Comparison of The Taste Compounds of Wild and Cultured Eel, Puffer and Snake head (천연산 및 양식산 뱀장어, 복어, 가물치의 맛 성분에 대한 연구)

  • In this study, eel, puffer and snake head fish, which are widely taken and highly valued for processing into baked fish, soup or fish juice were studied by classifying them into wild fish and cultured fish, and evaluated in terms of taste. The samples were gathered in the area including Kimhae, Samchonpo and Haman in Kyungsangnam-do, Kangsu-Gu in Pusan and Bukcheju-Gun in Cheju-do. Proximate compositions, the content of nucleotides and their related compounds, total and free amino acids, organic acids and fatty acids of the fishes were analysed. Generally, as for the compositions, wild fish had a higher crude fat content and a lower moisture content as compared with cultured fish, while there was no great difference between them in terms of crude protein and ash contents. Nucleotides and their related compounds including ATP, ADP, AMP, IMP, HxR, Hx were detected, and the amounts of which were nearly the same with respect to the growth conditions. IMP content was high in all of the samples, while ATP content was extremely low. Total 17 amino acids were detected from the samples, and most of the samples had high contents of glutamic acid, aspartic acid, lysine, leucine, glycine, alanine and arginine, and low contents of cystein, isoleucine and methionine. As for the other amino acids, generally same level of contents were detected for each samples. In cases of eel and snake head fish, the wild fish had higher total amino acid contents in comparison with cultured fish. And the result to the contrary was obtained for the case of puffer. Sample fishes had nearly the same compose distribution of free amino acid though the contents of which were a little different according to the samples. Each sample had high contents of taurine, hydroxyproline, glutamic acid, glycine, cystathionine, ${\beta}-aminoisobutyric$ acid and lysine. Total organic acid contents of cultured fish was higher than that of wild fish. All fishes commonly had high contents of lactic acid, iso-valeric acid, oxalic acid and fumaric acid. High levels of $C_{16:0},\;C_{16:1},\;C_{18:1(cis)},\;C_{18:3},\;and\;C_{22:6}$ were shown in all of the samples and ${\omega}-3$ polyunsaturated fatty acid content of cultured fish was higher than that of wild fish, and as for the essential fatty acid, wild fish has higher content in comparison with cultured fish.

Effects of Jeju Citrus unshiu Peel Extracts Before and After Bioconversion with Cytolase on Anti-Inflammatory Activity in RAW264.7 Cells (면역세포에서 Bioconversion 전후 제주 감귤 과피 추출물의 항염증 효과)

  • Citrus and its peels, which are by-products from juice and/or jam processing, have long been used in Asian folk medicine. Citrus peels show an abundant variety of flavanones, and these flavanones have glycone and aglycone forms. Aglycones are more potent than glycones with a variety of physiological functions since aglycone absorption is more efficient than glycones. Bioconversion with cytolase converted narirutin and naringin into naringenin and hesperidin into hesperetin. Therefore, this study aimed to investigate the anti-oxidant and anti-inflammatory effects of bioconversion of Citrus unshiu (CU) peel extracts with cytolase (CU-C) in RAW264.7 cells. HPLC chromatograms showed that CU and CU-C had 23.42% and 29.39% total flavonoids, respectively. There was substantial bioconversion of narirutin to naringenin and of hesperidin to hesperetin. All citrus peel extracts showed DPPH scavenging activities in a dose-dependent manner, and CU-C was more potent than intact CU. RAW264.7 cells were pre-treated with $0{\sim}500{\mu}g/mL$ of citrus peel extracts for 4 h and then stimulated by $1{\mu}g/mL$ of lipopolysaccharide (LPS) for 8 h. All citrus peel extracts showed decreased mRNA levels and protein expression of LPS-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in a dose-dependent manner. Especially, CU-C markedly inhibited mRNA and protein expression of iNOS and COX-2 compared to intact citrus peel extracts. All citrus peel extracts showed decreased NO production by iNOS activity. This result suggests that bioconversion of citrus peel extracts with cytolase may provide potent functional food materials for prevention of chronic diseases attributable to oxidation and inflammation by boosting the anti-inflammatory effects of citrus peels.

