호기/혐기성 조건에서의 BTEX 및 MTBE 동시 분해특성 (Microcosm Study on BTEX and MTBE (Methyl Tert-Butyl Ether) Biodegradation under Aerobic-Anaerobic Conditions)

  • 본 연구에서는 다양한 호기/혐기조건에서 유류오염물질인 및 MTBE의 생분해 특성을 비교하고, 특히 탈질 조건에서 질산염 영향을 조사하여 유류오염지역의 혐기적 자연정화방법의 적용 가능성을 평가하고자 한다. 단일기질 및 혼합기질 분해실험 결과, BTEX는 3가지 실험조건에서 차이는 있었으나 모두 분해가 일어났다. 그러나 benzene과 p-xylene은 호기성 조건에서 초기 공급된 용존 산소의 부족으로 인하여 분해가 지연되는 것으로 나타났다. 또한 혼합기질에서는 단일기질에 비해 BTEX 분해가 기질 경쟁관계로 인해 다소 지연되는 경향이 관찰되었다. MTBE는 탈질 조건에서만 생분해가 관찰되었으나, TBA 축적 없이 $CO_2$로 무기화되는 것으로 추정된다. 또한 BTEX 및 MTBE 분해에 대한 질산염 농도의 영향 실험 결과, 저농도(>50 mg/L)에서 BTEX 분해는 제한되었으며, 고농도 질산염(<200 mg/L) 조건하에서는 BTEX 분해가 억제되는 현상이 관찰되었다. 본 연구에서 도출된 결과는 유류오염지역의 경우 호기/혐기성 조건에서 자연 생분해를 유도할 수 있을 것으로 예상된다.

폐쇄형 식물생산시스템에서 광도, 수확 전 양액조성 및 양액결제시기가 잎상추의 체내 질산염 함량에 미치는 영향 (Effects of Light Intensity, Nutrient Solution Compositions before Harvest and the Time of Nutrient Solution Removal on Nitrate Contents in Hydroponically-Grown Leaf Lettuces in Closed Plant Production System)

  • 쌈채소 수경재배 시 문제가 되는 체내 질산염 함량을 효과적으로 저감시키기 위해 폐쇄형 생산시스템에서 1) 저광도($100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$내외) 조건에서의 수확 7일 전 양액조성 방법과 2) 질산염 저감을 위한 적정 광도 및 수확 전 적정 양액결제시기를 구명하기 위해 수행하였다. 청치마상추에서 수확 7일 전 양액조성 방법 중 양액결제(양액공급 중단) 처리와 양액내 질소 공급원으로 탄산암모늄[$(NH_4)_2CO_3$]을 사용한 처리는 처리기간 동안 체내 질산염 함량을 감소시켰으나, 생체중, 엽면적 등의 생육량도 감소되었다. 하지만 $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$조건에서 수확 7일전 양액내 $NO_3-N$ 비료를 결제하여 조성하거나 배양액 농도를 1/2배액으로 낮추어 공급함으로써 외적품질 및 생육량의 저하없이 질산염 함량은 감소되었다. 또한 수확 전 양액결제시기 및 적정 광도를 구명하기 위해 실험을 수행한 결과, 100, 200, 및 $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$의 3수준의 광조건 모두 수확 7일 전 양액결제시에 체내 질산염이 가장 낮게 나타났으며, 특히 $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$의 광도와 수확 7일 전 양액결제 처리시 식물체내 질산염의 빠른 저감효과를 볼 수 있었으나 생육량의 감소를 가져왔다. 수확 3일 전 양액결제 처리에 의해 $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 광도에서 체내 질산염 함량은 2,021ppm에서 480ppm로, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 광도에서 3,018ppm에서 1,035ppm로 감소되었으며, 외적 품질의 저하 없이 생육량이 높으면서 동시에 짧은 시간 동안 식물체내 질산염의 저감 효과를 볼 수 있었다. 수확 7일 전 양액결제 처리는 체내 질산염의 저감효과가 컸던 반면 엽중, 엽면적 등의 생육 감소를 가져왔는데, 200과 $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$의 광조건에서 수확7일 전 양액결제시 수확 3일 전 양액결제보다 엽중은 20~35%, 엽면적은 16~31% 낮은 값을 나타내었다. 광도 및 양액결제시기에 따른 비타민 C 함량을 분석한 결과 100, 200보다 $300{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 에서 수확 3일과 5일전 양액결제 처리시 비타민 C 함량이 가장 높게 나타났다.

