Bioprospecting of Novel and Bioactive Metabolites from Endophytic Fungi Isolated from Rubber Tree Ficus elastica Leaves

  • Endophytic fungi are an important component of plant microbiota, and have the excellent capacity for producing a broad variety of bioactive metabolites. These bioactive metabolites not only affect the survival of the host plant, but also provide valuable lead compounds for novel drug discovery. In this study, forty-two endophytic filamentous fungi were isolated from Ficus elastica leaves, and further identified as seven individual taxa by ITS-rDNA sequencing. The antimicrobial activity of these endophytic fungi was evaluated against five pathogenic microorganisms. Two strains, Fes1711 (Penicillium funiculosum) and Fes1712 (Trichoderma harzianum), displayed broad-spectrum bioactivities. Our following study emphasizes the isolation, identification and bioactivity testing of chemical metabolites produced by T. harzianum Fes1712. Two new isocoumarin derivatives (1 and 2), together with three known compounds (3-5) were isolated, and their structures were elucidated using NMR and MS. Compounds 1 and 2 exhibited inhibitory activity against Escherichia coli. Our findings reveal that endophytic fungi from the rubber tree F. elastica leaves exhibit unique characteristics and are potential producers of novel natural bioactive products.

Biological activities of Rosa multiflora Ethanol Extract as Cosmetic Material

  • The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity as a functional ingredient for cosmetic products have not yet been studied. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. The antioxidant activities were determined by DPPH. Additionally, the contents of total phenols and flavonoids were analyzed. Also, the phenolic compounds were detected using HPLC. The melanogenesis regulatory effect was evaluated using melanin content and cellular tyrosinase activity in B16F10 melanoma cells. The elastase inhibitory activity assay was performed for anti-wrinkle effect. The antimicrobial activity was assessed using the disc diffusion assay. The DPPH radical scavenging ability, denoted by the $SC_{50}$ value was found to be $123.1{\mu}g/ml$, whereas that of positive control (ascorbic acid) was $27.5{\mu}g/mL$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In addition, astragalin and gallic acid were identified in the extract. Also, the ethanol extract significantly inhibited ${\alpha}$-MSH-induced melanogenesis in B16F10 cells. For anti-wrinkle effect, elastase inhibition activity of the ethanol extract was 53.2% at a concentration of $100{\mu}g/ml$. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 0.5 - 5%, and no significant activity was noted against C. albicans. Therefore, the ethanol extract of Rosa multiflora can be used effectively for development of functional cosmetic materials.

찔레꽃 에탄올추출물의 생리활성과 화장품 방부효과 (Biological Activity and Cosmetic Preservative Effects of Rosa multiflora Ethanol Extracts)

  • Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the $SC_{50}$ (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be $131.63{\mu}g/m{\ell}$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA). Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.

천연물유래 여드름 치료제제의 항균활성 측정 (Antimicrobial Activities of the Anti-acne Compounds from Natural Sources)

  • The in vitro antibacterial activities of anti-acne agents prepared from the extracts of natural sources were investigated against several bacteria including antibiotic-susceptible and -resistant Propionibacterium acnes. SD-1 and SD-2 were prepared with different formulations and they showed strong antibacterial activities. The anti-acne agents completely inhibited the growth of the tested strains at the concentration of 0.5%. There was no difference in antibacterial activity between antibiotic-susceptible and -resistant P. acnes. The inhibitory activities of two agents showed time-dependent manner. In S. aureus, time-kill curve demonstrated 2.8- and 3.4-$log_{10}$-unit killing after 8 h with SD-1 and SD-2, respectively. In P. acnes, time-killing curve demonstrated 5.1- and 6.1-$log_{10}$-unit killing after 24 h with SD-1 and SD-2, respectively. SD-2 showed stronger antimicrobial activity than SD-1. From these results, we expect that SD-1 and SD-2 have strong antibacterial activities and have advantages for treating acne.

