Vitamin Contents in the Fruits of Rosa davurica PALL (생열귀나무 열매의 비타민 함량(含量))

  • Vitamin contents in three different types of the fruits and seeds of Rosa davurica Pall. were determined to examine their applicability for a new health food products as well as medical supplies. HPLC analysis of the fruits showed that circular(L) type fruits contained the highest ascorbic acid (AA) concentration (911mg/1000g). Elliptical and circular(S) type fruits contained a little less amounts of AA, their concentrations being 358mg/l00g and 344mg/100g, respectively. Spectrophotometric analysis of total ${\beta}-carotene$ contents in the fruits showed a similar amount of ${\beta}-carotene$ in elliptical (286mg/100g) and circular(L) (208mg/100g) type fruits, but a far less amount was shown in the circular(S) type fruits (24 mg/100g).

Variation of Homoharringtonine Contents from Eight Natural Populations of Korean Native Plumyew Trees(Cephalotaxus koreana) (한국산(韓國産) 개비자나무(Cephalotaxus koreana) 8개 천연집단(天然集團)의 Homoharringtonine 함량변이(含量變異))

  • Homoharringtonine isolated from the plumyew tree (Cephalotaxus koreana) is currently considered as one of the most promising chemotherapeutic agents for anti-cancer. Variation of homoharringonine contents from eight natural populations of Korean native plumyew trees was determined by HPLC (high performance liquid chromatography), and compared with the variation among populations, the variation of the different parts(needle, root, stem and seed), and the variation according to their ages and growth features. Homoharringonine contents among populations were significantly different with Mt. South Dukyu ($1,048{\mu}g/g$) having the highest and Mt. Obong having the lowest. The analyses of plant pans showed that the homoharringtonine contents were highest in plant needles ($874{\mu}g/g$), and followed by roots, stems and seeds. Homoharringtonine contents of the plumyew trees decreased about 30% in increment with their ages and growth.

Homologous Expression and Quantitative Analysis of T3SS-Dependent Secretion of TAP-Tagged XoAvrBs2 in Xanthomonas oryzae pv. oryzae Induced by Rice Leaf Extract

  • Xanthomonas oryzae pv. oryzae (Xoo) produces a putative effector, XoAvrBs2. We expressed XoAvrBs2 homologously in Xoo with a TAP-tag at the C-terminus to enable quantitative analysis of protein expression and secretion. Addition of rice leaf extracts from both Xoo-sensitive and Xoo-resistant rice cultivars to the Xoo cells induced expression of the XoAvrBs2 gene at the transcriptional and translational levels, and also stimulated a remarkable amount of XoAvrBs2 secretion into the medium. In a T3SS-defective Xoo mutant strain, secretion of the TAPtagged XoAvrBs2 was blocked. Thus, we elucidated the transcriptional and translational expressions of the XoAvrBs2 gene in Xoo was induced in vitro by the interaction with rice and the induced secretion of XoAvrBs2 was T3SSdependent. It is the first report to measure the homologous expression and secretion of XoAvrBs2 in vitro by rice leaf extract. Our system for the quantitative analysis of effector protein expression and secretion could be generally used for the study of host-pathogen interactions.

Extraction conditions for preparation of natural seasoning of red pepper seed (고추씨 향신조미료 제조를 위한 추출 조건)

  • The red pepper seeds(RPS), an industrial waste produced from red pepper powder industry, were investigated for its possible use as natural hot taste seasoning. The RPS was extracted with water with addition of salt, sugar, phosphate and citric acid at 70-100$^{\circ}C$ Effects of preheat treatments of steaming at 100$^{\circ}C$ and roasting at 215$^{\circ}C$ and 330$^{\circ}C$ were also studied on the flavor of the RPS extracts. The results showed that steaming and roasting increased the solid yield and reduced the turbidity. The hot flavor of RPS extracts was generally decreased by steaming and roasting. Extraction of RPS at the temperature range of 70-100$^{\circ}C$ for 10-60 minutes showed that solid yield were relatively high of 27% at 80$^{\circ}C$ for 30 minutes with the highest score of hot flavor. When the extraction was carried out with addition of NaCl, sucrose, Na$_2$HPO$_4$ and citric acid, the solid yield was little affected by their addition except a little increase by 0.5% NaCl and 0.2% Na$_2$HPO$_4$ and hot flavor was little affected.

  • Dill(Anethum graveolens L.) is a buttercup family, and flowers, leaves, stems, and seeds are all mainly used as spices. This study was carried out to discover and propagate genetic resources for the development of plant-derived medicinal fragrance materials, and to establish a database. In order to investigate the genetic characteristics of dill and to extract natural essential oils, 50 resources were parceled out the genetic resource center, sown, germinated, and then formally cultivated in the test research field. After sowing and propagation of 50 dill resources, the characteristics of each individual such as plant height, the diameter of polychasium during flowering, and the number of small inflorescences were investigated. The flowering period of dill was around May 31 to June 17, and about 10% of the proliferating population flowered around May 31. The plant length of dill was 32-14 lcm, and the length of petiole was 1~16cm, showing an average of 5cm. The color of the stem and leaf color was referred to the RHS Color chart, and the colors were classified as 44S, 19 V, and 75DI. Uses as many resources as possible with 35 resources equivalent to 19V. Inflorescences are lateral and opposite, polychasium inflorescences are somewhat flat at the upper part and have a diameter of 4.5~20cm, and divergent inflorescences are irradiated with 5~86 multiple flowers. The number of pedicels is 16-74, and the length of the pedicel is 1-18cm spread sideways or the length is different from each other.

