Effect of Activity Immersion Conflict Perceived by Rural Multicultural Couple on Their Self-esteem, and Happiness : Focusing on Analysis of Actor and Partner Effect Using APIM (농촌 다문화가족 부부의 활동몰입갈등이 자아존중감과 행복감에 미치는 효과 : APIM을 이용한 행위자와 파트너 효과 분석 중심)

  • This study examined the effects of the actor and partner according to the activity immersion conflict perceived by rural multicultural couples in terms of the self-esteem and happiness using Actor-Partner Interdependence Model (APIM). For this purpose, the data of 482 persons from 241 husband-immigrant women dyads were analyzed. As results of the analysis, only the effect of the actor among the actor and partner effect according to the activity immersion conflict affected the self-esteem significantly in both husband and immigrant women. Conflict, namely the actor and partner effect, affected the happiness in both the husband and immigrant women significantly. Second, the difference in the actor conflict level on activity immersion perceived by the couple according to the women's native country was significant in both husband and immigrant women. The level of actor conflict was highest in Vietnamese husbands and the Philippine women. On the other hand, the difference in the partner conflict level on the activity immersion according to the women's native country was significant only in the women, and the level of Vietnamese women was highest. Therefore, the characteristics in the level of actor and partner conflict on activity immersion was caused by the difference in the number of activity organizations participating between the husband and women. Based on the results, this study suggests some measures to reduce the conflict of couples through activity. In addition, some plans for a follow-up study using APIM are suggested.

Evaluating feed value of native Jeju bamboo (Sasa quelpaertensis Nakai) for beef cattle

  • Objective: Recently, indigenous Korean grass Sasa quelpaertensis Nakai (SQ) has garnered much interest as a roughage source for livestock to mitigate its adverse effects on habitat diversity. Thus, the objective of the present study was to evaluate the ruminal fermentation, palatability, and nutrient digestibility of SQ for Korean native beef cattle (Hanwoo) using in vitro rumen fermentation, in situ rumen degradability, and in vivo feeding trials. Methods: Using in vitro tests with rumen fluid as the inoculum for 48 h, ruminal fermentation of SQ was evaluated and compared with that of other roughage sources commonly used in Korea (i.e., rice straw, Timothy hay, and Italian ryegrass [IRG]). Additionally, an in situ trial 96 h was performed using three cannulated Hanwoo steers. Further, an in vivo trial was performed using eight Hanwoo steers to compare the palatability of SQ with rice straw in total mixed ration (TMR) and forage-concentrate separate feeding conditions. Finally, an in vivo digestibility trial of SQ fed as TMR of two particle sizes was performed with four Hanwoo steers. Results: In vitro and in situ trials revealed that SQ was comparable or superior to rice straw in terms of the ruminal fermentation characteristics of pH, gas production, total volatile fatty acid content, and effective ruminal dry matter digestibility (DMD), although its fermentability was lower than that of Timothy hay and IRG. In the palatability test, steers showed a greater preference for SQ when given as TMR. The total tract DMD of SQ fed as TMR was 75.9%±1.37%, and it did not differ by particle size. Conclusion: The feed value of SQ as a roughage source for Hanwoo steers is comparable or superior to that of rice straw, particularly when provided as TMR.

Importance and Priority of Indicators for Selection of Plant Species for Ecological Restoration (생태복원용 식물종 선정을 위한 지표의 중요도·우선순위)

  • Ecological restoration is considered a good means to prevent biodiversity loss in terms of the ecosystem's health and sustainability. However, there are difficulties in putting it into practice as there is no comprehensive and objective standard for the selection of plant species, such as environmental, ecological factors, and restoration goal setting. Therefore, this study developed an evaluation index necessary for selecting plant species for restoration using the Delphi method that synthesizes the opinions of the expert group. A survey with 38 questionnaires was conducted twice for experts in ecological restoration, etc., and the importance and priority of evaluation indicators were analyzed by dividing the restoration targets into inland and island regions. The result of the importance analysis showed that "native plants" had the highest average of 4.9 among the evaluation indices in both inland and island regions, followed by "seed security", "propagation", and "root growth rate". In the inland region, the index priority was analyzed in the order of "native plants", "appearance frequency", "root growth rate", "distribution range", and "seed security" in the island region, it was analyzed in the order of "native plants", "root growth rate", "appearance frequency", "distribution range", and "tolerance", showing slight differences between the two indicators. As a result of the importance and priority indicator analysis, we set the mean importance and priority of 4.1 and 2.9, respectively, in the inland region and 4.2 and 2.9, respectively, in the island region. As for the criteria of selecting plant species for ecological restoration, the "native plants" had the highest importance and priority. "Seed securing", 'viability", "topography", "proliferation", "tolerance", "soil conditions", "growth characteristics", "early succession", "distribution range", "appearance frequency", and "germination rate" were classified into subgroups of low importance and priority. The lowest indicators were "final stage of succession", "transition period", 'transition stage", "root", "reproduction", "soil", "appearance", "technology", "landscape", "climate", and "germination rate". We expected that the findings through objective verification in this study would be used as evaluation indicators for selecting native plant species for ecological restoration.

