Postoperative pain after endodontic treatment of necrotic teeth with large intentional foraminal enlargement

  • Objectives: To evaluate postoperative pain after endodontic treatment of necrotic teeth using large intentional foraminal enlargement (LIFE). Materials and Methods: The sample included 60 asymptomatic necrotic teeth (with or without chronic apical periodontitis), and a periodontal probing depth of 3 mm, previously accessed and referred to perform endodontic treatment. After previous procedures, the position and approximate size of the apical foramen (AF) were determined by using an apex locator and K flexo-files, respectively. The chemomechanical preparation was performed with Profile 04 files 2 mm beyond the AF to achieve the LIFE, using 2.5 mL of 2.5% NaOCl at each file change. The filling was performed by Tagger's hybrid technique and EndoFill sealer. Phone calls were made to all the patients at 24, 48 and 72 hours after treatment, to classify postoperative pain. Statistical analysis was performed by different tests with a significance level of 5%. Results: Age, gender, periradicular status and tooth type did not influence postoperative pain (p > 0.05). Only 1 patient (1.66%) reported severe pain after 72 hours. Moderate pain was reported by 7, 4 and 3 patients after 24, 48 and 72 hours, respectively (p = 0.0001). However, paired analyses showed a statistically significant difference only between 24 and 72 hours (p = 0.04). Sealer extrusion did not influence the postoperative pain (p > 0.05). Conclusions: Acute or moderate postoperative pain was uncommon after endodontic treatment of necrotic teeth with LIFE.

Genomic Regions associated with Necrotic Enteritis Resistance in Fayoumi and White Leghorn Chickens

  • In this study, we used two breeds of chicken to identify genomic regions corresponding to necrotic enteritis (NE) resistance. We scanned the genomes of a resistant and susceptible line of Fayoumi and White Leghorn chickens (20 birds/line) using a chicken 60 K Illumina SNP panel. A total of 235 loci with divergently fixed alleles were identified across the genome in both breeds; particularly, several clusters of multiple loci with fixed alleles were found in five narrow regions. Moreover, consensus 15-SNP haplotypes that were shared by the resistant lines of both breeds were identified on chromosomes 3, 7 and 9. Genes responsible for NE resistance were identified in chicken lines selected for resistance and susceptibility. Annotation of the regions spanning clustered divergently fixed regions revealed a set of interesting candidate genes such as phosphoinositide-3-kinase, regulatory subunit 5, p101 (PIK3R5) and inositol 1,4,5-trisphosphate receptor 1 (ITPR1), which participate in immune response. Consensus haplotypes were found in regions containing possibly relevant genes, such as myostatin and myosin, which play important roles in muscle development. Thus, genome scans of divergent selection in multiple chicken lines and breeds can be used to identify genomic regions associated with NE resistance.

Outbreak of Cucumber mosaic virus and Tomato spotted wilt virus on Bell Pepper Grown in Jeonnam Province in Korea

  • In August 2006, a severe disease incidence showing mosaic and/or necrotic symptoms on two bell pepper varieties including red-colored 'Special' and yellow-colored 'Fiesta' was observed in a greenhouse located in Gwangyang, Jeonnam province, Korea. To identify causal viruses, total RNAs were extracted from 11 fruit samples with and without symptoms. Specific oligonucleotide primers for Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Tomato spotted wilt virus (TSWV) and Pepper mild mottle virus (PMMoV) were designed based on the sequences available on GenBank. Database comparisons of the deduced amino acid sequences of each sequence produced 100% and 98% matches with nucleocapsid protein gene of TSWV (Acc. No. ABE11605) and coat protein gene of CMV (Acc. No. DQ018289), respectively, suggesting that the symptoms on bell pepper fruits might be caused by the infection of CMV and TSWV. To our knowledge this is the first report of necrotic as well as mosaic virus disease on bell pepper fruits by the infection of CMV and TSWV in Jeonnam province, Korea.

