The Egg Development of Korean Slender Gudgeon, Squalidus gracilis majimae (Cypriniforms: Cyprinidae) (한국산 긴몰개 (Squalidus gracilis majime, Cyprinidae)의 난발생)

  • This study was conducted, based on the field survey and laboratory observations, to elucidate egg developmental processes and their characteristics of the Korean slender gudgeon, Squalidus gracilis majimae. For the experiments, the mature adults were collected at the Woongcheon-Cheon Stream and Boreung Reservoir located in Boreung City, Chungnam Province and eggs were obtained from the natural spawning area. Morphological characteristics of the egg and embryonic development were summarized as follows: The shape of the fertilized egg was spherical, adhesive and transparent. The fertilized egg was 2.9${\pm}$0.3 mm (n = 30) in mean diameter under water temperature of $26{\pm}1.5^{\circ}C$, light white in color and had no oil droplets. After 20 minutes from the time of fertilization, a blastodisc was formed and divided into two cells at 48 minutes after fertilization. The blastular stage occurred at 5 hours 40 minutes after fertilization and the gastrular stage was detected at 8 hours 41 minutes after fertilization. The beginning of embryo formation was observed at 12 hours 58 minutes after fertilization and optic vesicles and 9 somites were discovered at 17 hours 05 minutes after fertilization. Differentiation of brains and embryo wiggling were observed at 37 hours 27 minutes after fertilization. Heart beating and the formation of melanophores in optic vesicles were detected at 44 hours 46 minutes after fertilization. The formation of pectoral fins and melanophores in the body were discovered at 50 hours 36 minutes after fertilization. Hatching occurred at 57 hours 49 minutes after fertilization. The newly hatched larvae were 3.3${\pm}$0.2 mm (n = 120) in total length. We believe that these results may contribute the species and population conservations under the situation of accelerated water pollution and the decreases of its diversity.

  • A review on the papers published in the Korean Journal of Air-Conditioning and Refrigerating Engineering in 2004 and 2005 has been done. Focus has been put on current status of research in the aspect of heating, cooling, air-conditioning, ventilation, sanitation and building environment. The conclusions are as follows. (1) Most of fundamental studies on fluid flow were related with heat transportation of facilities. Drop formation and rivulet flow on solid surfaces were interesting topics related with condensation augmentation. Research on micro environment considering flow, heat, humidity was also interesting for comfortable living environment. It can be extended considering biological aspects. Development of fans and blowers of high performance and low noise were continuing topics. Well developed CFD and flow visualization(PIV, PTV and LDV methods) technologies were widely applied for developing facilities and their systems. (2) The research trends of the previous two yews are surveyed as groups of natural convection, forced convection, electronic cooling, heat transfer enhancement, frosting and defrosting, thermal properties, etc. New research topics introduced include natural convection heat transfer enhancement using nanofluid, supercritical cooling performance or oil miscibility of $CO_2$, enthalpy heat exchanger for heat recovery, heat transfer enhancement in a plate heat exchanger using fluid resonance. (3) The literature for the last two years($2004{\sim}2005$) is reviewed in the areas of heat pump, ice and water storage, cycle analysis and reused energy including geothermal, solar and unused energy). The research on cycle analysis and experiments for $CO_2$ was extensively carried out to replace the Ozone depleting and global warming refrigerants such as HFC and HCFC refrigerants. From the year of 2005, the Gas Engine Heat Pump(GHP) has been paid attention from the viewpoint of the gas cooling application. The heat pipe was focused on the performance improvement by the parametric analysis and the heat recovery applications. The storage systems were studied on the performance enhancement of the storage tank and cost analysis for heating and cooling applications. In the area of unused energy, the hybrid systems were extensively introduced and the life cycle cost analysis(LCCA) for the unused energy systems was also intensively carried out. (4) Recent studies of various refrigeration and air-conditioning systems have focused on the system performance and efficiency enhancement. Heat transfer characteristics during evaporation and condensation are investigated for several tube shapes and of alternative refrigerants including carbon dioxide. Efficiency of various compressors and expansion devices are also dealt with for better modeling and, in particular, performance improvement. Thermoelectric module and cooling systems are analyzed theoretically and experimentally. (5) According to the review of recent studies on ventilation systems, an appropriate ventilation systems including machenical and natural are required to satisfied the level of IAQ. Also, an recent studies on air-conditioning and absorption refrigeration systems, it has mainly focused on distribution and dehumidification of indoor air to improve the performance were carried out. (6) Based on a review of recent studies on indoor environment and building service systems, it is noticed that research issues have mainly focused on optimal thermal comfort, improvement of indoor air Quality and many innovative systems such as air-barrier type perimeter-less system with UFAC, radiant floor heating and cooling system and etc. New approaches are highlighted for improving indoor environmental condition as well as minimizing energy consumption, various activities of building control and operation strategy and energy performance analysis for economic evaluation.