  • Recendtly an organic agricultured in Korea is getting a public attention not only for minimizing NO3- contamination of groundwater but also for producing of quility of agricultural products. But still there is not Basic Standards for Organic Agriculture in Korea and Japan since they just believe organic agriculture is one kind of the environmental friendly sustainable agriculture as written in western literature. In the paper it was discussed the core skills and doctrine of IFOAM Basic Standards compare to Korean Organic Farming Method in oder to point out thed disadvantages of overuse of organic fertilizer, 50-150MT/㏊ in each cultivation, intensive monocropping in glass-house without any rotation & legume, and without any green manure. Most korean or-ganic farmer believe that the more they apply organic fertilizer, the better they pro-duce high quility of crops and they practise organic agriculture completely. It was also suggested the overuse of organic fertilizer cause the accumulation of NO3- in rhizosphere and subsequently it might lead to drinkwater pollution by nitrate leaching. In conclusion it is suggested that for successful establishment of organic agriculture, The Association of Korean Organic Agriculture, The Society of Korean Organic Agri-culture and the certification body should be developed the Basic Standards which is acceptable by IFOAM Basic Standards.

Community structure analysis of nitrifying biofilms by 16S rRNA targeted probe and fluorescence in situ hybridization (FISH)

  • 질산화 생물여과 시스템 내 생물막 안에 존재하는 ammonia oxidizers 및 nitrite oxidizers의 군집 구조 및 공간적 분포를 조사하였다. FISH 분석 결과 생물막 내 숫적으로 우점종을 이루는 미생물은 ammonia oxidizer인 Nitrosomonas spp.로 나타났으며 nitrite oxidizer 인 Nilrospira spp.에 비해 3 ${\sim}$ 5 정도 더 많이 존재하였다. 이는 실협 기간동안 완전한 질산화를 보였지만 반응기가 2 년 이상 nitrite 축적을 위해 높은 free ammonia 농도 빛 낮은 용존 산소 상태에서 운선되어 nitrite oxidizers에 저해를 주었기 때문인 것으로 사료된다. FISH와 결합된 CLSM 관찰 결과 생물막 전체에 걸쳐 ammonia oxidizer가 분포하는 반면 안쪽으로 갈수록 nitrite oxidizers가 분포함을 보였다. 이는 폐수의 ammonium 을 생물막 내 ammon ia oxidizer가 먼저 nitrite로 산화시키고 이를 nitrite oxidizers가 곧바로 nitrate로 산화시키기 때문이다.

고온호기성 소화공정 배가스 처리를 위한 바이오필터 성능평가 (Performance Evaluation of Biofilter Treating Autothermal Thermophilic Aerobic Digestion Offgas)

  • Two combined autothermal thermophilic aerobic digestion (ATAD) and biofilter (BF) systems were operated to treat the piggery wastewater and the ammonia offgas. Experimental results indicated that the organic removal efficiency of ATAD-2, operated with oxygen, was higher than that of ATAD-1, operated with air. The concentration of ammonia in ATAD-2 offgas was higher compared to ATAD-1 offgas, but the total amount of ammonia produced from ATAD-2 was less than that from ATAD-1 due to the lower oxygen flowrate. The ammonia gas produced from both ATAD reactors was successfully removed by the BF. The BF-1, connected with ATAD-1, removed 93% of ammonia at the loading rate of $9.4g\;NH_3-N/m^3/hr$. The BF-2, connected with ATAD-2, removed 95% of ammonia gas at the loading rate of $8.1g\;NH_3-N/m^3/hr$. As the nitrification process continued, pH value of recirculating solution continuously decreased due to the accumulation of nitrate. When the ammonia loading rate was less than $22.7g\;NH_3-N/m^3/h$, the proper replacing cycle of recirculating solution was in the range of 10 to 11 days. Almost 90% of total mass of nitrogen fed into the each BF was confirmed from the mass balance on nitrogen.

Ecological and Biogeochemical Response of Antarctic Ecosystems to Iron Fertilization and Implications on Global Carbon Cycle

  • The European Iron Fertilization Experiment EIFEX studied the growth and decline of a phytoplankton bloom stimulated by fertilising $10km^2$ in the core of a mesoscale $(80{\times}120km)$ cyclonic eddy south of the Antarctic Polar Front with about 2 times 7 tonnes of iron sulphate. The phytoplankton accumulation induced by iron fertilization did not exceed $3{\mu}g\;chl\;a\;l^{-1}$ despite a draw down of $5{\mu}M$ of nitrate that should have resulted in at least double to triple the amount of phytoplankton biomass assuming regular Redfield-ratios for draw down after phytoplankton growth in the Southern Ocean. During EIFEX the fertilized core of the mesoscale eddy evolved to a hotspot for a variety of small and medium sized mesozooplankton copepods. In contrast to copepods, the biomass of salps (Salpa thompson)) that dominated zooplankton biomass before the onset of our experiment decreased to nearly extinction. Most of the species of the rnosozooplankton community showed extremely hiか feeding rates compared to literature values from Southern Ocean summer communities. At the end of the experiment, massive phytoplankton sedimentation reached the sea floor at about 3800m water depth.

