Present study was carried out for development of a new supplementary product with gastroprotective effect. Natural Products mentioned that have GI protective property in Dongeuibogam and reports were evaluated anti-bacterial activity against Helicobacter pylori, then five heres were selected. The material used for the test were water extract of Alpinia oxyphlla (AO), Astragalus membranaceus (AM), Cinnamomum loureirii(CL), Citrus aurantium(CA), Amomum villosum(AV). They were tested individually on HCI ethanol-induced gastric lesion in rats, AV CL, AO showed the most significant effectiveness, respectively. Then two mixture different in their content ratio (P020701-1,-2) and complex with P020701-1 (CP) were made, and tested on HCI·ethanol, indomethacin-induced gastric lesion, aspirin-ligature, Shay ulcer and gastric secretion. P020701-1, -2 and CP showed significant effect on HCI ethanol, indomethacin-induced gastric lesion, and Shay ulcer. It can be regarded that the antigasuitic and anti-ulcerative effects of P020701- 1, -2 and CP are originated from reduction of total acid output identified by gastric secretion test.
During liver fibrosis, hepatic stellate cells (HSCs) undergo a complex activation process characterized by increased proliferation and extracellular matrix deposition, which is the major pathological feature of hepatic cirrhosis. Therefore, suppression of HSCs activation has been proposed as therapeutic strategies for hepatic fibrosis. We tried to screen the antifibrotic activity of natural products employing HSC-T6, hepatic stellate cell lines as an in vitro assay system. In the present study, we investigated the antiproliferative activity of aerial parts and roots of Pulsatilla koreana Nakai (Ranunculaceae). Our present study shows that roots of P. koreana exerted more strong inhibitory activity compared to its aerial parts. In addition, among the fractions of the aqueous methanolic extract of P. koreana roots, both n-hexane and $CHCl_3$ fraction showed the strong inhibitory activity on HSC proliferation. Further study also demonstrated that the n-hexane and $CHCl_3$ fraction of P. koreana roots significantly inhibited the HSC proliferation in time- and concentration-related manners.
We investigated the anti-inflammatory effect of Pyunkang-tang extract (PGT), a complex herbal extract based on traditional Chinese medicine that is used in Korea for controlling diverse pulmonary diseases, on cigarette smoke-induced pulmonary pathology in a rat model of chronic obstructive pulmonary disease (COPD). The constituents of PGT were Lonicerae japonica, Liriope platyphylla, Adenophora triphilla, Xantium strumarinum, Selaginella tamariscina and Rehmannia glutinosa. Rats were exposed by inhalation to a mixture of cigarette smoke extract (CSE) and sulfur dioxide for three weeks to induce COPD-like pulmonary inflammation. PGT was administered orally to rats and pathological changes to the pulmonary system were examined in each group of animals through measurement of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) levels in bronchoalveolar lavage fluid (BALF) at 21 days post-CSE treatment. The effect of PGT on the hypersecretion of pulmonary mucin in rats was assessed by quantification of the amount of mucus secreted and by examining histopathologic changes in tracheal epithelium. Confluent NCI-H292 cells were pretreated with PGT for 30 min and then stimulated with CSE plus PMA (phorbol 12-myristate 13-acetate), for 24 h. The MUC5AC mucin gene expression was measured by RT-PCR. Production of MUC5AC mucin protein was measured by ELISA. The results were as follows: (1) PGT inhibited CSE-induced pulmonary inflammation as shown by decreased TNF-${\alpha}$ and IL-6 levels in BALF; (2) PGT inhibited the hypersecretion of pulmonary mucin and normalized the increased amount of mucosubstances in goblet cells of the CSE-induced COPD rat model; (3) PGT inhibited CSE-induced MUC5AC mucin production and gene expression in vitro in NCI-H292 cells, a human airway epithelial cell line. These results suggest that PGT might regulate the inflammatory aspects of COPD in a rat model.

The immune system is a complex process within the body that protects against disease. Recently, many studies have attempted to discover immunomodulative compounds from natural sources. Pinus koraiensis (PK) cone shell is a by-product of PK. One of the major compounds of PK cone shell is dehydroabietic acid, which has bioactivity, including antiulcer and anti-inflammatory activities. Therefore, this study was performed to examine the immunomodulative effects of PK cone shell. The immunomodulatory effects of PK cone shell extracted with 20% ethanol (EtOH) in vivo were examined initially by measuring the natural killer (NK) cell activity, phagocytic activity, Th1/Th2 cytokines release, serum immunoglobulin, and T/B cell proliferation. The NK cell activity and phagocytic activity were increased significantly by a treatment with a 20% EtOH extract of PK cone shell. Th1 type cytokine and T cell proliferation increased and Th2 type cytokine, B cell proliferation and serum immunoglobulin A, G, and E decreased after a treatment with PK cone shell extract. The 20% EtOH extract of the PK cone shell normalized the unbalanced production of Th1/Th2 type cytokine. This suggests that a 20% EtOH extract of PK cone shell has great potential as a health food.

