Anti-herpetic Activity of Various Medicinal Plant Extracts

  • In order to find antiviral compounds against Herpes simplex virus type I (HSV-1) and II (HSV2) from natural products, a convenient virus-induced cytopathic effect (CPE) inhibition assay was introduced. More than 300 fractions were prepared by solvent fractionation from sixty collected plants or purchased herbal medicines, and their anti-herpetic activities were evaluated. Among them, several medicinal plants showed potent anti-herpetic activity. Selective indexes (SI) of the EtOAc extract of Caraganae Radix (Caragana sinica) against HSV-1 and HSV-2 were more than 8.06 and 24.79, SI of the MeOH extract of Acer okamotoanum leaves were 3.92 and 3.51, SI of the $CH_{2}Cl_{2}$ extract of Veratri Rhizoma et Radix (Veratrum patulum) were 5.49 and 1.31 and SI of the MeOH extract of aerial part of Osmundae Rhizoma (Osmunda japonica) were more than 3.45 and 1.25, respectively.

저선량 CT를 이용한 폐암의 선별 검사 (Lung Cancer Screening with Low-dose Computed Tomography)

  • Lung cancer is the leading cause of cancer death for men and women in the industrialized world. It is desirable to detect disease at a stage when it is not causing symptoms and when control or cure is possible. If the screening test detects patients with the disease at an early stage, they can be examined to confirm the diagnosis and intervention can alter the natural history of the disease. The results of screening programs designed to detect early lung cancer using either conventional chest radiograph or sputum cytology are disappointing for a diagnostic screening test. Because of advances in helical CT imaging techniques, screening for lung cancer has been suggested as a possible method of improving outcome. Findings in recent publications suggest that substantial dose reduction is possible in chest CT. The advantages of low-dose CT are more sensitive than chest radiograph for detecting small pulmonary nodules that may be lung cancers, shorter scanning time than conventional chest CT scan without intravenous contrast injection, cheaper cost than standard CT, low radiation dose. However, the true clinical significance of the small tumors found by screening is still unknown, and their effect on mortality awaits future investigation. Furthermore, in addition to detecting an increased number of lung cancers, low-dose CT found at least one indeterminate nodule in many of all screened patients. The majority should be benign but evaluation of all these indeterminate nodules is not a trivial problem in routine practice. In conclusion, lung cancer screening with low-dose CT is a complex subject. The true effectiveness of lung cancer screening (a reduction in mortality from lung cancer) with low-dose CT can be determined through well-designed randomized control trials with enrolment of appropriate subjects.

식물(植物)의 사포닌 검색(檢索) (Screening of Saponins in the Plants)

  • One hundred and forty-four extracts have been tested for the presence of saponins, using the method of froth formation, precipitate formation by acid hydrolysis, Liebermann-Buchard reaction on the acid hydrolysate, and hemolytic activity of the extracts. Among the samples tested, thirteen extracts showed positive saponin reaction in all the tests employed and one hundred and thirty-six showed positive in one or more tests of saponins. The results are tabulated.

Synthesis of 2-Thio-4-aminopyrimidine Derivatives as Anti-cancer Agent

  • The screening of the anti-cancer activity of the chemical library provided 2-thio-4-aminopyrimidine as the initial hit. The confirmation of structure and biological effect of hit was performed by synthesis and biological evaluation. The optimization of hit was performed by derivatization of substituents while keeping the core structure. The evaluation of growth inhibitory activity was carried out using SRB assay against 6 human cancer cell lines and human fibroblast. The growth inhibitory activity of compounds showed substituent dependency and more than 5 compounds showed complete growth inhibition of cancer cell lines at 10 ${\mu}M$ concentration. Chemical library screening followed by synthetic modification provided possibility that 2-thio-4-aminopyrimidine can be used as a new scaffold for the development of anti-cancer agent.

A Study on Darrieus-type Hydroturbine toward Utilization of Extra-Low Head Natural Flow Streams

  • A two-dimensional Darrieus-type hydroturbine system, installed with a wear for flow streams such as small rivers and waterways, has been developed for hydropower utilization of extra-low head less than 2m. There are several problems such as flow rate change and flowing wastes to be solved for its practical use in natural flow streams. In the present study, at first, a design guideline in the case of overflow or bypass flow is shown by using simple flow model. Next, in order to avoid the unexpected obstacles flowing into the hydroturbine, an installation of waste screening system is examined. It is confirmed that the screen is effective with some amount of bypass flow rate, however the output power is remarkably deteriorated.

  • When three systems commonly used for screening(washed platelets suspended in uffer containing Ca2+, washed platelets in buffer without Ca2+and platelet rich plasma) are compared by studying the anti-aggregating capacity of garlic extract, platelet rich plasma was the least sensitive system. The most sensitive assay system, based on the IC50s of garlic extract and 2 other platelet aggregation-inhibiting agents(vitamin K3 and propranolol), was the platelet preparation without Ca2+in the suspension buffer. This system was confirmed as the most sensitive during subsequent investigation of garlic extract's capacity to inhibit platelet ATP release. These results suggest that applying the system with washed platelet without Ca2+is most effective to screen for the anti-affregating capacity of natural food.

Identification of Regulatory Role of KRAB Zinc Finger Protein ZNF 350 and Enolase-1 in RE-IIBP Mediated Transcriptional Repression

  • One of the WHSC1/MMSET/NSD2 variant RE-IIBP is a histone H3-K27 methyltransferase with transcriptional repression activity. Overexpression of RE-IIBP in various types of leukemia suggests it's role in leukemogenesis. Here we identify two proteins, KRAB zinc finger protein ZNF 350 and enolase-1 as RE-IIBP interacting proteins by yeast two-hybrid screening and confirmed direct interaction in vivo and in vitro. Both proteins have been known for their role in transcriptional repression. Reporter assays using transient transfection demonstrated that both ZNF 350 and enolase-1 proteins synergistically repressed transcription with RE-IIBP, respectively. These results indicate both proteins have roles in RE-IIBP mediated transcriptional repression by involving co-repressor complex.

  • The purpose of this study was to find the effect of 36 traditional medicinal plant species on hepatoprotective activity screening. For this study, carbon tetrachloride $(CCl_{4})$ intoxificated rats were used. Test extracts were made with the traditional medicinal plants refluxed in 95% MeOH and orally administered to the rats. Sixteen species, such as Mentha arvensis, Sophora japonica, Benincasa hispida, Lonicera japonica (Lonicerae Flos), Agaricus blazei, Epimedium koreanum, Aralia continentalis, Lithospermum erythrorhizon, Cimicifuga foetida, Gastrodia elata, Sanguisorba officinalis, Cephalonoplos segetum, Bupleurum falcatum, Alisma plantago-aquatica var. orientale, Lonicera japonica (Lonicerae Folium) and Sinomenium acutum showed protective effect against increased serum alanine aminotransferase (ALT) and/or serum asparteate aminotransferase (AST) activities.

  • The studies for antithrombotic substances from medicinal plants in my laboratory were started from the studies on PAF-antagonistic substances from Korean medicinal plants. The screening studies of PAF-receptor binding antagonistic activity were conducted on the extracts of 300 Korean medicinal plants, 37 tropical medicinal plants, 20 mushrooms, and 30 vegetables. From the results of screening studies, it was possible to select two Korean medicinal plants, i.e. 1) the leaf of Biota orientalis and 2) the seed of Arctium lappa, and two tropical medicinal plants, i.e. 3) the rhizome of Alpinia officinarum and 4) the leaf of Ardisia crispa as the candidates for the activity guided isolation of PAF-antagonistic substances.