Effects of Level of Dietary Protein or Carbohydrate on the Economic Characters and Amylase Activity of the Silkworm, Bombyx mori (인공사료의 단백질 및 탄수화물 수준이 가잠의 실용형질과 Amylase활성에 미치는 영향)

  • A series of experiment was conducted to investigate the effects of the level of dietary Protein (soybean meal) and carbohydrate (sucrose) on the growth rate, feed efficiency, nutrients digestibility and amylase activity of larvae at 5th instar. The results obtained from the experimentation are summarized as follow: (1) It was found that the body weight gain, cocoon quality and feed efficiency of the 5th instar larvae were significantly affected by the level of protein and carbohydrate in the artificial diet. Present data revealed that the proper level of dietary protein and carbohydrate would be the most important factor for the optimum growth of larvae. (2) In the amount of diet consumption, the group of larvae gained more body weight consumed more diet than the group gained less amount of body weight. (3) Digestibility of nutrients was improved as the level of protein in the diet increased, although the level of dietary carbohydrate did not exert any positive results. (4) It was observed that the amylase activity in digestive juice was stimulated by the increasement of dietary carbohydrate. However, no correlation between the level of dietary protein and carbohydrate was detected in the amylase activity. (5) Analytical data indicated that the amount of protein or carbohydrate excreted through feces and urine was proportional to the content of protein or carbohydrate in artificial diet. The group o( larvae consumed the diet contained high protein excreted more protein and less nitrogen free extract than did the group received low protein diet. Likewise, the group of larvae fed high dietary carbohydrate excreted more nitrogen free extract and less protein through excreta than the group consumed low carbohydrate diet group. (6) Although the amylase activity of the larvae received the artificial diet was higher than the larvae received natural diet (mulberry leaf), the rate of body weight gam and the quality of cocoon produced from the natural diet group was better than the artificial diet group. It is, thus, concluded that feeding the larvae by the artificial diet may net be recommendable. (7) It was determined that the best level of protein (soybean meal) and carbohydrate (sucrose) in artificial diet was found to be 40g and 0g, respectively when it was fed to the 5th instar larvae. (8) It may be concluded from the results obtained that the artificial diet that could stimulate the activity of amylase nay not be recommendable for the practical larvae feeding Purpose due simply to the Poor economic return from this diet than natural diet.

Production of Bioactive Yoghurt containing Cichorium intybus L. (Chicory) Extract - Preliminary Study

  • Cichorium intybus L. (chicory) roots and leaves are widely used in herbal preparations, which have beneficial effects on the stimulation of digestion and metabolism of food ingredients, gastric juice excretion, diuretic action, and bile excretion. Notably, chicory root is well known as a source of polyphenols, compounds with recognized value in health improvement. In this study, we examined the physicochemical characteristics (TA, pH, and sensory evaluation) of bioactive yoghurt containing different concentrations of chicory. With increasing incubation time (5 h), the TA of the yoghurt increased whereas the pH decreased, regardless of the amount of chicory. As the amount of chicory increased, the scores for color, flavor, taste, and overall acceptability generally decreased. Among the tested groups, yoghurt with the addition of 1% chicory attained the highest scores. Further studies on the production of bioactive yogurt with optimum chicory concentration are needed.

Calendar for the Prevalence of Honey Bee Diseases, with Studying the Role of Some Materials to Control Nosema (몇 가지 꿀벌 노제마병 방제물질의 평가와 꿀벌 질병의 발생시기 조사)

  • Nosema spp. (NS) causes severe problems to honey bee colonies including the death of the highly infected ones. Searching for effective materials to control this parasite is very important. The objectives of this study were to identify the calendar for the prevalence of NS and other bee diseases, and to test the efficacy of three materials: diluted honey mixed with lemon juice (M1), chamomile extract mixed with sugar syrup (M2) and sutrivet mixed with sugar syrup (M3) against Nosema. To realize these objectives, diseases of brood and adult honey bees were surveyed over one year. Also, the efficacy of M1, M2 and M3 against Nosema was evaluated under field and laboratory conditions. The results showed that few diseases for immature and mature stages of honey bees were recorded. NS was detected during winter and spring in link with low temperature and high relative humidity. Under field conditions, M2 reduced the infection by 36.66% while M3 by 23.33% and finally M1 by 13.33%. In the laboratory, the highest efficacy was to M2 followed by M1 and finally M3. The three materials impacted the percentage of survived bees significantly higher than infected bees without any treatments over the experimental period. The study suggests the potential role of chamomile as a natural material to control NS.

