천연 쑥 추출물의 약리 및 화학적 특성 연구 (A Study on the Pharmacetical and Chemical Characteristics of Natural Artemisia Extract)

  • Natural Artemisia extraction was extracted from Artemisia component using diethyl ether as a solvent, and we tested various pharmacetical and chemical characteristics of this extract. Characteristic experiments to use natural Artemisia extract tested antimicrobial experiment using microbe in pharmacetical material, and tested dye experiment using fiber in chemical material. From the result of characteristics experiment, some conclusions are obtained as follow. From the result of extraction experiment, it obtained about 10.4%-Artemisia extraction ratio as semi-solid state, and after dried in freezing from Artemisia extract of semi-solid state, it obtained about 10%-Artemisia extraction ratio as solid state of dark blue-green color. From result of antimicrobial experiment of Artemisia extract, number of staphylococcus aureus (ATCC-01) and aspergillus niger (ATCC-02) in microbe decreased more and more according to time passage. This phenomenon showed that Artemisia extract influences to antimicrobial effect. From the result of dye experiment of Artemisia extract, it appeared in direction of dark blue-green color after dyed to use cotton and silk with fiber to control in pH 7.5. Specially the result which confirmed dye of fiber with optical electron microscope (OEM), we could know that it appears darker silk than cotton.

  • Natural grape extraction was extracted from grape component using diethyl ether as a solvent, and we tested various pharmaceutical and chemical characteristics of this extract. Characteristic experiments to use natural grape extract tested antimicrobial experiment using microbe in pharmaceutical material, and tested dye experiment using fiber in chemical material.   • An Actinomycetes strain JB isolated from Mt. Hanla had a strong antimicrobial activity against gram positive bacteria and tumor cell growth inhibition. Especially, it couldn't degrade starch and casein as organic compounds. It was resist on lincomycin and rifampicin. The spore mass of strain JB which was arethrospore was white. DAP of the cell wall was L, L-DAP. Antimicrobial material was heat stable, dissolved in ethyl acetate, and not dissolved in butanol. In the pressnce of 0.1% phenol and 4% sodium chloride, strain JB could grow, but it didn't growth at below $10^{\circ}C$. Strain JB didn't use dextran, sodium acetate and sodium citrate as sole carbon source and L-cystein and L-thereonine as nitrogen source. The filtered broth of strain JB had the antimicrobial activity against gram positive bacteria, especially Staphylococcus aureus (ATCC 65389) and the growth inhibition of tumor cell line.

Nut류의 항산화 및 항균효과 (Antioxidative and Antimicrobial Effects of Nut Species)

  • It has been known that diseases related with aging or cancer result, at least in part, from free radicals, and antioxidants may reduce or prevent the abnormalities associated with free radical formation via its scavenging action. Thus, this study was carried out to investigate the antioxidative and antimicrobial activities of nut species for the purpose of developing antioxidant from natural products. Antioxidant activities of four kinds of nuts such as chestnut, groundnut, walnut and acorn were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The extracts from chestnut hull and acorn barnyard millet showed strong antioxidative activities among 10 samples tested. Because of their highest antioxidative activity among 10 samples, radical scavenging effects of 4 different extract compartments (Hexane, EtOAc, BuOH and $H_2O$ extracts) from chestnut hull and acorn barnyard millet, were further examined by DPPH method. EtOAc and BuOH extracts exhibited antioxidative activities similar to those of natural, tocopherol or synthetic antioxidants, BHA. These findings demonstrates that major fraction of the antioxidative activity of chestnut hull or acorn barnyard millet was the EtOAc and BuOH extract compartments. However, antimicrobial activities against food-related bacteria and yeasts was relatively weak.

한우의 자궁 내 분리 세균에 대한 천연물질 항균효과 (Antimicrobial effects of natural extracts on bacteria isolated from the uterus of Hanwoo (Korean native cattle))

  • Bacterial infection of the uterus in Hanwoo can kill embryos. Therefore, many antibiotics have been used to treat this infection in the uterus of repeat breeders. Incorrect use of antibiotics has led to resistance in bacteria. Natural compounds have used as substitutes for antibiotics because they are safe and have very mild side effects. This study was conducted to examine the antimicrobial effects of five extracts from medicinal plants including Humulus japonicas (Hj), Phelledendron amurense (Pa), Viola mandshurica (Vm), Carthamus tinctorius (Ct), and Chelidoni herba (Ch) on bacteria isolated from the uterus of Hanwoo using the paper disc diffusion method. Hj and Pa extracts had potent antimicrobial effects against Staphylococcus lentus, Streptococcus infantarius subsp. coli, and Bacillus pumilus. Pa had the greatest antimicrobial effect among the five medicinal plants and was effective against 19 types of bacteria from bovine uterus. Compared to Pa, Hj showed weaker antimicrobial effects on all the bacteria tested except Pseudomonas aeruginosa. Vm, Ct, and Ch also showed weak antimicrobial effects on the tested bacteria. The results obtained suggest that Hj and Pa are natural compounds suitable for treating bacterial infection in repeat breeders and improving conception rates of Hanwoo.

