Surface and size dependent effects on static, buckling, and vibration of micro composite beam under thermo-magnetic fields based on strain gradient theory

  • In this article, static, buckling and free vibration analyses of a sinusoidal micro composite beam reinforced by single-walled carbon nanotubes (SWCNTs) with considering temperature-dependent material properties embedded in an elastic medium in the presence of magnetic field under transverse uniform load are presented. This system is used at micro or sub micro scales to enhance the stiffness of micro composite structures such as bar, beam, plate and shell. In the present work, the size dependent effects based on surface stress effect and modified strain gradient theory (MSGT) are considered. The generalized rule of mixture is employed to predict temperature-dependent mechanical and thermal properties of micro composite beam. Then, the governing equations of motions are derived using Hamilton's principle and energy method. Numerical results are presented to investigate the influences of material length scale parameters, elastic foundation, composite fiber angle, magnetic intensity, temperature changes and carbon nanotubes volume fraction on the bending, buckling and free vibration behaviors of micro composite beam. There is a good agreement between the obtained results by this research and the literature results. The obtained results of this study demonstrate that the magnetic intensity, temperature changes, and two parameters elastic foundations have important effects on micro composite stiffness, while the magnetic field has greater effects on the bending, buckling and free vibration responses of micro composite beams. Moreover, it is shown that the effects of surface layers are important, and observed that the changes of carbon nanotubes volume fraction, beam length-to-thickness ratio and material length scale parameter have noticeable effects on the maximum deflection, critical buckling load and natural frequencies of micro composite beams.

한약재로부터 L1210 세포 생장 억제물질의 검색 (Primary Screening for Growth Inhibitors of L1210 Cells from Oriental Herbs.)

  • 천연물 가운데서 항암물질을 얻을 목적으로 동양에서 오랫동안 암이나 암에 비슷한 질병의 치료에 씌여온 한약재를 물이나 유기용매로서 추출하여 검색하였다. Fischer배지에 자라는 L1210 세포에 추출물을 첨가한 다음 48시간 후에 세포수를 헤아려 생장을 50% 억제할 수 있는 추출물의 농도를 나타내는 ED$_{50}$값을 측정하였다. 물로서 추출한 38종의 한약재 가운데서 6종이 낮은 ED$_{50}$값을 나타냈다. 이들을 다시 유기용매로서 추출하므로써 ED$_{50}$값이 더욱 떨어져 NCI의 품질관리범위 치역내에 들어갔다. 항암제원으로 유망시되는 한약재로서 Cinnamomum cassia, Citrus trifoliata, Coptis japonica, Panax ginseng, Phellodendron amurense, Scutellaria baikalensis를 들 수가 있다.

  • 광합성 세균의 수소생성 향상을 위한 노력의 일환으로 수소생성능이 좋은 Rhodopseudomonas E15-1 을 고정화하여 수소생성에 적당한 조건을 조사하였다. 담체로 4%의 alginate를 사용하였을 때 수소생성량이 많았으며 고정화함에 따라 산소. 질소에 대한 억제효과를 덜 받았다. Alginate 고정화 세포는 그 지름을 2mm 이하로 하는 것이 겔내로의 확산저항이 적으므로 적당했다. 여러가지 유기물을 활용하여 수소를 생성할 수 있었으며 특히 citrate, fumarate와 malate 같은 유기산의 경우 30mM의 농도가 수소생성에 있어서 적당하였다. 또한 고정화세포를 이용한 계속적인 수소생성에 있어서 20일부터 활성이 감소하기는 하나 겔의 구조적 안정성은 30일까지 유지되었다.

Iron Chelator-Inducible Expression System for Escherichia coli

  • The $P_{entC}$ promoter of the entCERA operon encoding enzymes for enterobactin biosynthesis in Escherichia coli is tightly regulated by the availability of iron in the culture medium. In iron-rich conditions, the $P_{entC}$ promoter activity is strongly repressed by the global transcription regulator Fur (ferric uptake regulator), which complexes with ferrous ions and binds to the Fur box 19-bp inverted repeat. In this study, we have constructed the expression vector pOS2 containing the $P_{entC}$ promoter and characterized its repression, induction, and modulation by quantifying the expression of the lacZ reporter gene encoding $\beta$-galactosidase. $\beta$-Galactosidase activities of E. coli transformants harboring pOS2-lacZ were highly induced in the presence of divalent metal ion chelators such as 2,2'-dipyridyl and EDTA, and were strongly repressed in the presence of excess iron. It was also shown that the basal level $\beta$-galactosidase expression by the $P_{entC}$ promoter was drastically decreased by incorporating the fur gene into the expression vector. Since the newly developed iron chelator-inducible expression system is efficient and cost-effective, it has wide applications in recombinant protein production.