배나무 잎검은점병의 피해상황과 방제 (Incidence of Pear Black Necrotic Leaf Spot and its Control by Top-Grafting)

  • 배나무 잎검은점병에 대한 방제대책을 검토하고자 시험을 수행하였다 이 병은 배 주산단지에 고루 발생하고 있으며 주 재배 품종인 신고에서 약 5.6% 정도 발생되었다. 농약에 의한 방제 효과는 없었고, 병 발생이 심한 나무는 수량 및 품질이 떨어지는 현상을 관찰할 수 있었다. 방제대책으로 경제적 갱신시기는 표준수확량의 70% 수준 이하일 때 즉 발병율이 80%이상일 때 저항성 품종으로 갱신하는 것이 효과적이었다. 이 병이 심하게 발생하고있는 신고품종에 저항성품종인 수황배와 화산배를 고접갱신 한 결과 2년 차까지 병 발생이 관찰되지 않았다.

Fumonisin $B_1$ induced renal toxicity in Sprague-Dawley rats

  • We investigated the effect of a single intravenous dose of Fumonisin $B_1(FB_1$) of rat kidney on the time sequence. Male Sprague-Dawley rats were intravenouslyin jected with FB$_1$at 1.25 mg/kg and were euthanized at 12 hrs, 1, 2, 4, and 6 days after the injection. In $FB_1$ treated rats, serum BUN and creatinine were elevated from 12 hrs. Microscopically, the initial target site was tubules of inner stripe, with mild degenerative and necrotic changes at 12 hrs, but the tubules recovered on day 4. In outer stripe, there were only a few scattered necrotic cells on day 1. These changes became more obvious over the time passed and most severe on day 4. On day 6, regeneration occurred, manifest as hypertrophic, basophilic tubular cells. The dying cells were proved to necrotic cells instead of apoptotic cells by TUNEL. Ultrastructural changes were cytoplasmic vacuole, dilated endoplasmic reticulum, swollen mitochondria, ballooned microvilli of the tubular cell in the outer stripe. These results showed that the renal tubules of outer medulla were the target to $FB_1$-induced nephrotoxicity in the rat. However, initial target was mner stnpe of medulla.

두가지 계통의 PVY가 잎담배 수량 및 내용성분에 미치는 영향 (EFFECT OF TWO PVY STRAINS ON YIELD AND CHEMICAL CONSTITUENTS OF TOBACCO PLANT)

  • Effect of the two PVY strains(PVY-VB and PVY-VN) on yield and chemical constituents of tobacco plants was determined in roar. NC2326 and Burley 21. The virus was inoculated 6 and 8 weeks after transplanting, respectively. The strains and time of inoculation were most critical factors for yield reduction. The greatest reduction was caused by the necrotic strain (PVY-VN) inoculated 6 weeks after transplanting, accounting for the loss of 40% in var. NC2326 and of 45% in var.. Burley21. When inoculated 8 weeks after transplanting with the necrotic strain, only 17% reduction was recorded in roar. Burley21, but no reduction occurred in roar. NC2326. Generally less reduction was caused by the PVY-VB strain, ranging 12-16% depending on the time of inoculation and/or variety. Compared to healthy tobacco, PVY infected tobacco contained higher concentrations of total nitrogen, protein nitrogen, nitrate nitrogen, and lower total sugar in cured leaf of roar. NC2326. Total alkaloid, P, Mg, Ca, and K levels were not altered. In var. Burley21, protein nitrogen and nitrate nitrogen increased, but other chemical components were not changed. Necrotic strain-infected tobacco with a severe symptom had higher nitrogen than did mild strain-infected tobacco.