Molluscicidal Effect of Eco-Friendly Agricultural Substances for Controlling Golden Apple Snails (Pomacea canaliculata, Lamarck)

  • The golden apple snail (Pomacea canaliculata) has been utilized as a natural and eco-friendly control of weeds in rice paddy fields. However, P. canaliculata can damage other crops. In this study, the effectiveness of plant extracts from various natural sources that are reportedly effective against pests in the control of P. canaliculata was investigated. The four plant extracts were effective against P. canaliculata and ranked in descending order as green tea seed (Camellia sinensis) > root of red spider lily (Lycoris radiata) > leaves of tobacco (Nicotiana tabacum) > root of sophora (Sophora flavescens). The mortality rate of P. canaliculata was increased using 200 to 2000 mg/kg of green tea seed powder. However, shrubby sophora root extract did not significantly increase the mortality rate. The LC50 and LC90 of green tea seed, tobacco leaves, shrubby sophora root, and red spider lily root were 900 and 2800 mg/L, 956 and 2320 mg/L, 2162 and 5325 mg/L, and 512 and 1054 mg/kg, respectively. The LC50 and LC90 of ground powder of C. sinensis, N. tabacum, S. flavescens and L. radiata were 248 and 646 mg/L, 403 and 733 mg/L, 409 and 905 mg/L, and 493 and 1141 mg/L, respectively. The findings indicate the remarkable control potency of green tea seeds against the golden apple snail. An organic material incorporating the four plant powders may help control green apple snail in an ecosystem-friendly manner.

Growth Characteristics According to Sowing Season and Seedling Duration of Perila of Perila(Perila Frutesces BRITTON var, Japonica HARA) (파종기 및 육묘기간에 따른 들깨의 생육 특성)

  • This experiment was conducted at Sangji university in Wonju, Korea from 2005 to 2006. The results obtained were summarized as follow; The Chung-ju local variety was tested to find out the possibility of transplanting of seedling as a after-barley crop. When transplanting of seedling for a after-barley crop was test, this short-day plant flowered from september 1 to september 3, showing no connection with sowing time nor seedling periods. The yield of seeds per 10a was affected by both the sowing time and the seedling periods and its peak in the plots sowed on May 15 and transplanted on June 24(40 days seedling).

Effect of the Volatile Oil of Nigella sativa Seeds and Its Components on Body Temperature of Mice: Elucidation of the Mechanisms of Action

  • The effect(s) of the volatile oil (VO) of Nigella sativa and its two components, ${\alpha}-pinene$ and ${\rho}-cymene$ on body temperature of male and female conscious mice were studied. Further investigations to delineate the mechanism(s) of action of the observed effect(s) by using various blockers involved in the central regulation of body temperature were made. VO and ${\alpha}-pinene$ caused significant reductions in rectal body temperature at is and 30 minute after treatment. ${\rho}-cymene$ had negligible effect on body temperature of mice. Cyproheptadine inhibited VO and ${\alpha}-pinene-induced$ hypothermia significantly. Nalbuphine inhibited ${\alpha}-pinene-induced$ hypothermia significantly but did not affect VO-induced hypothermia. Droperidol potentiated VO and ${\alpha}-pinene-induced$ hypothermia to a non-significant level; whereas atropine potentiated VO-induced hypothermia non-significantly. The study confirms further the role of serotoninergic receptors in the mechanism(s) of the observed pharmacological effects of the VO of Nigella sativa. It also indicated a possible role of opioid receptors in ${\alpha}-pinene-induced$ hypothermia.

Antineoplastic Natural Products and the Analogues (XI) -Cytotoxic Activity against L1210 Cell of Some Raw Drugs from the Oriental Medicine and Folklore- (항암성 천연물 및 그 유사체(XI) -한약재 및 민간약의 L1210세포에 대한 세포독성-)

  • Forty herbal drugs which are described to have potential antitumor activity were solvent-fractionated with petroleum ether, ether and ethyl acetate in sequence. The cytotoxic activity was mostly shown in the ether fraction(40.54%) and petroleum ether fraction (35.15%), but scarcely in the water phase (10.8%), meaning that most of the active components had less polar property. Twenty-seven percent of the drugs tested were active, which is higher value than 10.4% of the random sampled drugs The drugs possessing the $ED_{50}$ values less than $10{mu}g/ml$ were the roots of Lithospermum erythrorhizon, Curcuma domestica, Salvia miltiorrhiza, Astragalus membraneceus and Scutellaria indica, the leaves of Panax ginseng, S. indica and Liriodendron tulipifera, the barks of Picrasma ailanthoides and Rhus vernifera, the herbs of Agrimonia pilosa and Siegesbeckia pubescens the seeds of Tricosanthes kirilowii, P. ailanthoides, and the stem of P. ginseng.

Amygdalin Extract from Armeniacae Semen Induces Apoptosis through Bax-dependent Caspase-3 Activation in Human Cervical Cancer Cell Line ME-180

  • Objectives: Amygdalin is known to be a natural compound which has antitussive and anticancer activities. Amygdalin is abundant in the seeds of bitter almond and apricots of the Prunus genus, and other rosaceous plants. We investigated whether amygdalin induces apoptosis. Materials and Methods : 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)assay, terminal deoxynuclotidyl transferase (TdT)-mediated dUTP nick end-labeling (TUNEL) assay, 4,6-diamidino-2-phenylindole (DAFI) staining, flow cytometric analysis, DNA fragmentation assay, western blot, and caspase-3 enzyme assay were performed on ME-180 cervical cancer cells treated with amygdalin. Results: Through morphological and biochemical analyses, it was demonstrated that ME-180 cells treated with amygdalin exhibit several apoptotic features. It was shown that amygdalin induces increases in levels of Bax and caspase-3 and a decrease in Bcl-2 expression. Conclusions: These results suggest the possibility that amygdalin exerts an anti-tumor effect on human cervical cancer.