Effect of Cholecystokinin-pancreozymin on the Nitric Oxide Synthase Activity and Cyclic GMP Level in Rat Pancreatic Tis-sue

  • In pancreatic cells, NO formation is associated with increased levels of cGMP and endocrine/exocrine secretion. In the present study, the role of NO in the regulation of exocrine secretion was investigated in rat pancreatic tissues. Treatment of rat pancreatic tissue with sholecystokinin-pancreozymin (CCK-PZ) resulted in an significant increase in arginine conversion to citruline, the amount of nitrite/nitrate, the release of amylase, and the level of cGMP. Furthermore, CCK-PZ stimulated increase of amylase release and conversion of arginine to citrulline transformation were counteracted by the inhibitor of NO synthase, $N^G-nitro-L-arginine$ methyl ester. The results on the time course of CCK-PZ-induced citrulline formation within the first seconds of simulation. The kinetics of citrulline accumulation correlate well with those of cGMP rise, which further confirms the conclusion that NO mediates the response to CCK-PZ by cGMP.

전기분해법에 의한 해수내의 암모니아성 질소 제거 (Ammonia-nitrogen Removal in Sea Water by Using Electrolysis)

  • 전기분해를 이용한 해수의 암모니아성 질소 및 대장균 제거에 효과적이었다. 암모니아의 제거효율은 전류의 세기가 클수록, 반응 시간이 길수록, 극판간격이 좁을수록 증가하였다. 해수내 250/100 m\ell$의 대장균은 모든 실험 조건에서 검출되지 않아, 살균 목적으로 전기분해가 매우 효과적이었다. 암모니아의 제거 효율은 잔류 염소의 농도에 따라 증가하였다. 암모니아의 제거 효율과 잔류 염소와의 관계는 다음과 같다.$NH_4^+-N(%)=4cdotlog[Residual\;chlorine(mg/\ell)+28(r=0.892)$

Copper and Lead Concentrations in Water, Sediments, and Tissues of Asian Clams (Corbicula sp.) in Bung Boraphet Reservoir in Northern Thailand (2008)

  • Bung Boraphet is the largest freshwater reservoir in Thailand. This study examined the accumulation of copper (Cu) and lead (Pb) in water, sediment and tissues of Asian clams (Corbicula sp.) within Bung Boraphet to assess the possible polluting effect of soil erosion and the dissolution of water soluble salts from the Nan River. Samples were collected from 12 study sites within Bung Boraphet between February and December 2008. The physicochemical parameters of the water including temperature, pH, turbidity, ammonia nitrogen, nitrate nitrogen, orthophosphates, biochemical oxygen demand, dissolved oxygen, Cu, and Pb were measured. The water in Bung Boraphet was found to be medium clean according to the surface water quality standard of Thailand. The levels of Cu and Pb in the water were low but heavy metals were detected at higher levels in the sediment and tissues of Corbicula sp. In the near future, management practices and regulator approaches for Cu and Pb contamination will be needed to protect the water in Bung Boraphet.

하수처리를 위한 간헐 방류식 장기폭기 공정에서 아질산염의 축적에 영향을 미치는 인자 (Factors affecting nitrite build-up in an intermittently decanted extended aeration process for wastewater treatment)

  • An intermittently-aerated, intermittently-decanted single-reactor process (KIDEA process : KIST intermittently decanted extended aeration process) was applied for nitrogen removal from wastewater. Synthetic wastewater with chemical oxygen demand (COD): nitrogen (N) ratio of approximately 5.25: 1 was used. The average COD removal efficiency reached above 95%, and under optimal conditions nitrogen removal efficiency also reached above 90%. This process consisted of 72 minute aeration, 48 minute settling and 24 minute effluent decanting with continuous feeding of influent wastewater from the bottom of the reactor, and did not require a separate anoxic mixing-phase. In this process, nitritation ($1^{st}$ step of nitrification) was induced but nitratation($2^{nd}$ step of nitrification) was suppressed. Main factors responsible for the accumulation of nitrite ion in the experimental condition were free ammonium and dissolved oxygen. This condition of nitrite build-up accelerated by continuous feed flow in the bottom of the KIDEA reactor because of high concentration of ammonia nitrogen in the influent. This research provides one of answers to control nitrate build-up.