Studies on Discrimination between Organic Rice and Non-organic Rice using Natural Abundance of Stable Isotope Nitrogen($\delta^{15}N$) (질소 안정동위원소 자연존재비($\delta^{15}N$)를 이용한 유기벼와 일반벼 판별법 탐색)

  • To investigate the possibility of discrimination between organic and non-organic rice using stable isotope nitrogen of natural abundance, organic rice of 17 samples and non-organic rice of 13 samples grown at adjoining organic rice field were collected in 2008. Rice was grinded into brown rice, milled rice and hull, and samples were analysed for nitrogen and $\delta^{15}N$ at NICEM. Authors also made inquiries about N source for both farmers who conduct organic- and non-organic rice cultivation. In order to know whether the $\delta^{15}N$ can be used in discrimination between organic and non-organic rice, discriminant analysis were made with SPSS and logistic method. 1. Organic farmers used manure, rice bran, used mushroom culture, fermented fertilizer (company products), and oil cake, but non-organic farmers applied compound fertilizer. Rice straws were remained in organic rice field while moved out in non-organic field. 2. There were difference in $\delta^{15}N$ among organic rice and its byproduct(7.760????% in hull, 6.720????% in rice), but significant difference was not found between them. And the trend was same between province. Non-organic rice showed similar results. 3. Significant difference of $\delta^{15}N$ were found between organic rice and non-organic rice (p<0.01) and between hull of organic rice and that of non-organic rice hull (p<0.05). $\delta^{15}N$ seemed to be useful criteria for discrimination of organic and non-organic rice. 4. When applied discrimination analysis of SPSS and logistic, there were significant difference between organic rice, non-organic rice and its byproducts except brown rice and hull in SPSS method. Hull can be used as the most useful component for unknown sample prediction with 83.3% probability.

Study on the Biodegradability of Dispersants and Dispersant/Bunker-C Oil Mixtures and the Dissolved Oxygen Consumption in the Seawater(I) - The Biodegradability of Dispersants and the Dissolved Oxygen Consumption in the Seawater - (해수중에서 유처리제 및 유처리제/Bunker-C유 혼합물의 생분해도와 용존산소소비에 관한 연구(I) - 유처리제의 생분해도와 용존산소소비 -)

  • As the dispersants and the dispersant/oil mixtures are degraded naturally by the microorganisms in the seawater, the consumption of dissolved oxygen may cause marine organisms to be damaged especially in the waters where the dissolved oxygen level is low due to the pollution and the restriction of seawater flow. The biodegradation experiment, the TOD analysis and the element analysis for three dispersants(SG, GL and WC) and a nonionic surfactant(OA-5) were conducted for the purposes of evaluating the biodegradability of dispersants and studying the effect of dispersants on dissolved oxygen in the seawater. The results of biodegradation experiment showed 1mg of dispersants to be equivalent to $0.403{\sim}0.595mg$ of $BOD_5$ and to $0.703{\sim}0.855mg$ of $BOD_{20}$, and 1mg of nonionic surfactant to be equivalent to 0.50mg of $BOD_5$ and to 0.97mg of $BOD_{20}$ in the natural seawater. The results of TOD analysis showed 1mg of dispersants to be $2.37{\sim}2.80mg$ of TOD and 1mg of nonionic surfactant to be 2.45mg of TOD. The results of element analysis showed carbon content and hydrogen content to be $67.6{\sim}76.5\%$ and $10.2{\sim}12.2\%$ for dispersants, and $65.3\%$ and $10.3\%$ for nonionic surfactant, respectively. No nitrogen element was detected in dispersants and a nonionic surfactant. The biodegradability of dispersants shown as the ratio of $BOD_5/TOD$ was found to be in the range of $17{\sim}21\%$, and that of nonionic surfactant was found to be about $20\%$. This means that dispersants and nonionic surfactant belong in the organic matter group of middle-biodegradabilily. The deoxygenation rates($K_1$) and ultimate oxygen demands($L_o$) obtained through the biodegration experiment and Thomas slope method were found to be $0.121{\sim}0.171/day$ and $3.155{\sim}3.810mg/l$ for 4mg/l of dispersants and to be 0.181/day and 1.911mg/l for 2mg/l of nonionic surfactant in the seawater, respectively.

