Organic Analysis of Charred Residues on the Pottery in the Proto-Three Kingdom from Joong-do Site, Chuncheon (춘천 중도 유적에서 출토된 원삼국시대 토기 탄착물에 대한 화학적 분석)

  • We studied for the chemical characterizations of the charred residues obtained from the ancient potteries in the Proto-Three Kingdom period from archaeological sites in Joong-do, Chuncheon. Organic components of the charred residues were extracted and analysed using mass spectrometry and infrared spectroscopy. Lipid profiles from these samples were not identified in gas chromatography-mass spectrometry. Bulk stable isotope analyses of charred residues was used to infer an average values of the foods prepared. The average carbon isotope values (${\delta}^{13}C$) of the residues are $-14.7{\pm}2.8$‰ (ranging from -8.7‰ to -18.4‰, n=9), and nitrogen isotope values (${\delta}^{15}N$) are $6.2{\pm}1.1$‰ (ranging from -4.4‰ to -7.6‰, n=9). This is the first approach to analyse charred residues using stable isotopic method in Korea. Charred food residues on the interior surface of archaeological pottery can provide valuable information about pottery use and dietary habits of its population.

The Characteristics of Fish Community and Food Web in Eutrophic Agricultural Reservoir, Jeondae (부영양 농업용저수지인 전대지의 어류군집 및 먹이망 특성)

  • Fish community was investigated in eutrophic Jeondae agricultural reservoir from April to November 2012. The food web structure of major fish species of Jeondae reservoir showed that due to serious organic pollution, the hyper-eutrophic condition with high chemical oxygen demand (COD), chlorophyll-a (Chl-a), total nitrogen (TN) and total phosphorus (TP), as estimated by stable isotope analysis. Total of 679 individuals were collected from the reservoir, and 9 species were identified. Interestingly, Lepomis macrochirus and Hemiculter eigenmanni known as less sensitive to pollution and water quality deterioration, dominated the reservoir, and their relative abundances were 38% and 27%, respectively. Further, the analysis of the stomach contents revealed that main food source of dominant L. macrochirus was high amount of zooplankton, which includes copepoda and cladocera, while Cyprinidae species mainly consumed organic particles with zooplankton preys. Stable isotope analysis also suggested that L. macrochirus, Carassiusauratus, Pseudorasboraparva, and H. eigenmanni are major zooplantivorous group in the Jeondae Reservoir.

Stable Isotopic Reconstructions of Diets in Joseon Dynasty Using Human Remains from Myeongam-ri Site, Asan (조선시대 회곽묘 출토 인골의 안정동위원소 분석을 통한 아산 명암리 피장자의 식생활 복원)

  • This study reports on human dietary reconstruction of Joseon Dynasty using stable isotope analysis. The stable carbon and nitrogen isotope values were measured of bone collagen extracted from 25 human bones from archaeological site in Myeongam-ri, Asan. Average values of ${\delta}^{13}C$ and ${\delta}^{15}N$ are $-19.7{\pm}0.5$‰(n=23) and $9.6{\pm}1.1$‰(n=23), respectively. The isotopic data indicates that Myeongam-ri individuals consumed $C_3$ plants mainly and assumed of dietary resourced from a terrestrial animal protein. Previous isotopic studies of the Joseon era suggested that dietary composition of Joseon population was not influenced by region and burial type. Also comparison of the isotopic results of male and female from double burials showed sex-dependent dietary patterns among individuals living in the same circumstances.

Simultaneous Determiniation of Ar/$N_2$Ratios in Groundwater (지하수에 용해된 질소, 아르곤 가스의 동시측정)

