Objective: The objective of the experiment was to study yeast supplementation (yeast, Y) and dried kratom leaves (DKTL) on the digestibility, ruminal fermentation, blood metabolites and nitrogen balance in goats. Methods: Four of 7 to 8 months old male crossbred (50% Thai Native-Anglo Nubian) goats with average liveweight 20±0.13 kg were randomly assigned according to a 2×2 factorial arrangement in a 4×4 Latin square design to receive four diets ad libitum basis. The study investigated the effects of two levels of yeast (Y) supplementation (Y, 0 and 0.5g/kg dry matter [DM]) along with two levels of DKTL supplementation (DKTL, 0 and 4.44g/kg DM). The experimental groups were as follows: T1 = control group with 0Y+0DKTL, T2 = 0Y+4.44 DKTL, T3 = 0.5Y+0DKTL, and T4 = 0.5Y+4.44 DKTL. Results: The results showed that there were no interactions between Y levels and DKTL levels with respect to total DM intake, but there were significant effects (p<0.05) by levels of Y; goats receiving 0.05 g/kg DM Y had higher than goats fed 0.0 g/kg DM on average (kg/d). A percentage of body weight (% BW) and grams per kilogram of metallic weight (g/kg w0.75) had no influence on yeast levels and DKTL, but there was a difference (p<0.05) by yeast level Y at 0.5 g/kg DM, being higher compared to the non-supplemented group. Apparent digestibility coefficient of nutrition in the form of (DM, organic matter, crude protein, neutral detergent fiber, and acid detergent fiber) was an increased trend in the Y-level complementary group at 0.5 g/kg DM and DKTL at 4.44 g/kg DM, respectively. Protozoa populations decreased in the group receiving Y levels at 0.5 g/kg DM and DKTL levels at 4.44 g/kg DM when compared to group T1. The acetic acid concentration and methane gas generation decreased (p<0.05) in the group receiving Y levels of 0.5 g/kg DM and DKTL levels of 4.44 g/kg DM, while the amount of propionic acid increased (p<0.05). Conclusion: Effects of feeding combinations of Y and DKTL supplementation on feed showed no interaction effect (Y×DKTL) on feed intake, rumen fermentation, bacterial and fungi population. The effect on protozoal populations was lower in the group that was supplemented with DKTL at 4.44 g/kg DM related to synthetic CH4 was reduced.
인삼의 부산물중에서 박의 활용가능성을 조사하기 위하여 박의 구성성분, ext의 DPPH 환원력, 효모생육에 미치는 영향 및 효모균체내의 amino acid함량등에 미치는 영향등을 조사한 결과를 요약하면 다음과 같다. 박내에는 총당이 43~46%, 조단백이 14~15%, n-BuOH ext함량은 0.18% 였으며 alcohol박의 물ext는 alcohol ext에 비해 약 46%정도의 DPPH환원력이 남아있었으며 박ext를 첨가하고 효모를 배양시켰을 때 효모의 생육에 필요한 영양물질이나 생육을 촉진하는 물질이 박ext내에 존재하고 있다는 것을 알았으며 알콜박의 물추출물을 첨가하여 배양한 효모균체의 amino acid함량이 다른 ext를 첨가한 효모균체보다 많았다. 박에 cellulase를 0.1%, 20분간 처리로 박ext의 수율을 250%정도 증가시킬 수 있었다.
In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.
Heat shock proteins (Hsp) 70 are a ubiquitous family of molecular chaperones involved in many cellular processes. A yeast strain, ssa1/2, with two functionally redundant cytosolic Hsp70s (SSA1 and SSA2) deleted shows thermotolerance comparable to mildly heatshocked wild type yeast, as well as increased protein synthesis and ubiquitin-proteasome protein degradation. Since mRNA abundance does not always correlate well with protein expression levels it is essential to study proteins directly. We used a gel-based approach to identify stress-responsive proteins in the ssa1/2 mutant and identified 43 differentially expressed spots. These were trypsin-digested and analyzed by nano electrospray ionization liquid chromatography tandem mass spectrometry (nESI-LC-MS/MS). A total of 22 non-redundant proteins were identified, 11 of which were confirmed by N-terminal sequencing. Nine proteins, most of which were up-regulated (2-fold or more) in the ssa1/2 mutant, proved to be stress-inducible proteins such as molecular chaperones and anti-oxidant proteins, or proteins related to carbohydrate metabolism. Interestingly, a translational factor Hyp2p up-regulated in the mutant was also found to be highly phosphorylated. These results indicate that the cytosolic Hsp70s, Ssa1p and Ssa2p, regulate an abundance of proteins mainly involved in stress responses and protein synthesis.
