Effects of Non-protein Energy Intake on Whole Body Protein Synthesis, Nitrogen Retention and Glucose Turnover in Goats

  • The responses of whole body protein and glucose kinetics and of nitrogen (N) metabolism to non-protein energy intake (NPEI) were determined using an isotope dilution approach and measurement of N balance in three adult male goats. The diets containing 1.0, 1.5 and 2.0 times ME maintenance requirement, with fixed intake of CP (1.5 times maintenance) and percentage of hay (33%), were fed twice daily for each 21 d experimental period. After an adaptation period of 11 d, N balance was determined over 3 d. On day 17, whole body protein synthesis (WBPS) and glucose irreversible loss rate (ILR) were determined during the absorptive state by a primed-continuous infusion of [$^2H_5$]phenylalanine, [$^2H_2$]tyrosine, [$^2H_4$]tyrosine and [$^{13}C_6$]glucose, with simultaneous measurements of plasma concentrations of metabolites and insulin. Ruminal characteristics were also measured at 6 h after feeding over 3 d. Nitrogen retention tended to increase (p<0.10) with increasing NPEI, although digestible N decreased linearly (p<0.05). Increasing NPEI decreased (p<0.01) ammonia N concentration, but increased acetate (p<0.05) and propionate (p<0.05) concentrations in the rumen. Despite decreased plasma urea N concentration (p<0.01), increased plasma tyrosine concentration (p<0.05), and trends toward increased plasma total amino N (p<0.10) and phenylalanine concentrations (p<0.10) were found in response to increasing NPEI. Increasing NPEI increased ILR of both glucose (p<0.01) and phenylalanine (p<0.05), but did not affect ($p{\geq}0.10$) that of tyrosine. Whole body protein synthesis increased (p<0.05) in response to increasing NPEI, resulting from increased utilization rate for protein synthesis (p<0.05) and unchanged hydroxylation rate of phenylalanine ($p{\geq}0.10$). These results suggest that increasing NPEI may enhance WBPS and glucose turnover at the absorptive state and improve the efficiency of digestible N retention in goats, with possibly decreased ammonia and increased amino acid absorption. In addition, simultaneous increases in WBPS and glucose ILR suggest stimulatory effect of glucose availability on WBPS, especially when sufficient amino acid is supplied.

Comparisons of Decomposition Rates, Carbon and Nitrogen Dynamics of Branches in Pinus densiflora and Quercus variabilis Stands (소나무와 굴참나무 임분의 가지 분해율과 탄소 및 질소 동태 비교)

  • This study was carried out to understand how factors of decomposition such as quality of the substrate (tree species), forest types, and soil·meteorological conditions affect decomposition rates and dynamics of carbon (C) and nitrogen (N) of branches in adjacent Pinus densiflora and Quercus variabilis stands in Sancheong and J inju in Southern Korea. The branch bags (30 cm × 30 cm) with branch samples (a diameter of 2.0 - 4.0 cm and length of 10 cm) were placed on the forest floor at each stand in May 2018. The branch bags were collected in August and November 2018, February and May 2019, respectively. The decomposition rates of branches in P. densiflora stands were 4.49 % for Sancheong, and 5.75 % for Jinju. Whereas, the decomposition rates in Q. variabilis were 20.01 % for Sancheong, and 24.68 % for Jinju, respectively. The decomposition of branches was more rapid in Q. variabilis compared with P. densiflora in both regions. C and N in decomposed branches were more mineralized in Q. variabilis, whereas C and N were more accumulated in P. densiflora. These results indicated that the decomposition rates, C and N of decomposed branches may be affected by differences in substrate quality such as initial N concentration and C/N ratio rather than differences between both regions including different environmental factors.