Changes in the Sensitivity to Metalaxyl, Dimethomorph and Ethaboxam of Phytophthora infestans in Korea

  • Changes of control efficacy of chemical to potato late blight caused by Phytophthora infestans in potato fields from 2001 to 2004 were examined. Control efficacy of metalaxyl was suddenly decreased from 100% in 2002 to 50% in 2004 and that of dimethomorph also was similar to those of metalaxyl. However, the control efficacy of ethaboxam no great change. Both A1 and A2 mating type isolates were isolated from 2001 to 2004 in several areas in Korea. The majority of the P. infestans isolates were A1 mating type. Total 939 isolates of P. infestans obtained from several areas in Korea from 2001 to 2004 were examined for changes of sensitivity to metalaxyl. Frequencies of metalaxyl resistance isolates were gradually increased from 17% in 2001 to 84.2% in 2004, but isolation frequencies of metalaxyl sensitive and intermediate resistant isolate were decreased. Cause of decreasing control efficacy of metalaxyl was thought by increase of resistance isolates in A1 mating type population according to increasing metalaxyl use. Most isolates were grown at 0.5 ${\mu}g/ml of dimethomorph and isolates grown at 1 ${\mu}g/ml of dimethomorph were approximately 10.2-22.9%. However, no isolate was able to grow at 5.0 ${\mu}g/ml. Based on these results, minimum inhibitory concentrations (MIC) of dimethomorph to P. infestans were determined to be 0.5-1.0 ${\mu}g/ml. Our results indicated that the reason decreasing control efficacy of dimethomorph was not caused by occurrence of resistant isolates. About 5% and 12.1% isolates among the total isolates collected in 2003 and 2004 were grown on V-8 juice rye agar containing 1.0 ${\mu}g/ml ethaboxam. The 2.1 and 25.4% isolates had MICs of 0.2-0.4 ${\mu}g/ml, and MIC values of 87.9% and 74.3% isolates were less than 0.2 ${\mu}g/ml concentrations of ethaboxam. Therefore, resistance development by P. infestans to ethaboxam is not likely to occur in the natural condition.

Effects of Kalopanax Pictus Extracts and Their Related Origin on Gastric Lesions (해동피 및 유사생약 추출물의 위 손상에 대한 효과)

  • Kalopanax pictus has pharmacologically anti-inflammatory and analgesic effect and is known to respond to treatment of backache, knee pain and etc. In this study, we investigated the effects on gastric lesions of Kalopanax pictus both from Korea (KPK) and China (KPC) compared with their related origin, Znthoxylum ailanthoide both from Korea (ZAK) and China (ZAC), and Korean Bombax malabaricum (BMK). In preliminary screening, KPK and KPC shown effective inhibition of HCI EtOH-induced gastritis in rats. To elucidate their protective effects on gastric lesions, we assessed inhibition of H. pylori colonization, 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activities, reducing power test, and inhibition of lipid peroxidation. KPK was the most effective from antioxidant assays. KPK also shown the inhibition of indomethacin-induced gastric ulcer in rats. Gastric secretion in rats, KPK reduced the secretion of gastric juice and total acidity and raised pH. Therefore, it is possible that KPK can be developed as health functional food and natural medicine. In addition, it can contribute to the standardization with objectivity and reliability for KPK through the criteria establishment of the precise origin of medicine, the prevention of indiscriminate distribution of imitation, and the rising rate of dependence on imports of medicinal herbs, and mixing prevention of low-quality goods.

