Comparison of nitrogen transformation dynamics in non-irradiated and irradiated alfalfa and red clover during ensiling

  • Objective: To study the contribution of plant enzyme and microbial activities on protein degradation in silage, this study evaluated the nitrogen transformation dynamics during ensiling of non- and irradiated alfalfa (Medicago sativa L.) and red clover (Trifolium pratense L.). Methods: Alfalfa and red clover silages were prepared and equally divided into two groups. One group was exposed to ${\gamma}$-irradiation at a recommended dosage (25 Gky). Therefore, four types of silages were produced: i) non-irradiated alfalfa silage; ii) irradiated alfalfa silage; iii) non-irradiated red clover silage; and iv) irradiated red clover silage. These silages were opened for fermentation quality and nitrogen components analyses after 1, 4, 8, and 30 days, respectively. Results: The ${\gamma}$-irradiation successfully suppressed microbial activity, indicated by high pH and no apparent increases in fermentation end products in irradiated silages. All nitrogen components, except for peptide-N, increased throughout the ensiling process. Proteolysis less occurred in red clover silages compared with alfalfa silages, indicated by smaller (p<0.05) increment in peptide-N and free amino acid N (FAA-N) during early stage of ensiling. The ${\gamma}$-irradiation treatment increased (p<0.05) peptide-N and FAA-N in alfalfa silage at day 1, whereas not in red clover silage; these two nitrogen components were higher (p<0.05) between day 4 and day 30 in non-irradiated silages than the irradiated silages. The ammonia nitrogen and non-protein nitrogen were highest in non-irradiated alfalfa silage and lowest in irradiated red clover silage after ensiling. Conclusion: The result of this study indicate that red clover and alfalfa are two forages varying in their nitrogen transformation patterns, especially during early stages of ensiling. Microbial activity plays a certain role in the proteolysis and seems little affected by the presence of polyphenol oxidase in red clover compared with alfalfaa.

Nitrogen and Phosphorus Dynamics in an Salt Marsh in the Nakdong River Estuary (낙동강 하구 염습지 식물군락의 질소 및 인의 동태)

  • We studied primary production, nitrogen and phosphorus dynamics in a salt marsh of Okryutung at Nakdong River estuary. The standing biomass in Phragmites longivalvis, Carex scabrifolia and Zoysia sinica stand was $5.48kg/\textrm{m}^2,{\;}1.94kg/\textrm{m}^2{\;}and{\;}1.95kg/\textrm{m}^2$, respectively. The peak above-ground biomass in each stand was $1.99kg/\textrm{m}^2,{\;}0.74kg/\textrm{m}^2{\;}and{\;}1.03kg/\textrm{m}^2$, respectively. Soil nitrogen decreased from the onset of growing seson till July, and then increased. Seasonal patterns of soil phosphorus were different from stand to stand. Nitrogen concentrations of above-ground plant tissus were quite different among the plant species at the very beginning of the growing season, however, they became similar as the plants grow. Seasonal pattern of phosphrous in C. scabrifolia roots was quite different from those other two species. Nitrogen absorbed by plants during season in P. longivalvis, C. scabrifolia and Z. sinicia stand was 224kg/ha, 111kg/ha, 156kg/ha, respectively. Phosphorus taken up by plants was 22kg/ha, 29kg/ha and 21kg/ha, respectively. Because the vascular plants growing at salt marshes can immobilize large quantities of nitrogen and phosphorus, salt marsh vegetation can be sued for preventing the pollution of coastal sea water.

