A fungal isolate denoted NC14-264 was isolated from the soil in Jeongeup, Jeollabuk-do, Korea. Species in genus Plectosphaerella are pathogenic to several plant species, leading to fruit, root, and collar rot and collapse. In this study, a strain NC14-264 belonging to the Plectosphaerella was isolated from the soil and identified. Colonies were moderately grown, reaching 54 mm in diameter on potato dextrose agar, 49 mm on malt extract agar, and 55 mm on oatmeal agar at $25^{\circ}C$ after 10 days of incubation. Most Plectosphaerella species are distinguishable morphologically by irregular chlamydospores and different proportions of phialides and conidia. Based on morphological features and phylogenetic analysis using the internal transcribed spacer region and partial 28S rRNA gene sequences, the isolated fungus was identified as Plectosphaerella sinensis belonging to the Plectosphaerellaceae. This is the first report of P. sinensis in Korea.

In multi-cellular organisms, the control of gene expression is key not only for development, but also for adult cellular homeostasis, and gene expression has been observed to be deregulated with aging. In this review, we discuss the current knowledge on the transcriptional alterations that have been described to occur with age in metazoans. First, we discuss age-related transcriptional changes in protein-coding genes, the expected functional impact of such changes, and how known pro-longevity interventions impact these changes. Second, we discuss the changes and impact of emerging aspects of transcription in aging, including age-related changes in splicing, lncRNAs and circRNAs. Third, we discuss the changes and potential impact of transcription of transposable elements with aging. Fourth, we highlight small ncRNAs and their potential impact on the regulation of aging phenotypes. Understanding the aging transcriptome will be key to identify important regulatory targets, and ultimately slow-down or reverse aging and extend healthy lifespan in humans.

In this study, we investigated how Staufen1 influences the HIV-1 production. The overexpression of Staufen1 increased virus production without any negative affect on the viral infectivity. This increase was not caused by transcriptional activation; but by influencing post-transcriptional steps. Using multiple Gag protein derivatives, we confirmed that the zinc-finger domains of the HIV-1 nucleocapsid (NC) are important for its interaction with Staufen1. We also found that Staufen1 colocalized in stress granules with the mature form of the HIV-1 NC protein.
Recent clinical studies have shown that a high proportion of patients with acute promyelocytic leukemia (APL) achieve complete remission after treatment with all-trans retinoic acid (ATRA). However, most patients who receive continuous treatment with ATRA relapse and develop ATRA-resistant leukemia. Dendritic cells (DCs) are important antigen-presenting cells in the development of antileukemic T-cell responses. In this study, we investigated the strategies to overcome ATRA resistance of APL cells by inducing the differentiation of DCs from human leukemic cell lines for the developtment of adoptive immunotherapy. CD83 was used as a mature DC marker in this study and the expression of CD83 mRNA was determined by RT-PCR method. The promyelocytic leukemic cell line HL-60, B lymphoblast cell lines RPMI 7666 and NC-37 could be induced to dendritic cells in vitro. Treatment of HL-60 with phorbol 12-myristate 13-acetate (PMA) resulted in the expression of myeloid-related DC phenotypes, while treatment of RPMI 7666 with fms-like tyrosine kinase 3 ligand (Flt3-ligand, FL) and treatment of NC-37 with PMA and FL led to the expression of lymphoid-related DC phenotypes. In conclusion, myeloid-related DC phenotypes and lymphoid-related DC phenotypes could be generated from HL-60, NC-37 and RPMI 7666 cell lines, respectively. These DC phenotypes can potentially be used to generate antileukemic T cells in vitro for adoptive immunotherapy.
Objectives : In order to investigate the efficacy of BJBB on atopic dermatitis, various anti-inflammatory factors were studied. Methods : In-vitro, inflammatory mediators, such as MTT and nitric oxide were detected after the addition of LPS with or without BJBB in Raw 264.7 cells. In-vivo, in order to verify the effectiveness of BJBB in atopic dermatitis animal model, its role in inflammation factors and histological changes were observed in NC/Nga mice. Results : BJBB showed cell viability of 100% or higher in all concentration in Raw 264.7 cells. BJBB inhibited LPS-induced productions of inflammatory mediators nitric oxide in RAW 264.7cells. BJBB treated group showed significant decrease in the expression of IL-1b, IL-6 and TNF-a by 40%, 80% and 44% respectively. Also the group showed decrease in the transcription of IL-1b, IL-6 and TNF-a mRNA in spleen by 41%, 93% and 39% respectively. BJBB treated group showed significant decrease in WBC, neutrophil, lympocyte and monocytes immune cell ratio in blood by 54%, 63%, 57% and 86% respectively. BJBB treated group showed decrease in the expression of IgG by 39% respectively. Also, infiltration of adipocytes into skin was suppressed and the thickness of epidermis and dermis were relatively decreased in the BJBB treated group. Conclusion : BJBB has an anti-inflammatory effects in NC/Nga mouse. Thus, these results suggested a beneficial effect of BJBB in treatment with Atopic dermatitis and inflammatory.