발아효모 Saccharomyces cerevisiae의 핵공단백질인 Nic96 단백질과 유사성을 보이는 분열효모 Schizosaccharomyces pombe의 Nup97의 기능과 세포 내 위치를 조사하였다. nup97을 과 발현시켰을 때는 생장과 $poly(A)^{+}$ RNA의 분포에 별다른 이상을 보이지 않았다. 하지만, $kan^{r}$ 유전자를 이용하여 제작한 ${\Delta}nup97$ 결실돌연변이는 nup97의 발현이 억제되면, $poly(A)^{+}$ RNA가 핵 안에 축적되었고 비정상적인 DNA 분포를 보였으며 결국 생장하지 못했다. 한편, Nup97p의 N말단 또는 C말단에 GFP를 붙인 Nup97-GFP 융합단배질의 세포 내 위치를 확인하고자 하였다. 이러한 융합단백질들이 ${\Delta}nup97$ 결실돌연변이의 생장결함을 정상적으로 상보하는 것을 확인하고, nup97-GFP 유전자를 염색체의 nup97 유전자 위치에 삽입한 균주를 제작하였다. 자신의 프로모터에서 발현된 Nup97-GFP 융합단백질은 정상적인 기능을 보였으며, 핵막 주위에 점점 이 위치하였다. 이와 같은 결과들은 Nup97 단백질 역시 핵공단배질로 mRNA의 핵에서 세포질로의 이동에 중요하다는 것을 시사한다.
The yeast Candida albicans has a distinguishing feature, dimorphism, which is the ability to switch between two morphological forms: a budding yeast form and a multicellular invasive filamentous form. This ability has been postulated to contribute to the virulence of this organism. Previously, we reported that the yeast-to-hypha transition in this organism is suppressed by farnesoic acid, a morphogenic autoregulatory substance that accumulates in the medium as the cells proliferate. In this study, using a differential display reverse transcription polymerase chain reaction (DDRT-PCR) technique, we have identified several genes induced in C. albicans by farnesoic acid treatment. These observations indicate that farnesoic acid can alter the expressivity of multiple genes, including the DNA replication machinery and cell-cycle-control proteins.
Dimorphic yeast Candida albicans reversibly switches between the form of yeast and hyphae depending on external conditions. We investigated possible roles of the myosin family in the growth and dimorphic switches of C. albicans with a general myosin ATPase inhibitor, 2,3-butanedione-2-monoxime (BDM). Transition to hyphae as well as proliferation by budding was completely inhibited by BDM at 16 mM. Presence of 16 mM BDM did not affect hyphae-to-bud transition but it blocked budding. The effects of BDM on yeast growth and dimorphic switches were reversible. More than 70% of the BDM-treated cells demonstrated defects in the amount and the polarized localization of F-actin as well as in the shape and migration of the nucleus, suggesting that myosin activities are needed in these cellular processes of C. albicans.
Hrq1은 곰팡이 유전체에서 생물정보분석에 의해 발견된 새로운 RecQ helicase이다. 이 단백질은 인간의 RECQL4와 가장 상동성이 높으며 최근의 유전학적 생화학적 연구를 통해서 유전체 안정성을 유지하는데 어떤 역할을 할 것으로 예상되었다. 본 연구에서는 RECQL4와 상호작용하는 것으로 알려진 인간 유전자들과 상동성이 있는 효모 유전자들이 Hrq1과 상호작용하는지를 yeast two-hybrid assay를 이용하여 조사하였다. 총 11개의 유전자를 조사한 결과, nucleotide excision repair (NER) 인자 중의 하나인 Rad14이 Hrq1과 상호작용하는 것을 발견하였다. 또한 정제한 단백질을 이용한 pull-down assay로 Hrq1과 Rad14 사이의 직접적인 상호작용을 확인하였다. Hrq1과 Rad14 사이의 yeast two-hybrid 상호작용은 4-nitroquinoline-1-oxide에 의한 DNA 손상으로 더욱 증가하였으며, 이러한 상호작용의 증가는 또 다른 NER 인자인 Rad4에 의존적이었다. 이러한 결과들은 Hrq1이 Rad14과의 상호작용을 통하여 NER 과정에 어떤 역할을 할 가능성을 제시하고 있다.