Effect of Nitrogen on Cell Dynamics at Leaf Growth Zone in Two Rice Varieties

  • In plants, nitrogen is the major component for growth and development. Leaf growth is based on the division, elongation and maturation of cells, which are used for making of epidermis, mesophyll, bundle sheath, xylem, phloem and so on. Dynamics of these tissues with respect to nitrogen are required for better understanding. This experiment was conducted to evaluate effect of nitrogen on the elongation of epidermal and guard cell of two rice (Oryza sativa L.) varieties, Seoanbyeo and Dasanbyeo on May 2000 at Chungbuk national university in Cheongju. After transplaning the 20-day-old seedlings into a/5000 pots, the main characteristics related with cell elongation were investigated and evaluated. A maximum. leaf length reached at 7 or 8 days after emerging from the collar, and also the leaf elongation rates were greatly affected by the increase of N application rate. The initial and final cell length were about $17\mu\textrm{m}$ and $130\mu\textrm{m}$, respectively. Cell divisions occurred within 1.0mm from leaf base. With die higher nitrogen application rate of 22 kg-N $10\textrm{a}^{-1}$, cell division per hour was greater 1.5 to 1.9 and 1.2 to 1.3 fold as compared to the N application rate of 0 and 11 kg-N $10\textrm{a}^{-1}$, respectively. Cell enlargement of epidermal and guard cell under higher N application rate (22kg-N $10\textrm{a}^{-1}$) was finished within about 20 (Seoanbyeo) and 15 hours (Dasanbyeo), while it took much time, about 30 hours.

Analysis of Soil Total Nitrogen and Inorganic Nitrogen Content for Evaluating Nitrogen Dynamics

  • Various methods for assessing soil total nitrogen (TN) and inorganic N content have been developed to manage nutrient and to understand N cycle in soil. This paper address the technical procedures in arable soil samples to conduct soil sampling, sample preparation, and measuring total N and inorganic N. Among various methods for measuring soil total nitrogen contents, Kjeldahl distillation and Indophenol blue method have widely used due to reliability and economic advances. Also, two methods can analyze more samples at the same time compared with other nitrogen measuring methods. For evaluating inorganic N content, mainly in forms of nitrate-N ($NO_3{^-}-N$) and ammonium-N ($NH_4{^+}-N$), extraction with a single reagent such as 2M KCl has been employed, followed by Kjeldahl distillation or indophenol blue methods.

  • The dynamics of nitrogen compounds and RNA-based functional genes were characterized in the nitrification-denitrification coupling process. For the removal of residual ammonium, intermittent aeration was introduced in the denitrification reactor. N2O production was not observed in both reactors. In both reactors, the nitrifying genes (achaeal-amoA, bacterial-amoA and hor) and denitrifying genes (narG, nirK, norB and nosZ) had a copy number of 3.92 × 102-7.25 × 105 and 2.85 × 102-3.06 × 104 per ng of DNA, respectively. These results suggest that denitrification and nitrification reactions occur in both the nitrification and denitrification reactors, respectively. Therefore, the coupling process is a promising one for the conversion of ammonium to nitrogen without generating N2O.

QM/MM-MD 방법을 이용한 용액 속에서의 Potassium Thiocyanate의 Association/Dissociation Dynamics 연구

  • 본 논문에서는 양자 역학적 분자 동역학(Quantum Mechanical/Molecular Mechanical-Molecular Dynamics, QM/MM-MD)을 통해 수용액에 녹아 있는 Potassium Thiocyanate의 dynamics를 연구했다. Umbrella sampling technique을 활용하여 association/dissociation에 해당하는 Free energy surface를 구했다. 두 개의 Free energy minimum이 녹아 있는 두 이온의 center of mass 사이의 거리가 $4{\AA}$일 때와 $5{\sim}6{\AA}$ 부근일 때 나타났으며 $4{\AA}$일 때 더 안정 했다. 본 논문에서는 $4{\AA}$일 때를 Contact Ion Pair(CIP) $6{\AA}$일 때를 Dissociation Ion Pair(DlP)라고 칭했다. 이 minimum들이 무엇인 지를 밝혀 내기 위해 추가 연구를 수행하였다. Free energy 상에서 가장 안정 할 때(CIP) solute인 Potassium thiocyanate의 구조를 살펴 봤더니 Potassium ion은 Thiocyanate ion의 Sulfur보다 Nitrogen side를 선호하였다. 그 원인을 알아보기 위해 salvation shell의 구조를 Radial distribution function을 통해 살펴 봤더니 물 분자가 Nitrogen보다 Sulfur와 더 강한 상호작용을 하고 있었다. 그로 인해 Potassium ion이 Nitrogen을 선호한단 결과가 나온 것이다. 한편, 두 번째 minimum은 물 분자가 Potassium 이온과 Thiocyanate 이온 사이에 flexible하게 bridging을 하는 구조였다. 또한 단순 양자 계산을 통해서도 비슷한 구조를 얻을 수 있었다. 그러나 QM 계산은 0K에서 수행하는 것이기 때문에 엔트로피 효과가 없는 계산이지만 본 연구는 온도 300K로 실제 용매와 가깝게 수행함으로써 고정되어 있는 구조가 아니라 엔트로피와 엔탈피가 균형적으로 존재하는 실제 용액 속에서의 구조를 처음으로 보여주는 것이다.

