가시연꽃 종자 추출물의 화장품 원료로서의 특성 (Application as a Cosmeceutical Ingredient of Euryale ferox Seed Extract)

  • 천연물로부터 새로운 기능성 화장품 소재를 개발하기 위하여 국내외 천연물을 대상으로 항산화 활성을 평가한 결과, 가시연꽃의 종자 추출물이 우수한 활성을 나타내는 것을 발견하였다. 가시연꽃 종자의 추출물로부터 유효 성분을 분리하기 위하여 각종 컬럼 크로마토그래피 및 HPLC 등의 기법을 실시하여 순수하게 정제된 화합물 5종을 획득하였다. 각종 NMR 및 MS 스펙트럼 등을 분석하여 이들의 화학구조를 규명한 결과 fucosterol (1), 3-(4-hydroxy-3-methoxybenzyl)-4-[(7'R),5'-dihydroxy-3'-methoxybenzyl]tetrahydrofuran (2), resorcinol (3), pyrogallol (4) and 4-O-methylgallic acid (5)로 동정하였다. 이들 화합물의 화장품 원료로서의 특징을 조사하고자 각 화합물들을 대상으로 항산화 활성, 엘라스타아제 저해활성, 미백활성 등을 평가하였다. 이들 화합물의 자유라디칼소거활성은 $SC_{50}$ 값이 $17.0\;{\sim}\;100.2\;{\mu}M$이었고, 특히 화합물 4 및 5는 양성대조구인 ferulic acid보다 6배 이상 강력한 활성을 나타내었다. 또한 엘라스타아제 저해활성에 대한 $IC_{50}$ 값은 $18.8\;{\sim}\;78.2\;{\mu}M$로써 특히 화합물 4는 양성대조구인 EGCG와 비슷한 활성을 나타내었으며, B16F10 멜라노마 세포를 이용한 멜라닌 생성 저해활성에 대해 화합물 3은 양성대조구인 arbutin보다 약 2배 이상 강력한 미백활성을 나타내었다. 따라서 가시연꽃 종자 추출물은 강력한 항산화활성, 엘라스타아제 저해활성 및 미백활성을 나타내는 유효성분들을 다량 함유하므로, 복합 기능성을 나타내는 화장품 원료로 사용할 수 있을 것으로 사료된다.

Microbial Strains and Bioactive Exopolysaccharide Producers from Thai Water Kefir

  • The aims of this novel work were to determine the microbial strains and exopolysaccharide (EPS) producers in water kefir from Nakhon Ratchasima Province, Thailand. Thirty-three microbial strains were identified using 16S rRNA gene analysis consisting of 18 bacterial strains, as 9 strains of acetic acid bacteria (AAB), 9 strains of lactic acid bacteria (LAB), and 15 yeast strains. All bacteria were able to produce EPS with a diverse appearance on agar media containing different sugars at a concentration of 8%. Culture supernatants from AAB and LAB showed 31-64% 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity with the highest antioxidant activity of 64% from Acetobacter pasteurianus WS3 and WS6. Crude EPS from A. pasteurianus WS3 displayed the highest ferric reducing antioxidant power at 280 mM $FeSO_4/g$ EPS, greatest anti-tyrosinase activity at 20.35%, and highest EPS production of 1,505 mg EPS/L from 8% sucrose. These microbes offer beneficial health implications and their EPSs can be used as food additives and cosmetic ingredients.

Effect of facial cosmetic containing Prunus persica L. Batsch leaves extract on men's skin condition

  • Today, men's appearance is an important part of the global fashion industry, including Korea, with such fashion concepts as the Metrosexual and Ubersexual. Domestic and overseas beauty and cosmetics related organizations are using this phenomenon for marketing and researching to improve the function of cosmetics. The purpose of this study was to investigate the effect of Prunus persica L. Batsch leaf extract on the skin condition after clinical application to male subjects with oily skin to determine the possibility of functional cosmetics utilizing this extract. The cleansing bars were prepared by the MP method and the cleansing packs were prepared using natural ingredients. After 8 weeks of application, moisture, oil, erythema, melanin, pH, elasticity and tactile sensation were measured using MPA5. The results showed that there was a statistically significant difference in T-zone moisture and T-zone oil after using the natural cleansing bar, but there was no significant difference between the two groups for other items. However, the statistically significant changes were found in the individual skin condition change items of the subjects in each group, and it was judged that the two kinds of formulations could be used at the same time to have a higher skin improvement effect. Thus, Prunus persica L. Batsch leaf extract demonstrates potential to be used as a functional cosmetics item.