Acne, also known as Acne vulgaris, is a common disorder of human skin involving the sebaceous gland and Propionibacterium acnes (P. acnes). The purpose of this study was to demonstrate whether anti-acne herbal complex (AAHC), a functional extract from herbal complex can be used for acne treatment as a natural product. We first demonstrated anti-acne activity of AAHC in mouse acne model. Acne was induced by injecting P. acnes on the backside $2{\times}10^7$ CFUs in ICR mice and then the mice were treated with AAHC by dermal application once daily. ACFREE$^{(R)}$ (clindamicin phosphate) was used as a positive control. Treatment with AAHC decreased the P. acnes-induced skin swelling and inflammation. AAHC treatment significantly decreased serum DHT concentration in acne-induced mice. Especially, treatment of 20% AACH in mice was more effected than 40%. We next evaluated the antimicrobial property of AAHC against P. acnes, Staphylcococcus aureus (S.aureus), and Escherichia coli (E. coli). Incubation of P. acnes, S. aureus, and E. coli with AAHC showed minimal inhibitory concentration (MIC) values against the bacterial growth lower. Alamar blue method was also carried for the antibacterial activity. It was effectively MIC level at 6.25% of P. acnes. AAHC effectively inhibited the growth of S. aureus and E. coli at 0.097% on MIC level, respectively. Our results showed the potential of using AAHC as an alternative treatment for antibiotic therapy of acne and the application of AAHC as a herbal medicine for acne treatment.

The hydrolysate of desalinated tuna boiled extract (TBE) were prepared by continuous hydrolysis of TBE using a membrane reactor. TBE and tuna boiled extract hydrolysate (TBEH) were isolated depending on molecular weights. The major molecular weight distributions of TBEH-l0K, TBEH-5K and TBEH-lK were 9,800Da, 3,000Da and 990Da, respectively. The amounts of nucleotides and their related compounds of TBE were 3.47 $\mu$mole/g AMP, 23.75 $\mu$mole/g IMP, 9.07 $\mu$mole/g inosine and 1.89 $\mu$mole/g hypoxanthine. Total content of amino acids having desirable taste (glycine, glutamic acid, alanine, proline, aspartic acid, serine) was about $63\%$ of total amino acid from TBE and about $62\%$ from TBEH. The natural seasoninings were prepared with TBE and TBEH. From the results of sensory evaluations, complex seasoning containing TBEH-1K was almost equal to the shellfish complex seasoning obtained from the market. The mixed sauce which was made by mixing of $50\%$ TBEH sauce and $50\%$ fermented soy sauce was similar to the tradition soybean sauce in product quality and it showed the possibility to be used for the substitute product for acid hydrolyzed soysauce.

Skin aging is a complex biological process due to intrinsic and extrinsic factors. Free radical oxidative is one of extrinsic factors that induce activation of collagenase, elastase and hyaluronidase. Natural product from plants has been used as antioxidant and antiaging. This study aimed to evaluate antioxidant and antiaging properties of Hibiscus sabdariffa extract (HSE) and its compounds including myricetin, ascorbic acid, and ${\beta}$ carotene. The phytochemical of H. sabdariffa was determined using modified Farnsworth method and presence of phenols, flavonoids and tannins were in moderate content, whereas triterpenoids and alkaloids were in low content. Total phenolic content performed using Folin-Ciocalteu method, was $23.85{\mu}gGAE/mg$. Quantitative analysis of myricetin, ${\beta}-carotene$, and ascorbic acid of HSE was performed with Ultra-High Performance Liquid Chromatography (UHPLC) that shows $78.23{\mu}g/mg$ myricetin, $0.034{\mu}g/mg$${\beta}-carotene$, whilst ascorbic acid was not detected. HSE has lower activity on DPPH ($IC_{50}=195.73{\mu}g/mL$) compared to ${\beta}-carotene$, the lowest in ABTS assay ($IC_{50}=74.58{\mu}g/mL$) and low activity in FRAP assay ($46.24{\mu}MFe(II)/{\mu}g\;$) compared to myricetin, ${\beta}-carotene$. Antiaging was measured through inhibitory activity of collagenase, elastase, and hyaluronidase. HSE had weakest collagenase inhibitory activity ($IC_{50}=750.33{\mu}g/mL$), elastase inhibitory activity ($103.83{\mu}g/mL$), hyaluronidase inhibitory activity ($IC_{50}=619.43{\mu}g/mL$) compared to myricetin, ${\beta}-carotene$, and ascorbic acid. HSE contain higher myricetin compared to ${\beta}-carotene$. HSE has moderate antioxidants and lowest antiaging activities. Myricetin is the most active both antioxidant and antiaging activities.