Physical Properties of Octahedral Iron (Ⅱ) Complexes of Vicinal-Dioximes (팔면체형 철 (Ⅱ) Vicinal-디옥심 착물의 물리적 성질)

  • Six coordinate low spin iron(Ⅱ) complexes of vicinal-dioximes, [Fe$(DH)_2B_2$] and [Fe$(DH)_2$B(CO)]($(DH)_2$=$(CHDH)_2$(bis(1,2-cyclohexadinedioximato) bivalent anion), $(DPGH)_2$-(bis(diphenylglyoximato) bivalent anion), $(F{\alpha}DH)_2(bis(furil-{\alpha}-dioximato)$ bivalent anion); B = monodentate neutral nitrogen base), have been prepared and their physical properties have been investigated with respect to the effects of the equatorial ligands, $(DH)_2$. The order of ${\pi}$-acceptor CO stretching frequency for [Fe$(DH)_2$B(CO)] is $(F{\alpha}DH)_2$ > $(DPGH)_2$> $(CHDH)_2$, while non-${\pi}$-acceptor $NH_3$ stretching frequencies for [Fe$(DH)_2(NH_3)_2$] show the opposite order to that of CO, i.e., $(CHDH)_2$ > $(DPGH)_2$ > $(F{\alpha}DH)_2$. These infrared spectral data together with Fe-N (oxime) and Fe-N (oxime) and Fe-N (B) stretching frequency data suggest that the nitrogen atoms of $(CHDH)_2$ are more basic and poorer ${\pi}$-acceptors than the corresponding nitrogen atoms of $(DPGH)_2$ and $(F{\alpha}DH)_2$, respectively. It has been found from the electronic spectral data of [Fe$(DH)_2$] and [Fe$(DH)_2$B(CO)] that the energy of the charge transfer band from iron(Ⅱ) to $(DH)_2$ increases as the basicity of $(DH)_2$ increases.

Various Nitrogen Efficiencies and their Interrelation Among Rice Varieties (수도품종간(水稻品種間) 여러 질소효율(窒素効率)의 상호관계(相互關係))

  • The relationships between various nitrogen efficiencies among 57 rice cultivars grouped into three developmental categories were investigated by simple correlation with (12kg/10a) and without nitrogen fertilizer under the field condition. The relationship showed similar in a group or among all varieties of three groups. Yield among varieties showed highly significant correlation with Fe (fertilization efficiency: yield increment/nitrogen applied), E (nitrogen efficiency for yield), Ef(efficiency of nitrogen derived from fertilizer) and Eu (fertilizer use efficiency: nitrogen derived from fertilizer divided by total nitrogen applied). The E was correlated significantly and positively with harvest index (HI), percent translocation (T) of nitrogen to ear and negatively with nitrogen uptake amount (N), nitrogen concentration in grain (GN%) and in straw (SN%). The E depends almost on Ef and only inTongil group partly on Es (efficiency of nitrogen derived from soil). The Ef contributes to Fe more than Eu does (Fe = $Ef{\cdot}Eu$). It appears that Ef and Eu increased from the old group to the recommended local varieties, but only EF and Es increased markedly when rice was developed from recommended local group to Tongil line ($indica{\times}japonica$ hybrid selction). The fact that E and Fe depend more on Ef among rice varieties is very good contrast to the result that E and Fe depend more on Es and Eu respectively among soils in the previous investigatigation. The Ef appears as the most important parameter for rice varietal selection under fertilizer application system.

Nutrient regime, N:P ratios and suspended solids as key factors influencing fish tolerance, trophic compositions, and stream ecosystem health

