물 특히 벼와 고추가 가장 많았으나 포장시험까지 진행된 연구 성과는 아직 극히 저조한 것으로 밝혀졌다. 이러한 조사 결과는 국내에서 현재 실험실 또는 온실 단계에서 검정된 초기 단계의 GM식물의 조기상용화를 위하여 보다 많은 연구비가 투자되어야 할 것을 시사하였다.

Effects of Functional Electrical Stimulation (FES) on the Temporal-spatial Gait Parameters and Activities of Daily Living in Hemiplegic Stroke Patients

  • PURPOSE: This study examined the effects of functional electrical stimulation (FES) on temporal-spatial gait and the activities of daily living in hemiplegic stroke patients. METHODS: The subjects were 29 hemiplegic stroke patients (57.7 ± 10.3). The patients walked at a self-controlled speed in four states: (1) walking without FES (non-FES), (2) walking with FES on the gluteus medius in the stance phase (GM), (3) walking with FES on the common peroneal nerve and tibialis anterior in the swing phase (PT), (4) walking with both GM and PT. A GAITRite system, Timed-Functional Movements battery, and Timed UP and Go test were used to measure the variables. RESULTS: Significant improvements were observed in all variables of the GM+PT, GM, and PT states compared to the non-FES state (p < .05). There were significant improvements in the GM+PT state compared to GM and PT states (p < .05). Moreover, significant improvements were noted in the single support time on the affected side, backward walking 10ft, and side stepping 10ft on the affected side of the GM state compared to the PT state (p < .05). There were significant improvements in the stride length on the affected side and side stepping 10ft on the unaffected side of the PT state compared to the GM state (p < .05). CONCLUSION: FES is effective in improving the temporal-spatial gait and activities of daily living in hemiplegic stroke patients.

Assessment of Soil Microbial Communities in Carotenoid-Biofortified Rice Ecosystem

  • This study was conducted to investigate the effect of Psy-2A-CrtI (PAC), a genetically modified (GM) rice with enhanced ${\beta}$-carotene, on the soil microbial community. The soil used to cultivate GM rice and its wild-type, Nakdong, was analyzed for population density, denaturing gradient gel electrophoresis (DGGE), and pyrosequencing. It was found that the bacterial, fungal and actinomycetes population densities of the PAC soils were within the range of those of the non-GM rice cultivar, Nakdong. The DGGE banding patterns of the GM and non-GM soils were also similar, suggesting that the bacterial community structures were stable within a given month and were unaffected by the presence of a GM plant. The pyrosequencing result showed a temporal difference in microorganism taxon and distribution ratio, but no significant difference between GM and non-GM was found. The persistence of the transgene DNA in the plant and surrounding soil were investigated for different time periods. There were differences in the persistence within the plant depending on the gene, but they could not be detected after 5 weeks. Also the transgenes were not detected in the surrounding soil. These results indicate that soil microbial communities are unaffected by the cultivation of a PAC rice within the experimental time frame.

바이러스저항성 삼각주 재배가 토양 미생물상에 미치는 영향 (Impact of Virus-resistant Trigonal Cactus Cultivation on Soil Microbial Community)

  • 본 연구는 CVX 바이러스저항성 삼각주 재배가 토양 미생물에 미치는 영향과 수평적 유전자 이동성을 확인하기 위해 수행되었다. 생육시기별 토양 세균과 방선균 군집밀도는 형질전환 삼각주 재배 토양의 미생물 군집밀도와 비형질전환 삼각주 군집밀도가 유사하여 토양 미생물에 미치는 영향은 유사할 것으로 추정되었다. 토양 미생물의 우점종은 Proteobacteria, Uncultured archaeon와 Uncultured bacterium으로 나타났으며, 형질전환 삼각주 재배 토양의 우점종과 비율은 거의 일정하게 유지되었다. 근권 토양 DNA의 DGGE 분석을 통해 형질전환 삼각주와 비형질전환 삼각주 토양 미생물 군집의 profile 변화는 나타나지 않았다. 형질전환 삼각주와 비형질전환 삼각주 재배 토양의 화학성은 차이가 없었다. 형질전환 삼각주에 도입된 유전자로 토양 DNA에 대한 PCR 분석결과, 도입 유전자의 잔존성이 길지 않아 수평적 유전자 이동 가능성은 희박할 것으로 추정되었다.