몇종의 세균과 Saccharomyces cerevisiae에 대한 식품첨가물의 향균 특성 (Antimicrobial Activity of Some Food Additives against Bacteria and Saccharomyces cerevisiae)

  • 현재 식품첨가물로 많이 사용되고 있는 유기산과 안정제 및 색소 등의 각종 세균과 Sacch. cerevisiae 에 대한 향균 특성을 조사하였다. 산도 조절용으로 사용되고 있는 젖산, 사과산, 호박산 및 주석산은 L. acidophilus와 Sacch. cerevisiae에 대하여 향균성이 없었으나 B. subtilis등의 세균에 대하여는 향균성이 있었고 특히 사과산은 P. aeruginosa에 대하여 강한 향균력이 있었다.(최소생육저지농도 : 0.05%). 안정제로서의 알긴산과 펙틴은 B. subtilis, E. coli, P. aeruginosa에 대하여 비교적 강한 향균성을 보였고 L. acidophilus에 대하여는 향균성이 없었다. 황색 색소(홍화엘로우)와 적색 색소(Red powder-N)는 향균성이 없었고 표백제인 $NaHSO_3$의 세균에 대한 최소생육저지농도는 0.05%, Sacch. cerevisiae에 대하여는 0.5%로 향균성이 있었다.

천연물을 이용한 항균성 염료의 개발 (Development of Antimicrobial Dye for Natural Dyeing using Natural Substances)

  • 문헌조사를 바탕으로 천연 염색 시 염료로 사용이 기능할 것으로 판단되어지는 한약재와 천연물 중 72가지를 선발하고 메탄올 추출물을 이용한 1차 조사와 열수, 에탄올, 메탄올과 에틸아세테이트 추출물을 이용한 2차 조사 결과 전반적인 천연물에서 열수와 메탄올 추출물에서 상대적으로 유효한 항균성을 나타내는 것으로 확인되었다. 선별한 천연물 중 대황, 산사, 산수유, 소목, 오매, 오미자, 오배자와 황련 추출물에서 20 mm 이상의 생육저지환이 형성되었으나 항세균성에 대한 활성이 높은 것으로 조사되어 항진균성에 대한 연구가 보다 필요한 것으로 판단된다. 천연물 중 가장 우수한 항균성을 나타낸 황련 열수 추출물의 S. aureus ATCC 6538에 대한 MIC 값을 조사한 결과 110 mg/mL로 조사되었다. 또한 공시균주 외 다른 병원성 미생물에 대한 항균성을 조사한 결과 조사한 3종에 대하여도 양호한 항균성을 나타내는 것으로 조사되었다. 이에 선별된 천연물이 천연염색 뿐 아니라 다른 분야에서도 많이 적용될 수 있을 것으로 판단되었다. 그러나 용매에 따른 회수율과 항균성을 비교할 때 낮은 회수율에서도 높은 항균성을 나타내는 소목과 황련의 경우 회수율을 높일 수 있는 방법에 대한 연구가 진행된다면 보다 좋은 결과를 도출할 수 있을 것으로 판단되었다.

다가알코올로부터 추출된 황금, 아카시아, 한라봉 추출물의 천연 방부시스템 연구 (Study of Natural Preservative System Using the Mixture of Scutellariae radix, Acacia nilotica and Citrus reticulata Extracted from Polyhydric Alcohols)

  • The aim of this study was to develop a new natural preservative system to improve the weak points of natural polyhydric alcohols together with the efficiency of natural plants as a preservative. Polyhydric alcohols (glyceryl caprylate and ethylhexylglycerin) and antimicrobial plants (S. radix, A. nilotica and C. reticulata) were tested using the disc diffusion method and minimum inhibitory concentration (MIC) method for their antimicrobial activity against the common poultry pathogens, respectively. A study of the preservative efficacy of the cosmetic formulations containing the optimized preservative system demonstrated sufficient preservative efficacy against bacteria and eukaryotic test microbes. These results suggest that the natural preservative system including polyhydric alcohol extracts containing natural plants could be incorporated in cosmetic formulations.

Antimicrobial Effects of Lonicera japonica against Gram Positive and Gram Negative Anaerobic Bacteria

  • It has been shown that the butanol extract of Lonicera japonica has antimicrobial and other potentially useful biological activities. The purpose of this study was to determine the in vitro activity of Lonicera japonica compared to other antimicrobial agents against anaerobic bacteria. Specifically, the in vitro activity of the butanol extract was investigated against 104 clinical isolates of anaerobic bacteria using an agar dilution method and the results were compared to erythromycin, cefoxitin, imipenem, clindamycin, and metronidazole. It was found that Lonicera japonica and imipenem were the most active antimicrobial agents tested.

Antimicrobial Activity of Propolis Extract and Their Application as a Natural Preservative in Livestock Products: A Meta-Analysis

  • This study aimed to evaluate the effectiveness of propolis extract as a natural preservative for livestock products in term of chemical and microbiological characteristics by meta-analysis. The stages carried out in this study were identification, selection, checking suitability, and the resulting selected articles were used in the meta-analysis. The selection results obtained a total of 22 selected journal articles consisting of 9 articles for analysis of the antimicrobial activity of propolis extract and 13 articles for analysis of the chemical and mirobiological characteristics of livestock products. The articles were obtained from electronic databases, namely Science Direct and Google Scholar. The model used in this study is the random-effect model involving two groups, control and experimental. Heterogeneity and effect size values were carried out in this study using Hedge's obtained through openMEE software. Forest plot tests and data validation on publication bias was obtained using Kendall's test throught JASP 0.14.1 software. The results showed that there is a significant relationship between propolis extract with the results of the antimicrobial activity (p<0.05). In addition, the results of the application of propolis extract on the livestock products for the test microbes and the value of thiobarbituric acid reactive substances (TBARs) showed significant results (p<0.05). Conclusion based on the random-effect model on the effectiveness of antimicrobial activity of propolis extract and their apllication as a natural preservative of the chemical and microbiological characteristics of livestock products is valid by Kendall's test (p>0.05). Propolis in this case effectively used as natural preservatives in livestock products.