Study of Trans Fatty Acids and Saturated Fatty Acids in Child-favored Foods (어린이 기호식품 중 트랜스지방 및 포화지방 실태조사)

  • We studied the amounts of trans fatty acids and saturated fatty acids in child-favored foods. Confectioneries, breads, donuts and hamburgers were included in this study. The total fatty acid content and the amount of each type of fatty acid were determined by gas chromatography with a flame-ionized detector. Confectioneries were shown to have the highest content of crude fat. The mean content was 24.2${\pm}$6.9 (range: 4.6~41.1) g/100 g food. The mean content of crude fat in donuts, bread and hamburgers was 23.9${\pm}$5.8 (range: 14.1~39.5) g/100 g food, 15.7${\pm}$7.9 (range: 1.4~30.0) g/100 g food, and 9.5${\pm}$3.4 (range: 4.5~18.5) g/100 g food, respectively. Bread had the most trans fatty acids at 1.3 g/100 g food. This result inferred that manufacturers have tried to reduce the trans fat content. The mean content of saturated fatty acids in confectioneries, donuts, bread, and hamburgers was 11.6${\pm}$4.8 (range: 2.0~22.7) g/100 g food, 11.2${\pm}$4.0 (range: 4.8~23.2) g/100 g food, 6.9${\pm}$4.1 (range: 0.6~15.4) g/100 g food, 3.0${\pm}$1.0 (range: 1.0~5.8) g/100 g food, respectively. This content depended on crude fat. The composition of fatty acids varied according to the oil and fat used in the manufacturing process of each food, and the natural content. Foods that were fried in vegetable oil tended to be especially low in saturated fat.

Enhancing the Effects of Zerumbone on THP-1 Cell Activation (단핵구세포주의 활성에 미치는 Zerumbone의 영향)

  • Zerumbone is a major component of the essential oil from Zingiber zerumbet Smith, which is a kind of wild ginger. In addition, various biological functions, such as liver protection, pain relief, atherosclerosis, and antimicrobial activity have been reported. It is also known to be effective in the proliferation of immune cells and the expression of cytokines. In this study, we investigated the effects of zerumbone on monocyte activation. First, it was confirmed that the proliferation of THP-1 cells was increased by zerumbone. The strongest increase in THP-1 proliferation after lipopolysaccharide treatment was observed at $5{\mu}M$ zerumbone treatment, and the increase of cell proliferation without lipopolysaccharide was the highest at $10{\mu}M$. Conversely, when treated with $50{\mu}M$ zerumbone, a rapid decrease of proliferation was observed regardless of the presence of lipopolysaccharide (LPS). The phosphorylation of signaling protein, Erk, induced by LPS was also increased by zerumbone. The strongest increase in phosphorylation was observed when treated with $50{\mu}M$ of zerumbone with reduced proliferation. The activity of transcription factor $NF-{\kappa}B$ was not significantly altered by zerumbone alone, but increased when treated with lipopolysaccharide. Furthermore, the transcription of the inflammatory cytokines $TNF-{\alpha}$ and IL-8, which are regulated by $NF-{\kappa}B$, is also increased by zerumbone. These results suggest that zerumbone can enhance the proliferation and activity of monocytes. Furthermore, it is believed that zerumbone can enhance rthe immune responses through increased monocyte activity in bacterial infections with LPS, thereby helping to treat effective bacteria.