Comparison of nutrition, anti-nutritional factors of rice straw and microbial composition in soil according to GM and non-GM rice field

  • The study was conducted to evaluate differences of growth characteristics of rice cultivated in two different regions (Cheonan and Jeonju). It focused on nutritional composition and anti-nutritional factors of rice straw produced from 21 rice varieties including GM rice (Iksan 483). The range of general nutrition ingredient is that crude was 0.97 ~ 3.2 %, carbohydrate was 67.45 ~ 80.01 %, crude protein was 1.46 ~ 4.81 %, crude ash was 6.52 ~ 18.96 %, crude fiber was 25.77 ~ 40.02 %, NDF was 51.84 ~ 67.77 %, ADF was 27.11 ~ 40.44 %, calcium was 0.49 ~ 5.18 mg/g and phosphorous was 0.26 ~ 2.77 mg/g. The general nutritional contents of GM rice were included above range. The range of phytic acid of rice straws cultivated in Cheonan and Jeonju was 0 ~ 0.056 mg/ml and 0 ~ 0.059 mg/ml, respectively. The phytic acid content of GM was 0.033 mg/ml, which was in the range of the content of rice straw in Cheonan and Jeonju. The range of trypsin inhibitor of rice straws cultivated in Cheonan and Jeonju was 0.061 ~ 0.461 TIU/mg and 0 ~ 1.278 TIU/mg, respectively. The trypsin acid content of GM was 0.461 TIU/mg, which was in the range of the content of rice straw in Cheonan and Jeonju. In addition, we investigated microbial community from each soil sample by using metagenomics sequencing based on rRNA microbial diversity in order to inspect indirect changes of soil environment with cultivation of GM rice. Metagenomics analysis was carried out using soil samples cultivated with GM and non-GM rice for before transplanting, young panicle differentiation stage, heading stage, and ripening stage. Beta diversity of microbial community in both soil environments were calculated by using Bray-Curtis distance method and showed low value with an average of 0.24 (dissimilarity = 1). As a result, it was confirmed that the cultivation of GM does not give a significant effect on the change of microbial composition in soil. Therefore, Our study demonstrates that there is no difference in the composition of soil microorganism due to GM and non-GM rice.

Molecular Analysis of Bacterial Community Structures in Paddy Soils for Environmental Risk Assessment with Two Varieties of Genetically Modified Rice, Iksan 483 and Milyang 204

  • The impacts of planted transgenic rice varieties on bacterial communities in paddy soils were monitored using both cultivation and molecular methods. The rice field plot consisted of eighteen subplots planted with two genetically modified (GM) rice and four non-GM rice plants in three replicates. Analysis with denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S rRNA genes revealed that the bacterial community structures were quite similar to each other in a given month, suggesting that there were no significant differences in bacterial communities between GM and non-GM rice soils. The bacterial community structures appeared to be generally stable with the seasons, as shown by a slight variation of microbial population levels and DGGE banding patterns over the year. Comparison analysis of 16S rDNA clone libraries constructed from soil bacterial DNA showed that there were no significant differences between GM and non-GM soil libraries but revealed seasonal differences of phyla distribution between August and December. The composition profile of phospholipid fatty acids (PLFA) between GM and non-GM soils also was not significantly different to each other. When soil DNAs were analyzed with PCR by using primers for the bar gene, which was introduced into GM rice, positive DNA bands were found in October and December soils. However, no bar gene sequence was detected in PCR analysis with DNAs extracted from both cultured and uncultured soil bacterial fractions. The result of this study suggested that, in spite of seasonal variations of bacterial communities and persistence of the bar gene, the bacterial communities of the experimental rice field were not significantly affected by cultivation of GM rice varieties.

유전자변형 배추를 섭취한 마우스 장기에서의 Housekeeping Gene의 발현 분석 (Analysis of Housekeeping Gene Expression in Mice Administered to GM and non-GM Cabbage)

  • 유전자 변형 작물은 생산성 측면에서 많은 장점이 있지만 이를 섭취할 경우 잠재적인 위험 요소들에 의해 많은 문제가 대두대고 있다. 본 연구는 저항성유전자를 이입한 배추에서 Profillin, Tubulin-${\alpha}$ (Tub-${\alpha}1$), Heat-shock protein (Bchsp 17.6) and Ubiquitin conjugating enzyme (UBE)의 발현과 이를 30일간 섭취한 마우스에서 ${\beta}$-actin(${\beta}$-act), ${\beta}$-2-microglobulin (B2m), Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and ${\beta}$-glucuronidase (Gus)의 발현 정도를 RT-PCR을 통해 알아보았다. 실험 결과 저항성유전자를 이입한 배추와 그렇지 않은 배추의 유전자 발현 패턴은 큰 차이를 보이지 앓았으며, 이를 섭취한 마우스 장기에서도 발현에 따른 큰 차이는 나타나지 않았다.