  • Stability of the $\beta$-sitosterol glycoside from Schima wallichii sp. at various physical conditions were investigated, mutagenecity of the steroid saponin was determined by Ames test. When exposed in pH 3 to pH 8, the $\beta$-sitosterol glycoside was stable on antimicrobial activity against yeasts. The antimicrobial activity of the $\beta$-sitosterol glycoside also stable in high temperature, $N_2$, $O_2$ gas and light exposure, and metal ion. Ames test result revealed that $\beta$-sitosterol glycoside did not have any mutagenic activity. These results suggest that the $\beta$-sitosterol glycoside might be a promising candidate as a natural antimicrobial compound.

Carvacrol과 thymol을 병행처리한 키토산 코팅이 냉장저장 시 흰다리 새우의 미생물 및 품질 특성에 미치는 효과 (The Effect of Chitosan Coating Combined with Carvacrol and Thymol on Microbial and Quality Characteristics of Litopenaeus vannamei during Cold Storage)

  • 본 실험에서는 냉동 새우(Litopenaeus vannamei)를 $4^{\circ}C$에서 12일동안 저온 저장하며, 키토산 코팅과 천연보존제의 병행처리(0.05% carvacrol, 0.05% thymol)가 미생물(중온균, 저온균, Pseudomonas spp., $H_2S$ 생성균)의 성장과 새우의 이화학적 특징(총 휘발성 질소화합물, pH) 및 관능적 품질(외관, 냄새, 전반적 수용도)에 미치는 영향을 살펴보았다. 키토산 코팅과 carvacrol 병행 처리, 키토산 코팅과 thymol 병행 처리는 모든 대상 미생물의 성장을 억제하였고 특히 중온균, 저온균, $H_2S$ 생성균의 경우 실험의 마지막 날인 12일째까지 유의적인 항균효과(p < 0.05)를 보였다. 이 처리구들은 총 휘발성 질소화합물과 관능적 특성(외관, 냄새, 전반적 수용도)에서도 12일째까지 유의적인 차이(p < 0.05)를 보였으나 pH에서는 유의적 차이가 없었다. 따라서 키토산 코팅과 carvacrol 병행 처리, 키토산 코팅과 thymol 병행 처리는 새우의 주요 부패 미생물의 성장을 효과적으로 억제하는 것으로 보이며 냉장 새우의 유통기간 연장에 활용할 수 있을 것으로 보여진다.

김치의 선도유지를 위한 천연보존제의 탐색 (Screening of Natural Preservatives to Inhibit Kimchi Fermentation)

  • As a primary step to develop natural preservative for extending the shelf-life of kimchi, the effect of 102 edible plants, 21 antimicrobial agents and related compounds on kimchi fermentation was studied. Among 42 oriental medicinal plants tested, Baical skullcap and Assam indigo were found to be highly effective for maintaining the fresh state of kimchi. Although Bugbane, Red mangolia, Bushy sophora, Szechuan pepper, Chinese quince and Scisandre significantly inhibit the growth of Lactobacilli, their effect was not high enough to be used as raw materials for kimchi preservative. When the effect of 32 herbs and spices was tested, peppermint, cinnamon, lemon balm, clove, hop, rosemary, sage, horseradish and thyme showed high antimicrobial activity against kimchi microorganisms. Among them, the effect of clove ranked top. When it was added to fresh kimchi, initial cfu value ($2.4{\times}10^{6}cfu/g$) changed little even after 2 day's fermentation ($2.6{\times}10^{6}cfu/g$). Sensory test was not a good criteria to evaluate the effect of herbs and spices, since their highly specific flavors affected the taste of kimchies. Twenty eight fruits, vegetables and related plants were tested, but only leaves of pine tree, persimmon and oak leaves showed a significant bactericidal effect, finally contributing to the storage of kimchi. In addition, when 21 natural preservatives and other compounds were added individually to fresh kimchi, nisin and caffeic acid could inhibit fermentation.

Synthesis and Biological Activity of Geranyloxy Compounds

  • Disk assays on the compounds (10 and 12) showed both to have antifungal activity against the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (1 and 3 mm inhibition zones at $60\;{\mu}g/disc$), but not against the Gram-positive bacterium B. subtilis or the Gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa or fungi Cladosporium resinae and Candida albicans. However, the compound (13) did not show against antifungal activity. The geranyloxy compounds (10, 12, and 13) were cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_50$ >6,250 ng/mL at $7.5\;{\mu}g/disc$). These results suggest that The geranyloxy compounds possesses antimicrobial and antitumor activities.