Effect of the Various Sources of Dietary Additives on Growth, Body Composition and Shell Color of Abalone Haliotis discus hannai (다양한 원료의 사료첨가제가 전복의 성장, 체조성 및 패각 색채에 미치는 영향)

  • Effect of the various sources of dietary additives on growth, body composition and shell color of abalone Haliotis discus hannai was investigated for 16 weeks. Forty juvenile abalone averaging 13.5 g were randomly stocked into 21 of 50 L plastic rectangular containers each. Eight kinds of additives were prepared for this study: four commercially available microalgae [Haeatococcus (Hae), Isochrysis galbana (Iso), Shizochytrium (Sch) and Spirulina (Spi)], three crustacean meals [krill meal (KM), shrimp head meal (Shm) and red crab meal (Rcm)], and green tea by-product (Gre). In addition, dry sea tangle (Dst), Laminaria japonica, as a control, was prepared. Casein, dextrin and a mixture corn oil and fish oil was protein, carbohydrate and lipid sources, respectively, in the experimental diets. The 2% each additive was included into the experimental diets. The experimental diets were fed to abalone once a day at the ratio of $1.5{\sim}2.0%$ total biomass of abalone with a little leftover throughout the 16-week feeding trial. Survival of abalone was not significantly (P>0.05) affected by the experimental diets. However, weight gain of abalone fed the all experimental diets containing the various sources of additives was significantly (P<0.05) higher than that of abalone fed the Dst diet. Weight gain of abalone fed the Spi diet was highest and Shi, KM and Iso diets in order. Shell length and the ratio of soft body weight to body weight of abalone was not significantly (P>0.05) affected by the experimental diets. However, shell width of abalone fed the all experimental diets containing the various sources of additives was significantly (P<0.05) higher than that of abalone fed the Dst diet. The shell color of abalone fed the Spi diet was improved the most distinctively and similar to that of natural abalone. Therefore, it can be concluded that the experimental diets with the various sources of additives (microalgae and crustacean meals) was effective to improve growth of abalone and dietary inclusion of Spirulina was most effective to improve shell color of abalone.

Changes in Physicochemical Properties and Microbial Population during Fermenting Process of Organic Fertilizer (혼합발효 유기질비료의 발효과정 중 이화학성 및 미생물밀도 변화)

  • This study was conducted to investigate the changes in physicochemical and microbiological properties during fermenting process of organic fertilizer which was made from the mixture of organic materials such as sesame oil cake, fish meal, blood meal, rice bran, ground bone meal, and natural minerals such as illite, crusted oyster shell and loess. They were mixed and fermented for 70 days. The sesame oil cake and rice bran, major ingredients for organic fertilizers, consisted of 7.6 and 2.6% total nitrogen, 3.6 and 4.6% $P_2O_5$, 1.4 and 2.2% $K_2O$, respectively. The ground bone meal included 29.2% $P_2O_5$ and illite included 3.8% $K_2O$. Temperature of organic fertilizer during the fermentation rapidly increased over $50^{\circ}C$ within 2 days after mixing and stabilized similar to outdoor temperature after 40 days. Moisture content decreased from 36.3 to 16.0% after 1 month. C/N ratio of organic fertilizer slightly increased until 30 days and thereafter, it slowly decreased, It resulted from the faster decrease of total nitrogen concentration compared with organic matter. Concentration of $NH_4-N$ in organic fertilizer rapidly increased from 1,504 to $5,530mg\;kg^{-1}$, the highest concentration after 10 days. Meantime, $NO_3-N$ concentration was low and constant about $150mg\;kg^{-1}$ over the whole fermenting period. This result seemed to be due to the high pH. The organic ferfilizer fermented for 70 days was composed of 2.7% N, 2.8% $P_2O_5$, 1.8% $K_2O$, and 35.9% organic matter. Total populations of aerobic bacteria, Bacillus sp. and actinomycetes, after fermenting process, were $12.5{\times}10^{10}$, $45.5{\times}10^{5}$ and $13.6{\times}10^{5}cfu\;g^{-1}$ respectively. Pseudomonas sp. was $71.9{\times}10^{7}cfu\;g^{-1}$ at first, but it rapidly decreased according to the rise of temperature. Yeasts played an important role in the early stage of fermentation and molds did in the late stage.