Lipoxygenase is an enzyme, mainly produced by plants, capable of converting unsaturated fatty acids to fatty acids. It has vast application potential in the food, pharmaceutical and agricultural industries. The aim of this study was to isolate novel lipoxygenase-producing bacteria from the environment and to investigate the lipoxygenase enzymatic properties for industrial production. The strain, NC1, isolated from cultivation soils, was identified as Bacillus subtilis based on the phenotypic characteristics and 16S rRNA gene sequencing. This strain formed a pink color around the colony when cultured on indamine dye formation plates. The production of lipoxygenase by B. subtilis NC1 was influenced by the composition of the medium and linoleic acid concentrations. The optimum temperature and pH for lipoxygenase activity was determined to be 40 ℃ and pH 6, respectively. The enzyme showed relatively high stability at temperatures ranging from 20-50 ℃ and acid-neutral regions. In addition, the lipoxygenase produced by B. subtilis NC1 was able to degrade commercially available oils including sunflower seed oil and Perilla oil. In this study, a useful indigenous bacterium was isolated, and the fundamental physicochemical data of bacterial lipoxygenase giving it industrial potential are presented.
In order to improve efficacy of oriental medicines and to study the application of fermented oriental medicine in clinicals, the efficacy of CYT and CYTBH on atopic dermatitis were evaluated. The results and conclusions are as follows. CYT and CYTBH significantly improved the atopic dermatitis symptoms in NC/Nga mice by naked eye evaluation and significantly decreased clinical index in both groups. CYT and CYTBH both decreased the cell numbers of CD3+, CD11b+Gr-1+ cells in dorsal skin. Of the cells, CYT significantly decreased CD11b+Gr-1+ cells whereas CYTBH significantly decreased all immune cells. CYT and CYTBH both decreased the production rate of IL-4 and IFN-${\gamma}$ activated by CD3/CD28. In the case of CYTBH, significant decrease in all cases was observed. CYT and CYTBH decreased the production rate of IL-5, IL-13 and IL-17 in serum. Significant decrease of IL-5 in the case of CYT and IL-5 and IL-13 in the case of CYTBH were observed. CYT and CYTBH significantly decreased transcription of IL-5 mRNA and IL-13 mRNA in skin. Significant decrease in IgG1 and IgE immunoglobulins in serum were oberved in both groups. Significant decrease was only observed in the case of CYTBH. Both CYT and CYTBH significantly decreased the secretion of histamine. Both CYT and CYTBH suppressed erythema, hemorrhage, edema, excoriation, erosion of skin tissues of NC/Nga mice resulting in the decrease of thickness of epidermis. Significant decrease of infiltration of obese cells was also observed. The results above indicated that both CYT and CYTBH had significant efficacy in the treatment of atopic dermatitis through immune modulation. Animal studies showed that CYTBH had superior activity than that of CYT suggesting further and continuous studies on the changes in ingredients or absorption improvement by fermentation should follow.

Atopic dermatitis(AD) is a chronic inflammatory skin disease. AD has increased gradually, many people are tortured with AD. Chunggi-san(CG) and Samhwangseze-gamibang(SG) has been used for many kinds of skin disease in the Oriental medicine. But reports about the effect of CG and SG are insufficient. So, author investigated the effect of CG and SG on NC/Nga atopic mice. Major findings are summarized as follows: The clinical skin severity scores of experimental group in 13 and 16 week were decreased by 42% and 50% compared to the control group. Serum IgE, IL-4, IL-5, IL-6, IgM, IgGI levels of experimental group were significantly decreased compared to the control group. Serum $IFN-\nu$ was significantly increased in the experimental group compared to the control group. mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of experimental group were significantly decreased, and expression level of IL-6 in the skin tissues of experimental group was significantly decreased compared to the control group. $IFN-\nu$ mRNA expression levels was increased compared to the control group. According to biopsy reports of the ear and skin tissues showed that the tissue damage, experimental group were highly reduced compared to the control group. Judging from that $IL-1{\beta}$, $TNF-{\alpha}$, IL-6 express of gene, the effects of inflammatory cytokines revelation were significantly decreased compared to the control group. Depending on the density of CG, inflammatory RAW 264.7 in the serum of CG were significantly inhibited compared to the control serum that leaded a COX-2 activity model.

Atopic dermatitis (AD) usually develops in patients with an individual or family history of allergic diseases, and is characterized by chronic relapsing inflammation seen specially in childhood, association with IgE hyperproduction and precipitation by environmental factors. However, the exact etiology of AD has been unclear. To further explore the pathogenesis and treatment of AD, a suitable animal model is required. We found that skin lesions, which were clinically and histologically very similar to human AD, mite antigen-induced dermatitis on the face, neck, ears and dorsal skin of inbred NC/Nga mice. The aim of this study is to investigate the effect of Zizyphus jujuba var. spinosa Composite oil(ZjJ-C_oil) and Aralia cordata var. continentalis Composite oil(ARC-C_oil) to NC/Nga mice induced in Atopic dermatitis-like skin lesions by DNCB. NC/Nga mice which have been induced to Atopic dermatitis-like skin lesions by DNCB are divided into 4 groups, the first is the mice which have been spread with izyphus jujuba var. spinosa Composite oil(ZjJ-C_oil), the second is the mice which have been spread with Aralia cordata var. continentalis Composite oil(ARC-C_oil), the third is which have been spread with dexamethasone (Dexa.) 0.5% on their Atopic lesion, the last is the control group. Then We analyzed skin clinical score, blood sample of each group of measure state of the dorsal skin, the number of immunocytes, and resect the skin lesion to anlayze the state of cells. There are meaningful results of measuring the number of IgE, IL-4, IL-13, $IFN-{\gamma}$, IL-5, the total cells in ALN, dorsal skin, CD4+ Th, CD11b+/Gr-1+ in PBMCs, CD4+ Th, B220+/CD23+ in ALN, $CD4^+$, $CD8^+$ Th in dorsal skin, the level of COX-2, $TNF-{\alpha}$, $TGF-{\beta}1$, IL-4, IL-13 mRNA, the state of the skin lesion and cells in the group with ZjJ-C_oil, ARC-C_oil cream in comprarison with the control group.
