Functional Characterizations of Extruded White Ginseng Extracts

  • The antibacterial and antioxidant potentials of extruded ginseng extract (EGE) with 60% ethanol and methanol were investigated. The inhibitory activity of the EGE in Gram-positive bacteria was significantly higher than in Gram-negative bacteria. Higher antibacterial activity was observed with methanol ginseng extract when moisture content and barrel temperature were 20% and $115^{\circ}C$, respectively, that diameter of inhibition zone at 1,500 mg/mL was $15.40{\pm}0.13\;mm$ for Bacillus subtilis and $9.31{\pm}0.05\;mm$ for Salmonella typhimurium. The amount of total phenolics was highest in extruded ginseng at 20% moisture content and $115^{\circ}C$ barrel temperature. Especially, a positive correlation was observed between the total phenolic content and antioxidant activity of the extracts. In the 2,2-diphenyl-1-picryhydrazyl radical (DPPH) system, all tested extract of extruded ginseng at 20% moisture content exhibited very strong antioxidant properties when compared to red ginseng with percent scavenging effect of 23-35% at 20mg/mL. In conclusion, it can be said that the extracts of extruded ginseng could be used as natural antimicrobial and antioxidant agents in the food preservation.

잘피(Zostera marina) 에탄올 및 물 추출물의 항산화 효과 (Antioxidant Effect of Zostera marina Ethanol and Water Extracts)

  • 본 연구는 잘피 추출물로부터의 항산화 효과를 알아보았다. 총 폴리페놀 함량에서는 에탄올 추출물과 물 추출물이 각각 2.12 mg/g, 3.88 mg/g으로 측정되었다. DPPH 라디칼 소거능의 경우 에탄올 추출물이 1 mg/ml농도에서 94%로 물 추출물보다 더 높았으며 환원력은 두 추출물 모두 농도의존적인 경향을 나타냈다. 금속봉쇄력의 경우 물 추출물이 에탄올 추출물 보다 더 높았고 rancimat에 의한 산화도 측정 시 에탄올 추출물이 1 mg/ml농도에서 66%로 물 추출물보다 더 높았음을 알 수 있었다. 이러한 결과는 잘피 추출물이 식품산업에서 천연항산화제로서 사용이 될 수 있음을 보여준다.

Effects of Lotus (Nelumbo nucifera) Leaf on Quality and Antioxidant Activity of Yogurt during Refrigerated Storage

  • The supplementation with natural ingredients that are rich in polyphenols could improve the quality and functionality of yogurt. Lotus leaf (LL) are abundant in phenolic compounds. We aimed to investigate the effects of LL powder on the quality properties, total phenolic content (TPC), and antioxidant activity of yogurt. Yogurt was supplemented with four different concentrations (0%, 0.2%, 0.5%, and 1%) of LL powder and evaluated for 14 d refrigerated storage. During storage, the titratable acidity (TA) of all LL yogurt groups was higher than that of the control (p<0.05). The $L^*$ and $a^*$ values decreased by LL addition to the yogurts, while the $b^*$ values increased. All LL yogurt groups indicated better viscosity than that of control, and among them, 0.2% LL yogurts had the highest viscosity without significant decrease until the end of the storage duration. The addition of LL into yogurt enhanced the water-holding capacity (WHC) by at least 1.5-fold than that of control for the entire storage duration. The TPC of yogurts gradually increased with the addition of LL (p<0.05) and continued to increase during storage; the 1% LL yogurt at 14 d showed the highest value of $61.94{\pm}1.68{\mu}g$ GAE/g. The DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and reducing power of the yogurts were also significantly enhanced with increasing LL concentrations (p<0.05). These results suggest that lotus leaf may be useful as a natural ingredient for improving the quality and antioxidant activity of yogurt.

  • The root of Aralia continentalis Kitagawa (Araliaceae) have been used as an analgesic and fever remedy, and for treatment of rheumatism in Chinese medicine, whereas the young leaves are used for ingredient of salad. Antioxidant activity of the young leaves of A. continentails was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA) and by evaluation the radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Chromatographic separation of active fraction led to the isolation of six flavonoids, among which quercetin, hyperoside and kaempferol showed strong antioxidant activities, while 6"-O-acetyl astragalin, astragalin and trifolin were inactive. Adenosine, oleanolic acid 28-O-glucosyl ester and salsoloside C methyl ester isolated from the somewhat active BuOH fraction exhibited no antioxidant activities.

Phenolic Compounds with Antioxidant Activity on DPPH Free Radical Scavenging and Inhibition of Xanthine/Xanthine Oxidase from the Flowers of Chrysanthemum morifolium

  • The flowers of Chrysanthemum morifolium (Compositae) were investigated for their biofunctional components. Antioxidant compounds were identified from the flower of Chrysanthemum morifolium, through activity-guided fractionation and repeated column chromatographic separations. The structures of the isolated compounds were characterized as chlorogenic acid (1), luteolin (2) and quercitrin (3) by the analysis of physico-chemical and spectral data. Their antioxidant effects were evaluated by assaying for 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity and inhibition of xanthine/xanthine oxidase activity.