Effects of Dietary Fatty Acids on fatty Acid Pattern in Development Rat Brain Phospholipids - Effects on P/M/S and $\omega$3/$\omega$6 Fatty Acid Ratios -

  • Docosahexaenoic acid(DHA), a $\omega$3 series fatty acid and arachidonic acid(AA). a $\omega$6 series fatty acid were found in relatively high concentrations in the phospholipids(PLs) of cell membranes of nerve tissues, and they can be affected by various factors. The present study examined the effects of dietary $\omega$6 and $\omega$3 fatty acid composition on P/M/S and on $\omega$3/$\omega$6 fatty acid ratios in brain PLs of 2nd generation rats. The expeimental diets consisted of 10% fat(by wt), which were computer- searched mixed oil('M') with P/M/S ratio, 1 : 1.4 : 1 and $\omega$6/$\omega$3 ratio, 6 : 1 and safflower oil('S') poor in $\omega$3 fatty acids. The experimental diets were started 3-4 wks prior to conception. During the lactation period, the feeding mothers were switched 1 wk after birth and provided the pups for 2 wks with milk which had compositions different from that of their natural mother. The same diet as their mothers was provided from weaning to 9 wks of age. The 'M'and 'S' rats were again subdivided into MM, MS, SS, SM rats according to diet which their lactating mothers were fed from the begining of the experiment. The relative percentage of P/M/S fatty acids in brain PLs in all experimental groups converged to a very similar value at 9 wks of age, indicating the existence of a control mechanism for the degree of fatty acids, unsaturation. The $\omega$3/$\omega$6 fatty acid ratios of brain PLs converged to about 1.0 in MM & SM groups and to 0.7 in SS & MS groups, suggesting also the existence of some balance between $\omega$3 and $\omega$6 fatty acids in developing rat brain. The concentrations of $\omega$3 fatty acids, especially DHA, in the SM group were increased and became similar to those in MM group at 9 wks of age. The increase in DHA of brain PLs was counterbalanced b)r a decrease in 22 5$\omega$6. Therefore, the ratios of 22 : 6$\omega$6/22 : 5$\omega$6 were higher in both MM & SM groups than those of SS & MS groups at 9 wak of age. Although dietary $\omega$3 and $\omega$6 fatty acids affected 22 : 6$\omega$S and 22 : 5$\omega$6 contained in rat brain PLs reciprocally, the relative percentage of AA did not appear to be significantly influenced by the diet in all groups at 9 wks of age, suggesting that a mechanism for the maintenance of a certain level of AA in brain PLs exists. In conclusion, the $\omega$3/$\omega$6 fatty acid and 22 : 6$\omega$3/22 : 5$\omega$6 ratios, but not P/M/S ratio, of rat brain PLs were affected by the postnatal dietary changes. Futher studies are required to clarify the mechanism(S) of ensuring a certain level of DHA and of maintaining a similar level of AA in rat brain PLs after. weaning(9 wk) regardless of prenatal and postnatal dietary changes. Estrogen Replacement Effect of Korean Ginseng Saponin on Learning and Memory of Ovariectomized Mice

  • Estrogen can influence on the expression of behaviors not associated directly with reproduction, including learning and memory. Recently estrogen has received considerable attention for its effects on neuroprotection and neural circuits in brain areas associated with cognition. Although estrogen replacement therapy may be helpful to postmenopausal women, it also results in a number of harmful side effects. Ginseng also has steroidal qualities and contains several ginsenoside components which have similar backbone structure to estrogen. The objectives of this experiment were 1) to examine the effects of estrogen and 2) to investigate the effects of ginsenosides as estrogenic agent on learning and memory using the Morris water maze, a traditional experimental task for spatial memory. In the experiments designed here, ovariectomized mice were implanted subcutaneously with Sila, itic capsules containing 17${\beta}$-estradiol (100∼250 $\mu\textrm{g}$/$m\ell$), panaxadiol (PD) and panaxatriol (PT) saponins (15∼100 $\mu\textrm{g}$/$m\ell$) diluted with sesame oil. In the first set of experiment, the effects of estradiol on learning and memory during the Morris water maze was examined. When estradiol was delivered via Silastic capsules following training improved spatial memory performance in ovariectomized female mice. In the second set of experiment, three different PD and PT saponin concentrations were delivered via Silastic implants to ovariectomized female mice and their effects were compared with estrogenic effects. Results of three separate experiments demonstrated that estradiol, PD and PT administrated by Silastic implants for 2 weeks prior to water maze training significantly improved spatial memory performance compared to ovariectomized (OVX) mice, as indicated by lower escape latency over trial. The positive effect of estradiol suggests that estrogen can affect performance on learning and memory. In addition, the positive effect of PD and PT saponins suggest that ginsenosides have an estrogen-like effects in mediating learning and memory related behavior action.