등대풀(Euphorbia helioscopia) 용매 추출물의 항산화 및 항균활성 (Antioxidative and Antimicrobial Activities of Euphorbia helioscopia Extracts)

  • 본 연구는 대극과 식물인 등대풀(Euphorbia helioscopia)을 대상으로 에탄올 추출 및 용매분획하여 항산화 활성 및 항균활성을 비교 분석하였으며, 아울러 총 폴리페놀 및 플라보노이드 함량변화를 조사 비교함으로써 식품 저장성이나 안전성을 향상하기 위한 식품첨가제나 보존제로서의 개발 가능성을 알아보고자 하였다. 그 결과 에탄올 추출물 및 순차적 분획물의 항산화 활성은 처리 농도가 높아질수록 항산화 활성도 비례해서 증가되었으며, 특히 순차적 용매분획물 중 에틸아세테이트 분획물에서 항산화 활성이 가장 우수하게 나타났다. 또한 항균활성은 순차적 용매분획물 중에 에틸아세테이트 분획물이 본 실험에 사용된 7종의 균주 모두에서 가장 효율적인 항균활성을 보여 주었다. 이러한 등대풀 에탄올 추출물과 순차적 분획물별 항산화 및 항균활성 결과는 총 폴리페놀 및 플라보노이드의 함량과의 비례 상관관계를 보여주었으며, 한편 총 폴리페놀 함량은 에틸아세테이트 분획물>부탄올 분획물>디클로로메탄 분획물>에탄올 추출물>수용성 분획물>헥산 분획물 순으로, 총 플라보노이드 함량은 디클로로메탄 분획물>에틸아세테이트 분획물>에탄올 추출물>헥산 분획물>부탄올 분획물>수용성 분획물 순으로 나타났다.

프로폴리스 추출물의 항균 활성에 대한 연구 (Studies on the Antimicrobial Effect of Extracts of Propolis)

  • 기존에 식품의 저장성 향상을 우해 사용되어졌던 방법들의 문제점들을 최소화하고 보다 효과적인 식품저장을 위해 전통적으로 식용되어 왔으며 그 안전성이 확보된 프로폴리스를 대상으로 대표적인 식품 부패 미생물에 대한 생육억제활성을 식용되어 왔으며 그 안전성이 확보된 프로폴리스를 대상으로 대표적인 식품 부패 미생물에 대한 생육억제활성을 탐색하고 건강 지향적인 식품보존제로서 이를 식품에 적용하여 보다 좋은 품질의 가공식품개발에 활용코자 본 연구를 진행하였다. 프로폴리스 에탄올 추출물의 일반성분 분석 결과를 보면 19.41% 수분, 0.39% 조단백, 5.12% 조지방, 0.15% 조회분의 결과를 보여주었다. 프로폴리스는 에탄올 60%, 에탄올 80% 추출물에서는 높은 추출율을 보였다. 프로폴리스 에탄올 추출물에 의한 항균성은 100% 에탄올 추출물로 실험한 처리구에서 높은 항균 효과를 나타내어 이를 사용하여 유해 미생물들에 대한 농도별 항균 효과를 살펴보았다. 프로폴리스 추출물로 유해미생물을 처리했을 때 Bacillus subtilis, Micrococcus luteus는 1~2 mg/mL로 처리 시 생균수가 1~4log 감소하였고, E. coli는 3mg/mL로 처리 시, Bacillus cereus는 1~2 mg/mL의 노도로 처리 시 대조군에 비해 생균수가 2log 감소하였다. Staphylococcus aureus와 Shigella sonnei의 사멸효과는 0.1~3 mg/mL의 농도에서 생균수가 2~4log 감소하였다. Erwinia rhapontici의 사멸효과는 2 mg/mL 농도로 실험했을 때 2 log 정도 생ㅇ육저해효과를 보였고, Salmonella choleraesuis, Vibrio parahaemolyticus에서의 사멸효가는 크게 관찰할 수 없었다. 프로폴리스는 그램양성균에 대한 사멸효과가 그램음성균에 대한 효과 보다 더 우수하였다.