  • The objectives of this study were to determine the effects of stream nutrient regime, N:P ratios and suspended solids on fish tolerance/trophic compositions and stream ecosystem health, based on multi-metric model, during 2008-2013. Also, stream ecosystem health was evaluated in relation to chlorophyll-a (CHL) as a measure of algal productivity or indicators of trophic state to water chemical parameters. Total number of sampled fish species were 50 and showed a decreasing trend from 2008 to 2013. The minnow of Zacco platypus, based on the catch per unit effort (CPUE), was the most dominant species (25.9%) among the all species. Spatial heterogeneity was evident in the fish tolerance guilds that showed the dominance of sensitive species (89%) in the headwaters (S1) and the dominance of tolerant species (57%) in the urban. These conditions were directly influenced by concentrations of nutrients and organic matter (COD). The N:P ratios, as a barometer of water pollution, had a negative linear function (R2 = 0.40, P < 0.01) with CHL, and the ratios had an important role in changes of COD concentration (R2 = 0.40, P < 0.01). Under the circumstances, the N:P ratio directly influenced the relative proportions of fish trophic/tolerance compositions. According to the regression analysis of omnivore (Om) and insectivore sp. (In) on total nitrogen and total phosphorus, nitrogen had no significant influences (P > 0.05) to the two compositions, but phosphorus influenced directly the two guilds [slope (a) = -32.3, R2 = 0.25, P < 0.01 in the In; a = 40.7, R2 = 0.19, P < 0.01 in the Om]. Such water chemistry and fish trophic guilds determined the stream ecosystem health, based on the multi-metric fish model.

Adsorption Characteristics and Kinetic Models of Ammonium Nitrogen using Biochar from Rice Hull in Sandy Loam Soil

  • Objective of this study was to investigate adsorption characteristics and kinetic models of $NH_4-N$ to biochar produced from rice hull in respective to mitigation of greenhouse gases. $NH_4-N$ concentration was analyzed by UV Spectrophotometer. For the experiment, the soil texture used in this study was sandy loam soil, and application rates of chemical fertilizer and pig compost were $420-200-370kgha^{-1}$ (N-P-K) and $5,500kgha^{-1}$ as recommended amount after soil test for corn cultivation. Biochar treatments were 0.2-5% to soil weight. Its adsorption characteristic was investigated with application of Langmuir isotherm, and pseudo-first order kinetic model and pseudo-second order kinetic model were used as kinetic models. Adsorption amount and removal rates of $NH_4-N$ were $39.3mg^{-1}$ and 28.0% in 0.2% biochar treatment, respectively. The sorption of $NH_4-N$ to biochar was fitted well by Langmiur model because it was observed that dimensionless constant ($R_L$) was 0.48. The maximum adsorption amount ($q_m$) and binding strength constant (b) were calculated as $4.1mgg^{-1}$ and $0.01Lmg^{-1}$ in Langmuir isotherm, respectively. The pseudo-second order kinetic model was more appropriate than pseudo-first order kinetic model for high correlation coefficient ($r^2$) of pseudo-second order kinetic model. Therefore, biochar produced from rice hull could reduce $N_2O$ by adsorbing $NH_4-N$ to biochar cooperated in sandy loam soil.

Physical Property of W-C-N Diffusion Barrier through Stress-Strain curve (Stress-Strain curve를 이용한 W-C-N 확산방지막 물성 특성 연구)

  • This paper suggest tungsten (W)-carbon (C)-nitrogen (N) thin films for diffusion barrier that W is main material and C and N are additives. W-C-N thin films are deposited with fixed rates of W and C but with a variation of $N_2$ gas flow and W-C-N thin films are heated at $600^{\circ}C$. From the experimental results, the variation of elastoplastic region for W-C-N thin film measured by tribological property is larger than that of elastic region with a variation of $N_2$ gas flow. These results show that the $N_2$ gas flow is more directly related with the elastoplastic region of W-C-N thin film. Nanoindenting test executed 16 times consecutively and we got the stress-strain curve graphs and hardness datas at each sample. Through the stress-strain curve graphs, the standard diviation of stress-strain curve for $N_2$ gas flow rate of 2.0 sccm is smaller than that of 0, 0.5, 1.5 sccm. Consequently, the physical stability of W-C-N thin film depends on the flow rate of $N_2$ gas.