Complete Sequence Analysis of a Korean Isolate of Chinese Yam Necrotic Mosaic Virus and Generation of the Virus Specific Primers for Molecular Detection

  • Chinese yam necrotic mosaic virus (CYNMV) is one of the most widespread viruses in Chinese yam (Dioscorea opposita Thunb.) and causes serious yield losses. Currently, genetic information of CYNMV is very restricted and complete genome sequences of only two isolates (one from Japan and another from China) have been reported. In this study, we determined complete genome sequence of the CYNMV isolate AD collected from Andong, Korea. Genetic analysis of the polyprotein amino acid sequence revealed that the Korean isolate AD has high similarity with the Japanese isolate PES3 (97%) but relatively low similarity with the Chinese isolate FX1 (78%). Phylogenetic analysis using the CYNMV 3' proximal nucleotide sequences harboring the coat protein and 3' untranslated region further supported genetic relationship among the CYNMV isolates. Based on comparative analysis of the CYNMV genome sequences determined in this study and other previous studies, we generated molecular detection primers that are highly specific and efficient for CYNMV diagnosis.

  • This study was taken to clarify the histopathological changes of pigs naturally infected with hog cholera. Microscopic observations of the necrotic lesion and inclusion body in the lymphoid organs were carried out in the natural cases of hog cholera and experimental cases inoculated with ALD virus and isolated virus strains. Electron microscopic findings of the intranuclear inclusion bodies in the reticular cell of spleen and lymph node were also observed in the experimental cases. The results obtained are as follow, As the histological findings necrosis of lymphoid organs was observed mainly in the lymph follicle. The necrotic lymphoid organs were found to contain 35.0% in the natural and 37.5% in the experimental cases. Intranuclear inclusion bodies were found mainly in the reticular cells of lymphoid organ, the epithelium of bronchiole and alevolus, and the vascular endothelium of brain. These inclusion bodies were seen in 40.0% of the natural cases and all of the experiment. The inclusion body was appeared to compose of activated nucleoli and chromatin granules (interchromatin and perichromatin) by electron microscopy.

Graft Transmission and Cytopathology of Pear Black Necrotic Leaf Spot (PBNLS) Disease

  • Graft transmission and cytopathological studies of a severe pear disease, pear black necrotic leafspot(PBNLS), were carried out to determine the causal agent of the disease. No evidence was found that a fungal or bacterial pathogen could be the causal agent of the disease. Attempts to transmit the agent by sap-inoculation to other plants including herbaceous hosts failed. How-ever, the pathogen was readily graft-transmitted from symptomatic diseased pears to healthy pears. Graft transmission of the pathogen was also demonstrated by using an indicator plant, PS-95, developed in the laboratory through various grafting methods. Ultrastructural study of the disease revealed the consistent presence of flexuous rod-shaped virus-like particles (VLP) in the symptomatic leaves of both Niitaka cultivar and indicator pear, PS-95. The particles, approximately 12 nm in diameter with undetermined length, occurred in the cytoplasm of mesophyll parenchyma cells. Cells with VLPs also contained fibril-containing vesicles, which are common in cells infected with plant viruses with ssRNA genome. The vesicles were formed at the tonoplast. Based on the symptomatology, the presence of fibril-containing vesicles, and graft-transmissibility, it is believed that the VLPs that occurred on symptomatic leaves of black necrotic leafspot of pear are viral in nature, possibly those of a capillovirus.

Pontine Necrosis Related with Radiation Therapy, Complicated with Spontaneous Hemorrhage

  • The brain necrosis induced by radiation therapy (RT) is an uncommon pathology of brain. A case of spontaneous hemorrhage at necrotic brain is also rare. A 52-year-old man who had nasopharyngeal carcinoma and had been treated with RT, presented with gait disturbance, dizziness, ataxia, dysarthria, and dysphagia. Magnetic resonance imaging (MRI) demonstrated progressed radiation necrosis of pons, and spontaneous hemorrhage at the site of necrosis. The hematoma was diminished by conservative treatment. However, the patient's neurologic symptoms did not recover. Two years later, spontaneous bleeding recurred at necrotic brain. His neurologic symptoms worsened. One year later, his neurologic symptoms were more progressed. He showed severe dysphagia, profound weakness and respiratory failure. This case provides the description of relapsed spontaneous hemorrhage and medullary dysfunction caused by pontine necrosis and progressed post-radiation injury, complicated with hemorrhage, and urges caution in that the necrotic brain tissue may be vulnerable to bleeding.