천연물을 이용한 자외선차단제의 자외선차단지수(SPF) 평가 (Sun Protection Factor (SPF) Assessment of the Sunscreen Composed of Natural Substances)

  • 자외선 노출 증가로 인한 자외선의 유해성을 방지하기 위해 자외선차단제의 사용이 보편화되었다. 이러한 추세에 맞게 화장품 업계에서는 서로 다른 성분을 혼합하거나 천연물에서 유효성분을 추출하는 연구를 활발하게 진행하고 있다. 본 연구에서는 황금에서 추출한 천연성분인 황금추출물을 함유하는 자외선차단제품의 자외선 차단효과를 확인해보고자 하였다. 총 10명의 지원자를 대상으로 각 제품의 최소홍반량을 측정하였고 자외선차단지수를 구하였다. 그 결과 황금추출물을 함유하는 자외선차단제품과 함유하지 않는 자외선차단제품의 자외선차단지수는 각각 $34.52{\pm}2.13$, $32.67{\pm}1.44$였다. 두 제품 간의 자외선차단지수 차이는 통계적으로 유의하지는 않았지만, 인체를 대상으로 천연물 자외선차단제의 임상적 효과를 평가하였고 제품도포로 인한 이상반응이 없었다는 점에서 의미 있는 시도였다고 생각된다.

  • 최근 환경오염으로 인한 지구오존층의 파괴로 자외선의 지표면의 도달 량이 증가하고 그로 인한 인간에 대한 피해가 증가하고 있다 . 과도한 자외선의 피부에 대한 직접적인 노출은 피부에 많은 문제점을 야기하므로 일차적으로 자외선차단제를 이용하여 인체에 대한 직접적인 피해를 최소화 해야한다 . 현재 자외선 차단제는 유기자외선흡수제와 무기자외선산란제가 많이 사용이 되는데 적은량으로도 효과가 높은 유기자외선차단제는 사용 시 주의를 요하므로 국가별로 사용량과 사용여부에 대하여 규제를 하고 있다. 본 연구에서는 플라보노이드류 ,알카로이드류 같은 자외선 흡수 성분을 함유한 식물중 UV/vis spectrophotometer 에서 자외선 흡수 peak을 나타내는 금은화 , 포공영, 녹차 , 황금추출물을 이용하여 화장품의 자외선 흡수제로서의 사용가능성여부를 조사하였다. 또한 식물추출물이 가진 자극성 성분과 오염성, 그들이 가진 유효성분들을 보호하고자 실리콘을 이용한 겔화를 시도하였다 . 자연친화적인 천연물로써 식물추출성분을 이용 유기자외선흡수제의 사용량을 줄이는 자외선홉수보조제로서 가능성을 보여주었다.

Franz Diffusion Cell을 이용한 문신용 염료 내 유해물질의 피부 투과특성 연구 (Permeation Characteristics of Hazardous Substances in Tattoo Dye using Franz Diffusion Cells)

  • Objectives: The purpose of this study is to determine the exposure risk to tattoo components by analyzing skin absorption using the in vitro method. Tattoos are commonly used for cosmetic purposes, and the skin of not only the operator but of the people who are undergoing the cosmetic procedure is continuously exposed to hazardous chemicals. Methods: Skin permeation risk determination was conducted by the in vitro Franz diffusion cell method according to the ingredient types of tattoo dyes, such as volatile organic compounds (VOCs), non-volatile organic compounds and heavy metals, using hairless mouse full skin and human cadaver epidermis. Results: The major components with good skin penetration for each type of tattoo dye ingredient were clarified. Among the tatto dye ingredients, 1,2-Dichlorobenzene, Zn, Al, Pb and Ti showed good skin penetration. Most of the skin transmission rates were higher in hairless mouse full skin than in human cadaver epidermis. Conclusion: A possible exposure risk to hazardous substances in tattoo dyes was confirmed from this study. These results are expected to provide a positive contribution to the establishment of management regulations for tattoo dyes.

