Numerical Study On Combined Natural Convection-Radiation In Partially Open Square Compartments with A Heater (발열체가 있는 열린 공간내에서의 자연대류-복사열전달 현상에 관한 수치적 연구)

  • Study on combined natural convection-radiation In partially open square enclosures filled with absorbing-anisotropic scattering media is performed. A heater block located in the enclosure causes the natural circulation of the fluid in the enclosure which results In significant in-flow of the cold fluid through the partially open wall. Four different locations of the heater are considered to observe the effect of the heater locations on the resulting heat transfer. Results obtained from the combined convection-radiation analyses show much stronger circulation of t he fluid inside the enclosure as compared to those obtained from the pure convection analyses. As the ratio of the open area is Increased, the inflow of the cold fluid and the circulation of the fluid inside the enclosure is increased causing lower fluid temperature Inside the enclosure. It is shown that the location of the heater influences the circulation and heat transfer significantly by showing stronger circulations and more uniform temperature distributions for the cases where the heater is located on the bottom wall as compared to those for the cases where the heater is located on the upper part wall of the enclosure. For pure absorbing medium, the expected circulation in the fluid is relatively week as compared to those with absorbing-scattering medium due to the smaller wall heating as the radiant heat is used to heat the fluid instead. The forward anisotropic scattering phase function is shown to increase the fluid circulation further as compared to the isotropic scattering medium.

  • One of the most important challenges facing the Spirulina mass cultivation industry is to find a way to reduce the high production costs involved in production. Although the most commercial medium (Zarrouk's medium) for Spirulina cultivation is too expensive to use, it contains higher amount of $NaHCO_3$ (16.80 g $L^{-1}$), trace metals and vitamin solutions. The purpose of this study was to increase the efficiency of Spirulina platensis biomass production by developing a low-cost culture medium at an isolated tropical island such as Chuuk State, Federated States of Micronesia (FSM). This study set out to formulate a lowcost medium for the culture of S. platensis, by substituting nutrients of Zarrouk's medium using fertilizer- grade urea and soil extract with a different concentration of carbon source under natural weather condition. In order to select a low-cost culture medium of S. platensis, 10 culture media were prepared with different concentrations of nitrogen (urea and $NaNO_3$) and $NaHCO_3$. The highest maximum specific growth rate (${\mu}max$) and mass production were 0.50 $day^{-1}$ and 1.05 g $L^{-1}$ in modified medium ($NaHCO_3$ 7.50 g $L^{-1}$, urea 2.00 g $L^{-1}$ without $NaNO_3$) among all the synthesized media. Protein (56.14%) and carbohydrate (16.21%) concentrations of the lyophilized standard samples were estimated with highest concentration of glutamic acid (14.93%). This study revealed that the use of a low concentration of urea and $NaHCO_3$ with soil extract was an affordable medium for natural mass cultivation in the FSM.

  • In batch culture of A. pullulans HP-2001, maximal production of pullulan was 57.90 g/l at 96 hr. The maximal production of pullulan with 76.55 g/l occurred when substituted solution was 10% (w/v) sucrose, 0.25% (w/v) yeast extract and mineral salts. The maximal production of pullulan was 75.00 g/l with a dilution rate of $0.015\;h^{-1}$. Productivity of pullulan in the continuous culture fed with the fresh medium was higher than that fed with the fresh medium without nitrogen source.

  • Streptomyces sp. MCY-846 selected by in vitro cytotoxicity assay produced elaiophylin. Individual characteristics of the strains such as spore morphology, and physiological characteristics indicated that the strain is resembled to Streptomyces hygroscopicus. The time course of cell growth and antibiotic production was observed in the medium containing 0.5% trehalose and 0.5% soybean meal as carbon and nitrogen sources, respectively. The optimum production of elaiophylin was tested with different combinations of carbon and nitrogen sources and reached a maxima of $470{\mu}$/ml in the PC-II medium.