  • Previously reported Ar/N$_2$ratios in groundwater have been measured by single ion monitoring (Barnes et al., 1975; Vogel et al., 1981; Mariotti et al., 1988). The detector geometry and flared flight tube in VG Optima isotopic ratio mass spectrometer appeared to be fortuitously aligned for the simultaneous measurement of Ar/N$_2$ratios. Method development included mechanical adjustments to optimize the mass spectrometer for Ar/N$_2$ratio measurements followed by development of a preparation system for the extraction of air-saturated water samples. Samples containing known Ar/N$_2$ratios were used to assess accuracy and precision, and to test the applicability of methods for measurements of aqueous Ar/N$_2$ratios. The results indicated that the prepared air-saturated water samples were almost identical to the predicted Ar/N$_2$ratios (p <0.001). Groundwater samples were collected from on-going research sites, Shelton and Grand Island, Nebraska. Samples from the Grand Island sludge injection site form a lower boundary for worldwide reported Ar/N$_2$ratios. These lower Ar/N$_2$ratios can be explained by the production of nitrogen gas from this site, where denitrification was reported previously.

  • Nitrogenous new production and regenerated production were measured in the southern part of the Yellow Sea (Hwanghae) using a stable isotope /SUP 15/N nitrate and ammonia between April 25∼30, 1993. Nitrogen production varied between 155 and 688 mg N m/SUP -2/ d/SUP -1/, which belongs to meso to eutrophic area values. This is equivalent to 881∼3909 mg C m/SUP -2/ d/SUP -1/, assuming the Redfield ratio for C:N of 5.7:1 (by weight). the f0ratio which is the fraction of new production from primary production, varied between 0.12 and 0.26, indicating that 74 to 88% of primary production was supported by the regeneration of nutrients within the euphoric zone. This low f0ratio is the characteristics of the oligo- to mesotrophic area. Contrary to the expected, the ambient nutrient concentration was not an important factor for controlling productivity in this area during the study period. The difference in productivity among stations was mainly due to the variations in phytoplankton biomass in different water masses.

Comparison of Morphological Characters and Stable Isotopes of Seeds between Wild Simulated Ginseng and Cultivated Field Ginseng (국내 산양삼과 인삼 종자의 형태적 형질과 안정성 동위원소 비교)

  • We compared the morphological characters and stable isotopes of seeds of wild simulated ginseng with those of cultivated field ginseng. Seeds were collected from ten areas for wild simulated ginseng and two areas for cultivated field ginseng. The length, width, thickness and number of seeds per gram were measured and the ratio of stable isotopes of carbon and nitrogen was analysed as well. There was a overlapping variation of morphological characters and the ratio of stable isotope of the wild simulated ginseng among study areas. In Geumsan area, compared with the seed of cultivated field ginseng, the seed of wild simulated ginseng was significantly small and light, and the ${\delta}^{15}N$(‰) of wild simulated ginseng was lower than that of cultivated field ginseng. However, it is somewhat impossible to compare the regional variation of the unique value of ${\delta}^{15}N$(‰) among study areas of wild simulated ginseng. We can suggest the value of ${\delta}^{15}N$(‰) can be used as detection factor for cultivation regime like chemical fertilization and organic farming.

Studies on Discrimination between Organic Rice and Non-organic Rice using Natural Abundance of Stable Isotope Nitrogen($\delta^{15}N$) (질소 안정동위원소 자연존재비($\delta^{15}N$)를 이용한 유기벼와 일반벼 판별법 탐색)

  • To investigate the possibility of discrimination between organic and non-organic rice using stable isotope nitrogen of natural abundance, organic rice of 17 samples and non-organic rice of 13 samples grown at adjoining organic rice field were collected in 2008. Rice was grinded into brown rice, milled rice and hull, and samples were analysed for nitrogen and $\delta^{15}N$ at NICEM. Authors also made inquiries about N source for both farmers who conduct organic- and non-organic rice cultivation. In order to know whether the $\delta^{15}N$ can be used in discrimination between organic and non-organic rice, discriminant analysis were made with SPSS and logistic method. 1. Organic farmers used manure, rice bran, used mushroom culture, fermented fertilizer (company products), and oil cake, but non-organic farmers applied compound fertilizer. Rice straws were remained in organic rice field while moved out in non-organic field. 2. There were difference in $\delta^{15}N$ among organic rice and its byproduct(7.760????% in hull, 6.720????% in rice), but significant difference was not found between them. And the trend was same between province. Non-organic rice showed similar results. 3. Significant difference of $\delta^{15}N$ were found between organic rice and non-organic rice (p<0.01) and between hull of organic rice and that of non-organic rice hull (p<0.05). $\delta^{15}N$ seemed to be useful criteria for discrimination of organic and non-organic rice. 4. When applied discrimination analysis of SPSS and logistic, there were significant difference between organic rice, non-organic rice and its byproducts except brown rice and hull in SPSS method. Hull can be used as the most useful component for unknown sample prediction with 83.3% probability.