황기, 백출 및 오가피의 항산화성 및 미백효과에 관한 연구 (Antioxidative Properties and Whitening Effects of the Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex)

  • In an attempt to find natural sources of antioxidants and whitening agents, Comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex were carried out. Comparison of the four ethanol extracts revealed that, Astragali Radix had the highest electron-donating ability(72.5%) and the highest SOD-like ability(26.1%). The xanthine oxidase experiment exhibited a hindrance effect of 88.5% in Atractylodis Rhizoma Alba, 81.1% in Acanthopanacis Cortex, 75.8% in Astragali Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 42.1% in the Acanthopanacis Cortex, 37.2% in the Atractylodis Rhizoma Alba, 6.0% in the Astragali Radix. Based on these results, we suggest that the ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex can be used as food and cosmetic ingredients.

Evaluation of Senescence Induced Prematurely by Stress. Application for cosmetic active ingredients

  • Living cells are continuously subject to all sorts of stress such as ultraviolet rays on skin cells. Tests made in various laboratories show that when young fibroblasts (Le. at the beginning of their proliferate life) were repeatedly put under stress at subletal doses, they acquired a phenotype similar to Senescence Induced Prematurely by Stress (SIPS). The work presented hereafter was made on a new model of senescence induced prematurely by stress from ultraviolet Brays (UVB). The human fibroblast model was put under repeated UVB stress, causing SIPS. Several ageing biomarkers were used in order to characterise the cells that underwent stress:. an increase in the proportion of positive cells with senescence associated $\beta$-galactosidase activity (SA $\beta$-gal) measured by a specific coloration,. the proportion in the different morphological stages that fibroblasts undergo during culture visualised by microscopic observation,. the expression of genes known for overexpressing during senescence, particularly fibronectin and apolipoprotein J, measured by Real Time-PCR,. the common deletion of 4,977 bp in mitochondrial DNA, evaluated by nested PCR. Studying the variation of these 4 biomarkers, we have evaluated the protective effect of a Laminaria digitata extract (LDE) that can be used as a natural active ingredient for anti-ageing cosmetics.

토사자, 복분자 및 작약의 항산화성 및 미백효과에 관한 연구 (Antioxidative Properties and Whitening Effects of the Cuscutae Semen, Rubi Fructus and Paeoniae Radix)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix were carried out. Comparison of the three ethanol extracts revealed that, Paeoniae Radix had the highest electron-donating ability(79.3%),; however, Rubi Fructus had the highest SOD-like ability(31.1%). The xanthine oxidase experiment exhibited a hindrance effect of 74.3% in Cuscutae Semen, 80.4% in Rubi Fructus and 60.8% in Paeoniae Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 20.1% in the Cuscutae Semen, 54.2% in the Rubi Fructus, 56.3% in the Paeoniae Radix. Based on these results, we suggest that the ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix can be used as food and cosmetic ingredients.

Physiological and Whitening Effects of Morus alba Extracts

  • Mulberry extracts can be incorporated into skin-whitening products. The compound attributed to lighten the skin is arbutin, a form of hydroquinone that inhibits melanin release by suppressing the tyrosinase enzyme. For the cosmetic applications, the physiological effects of mulberry (Morus alba) extracts were investigated. The water soluble fraction of mulberry contains higher amount of protein (16.28~4.47%) in contrast to fat (1.55~1.41%). In addition, the fraction abundantly contains succinic acid (972.4-275.8 mg/g) and phosphoric acid (1,628.4-121.9 mg/g) in different parts of mulberry. The free radical scavenging ability in water soluble fraction was found to display remarkable effects in comparison with methanol and ethyl acetate fraction. The ethyl acetate-soluble of root and leaf showed remarkable tyrosinase inhibition activity by IC 50 (${\mu}g/ml$). The anticancer activity of methanol fraction obtained from mulberry using human cancer cell lines showed growth inhibition effect (270.14 mg/ml in Calu-6 cells, 295.29 mg/ml in HCT-116, and 332.29 mg/ml in MCF-7 cells, respectively). Based on the results, Morus alba extracts include cosmetic ingredients with antioxidizing and whitening properties.