  • On the whole the various medium colors were developed by repeat dyeing with different colorants after dyeing with one colorant in case of natural dyeing. In this study, however in order to obtain various colors on silk fabrics with natural dyes, pre-mordanting and combination dyeing were carried out. Metal compounds containing alum, copper and iron were used for pre-mordants. Gardenia and Sappan wood were used as colorants for combination dyeing. In combination dyeing of Gardenia and Sappan wood, these two natural colorants showed good miscibility in dye bath. The various colors were obtained according to various metal compounds for pre-mordants and various mixing portion of Gardenia and Sappan wood.

  • The rate of growth and production of bioactive substances from Rehmannia glutinosa Liboschitz (Scrophulariaceae) were studied with the variation on the constituents of the culture media. The best growth was observed from MS basal medium containing 3.0 ppm NAA and 2.0 ppm kinetin. Carbohydrates (fructose, glucose and sucrose), phytosterols(${\beta}-sitosterol$, campesterol and stigmasterol) and carotenoid like substances were identified by GC-MS and TLC from the callus mass. However, catalpol was not detected from both solid and cell suspension cultures containing geraniol. Callus cultured Rehmannia glutinosa in the MS basal medium containing 0.1 ppm NAA and 0.1 ppm kinetin become differentiated to root.

  • The problem of natural convection from a vertical fin is solved by coupling the thermal diffusion equation in the fin to the constitutive equations of the ambient medium involving the radiation of the medium. The analysis is accomplished by employing an integral method. The governing equations for the problem are solved by shooting method based on the Runge-Kutta Scheme at Pr= 0.7. For the range of values of the fin parameter and the radiation-conduction parameter in the analysis, the numerical results show that the radiation effects play an important role in the heat transfer and enhance the heat transfer.

  • Cirsium setidens nakai belonging to cirsium has been reported to have various physiological activities including anticancer activity because it contains polyphenols, dietary fiber, minerals and vitamins. Despite these positive positive efficacies, however, no studies have studied cirsium setidens nakai products as biomaterials such as cellular metabolism and bone formation. Thus, the aim of this study was evaluate of osteogenesis differentiation a natural material extracted from cirsium setidens nakai. The natural materials in this studys in this studywere created by 40% ethanol extraction process and then dried. FabricatedFabricatedpowders were added to a medium at various concentrations (0.01, 0.05, 0.1, 0.2, and $0.25{\mu}g/mL$), and pure medium was used as a control. The natural material caused positive increases in cell metabolic activity and mineralized bone formation without cytotoxicity. In addition, we observed higher expression of genes such as ALP, BSP, Runx2 and COL1 in cirsium setidens nakai treatment cells. As a result, this study produced and investigated cirsium setidens nakai extracts and the natural materials showed potential biomaterials. In this research indicated that the cirsium setidens nakai extracts might have promising applications in areas of agricultural, biological and food engineering as a biomaterial.

Molecular Structure of PCR Cloned PHA Synthase Genes of Pseudomonas putida KT2440 and Its Utilization for Medium-Chain Length Polyhydroxyalkanoate Production

  • A new phaC gene cluster encoding polyhydroxyalkanoate (PHA) synthase I PHA depolymerase, and PHA synthase II was cloned using the touchdown PCR method, from medium-chain length (mcl-) PHA-producing strain Pseudomonas putida KT2440. The molecular structure of the cloned phaCl gene was analyzed, and the phylogenic relationship was compared with other phaCl genes cloned from Pseudomonas species. The cloned phaCl gene was expressed in a recombinant E. coli to the similar level of PHA synthase in the parent strain P. putida KT2440, but no significant amount of mcl-PHA was accumulated. The isolated phaCl gene was re-introduced into the parent strain P. putida KT2440 to amplify the PHA synthase I activity, and the recombinant P. purida accumulated mcl-PHA more effectively, increasing from 26.6 to $43.5\%$. The monomer compositions of 3-hydroxylalkanoates in mcl-PHA were also modified significantly in the recombinant P. putida enforcing the cloned phaCl gene.