We evaluated the anti-arthritic effect of a new diet-supplement product containing red ginseng, glucosamine, shark cartilage, ascorbic acid and manganese chloride for the relieving arthritic symptoms. Anti-inflammatory activities of the aqueous extract of red ginseng (250 and 500 mg/kg), glucosamine (240 mg/kg) and shark cartilage (240 mg/kg) were tested individually on vascular permeability and carrageenan-induced paw edema. Glucosamine and shark cartilage showed the inhibition of vascular permeability by 29.6 and 32.9%, respectively. Red ginseng (500 mg/kg) and shark cartilage showed the inhibition of carrageenan-induced paw edema at 0.5, 1, 2 and 3 hr. The supplement (red ginseng mixture: RGM) composed of red ginseng (43.5%), glucosamine (25.0%), shark cartilage (25.0%), ascorbic acid (5.0%) and manganese chloride (1.5%) was prepared and its inhibitory activities including vascular permeability and carrageenan-induced paw edema were comparable to anti-inflammatory drugs such as diclofenac and ibuprofen. It was also tested on adjuvant-induced arthritis in rats as one of chronic arthritic tests and Randall-Selitto assay as an analgesic test. RGM showed the inhibition against the swelling of rat paws induced by Mycobacterium tuberculosis at a dose of 1,500 mg/kg. Determination of cytokines of the sera sampled from arthritis-induced animals indicated that RGM increased the levels of $interferon-{\gamma}$ and interleukin-6, representing the immunostimulatory effect by red ginseng. RGM treatment moderately reduced the production of NO in RAW 264.7 cells in a dose-dependent manner. Taken together, these results support that RGM can be applicable for the improvement of arthritic as a new diet-supplement.
Cimicifuga heracleifolia extract (CHE) was investigated for its effects on the release of nitric oxide (NO) and at the level of inducible nitric oxide synthase (iNOS) gene expression in mouse macrophages. We found that C. heracleifolia elicited a dose-dependent increase in NO production and the level of iNOS mRNA. Since, iNOS transcription has been shown to be under the control of the transcription factor $NF-{\kappa}B$, the effects of CHE on $NF-{\kappa}B$ activation were examined. Transient expression assays with $NF-{\kappa}B$ binding sites linked to the luciferase gene revealed that the increased level of iNOS mRNA, induced by CHE, was mediated by the $NF-{\kappa}B$ transcription factor complex. By using DNA fragments containing the $NF-{\kappa}B$ binding sequence, CHE was shown to activate the protein/DNA binding of $NF-{\kappa}B$ to its cognate site, as measured by electrophoretic mobility shift assay. These results demonstrate that C. heracleifolia stimulates NO production and is able to up-regulate iNOS expression through $NF-{\kappa}B$ transactivation.
Hygrophila auriculata (K. Schum.) Heine is a wild herb commonly found in moist places on the banks of tanks, ditches and paddy fields throughout India and is one of the main sources of Ayurvedic 'Rasayana' drug. The free radical scavenging potential of 50% aqueous alcoholic extract of H. auriculata (HAEt) was studied for DPPH scavenging activity, nitric oxide, hydroxyl radical and ferryl bipyridyl complex scavenging activity along with lipid peroxidation and total antioxidant capacity inhibition using Thiobarbituric acid reactive substances (TBARS) in rat liver homogenate. Integral antioxidative capacity was determined by photochemiluminescence assay. HAEt showed good radical scavenging activity at various concentrations $(200-1000\;{\mu}g/ml)$ against 1,1 diphenyl, 2-picrylhydrazyl (DPPH) (32.32-77.02%) with moderate scavenging activity against Nitric oxide (12.46-52.84%), hydroxyl radical (11.69-55.26%), ferryl bipyridyl complex (17.66-58.67%) and lipid peroxidation (0.829-0.416nmoles/mg protein). The above results indicate HAEt to be very effective antioxidant. It was suggested that the varied therapeutic activities claimed for the plant in the Indian indigenous systems of medicine may be in the part due to the free radical scavenging and/ or potent antioxidant activity.
