In order to maximize the secretory expression of human serum albumin (HSA) in the yeast Saccharomyces cerevisiae, a series of HSA expression vectors were constructed with a combination of different promoters, 5' untranslated regions (5'UTR), and secretion signal sequences. The expression vector composed of the galactose-inducible promoter GALl0, the natural 5'UTR, and the natural signal sequence of HSA directed the most efficient expression and secretion of HSA among the constructed vectors when introduced into several S. cerevisiae strains. Although the major form of HSA expressed and secreted in the yeast transformants was the mature form of 66 kDa, the truncated form of 45 kDa was also detected both in the cell extract and in the culture supernatant. The level of the intact HSA protein in the culture supernatant reached up to 30 mg/l at 24 h of cultivation in a shake-flask culture but began to decrease afterwards, indicating that the secreted HSA protein was unstable in a prolonged culture of yeast.
In 2021, seven yeast strains were isolated from earthworm (Eisenia andrei) gut samples collected from the Nanji Water Regeneration Center in Goyang City, Gyeonggi Province, Korea. A total of seven yeasts were isolated, of which three strains have not been previously reported in Korea. To identify the yeasts, pairwise sequence comparisons of large subunit (LSU) rDNA sequences were performed using the basic local alignment search tool (BLAST). Assimilation test and cell morphology analysis were performed using the API 20C AUX kit and phase contrast microscope, respectively. Five of the seven strains were assigned to the genus Candida of the order Saccharomycetales of the class Saccharomycetes, and two to the genus Apiotrichum of the order Trichosporonales of the class Tremellomycetes. The yeast strain Candida sojae E2 belongs to the family Debaryomycetaceae, and Apiotrichum laibachii E8 and A. laibachii E9 belong to the family Trichosporonaceae. All strains were cultured in yeast mold agar for three days and showed different colony forms. C. sojae E2 was round and entire shaped, while A. laibachii E8 and A. laibachii E9 was round and convex shaped. This study focuses on the description of the three yeast strains that have not been officially reported in Korea.

The production of pullulan by Aureobasidium pullulans HP-2001 was investigated under various ratios of glucose as carbon source and yeast extract as the nitrogen source, Highest conversion rate (productivity) of glucose to pullulan was 40.0% when concentrations of glucose and yeast extract were 5% and 0.15%, respectively. Maximal production of pullulan was 29.3g/1 when the concentration of glucose was 8%(w/v) and that of yeast extract was 40:1. On basis of the result that production of pullulan was found in a medium which concentration of glucose as carbon source was up to 20%(w/v), Aureobasidium pullulans HP-2001 seemed to overcome the catabolite repression. Conversion rate of pullulan from 20%(w/v) of glucose was 11.1%.
In 2020, 11 Basidiomycetous yeast strains were isolated from soil samples collected from the forests of Namhansanseong in Korea. Among them, seven species were reported, but four species were unreported in Korea. To identify wild yeasts, pairwise sequence comparisons of D1/D2 domain of the 26S rRNA were performed using Basic Local Alignment Search Tool (BLAST). The cell morphologies and assimilation test are observed by phase contrast microscope and API 20C AUX kit, respectively. The 11 strains were assigned to the genera Rhodotorula (4 strains) of the order Sporidiobolales of the class Microbotryomycetes; and Cryptococcus(2 strains), Goffeauzyma (1 strains), Naganishia (2 strains) of the order Filobasidiales and Saitozyma (2 strains) of the order Tremellales of the class Tremellomycetes in the phylum Basidiomycota. The unreported yeast strains Cryptococcus gastricus 20n5-2, Goffeauzyma gilvescens 20n2-7, Naganishia adeliensis 20n8-1, and Naganishia friedmannii 20n24-1 belong to the family Filobasidiaceae. All strains had oval shaped cells and cream-colored colonies cultured on on YM agar for 3 days. In this study, we focus on the description of four unreported yeast species in Korea.
The purpose of this study was to isolate and identify wild yeasts from the soil collected in Gwangju and Pocheon City, Gyeonggi Province, Republic of Korea. Among 10 strains, six strains were already reported, but four strains were unrecorded in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were performed using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation tests were carried out using API 20C AUX kit. The 10 strains were assigned to the phyla Basidiomycota (8 strains) and Ascomycota (2strains). The unrecorded four yeast strains, NH33, NH19, NH20, and YP416, belong to the phylum Basidiomycota and the genera Buckleyzyma, Leucosporidium, Holtermanniales, and Mrakia, respectively. All strains had oval-shaped and polar budding cells. In this research, the morphological and biochemical properties of four unreported yeast species were characterized intensively, which were not officially reported in Korea.
The objective of this study was to determine the effects of yeast cultures (Candida zeylanoides and Debaryomyces hansenii) isolated from traditionally dry fermented Turkish sucuks, on some physicochemical and microbiological properties of the product. Eight different batches of the sucuks were produced by the inoculation of yeast and lactic acid bacteria (LAB) cultures (Lactobacillus curvatus, Lactobacillus plantarum and Lactobacillus sakei) in different combinations. The sucuks were ripened for 12 days and analyzed at 1st, 6th, and 12th days of ripening. Percent moisture content, pH, water activity (aw) and residual nitrite values of the sucuk inoculated with the yeast cultures were higher at the end of the ripening. The use of yeast cultures decreased hardness, gumminess, and chewiness values of the sucuk while increased adhesiveness values. Major volatile groups were aldehydes, terpenes, and sulphur compounds in the sucuk samples. The most noticeable results were for sensory properties of the sucuk that were positively improved by the yeast cultures.