Gene Cloning, Expression, and Characterization of a $\beta$-Agarase, AgaB34, from Agarivorans albus YKW-34

  • A $\beta$-agarase gene, agaB34, was functionally cloned from the genomic DNA of a marine bacterium, Agarivorans albus YKW-34. The open reading frame of agaB34 consisted of 1,362 bp encoding 453 amino acids. The deduced amino acid sequence, consisting of a typical N-terminal signal peptide followed by a catalytic domain of glycoside hydrolase family 16 (GH-16) and a carbohydrate-binding module (CBM), showed 37-86% identity to those of agarases belonging to family GH-16. The recombinant enzyme (rAgaB34) with a molecular mass of 49 kDa was produced extracellularly using Escherichia coli $DH5{\alpha}$ as a host. The purified rAgaB34 was a $\beta$-agarase yielding neoagarotetraose (NA4) as the main product. It acted on neoagarohexaose to produce NA4 and neoagarobiose, but it could not further degrade NA4. The maximal activity of rAgaB34 was observed at $30^{\circ}C$ and pH 7.0. It was stable over pH 5.0-9.0 and at temperatures up to $50^{\circ}C$. Its specific activity and $k_{cat}/K_m$ value for agarose were 242 U/mg and $1.7{\times}10^6/sM$, respectively. The activity of rAgaB34 was not affected by metal ions commonly existing in seawater. It was resistant to chelating reagents (EDTA, EGTA), reducing reagents (DTT, $\beta$-mercaptoethanol), and denaturing reagents (SDS and urea). The E. coli cell harboring the pUC18-derived agarase expression vector was able to efficiently excrete agarase into the culture medium. Hence, this expression system might be used to express secretory proteins.

동결보존된 생쥐 고환조직 세포의 광학 및 전자현미경적 관찰 (Light and Electron Microscopic Observation in the Frozen-thawed Mouse Testicular Tissues)

  • Objective: The aim of this study was to investigate the morphological aspects of testicular tissue before and after freezing-thawing by light and transmission electron microscopy. Methods: Tissue biopsies were carried out on mouse testis for freezing. Samples in medium containing 20% glycerol were frozen by computer-controlled freezing program. The effect of freezing-thawing on the structural change of testicular tissues were examined by light and electron microscopy. Results: The freezing-thawing procedure had no significant effect on tubular diameter. However, it caused folding of the lamina propria, and notable damage to Sertoli cells, spermatogonia and spermatocytes. The cells were detached, desquamated from the basal lamina and had increased vacuolization. Round spermatids, elongated spermatids and spermatozoa were less affected, and most of them maintained their normal structure. Conclusions: The structure of spermatogonia, spermatocyte and basal compartments in seminiferous epithelium was significantly altered by freezing-thawing procedure of mouse testicular tissues. Thus, we need to develop a more reliable method for the cryopreservation of testicular tissues.

Photodegradation of 17α-ethynylestradiol in nitrate aqueous solutions

  • $17{\alpha}$-Ethynylestradiol (EE2) has gotten growing concerns due to its widely detected in the environment and high estrogenic potency. However, the knowledge on the photochemical behaviors of EE2 in natural waters is still limited. Herein, the photodegradation and estrogenic potency variation of EE2 induced by nitrate were studied using a sunlight simulator consisted by a 300 W medium pressure mercury lamp and 290 nm cut-off filters. It was found that EE2 could be photodegraded at a rate of $0.0193h^{-1}$ in pure aqueous solutions, and the photodegradation of EE2 could be significantly promoted by nitrate. The photodegradation removal rate of EE2 was increased from 9% in Milli-Q water to 85% in 2.0 mM nitrate solutions. Reactive species scavenging experiments demonstrated that the photogenerated $HO{\bullet}$ contributed about 55% to EE2 degradation. Fe(III), Cl- and dissolved humic acid (DHA) could inhibit the photodegradation of EE2 by competing the incident light and photogenerated $HO{\bullet}$, while $HCO_3{^-}$ had no influence on EE2 photodegradation. EE2 was determined to be phototransformed into organic chemicals without estrogenic potency by GC-MS and MCF-7 cell proliferation toxicity tests. These findings could extend our knowledge on the photochemical behaviors of steroid estrogens and provide information for ecological risk assessment.