Effects of steam blanching pretreatment on quality of spray-dried powders prepared from pressed juice of garlic chives (부추의 증숙처리가 착즙액 분무건조 분말의 품질에 미치는 영향)

  • This study was conducted to develop a spray-dried garlic chives (Allium tuberosum) powder and to evaluate its quality characteristics depending on the treatment of steam blanching pretreatment $100^{\circ}C$, 3 min) and the addition of forming agents (dextrin (DE=10), ${\beta}$-cyclodextrin) during process. The steam blanching pretreatment showed an increase in $L^*$ value while a decrease in $-a^*$, $b^*$, $C^*$, and $h^o$ values of the powder. Moisture content and water soluble index were not affected by the treatment of steam blanching and the addition of forming agents, whereas the particle diameter was the smallest in the steam blanching treatment and dextrin addition. Chlorophyll, phenolic compound, and vitamin C content, and DPPH radical scavenging activity of non-pretreated powder were significantly higher than those of the steam blanching treated powders. However, there was no significant difference between the two forming agents. The sensory acceptability (color, smell, and overall acceptability) of powder treated with steam blanching were significantly higher than those of non-pretreated powders. Therefore, the steam blanching pretreatment of fresh garlic chives affected on the better quality characteristics of the spray-dried powders when compared with non-pretreated powder though it adversely affected the natural chemical quality of fresh garlic chives.

Factors Affecting Color Loss in Plum Nectar (자두 Nectar의 퇴색(褪色)에 미치는 영향인자(影響因子))

  • The effects of various factors on the color stability of plum nectar were studied. The data showed that pH and temperature were the most important factors affecting the rate of color loss. The concentration of ascorbic acid affected the rate of color loss in plum nectar. Although the color loss could be influenced by the presence of sugars and their degradation products such as 5-hydroxymethyl-2-furfural(HMF), HMF did not accumulate in sufficient amounts during the storage in the plum nectar to affect appreciably the degradation rate. The pigment was very unstable in the fermented juice and its. spectrophotometric spectrum was changed differently from that of natural plum color material. Materials present in plum nectar itself apparently have an appreciable effect on the rate of color loss.

The Studies on Search of Antifibrotic Effect by Several Herbs Extract in Rat Liver Tissue (수종(數種)의 한약재(韓藥材) 전탕액(煎湯液)이 동물 간조직내 항섬유효능검색에 관한 실험적 연구)

  • Antifibrotic drugs could be delayed or inhibited massive collagen deposition in liver tissue or inhibited collagen synthesis. we investigated antifibrotic effects by several herbs(Schisnadra chinensis, Ganoderma japonicum, Sedum sarmentosum, Alisma canaliculatum, Plantago asiatica) extract with observations of collagen accumulation in liver tissue and collagen synthesis in sera. Rats were used for experimental animal that were devided 3 groups(sham control, BDL/S, BDL/S-ER). Rats were operated for inducing liver fibrosis(cirrhosis) by bile duct obstruction. Several herbs were prepared by water extraction and were applicated p. o. $3ml/day$ during 4 weeks. After observation period, rats were sacrificed and liver tissue and sera were collected. In result, the mortality of rats was 35% in BDL/S group and 20% in BDL/S-ER The color of bile juice in BDL/S-ER was bright yellow and murky yellow in BDL/S group. The significantly lower weight of liver($16.21g{\pm}5.3,\;20.58{\pm}2.4$) and spleen($1.96g{\pm}0.96,\;3.93{\pm}0.21$) were shown in BDL/S-ER than that of BDL/S(p<0.05) group. The value of collagen in liver tissue(25.7%) in BDL/S-ER was observed significantly lower than that of BDL/S group (Tab. 2). AST. ALT, ALP, t-bilirubin, BUN levels were low in BDL/S-ER as compared with those of BDL/S group, but the significance was not proven. The trichrome stained liver tissue in BDL/S-ER group was observed mild bile duct proliferation and fibrosis compared with BDL/S group. In conclusion, natural products inhibited new collagen synthesis and delayed massive collagen deposition in liver tissue, so that they have noticeable antifibrotic effects in experimental liver fibrosis(cirrhosis).