The effect of water turnover time on decomposition of wild rice (Zizania latifolia) and nutrient dynamics in an artificial wetland system

  • The effect of different turnover time of water on the decomposition of emergent macrophyte litter (Zizania latifolia Griseb.) was investigated using a microcosm experiment. Microcosm treatment represented different turnover time of water; 1, 2, 4 and 8 weeks. The litterbags from each treatment were retrieved every 2 weeks until the 8th week and the water simultaneously sampled with the litterbag. The dry weight and the content of major cations in the litter, and the content of available N, P, and major cations in the water were analyzed. Dry weight loss after 8 weeks indicated the lower decay rates under the condition of short turnover time of water. Major cations from the litter and the water showed that the leached amounts of K and Mg from the litter were highest in the 2nd week and dramatically decreased from the 4th week. The dynamics of available nitrogen and phosphorus in the water showed that as the water turnover time was getting longer, the amounts of available nitrogen and phosphorus remained higher. These results suggest that wetlands with longer turnover time of water could maintain the increased nitrogen and phosphorus and no outflow of the nutrients could cause eutrophication problem.

Modeling the Relationship between Land Cover and River Water Quality in the Yamaguchi Prefecture of Japan

  • This study investigated the relationship between land cover and the water quality variables in the rivers, which are located in the Yamaguchi prefecture of West Japan. The study area included 12 catchments covering $5,809\;Km^2$. pH, dissolved oxygen, suspended solid, E. coli, total nitrogen and total phosphorus were considered as river water quality variables. Satellite data was applied to generate land cover map. For linking alterations in land cover (at whole catchment and buffer zone levels) and the river water quality variables, multiple regression modeling was applied. The results indicated that non-spatial attribute (%) of land cover types (at whole catchment level) consistently explained high amounts of variation in biological oxygen demand (72%), suspended solid (72%) and total nitrogen (87%). At buffer zone-scale, multiple regression models that were developed to represent the linkage between the alterations of land cover and the river water quality variables could also explain high level of total variations in suspended solid (86%) and total nitrogen (91%).

Nitrogen Mineralization and Dynamics in the Forest Soil (삼림토양의 질소 무기화와 무기질소의 동태)

  • Mineral nitrogen dynamics and net mineralization of nitrogen in oak(quercus accutissima) and pine(pinus rigida) forest soils were studied. Nitrogen mineralization was determined over 8-week period by incubation method at laboratory. Initial water content of incubating soils was adjusted by applying suction(30mmhg), and lossof water during incubation was recovered with deionized water using syringe at every 3 or 4days. Temperature of incubator was maintained with 35+0.3c during the incubation period. Content of organic matter, total nitrogen, nh4-n and no3-n in soils in oak stand were significantly highter than those in pine stand. soil ph was lower in pine stand than in oak stand. initial nh4-n and no3-n of soils used in incubation experiment were 12.6 ug/g and 6.5 ug/g for oak stand, and 5.3ug/g and 5.1 ug/g for pine stand, respectively. Production of nh4-n increased from the beginning st both stands, and showed a peak at 5th week in oak stand(28.5 ug/g) and 6th week in pine stand(16.7 ug/g), and then decreased. intial no3-n of soils in oak(6.5 ug/g) and pine(5.1ug/g)stands, increased to 36.2 ug/g in soils of oak stand(5th week) and 13.4 ug/g in pine stand(4th week), respectively. The low values of no3-n of the field soil in the growing season compared with those of incubating soils at both stands indicate that considerable amount of nh4-n and no3-n produced in soils of oak and pine stands during two-months incubation were 59.7 and 141.6mg/kg soil, and 51.9 and 41.2mg/kg soil, respectively.
