Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.

Molecular Structure of the PHA Synthesis Gene Cluster from New mcl-PHA Producer Pseudomonas putida KCTC1639

  • Pseudomonas putida KCTC 1639 was newly identified as a potential producer of biodegradable medium chain length polyhydroxyalkanoates. It exhibited a carbon assimilation pattern quite different from other known P. putida strains, but a more similar pattern with P. oleovorans, which assimilates the carbon sources mainly through ${\beta}$-oxidation rather than the fatty acid biosynthesis pathway. The PHA synthesis gene cluster from P. putida KCTC1639 was composed of two gene loci; the PHA synthase gene locus and granule-associated gene locus, which were cloned and deposited in the GenBank under accession numbers AY286491 and AY750858 as a new nucleotide sequence, respectively. The molecular structure and amino acid homology of the new gene cluster were compared with those from Pseudomonas species, including other P. putida strains and P. oleovorans, and a higher than $90\%$ homology was observed.

Biodegradation of Phenol by a Trichloroethylene-cometabolizing Bacterium

  • A microorganism which degrades phenol and co-metabolizes trichloroethylene (TCE) was isolated from Yangsan stream after enrichment in a medium containing phenol as the sole carbon source. The isolate EL-43P was identified as the genus Rhodococcus by its morphological, cultural and physiological characteristics. Phenol-induced cells of Rhodococcus sp. EL-43P degraded TCE. Toluene and nutrient broth could not replace the phenol requirement. The optimal conditions of initial pH and temperature of media for growth were 7.0~9.0 and $30~50^{\circ}C$, respectively. Rhodococcus sp. EL-43P could grow with phenol up to 1,000 ppm. Growth was inhibited by phenol at a concentration above 1,500 ppm. It was observed that Rhodococcus sp. EL-43P was able to degrade 90% of phenol (1,000 ppm) after 40 h in a culture. Phenol-induced cells of Rhodococcus sp. EL-43P degraded 95% of $5{\mu}M$ TCE in 6 h. Rhodococcus sp. EL-43P hardly degraded TCE above $100{\mu}M$.

미생물 violacein 색소의 다섬교직포에서의 염색성 (Dyeing Properties of Microbial Violacein on Mutifiber Fabrics)

  • Dyeability of microbial violacein produced from Chromobacterum violaceum CV107 on to multifiber fabrics has been studied. The bluish-purple colourants were produced by cultivation of Chromobacterum violaceum using LB liquid medium for 2 days. The colourant was extracted with 80% acetone and identified as violacein by LC/MS analysis. The violacein could be dyed on not only natural fibers such as Cotton, Silk and Wool but also synthetic fibers such as Diacetate, Triacetate, Creslan 61 and Nylon 66. Maximum K/S values were shown at 540-580 nm according to different fiber with color appearance of purple or blue. An optimum pH and temperature under dyeing condition were 10 and $70^{\circ}C$, respectively. Any mordants were not improved colour density and quality on various fabrics. From this studies, pigments produced microbe have a high potentials for natural dyeing on fabrics. Finally, development of new colourants from microbe has made a possible change for new dyeing field in respects of eco-friend and repeatability of natural dyeing for apparels.

  • In recent years, Floating Liquefied Natural Gas (FLNG) Unit have attracted considerable attention. Generally, liquefied natural gas (LNG) units are produced in onshore liquefaction terminals from gas supplied from onshore gas fields or large-scale offshore gas fields near the coast. However, the development of these gas fields has approached saturation. Large-scale offshore gas fields far from the coast, as well as undeveloped medium- and small-scale offshore gas fields, have recently attracted attention. Among several proposed concepts, the floating LNG plant in the form of the FLNG system was chosen for further evaluation and development, considering worldwide receiving infrastructure. The design of a 2.5 million tonne per annum FLNG unit has been completed with a capacity corresponding to that of modern onshore liquefaction plants. Various simulation tests were performed to evaluate the performance of the electrical power plant, focusing on the efficiency of the electrical system to secure the aspects of plant safety. This design study analyzes the electrical system for the FLNG unit to improve the safety of operation and maintenance in the field.