The Study of Cyclophosphamide Metabolite $^{15}N$ and $^{17}O$ Phosphoramide Mustards (항암제인 Cyclophosphamide의 중간체인 $^{15}N$$^{17}O$-phosphoramide Mustards의 합성)

  • Each nitrogen and oxygen site isotope enriched the cyclophosphamide metabolite phosphoramide mustard was synthesized. Reaction of N,N-bis(2-chloroethyl)phosphoramidic dichloride$[Cl_2P(O)N(CH_2CH_2Cl)_2]$ with benzyl alcohol and ammonia gave N,N-bis(2-chloroethyl)phosphorodiamidic acid phenylmethyl ester $[BzO(H_2N)P(O)N(CH_2CH_2Cl)_2]$. Catalytic hydrogenation of this benzyl ester followed by the addition of cyclohexylamine provided PM. Incorporation of $^{15}NH_3$ into this general scheme gave PM with a $^{15}NH_2$ moiety. Glycine-$^{15}N$ was converted to bis(2-chloroethyl)amine-$^{15}N$ hydrochloride which, in turn, provided for N,N-bis(2-chloroethyl)phosphorodiamidic-$^{15}N$ dichloride. Use of this compound in the general synthetic pathway yielded PM CHA with $^{15}N$ in the mustard moiety. $^{17}O$-Enriched PM was generated through the use of benzyl alcohol-$^{17}O$. To obtain the alcohol, labelled benzaldehyde was made by exchange with $^{17}OH_2$ and was then reduced with sodium borohydride.

Effects of Water Deficit on Biomass Accumulation and Water Use Efficiency in Soybean during Vegetative Growth Period

  • Water deficit is the primary constraint of soybean [Glycine max (L.) Merr.] yield, and a physiological understanding of processes affected by water deficit is a key step in identifying and improving drought tolerance in soybean. The objectives of this research were to evaluate biomass and nitrogen accumulation patterns and water use efficiency (WUE) as possible mechanisms associated with the drought tolerance of Jackson. Biomass accumulation of Jackson was contrasted with the PI416937, which also has demonstrated tolerance to drought. For water-deficit treatment, total biomass accumulation was negligible for PI416937, but biomass accumulation continued at approximately 64 % of the well-watered treatment of Jackson. Transpirational losses for Jackson and PI416937 were approximately the same for the water-deficit treatment, indicating that Jackson had superior WUE. Isotopic discrimination of $^{13}$ C relative to $^{12}$ C also indicated that Jackson had higher WUE. Results indicated that increased WUE for Jackson under water deficit showed it was tolerant to drought rather than had an avoidance mechanism.

Isotopic Assessment of Ecological Function of a Transplanted Macroalgal Habitat (Grateloupia asiatica) (안정동위원소 분석을 이용한 이식된 지네지누아리의 생태학적 기능 평가)

  • To assess the trophic function of an artificial macroalgal habitat, we compared the stable carbon and nitrogen isotope ratios of two amphipods (Caprella sp. and Ampithoe sp.) and their potential food sources in a transplanted macroalgal habitat (Grateloupia asiatica) with those in a natural habitat. There were no significant differences in the isotopic values of both consumers and their potential food sources between the transplanted and natural habitats. Such isotopic similarities between the two sites indicate a comparable resource consumer relationship. Additionally, our results showed similar isotopic niche areas and high dietary overlap (>65%) of the two amphipods between the transplanted and natural sites, suggesting that the transplanted habitat plays ecological roles similar to the natural habitat. Overall, isotopic assessment can provide information on the trophic function of diverse transplanted macroalgal habitats and improve post-monitoring efforts in the management of artificial ecosystems.