혼합응집제에 의한 자연유기물질 제거에 미치는 영향 인자 (Influencing Factors on NOM Removal using Blended Coagulants)

  • This study was carried out to investigate the major factors for the removal of NOMs (Natural Organic Matters) by alum ferric chloride and blended coagulants that consisted of alum and ferric chloride. Investigated factors were pH, the dosage of coagulant, alkalinity, hardness and bloc strength. The particle size contained in the test water came from the Han River was also measured. DOC(Dissolved Organic Carbon) removal at pH 6 was two to three times higher than at pH 8.5. The blended coagulant showed 9 to 10 percent higher DOC removal efficiency and 2 to 4 percent higher turbidity under the same condition. Alkalinity consumption of alum, ferric chloride and blended coagulant was 81%, 90% and 86% of theoretical value, respectively. The limit concentration of alkalinity to avoid pin floe was 10 mg $CaCO_3/L$ when alum was used. Hardness had no apparent effect on coagulation. The residual turbidity and $UV_{254}$ showed a tendency of increasing with floc strength($sec^{-1}$) increase. The order of floe strength was the following; alum >blended coagulant > ferric chloride. The particle counter test showed 89 percent of the small particle size(SPS, $1~5{\;}{\mu}textrm{m}$) and 11 percent of the medium to large particle size(M.LPS, $5~125{\;}{\mu}textrm{m}$). At PH7.85, the particle removal efficiencies of SPS($1~5{\;}{\mu}textrm{m}$) and M.LPS($5~125{\;}{\mu}textrm{m}$) in the coagulation process were 81% and 95%, respectively.

실선계측 데이터 대체를 위한 AIS 및 ECMWF 데이터베이스 조합을 이용한 LNGC의 분당 회전수 및 동력 추정에 관한 타당성 연구 (A Feasibility Study on the RPM and Engine Power Estimation Based on the Combination of AIS and ECMWF Database to Replace the Full-scale Measurement)

  • In the previous research, a study was carried out to estimate the actual performance such as the propeller Revolution Per Minute (RPM) and engine power of a Liquefied Natural Gas Carrier (LNGC) using the full-scale measurement data. After the predicted RPM and engine power were verified by comparing those with the measured values, the suggested method was regarded to be acceptable. However, there was a limitation to apply the method on the prediction of the RPM and engine power of a ship. Since the information of route, speed, and environmental conditions required for estimating the RPM and engine power is generally regarded as the intellectual property of a shipping company, it is difficult to secure the information on a shipyard. In this paper, the RPM and engine power of the 151K LNGC was estimated using the combination of Automatic Identification System (AIS) and European Centre for Medium-Range Weather Forecasts (ECMWF) database in order to replace the full-scale measurement. The simulation approach, which was suggested in the previous research, was identically applied to the prediction of RPM and engine power. After the results based on the AIS and ECMWF database were compared with those obtained from the full-scale measurement data, the feasibility was briefly reviewed.

수리역학적 상호작용을 고려한 균열암반매질에서의 수리학적 거동에 대한 수치적 연구 (A Numerical Study on Hydraulic Behavior in a Fractured Rock Medium with Hydromechanical Interaction)

  • 본 논문은 수리학적 현장 주입시험 기간 동안 고려될 필요가 있는 수리역학적 상호작용에 따른 균열암반매질의 수리학적 거동에 대한 수치적 연구이다. 이러한 주입시험은 굴착정 내에 설치된 개구간(open hole section)을 따라 높은 압력을 가진 유량을 주입하며, 이를 통해 굴착정을 가로지르는 균열로부터 유량을 측정하는 것이다. 시간에 따라 변화되는 유량측정결과는 수리역학적 상호작용에 대한 분석 및 예측을 위해 개발된 수치모형의 현장 적용성을 평가하기 위해 사용되었다. 유량측정결과 전도성이 있는 균열들은 상호의존적인 균열망을 형성하며, 이로 인해 균열망을 구성하는 개별적인 균열요소들은 독립적인 시스템으로서 적절하게 모형화 될 수 없음을 보여주었다. 또한 간극수압이 굴착정 주위에 작용하는 최소주응력을 초과할 때 새로운 유체유입영역이 발생되며, 이러한 최소주응력보다 훨씬 큰 간극수압은 굴착정 주위의 균열들에 의해 유지될 수 있다는 것을 보여주었다. 본 연구에서 이러한 특성들이 자연 상태의 균열망의 구성형태에 따라 어떻게 영향을 미치게 되는지 이산 균열망 모형을 통해 수치적으로 분석되었다.