Emulsification of Crude Oil by Acinetobacter sp. SH-14

  • As basic study to evaluate the treatability of oil-contaminated environment with bacteria, isolation and characterization of crude oil-degrading bacterium were carried out. A bacterial strain SH-14 capable of degrading crude oil was isolated from contaminated soils by enrichment culture technique and identified as Acinetobacter sp. by morphological, cultural and biochemical characteristics, and so named Acinetobacter sp. SH-14. The optimal medium composition and cultural conditions for the growth and emulsification of crude oil by Acinetobacter sp. SH-14 used were crude oil of 2.0%, $KNO_3$ of 0.2%, $K_2HPO_4$ of 0.05%, and $MgSO_4\;{\cdot}\;7H_2O$ of 1.0%, along with initial pH 7.0 at $30^{\circ}C$. Acinetobacter sp. SH-14 showed to be resistant to chloramphenicol and utilized various hydrocarbons such as dodecane, hexadecane, isooctane, cyclo-hexane etc., as a sole carbon source. Acinetobacter sp. SH-14 harbored a single plasmid. By agarose gel electrophoresis and curing experiment it was found that the genes for crude oil components degradation were encoded on the plasmid.

Opitimization of culture conditions involved in dissolved oxygen for production of pullulan by Aureosidium pullulan

  • Effect of carbon source and culture conditions involved in the concentration of dissolved oxygen on cell growth and the production of pullulan by A. pullulans HP2001 were investigated. Among those carbon sources, glucose was found to be the best carbon source for the production of pullulan by A. pullulans HP2001. Maximal production of pullulan by A. pullulans HP2001 was 26.6 g/ f when concentrations of glucose and yeast extract were 8% (w/v) and 0.25% (w/v), respectively. It was found that aeration rate, agitation speed and inner pressure of a bioreactor, which were some of physiological factors involved in the dissolved oxygen in the medium may affect cell growth and the production of pullulan by A. pullulans HP2001.

키토산과 천연색소를 이용한 Eco-Printing(제1보) (Eco-printing Using Chitosan and Natural Colorants(1))

  • The aim of this study is to develope eco-printing method using natural pigments and chitosan as a natural binder. Three chitosans with different molecular weights were employed to find appropriate conditions including chitosan concentration and pigment/binder ratio. Dye uptake, color and fastnesses of the printed fabrics were evaluated to find optimum conditions within the range of experiments carried out in this study. The effectiveness of chitosan as a printing binder was examined in comparison with color, dye uptake, and fastnesses of conventional synthetic binder and guar gum. It was found that chitosans with low or medium molecular weight were appropriate. Using low molecular weight chitosan, optimum concentrations were 1.7% for charcoal, madder and chlorophyll, whereas 2.2% for ocher, yellow soil, indigo and cochineal. Regardless of molecular weight and concentration of chitosan, the color fastnesess of fabrics printed with mineral pigments were superior to those of the fabrics printed with plant and animal pigments. As pigment/chitosan ratio became higher, rubbing fastness was decreased by 1-3 grade. The colorfastness of printed fabric with chitosan binder was similar to that with synthetic binder, which was higher than that with guar gum.

Protein Kinase C-delta Stimulates Haptoglobin Secretion

  • Haptoglobin (Hp) is a glycoprotein that is produced by hepatic cells and secreted into the circulation. While studying the physiologic functions of Hp, we found that Hp synthesized in THP-1 monocytic cells was largely retained within cells, although Hp is considered a secretory protein. To investigate the molecular mechanism on Hp secretion in THP-1 cells, in the present study, we examined the effect of protein kinase C (PKC) on Hp secretion. When several inhibitors of PKC isoforms were tested, only Rottlerin, a specific inhibitor of PKC-$\delta$, completely blocked Hp secretion from cells to culture medium. To confirm the role of PKC-$\delta$ in Hp secretion, Hp-overexpressing COS7 cells were transiently transfected with a wild-type or a dominantnegative mutant of the PKC-$\delta$ gene. Mutant PKC-$\delta$ significantly inhibited Hp secretion, whereas the wild-type gene slightly increased Hp secretion. These results demonstrate that the PKC-$\delta$ signal is involved in Hp secretion.

트윈롤 시스템을 이용한 천연가스 하이드레이트 펠릿의 연속성형 (Study on the Continuous Forming of Natural Gas Hydrate Pellet using Twin Roll System)

  • This study investigates compressive strength of ice pellet strip which is potential medium for Natural Gas Hydrate(NGH) extruded from die holes of Twin-roll Press for Continuous Pelletizing(TPCP). Recently, the prototype of TPCP is newly developed where ice powder is continuously fed and extruded into strip-type pellet between twin rolls. The system is specifically designed for future expansion towards mass-production of ice pellet strips or solid form of natural gas hydrate. It is shown that the compressive strength of pellet strip heavily depends on factors in extrusion process such as disk size, surface smoothness, ring size, taper shape, feeding mechanism, and rotational speed. Here, the mechanism of TPCP, along with compressive strength of pellets is discussed in terms of its feasibility for producing NGH pellets in the future.