Effects of different nitrogen doses and cultivars on fermentation quality and nutritive value of Italian ryegrass (Lolium multiflorum Lam.) silages

  • Objective: The fermentation profile and silage quality of 3 Italian ryegrass (Lolium multiflorum Lam.) cultivars (cvs. Devis, Hellen, and Trinova) treated with 5 nitrogen doses (0, 50, 100, 150, and 200 kg/ha) were evaluated. Methods: The experiment was laid out in split plot in randomized complete block design with three replications. Annual ryegrass cultivars used in this study have been commonly grown in Turkey. Nitrogen doses were set in main plot and cultivars in split plot in the field. Plants were harvested at full-flowering stage with dry matter content about 220 g/kg for first cutting and 260 g/kg for second cutting. Harvested plants were chopped theoretically into 2 to 3 cm lengths for ensiling. Chopped fresh materials were ensilaged by compressing in 2 L plastic jars about 3±0.1 kg. Results: Effects of N doses on dry matter, neutral detergent fiber, acid detergent fiber, dry matter digestibility, relative feed value, crude protein, pH, ammonia nitrogen, lactic acid, acetic acid, and lactic acid/acetic acid were statistically significant while water soluble carbohydrate, ash and organic matter were not statistically different. Ammonia nitrogen, crude protein, ash, organic matter, lactic acid, and lactic acid/acetic acid were affected by cultivars, but the other parameters were not. Increasing nitrogen applications positively affected the chemical composition of annual ryegrass silage. The significant increase in protein content was remarkable, however, silage fermentation properties were adversely affected by the increasing nitrogen dose. Conclusion: It can be recommended 150 kg/ha nitrogen dose for annual ryegrass harvested at full blooming stage. Even though the silage fermentation properties of the used cultivars were similar, cv. Devis gave better results than the others in terms of silage pH and relative feed value.

Varietal Differences in Physiological Characteristics Focussed on N. P. K and Si Uptake under Cold Temperature at Seedling Stage (묘대기(苗垈期) 수도품종별(水稻品種別) N, P, K, Si 함량(含量)에 미치는 저온처리와 질소(窒素) 및 인산증시의 영향(影響))

  • The investigate the varietal differences is physiological characteristics related to nutrient uptake and response to nitrogen and phosphorus, a pot experiment was conducted in a temperature control chamber. Three varieties with different cold resistances were grown under normal ($23^{\circ}-25^{\circ}C$) and cold ($15^{\circ}C$) conditions with three nitrogen and phosphorus application levels. The cold temperature was treated for twelve days after thirty days from seeding. The results were summarized as follows: 1. Dry weight and nutrient contents were reduced in cold temperature treatment compared with control and the reduced ratio by low temperature were in order of dry weight 29.4% $P_2O_5$ 21.3%, N 15.3%, $K_2O$ 10.1%, $SiO_2$ 8.7%. 2. The reduced ratio of phosphorus contents of each varieties to normal condition were 13.5% in resistant, 19.5% in medium, 31.1% in susceptible variety, respectively. 3. The phosphorus and silicate contents of plants showed a good relationship with varietal differences in cold resistance but nitrogen and potassium didn't showed, therefore, phosphorus and silicate contents might be used as the index for physiological characteristics related with cold resistance. 4. Phosphorus content slightly increased with nitrogen application but silicate content decreased. Upon phosphorus application, nitrogen and silicate contents were increased.

  • This study was carried out to find out the effects of nitrogen dioxide gas on vegetable crops. Chinese cabbage, radish, tomato and cucumber were used for this experiment, and these crops were fumigated with various concentration of nitrogen dixoide gas for 2 hours under the different condition of light and soil water at the vegetable growth stage. The results obtained were as follows. The crops damages measured by percentage of leaf destruction were more severe at the night time fumigation than at the day time, under the full sun condition than under the shading, and at the higher soil water contents than at the low soil water contents. The cucumber plant showed the highest resistance to nitrogen dioxide gas and tomato plant showed the least resistance. As the nitrogen dioxide concentration increased from $0.05mg/{\ell}$ to $0.2mg/{\ell}$, the N contents in leaves of each crops were increased, and the increment of N contents in leaves was higher by fumigation at the night than at the day time. The increment of N contents in leaves showed possive correlation with the percentage of leaf destruction significantly.