Synergistic Antioxidant Effects of Lycopene and Other Antioxidants on Methyl Linoleate Autooxidation

  • The beneficial effects derived from consuming natural antioxidants may not depend on the action of an individual antioxidant, but rather on the concerted action of several antioxidants naturally present. The aim of this study was to determine the concentrations and combinations of antioxidants that can produce synergistic effects (SyEs). Quantification of the lipoperoxyl radical scavenging capacity of antioxidants was carried out in a homogeneous model system where the free radicals were produced by the oxidation of methyl linoleate, initiated by the 2,2'-azobis (2,4-dimethylvaleronitrile). The greatest SyE (2.21, p<0.05) was seen in mixtures of all 4 antioxidants when used with concentrations of 15 ${\mu}M$ lycopene, 2.5 ${\mu}M$ vitamin E, 0.16 ${\mu}M$ vitamin C, and 10 ${\mu}M$ ${\beta}-carotene$. Doubling the vitamin E concentration from 2.5 to 5.0 ${\mu}M$ in the mixture with all 4 antioxidant reduced the SyE to 1.69 (p<0.05). Other combinations produced synergistic effects that ranged from 1.28 to 1.41.

추출방법에 따른 소목 심재의 항산화 및 항당뇨 활성 평가 (Evaluation of Antioxidant and Anti-diabetic Effects of Sappan Lignum by Extraction Method)

  • Objectives : The heartwood of Sappan Lignum has been used since ancient times as an ingredient in folk medicines against anti-bacterial and anti-anemia purposes. Many bioactive constituents have been derived from this biomass such as chalcones and homoisoflavonoids. In the current investigation, the antioxidant and anti-diabetic properties using DPPH and $ABTS^+$ radicals scavenging, ${\alpha}-glucosidase$, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Sappan Lignum. Methods : In our continuing investigation for bioactive natural ingredients, the antioxidant and ${\alpha}-glucosidase$ inhibitory properties of Sappan Lignum extracts were prepared from different extraction methods and the biological efficacies were investigated in vitro. The antioxidant properties were evaluated employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effects of Sappan Lignum extracts were tested via ${\alpha}-glucosidase$ and AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested samples showed dose-dependent radical scavenging and ${\alpha}-glucosidase$ inhibitory activities. Among the tested extracts, the 80% methanolic extract of Sappan Lignum was showed the most potent activity with an $IC_{50}$ value of $82.3{\pm}1.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. While, $ABTS^+$ radical scavenging activity of 80% methanolic extract was higher than those of other extracts. Also, ${\alpha}-glucosidase$ inhibitory and AGEs formation effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Sappan Lignum can be considered as a new effective source of natural antioxidant and anti-diabetic materials.

Antioxidant Activity of Roasted Defatted Perilla Seed

  • The antioxidant activity of roasted defatted perilla (Perilla frutescens) seed was determined by measuring its radical scavenging effect on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals, inhibitory activity on total reactive oxygen species generation in kidney homogenates using 2',7'-dichlorodihydro-fluorescein diacetate, and scavenging effect on authentic peroxynitrites. The methanolic extract of roasted defatted perilla seed showed strong scavenging activity in both DPPH and peroxynitrite radicals, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate>n-butanol>dichloromethane>water>n-hexane fraction. The ethyl acetate soluble fraction exhibiting strong antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Luteolin was isolated as one of the active principles from the ethyl acetate fraction, together with the inactive chrysoeriol and apigenin.

Antioxidant Activity of Tomato Powders as Affected by Water Solubility and Application to the Pork Sausages

  • This study was conducted to evaluate the antioxidant activity of oven-dried ($60^{\circ}C$) tomato powder (TP) as affected by water solubility and to evaluate the effects of TP on the physico-chemical properties, and antioxidant and antimicrobial activities of emulsified pork sausages with two levels of TP (1% and 2 %). After fresh tomatoes were homogenized and dried at $60^{\circ}C$ oven, they were extracted by stirring with water. Then, the aqueous solution was filtered, and water soluble and insoluble tomato powders were obtained with freeze drying. In experiment I, total phenolic contents in oven-dried tomato powder, water soluble and insoluble powder were more than 2 g/100 g. The radical scavenging activity and iron-chelation ability of the water-insoluble extracts were higher than those of dried TP and water-soluble extracts. In experiment II, pH values and Hunter L values of pork sausages formulated with TP were reduced. However, hunter a and b values were higher than those of the control, and those effects were increased with increased levels of TP. 2-Thiobarbituric acid reactive substance values of pork sausages containing TP were lower than that of the control during refrigerated storage, regardless of the TP level. These results indicated that TP could be used as a natural color agent and antioxidant in meat products without defects.
