The Hypernodulating nts Mutation Induces Jasmonate Synthetic Pathway in Soybean Leaves

  • Symbiotic nitrogen fixation with nitrogen-fixing bacteria in the root nodules is a distinctly beneficial metabolic process in legume plants. Legumes control the nodule number and nodulation zone through a systemic negative regulatory system between shoot and root. Mutation in the soybean NTS gene encoding GmNARK, a CLAVATA1-like serine/threonine receptor-like kinase, causes excessive nodule development called hypernodulation. To examine the effect of nts mutation on the gene expression profile in the leaves, suppression subtractive hybridization was performed with the trifoliate leaves of nts mutant 'SS2-2' and the wild-type (WT) parent 'Sinpaldalkong2', and 75 EST clones that were highly expressed in the leaves of the SS2-2 mutant were identified. Interestingly, the expression of jasmonate (JA)-responsive genes such as vspA, vspB, and Lox2 were upregulated, whereas that of a salicylate-responsive gene PR1a was suppressed in the SS2-2 mutant. In addition, the level of JA was about two-fold higher in the leaves of the SS2-2 mutant than in those of the WT under natural growth conditions. Moreover, the JA-responsive gene expression persists in the leaves of SS2-2 mutant without rhizobia infection in the roots. Taken together, our results suggest that the nts mutation increases JA synthesis in mature leaves and consequently leads to constitutive expression of JA-responsive genes which is irrelevant to hypernodulation in the root.

Influence of soybean cyst nematode on growth and yield of soybean (콩씨스트선충이 콩생육 및 수량에 미치는 영향)

  • In pot experiment, relations between soybean cyst nematode, Heterodera glycines, and growth and yield of soybean were followed. Soybean growth and nodule development in plots which inoculated more than 16 cysts were reduced greatly from two month after inoculation. Tolerance limit was about 20 cysts per 500g of soil at seeding period. There was $60\%$ loss of grain yield 낀hen nematode population was 1 cyst/g of soil at seeding period. Reproduction of soybean cyst :nematode was good when initial population had been low. So, the final population was no difference depend on inoculation density.

  • A putative protein encoded by Arabidopsis AtMTN3 gene, a homologue of Medicago truncatula MTN3, consists of 285 amino acid residues, and has a predicted molecular mass of 31.5 kDa and a calculated pI of 9.1. Primary amino acid sequence analyses have revealed that the protein contains seven putative transmembrane regions with N-terminus oriented to the outside of the membrane. The AtMTN3 protein shows overall 16.4% of amino acid identity with the rat GALR3 protein, known to be a G-protein-coupled receptor. The gene is present as a single copy in the Arabidopsis genome, and expressed in aerial parts but not in roots of Arabidopsis. Therefore, AtMTN3 appears not to be specifically involved in Rhizobium-induced nodule development, as was predicted for the MTN3 gene. These proteins possibly mediate signal transmission through G-protein-coupled pathways during general interactions between plants and symbiotic or pathogenic microbes.

  • Air-operated valve is one of principal valves that are using to control fluid flow. A period diagnosis for safety of power plants is necessary. But there are many difficulties such as economic loss caused by intone of high cost devices and a matter hard to deal with users. In this study we developed the diagnosis system that usersofpower plants are easy to handle.   • A 1.2KW Proton Exchange Membrane Fuel Cell(PEMFC) stack(Nexa Power Module) is installed to motor driven general scooter. The 24V, 650W BLDC motor is used for actuator and 2nd battery(24V, 4Ah) is used for stable starting of this motor. The operating information of Nexa Power Nodule is transmitted by RS-232 serial communication to notebook PC, and that data is analysed and displayed by the LabVIEW monitoring system.

  • This paper resents a virtual forming system to simulate deep drawing process for stress-strain information by utilizing virtual system designed using Virtual Reality Modeling Language (VRML) and computer aided analysis (CAE) tool. The CAE tool to calculate stress, strain, and deformation is designed using Finite Element Method. Stress distributions and deformation profiles as well as the operation of forming machine can be simulated and visualized in the web. The developed system consists of three modules, input module, virtual forming machine module, and output module. The input nodule was designed using HTML and ASP. The input data for FEM calculation is directed to the forming machine module for calculation. The results from the forming machine module can be visualized through output module as well as the forming process simulation.

  • Legume plants develop root nodules to recruit nitrogen-fixing bacteria called rhizobia. This symbiotic relationship allows the host plants to grow even under nitrogen limiting environment. Since nodule development is an energetically expensive process, the number of nodules should be tightly controlled by the host plants. For this purpose, legume plants utilize a long-distance signaling known as autoregulation of nodulation (AON). AON signaling in legumes has been extensively studied over decades but the underlying molecular mechanism had been largely unclear until recently. With the advent of the model legumes, L. japonicus and M. truncatula, we have been seeing a great progress including isolation of the AON-associated receptor kinase. Here, we summarize recent studies on AON and discuss an updated view of the long-distance control of nodulation.

  • The effect of Ganoderma lucidum (Polyporaceae) extract on initiation and promotion of liver preneoplastic lesions in chemical carcinogenesis were examined in male rats. Male Fisher 344 rats, 8 weeks old, were injected i.p. with diethylnitro-samine(DENA` 200 mg/kg) as an initiator and were fed on diet containing 0.02%-acetaminofluorene (2-AAF) as a promoter for 2 weeks (group 1, 2` 2-4 week) and were fed on basal diet (group 3 and 4). The rats in test groups(group 1 and 3) were fed on diet containing 0.1% of Ganoderma lucidum extract for 6 weeks (2-8 week).

  • Aqualyx (Marllor International Ltd, Rimini, Italy) was originally developed in Italy by Professor Pasquale Motolese and has been commercially available since 2009. It is a deoxycholate, aqueous gelatinous solution mixed with saline and buffering compounds. It is the only drug approved by the European Union for the reduction in localized fat. Aqualyx is sold exclusively to doctors and nurses trained in intralipotherapy. In the case of our patient, the product administered was advertised as Aqualyx, but was not administered by a trained health professional and was administered too superficially. The patient developed severe pain following the injection and was unable to sit for several weeks. There was localized skin necrosis, and palpable collections where the injection was administered. Our initial suspicion was development of an abscess or hematoma. To characterize further, we arranged an ultrasound scan that showed a "superficial hypoechoic lesion" but no deeper infection or spread. The numerous painful nodules ruptured onto the skin surface, resulting in purulent and bleeding lesions. This case demonstrates the importance of appropriate training and competence in performing cosmetic procedures including injections and fat dissolving treatments.