Green tea (Camellia sinensis) seed extract (GTSE) was prepared under various pretreatment conditions and used to investigate its extraction yield and anti-yeast activity. Anti-yeast activity of GTSE from seeds with or without the coats was the same, whereas the extraction yield was slightly higher in extract from seeds without the coat. Anti-yeast activity of GTSE from seeds with different water contents or particle sizes was the same, whereas the extraction yield was highest in extract from seeds with 7.3% moisture or a smaller particle size. Anti-yeast activity of defatted green tea seed extract (DGTSE) was the same as that of GTSE. Extraction yield was higher in DGTSE from defatted seeds by the oil press machine compared to hexane extraction. Defatted green tea seed (DGTS), a by-product from the oil extraction process, is a good natural source of anti-yeast preservative. The extraction yield and anti-yeast activity of DGTSE were investigated using various extraction solvents, temperatures, and times. The results show that water was an economic extraction solvent, and anti-yeast activity of DGTSE was unstable at $90^{\circ}C$. These results suggest that water, extraction temperature of $50^{\circ}C$, and extraction time of 4 h were the most efficient for extraction of anti-yeast compounds from DGTS.

The influence of agitation speed on the yeast growth was investigated in the production of probiotic feed from pulverized liquified food wastes by aerobic fermentation. A yeast Kluyvermyces marxianus was selected through a preliminary screening. The yeast was cultured by 2liter jar fermenter. in 10% solid(w/v) substrate of liquified food waste at $35^{\circ}C$ with each different agitation speed of 500, 900 and 1200 rpm. For the acceleration of enzyme excretion mixed culture with Aspergillus oryzae was also attempted and the results were compared to those of single culture. As results the viable cell number was increased by increasing agitation speed. But it showed highest value in 900rpm and then decreased in 1200rpm. The mixed culture increased amylase activity and growth rate, but did not seem to enhance the highest viable cell count in the final fermentation stage.
With 156 water samples collected from 39 locations in the Yeong San River estuary during the 12month period from March 1976 to February 1977, the seasonal distribution of yeast and the distributional pattern of yeast on salinity gradient have been investigated. An overall average number of yeast ranged from 52 to 487 viable cells (c.f.u.) per 100ml water sample. The highest count of yeast was obtained in spring while the lowest value came in summer. 933 yeast and one yeast-like fungus pertaining to 14 genera and 83 species were recovered, of which Candida were 29%, Debaryomyces 17.3%, Rhodotorula Glutinis were dominant forms in all locations as well as throughout the year. The population size of total aerobic bacteria, the amount of terrestrial imputs, and some of geographical and/or climatic factor appear to reflect the seasonal distribution of yeast as well as the composition of yeast species in an estuarine environ. Average number of yeast, species diversity, and particularly the number of fermentative and pseudomycelium-producing yeasts increased with decreasing salinity whereas nitrate-utilizing yeasts showed opposite trend, suggesting that salinity gradient can be used as a feasible detector for the distributional pattern of yeast in estuarine habitat.