Establishment of Korean Native Chicken Auto-Sexing Lines Using Sex-Linked Feathering Gene (한국토종닭의 깃털 발육성 반성 유전자를 이용한 자가성감별 계통 조성)

  • Although feather-sexing using sex-linked genes related to feather development is a widely used chick sexing method in the poultry industry, the feather-sexing method has yet to be used for Korean native chickens (KNCs). The purpose of this study was to construct a KNC feather-sexing line using early-feathering (EF) and late-feathering (LF) genes for industrial application. Using 557 reddish-brown KNCs as the basal flock, frequencies of the EF (k) and LF (K) genes were estimated to be 0.814 and 0.186, respectively. This indicating that it would be feasible to construct a feather-sexing line using this chicken group, and we accordingly constructed EF paternal and LF maternal lines. On the basis of test-cross for the selection of LF homozygous (KK) males in the maternal line, we confirmed that three of 40 chickens were homozygous males. The survival rate, body weight, days at first egg-laying, hen-day egg production, and egg weight were analyzed to compare the production performance of EF and LF chickens. The results revealed that EF chickens were characterized by a superior survival rate, whereas LF chickens were superior in terms of egg production rate. However, no differences between LF and EF chickens were detected with respect to other production performance parameters. In addition, assessment of the fitness of sexed chicks produced in the established KNC feather-sexing lines revealed that the accuracy of sexing was 98.6%. Collectively, these findings indicate the feasibility of constructing effective KNC feather-sexing lines with potential industrial application.

Biosynthesis of a Biological Active Single Chain Equine Chorionic Gonado-tropin

  • The equine chorionic gonadotropin (eCG) subunits $\alpha$ and ${\beta}$ are transcribed from different genes and associate noncovalently to form the bioactive eCG heterodimer. Dimerization is rate limiting for eCG secretion, and dissociation leads to hormone inactivation. The correct conformation of the heterodimer is alto important for efficient secretion, hormone-specific post-translational modifications, receptor binding and signal transduction. To determine whether ${\alpha}$ and ${\beta}$ subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule by fusing the carboxyl terminus of the eCG ${\beta}$-subunit to the amino terminus of the af-subunit was construe-ted and transfected into chinese hamster ovary (CHO-Kl) cells. LH- and FSH-like activities were assayed in terms of testosterone production and aromatase activity in primary cultured rat Leydig cells and granulosa cells, respectively. The tethered-eCG was efficiently secreted and showed similar LH-like activity to the dimeric eCG ${\alpha}$/${\beta}$ and native eCG. FSH-like activity of the tethered-eCG was also shown similarly in comparison with the native and wild type eCG ${\alpha}$/${\beta}$. Our data for the first time suggest that the tethered-eCG can be expressed efficiently and the produced product by the CHO-K1 cells is fully LH- and FSH-like activities in rat in vitro bioassay system. Our results also suggest that this molecular can imply particular models of FSH-like activity not LH-like activity in the eCG. Taken together, these data indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion.

Comparison of Hemodynamic Energy between Expanded Polytetrafluoroethylene and Dacron Artificial Vessels

  • Background: Artificial grafts such as polyethylene terephthalate (Dacron) and expanded polytetrafluoroethylene (ePTFE) are used for various cardiovascular surgical procedures. The compliance properties of prosthetic grafts could affect hemodynamic energy, which can be measured using the energy-equivalent pressure (EEP) and surplus hemodynamic energy (SHE). We investigated changes in the hemodynamic energy of prosthetic grafts. Methods: In a simulation test, the changes in EEP for these grafts were estimated using COMSOL MULTIPHYSICS. The Young modulus, Poisson ratio, and density were used to analyze the grafts' material properties, and pre- and post-graft EEP values were obtained by computing the product of the pressure and velocity. In an in vivo study, Dacron and ePTFE grafts were anastomosed in an end-to-side fashion on the descending thoracic aorta of swine. The pulsatile pump flow was fixed at 2 L/min. Real-time flow and pressure were measured at the distal part of each graft, while clamping the other graft and the descending thoracic aorta. EEP and SHE were calculated and compared. Results: In the simulation test, the mean arterial pressure decreased by 39% for all simulations. EEP decreased by 42% for both grafts, and by around 55% for the native blood vessels after grafting. The in vivo test showed no significant difference between both grafts in terms of EEP and SHE. Conclusion: The post-graft hemodynamic energy was not different between the Dacron and ePTFE grafts. Artificial grafts are less compliant than native blood vessels; however, they can deliver pulsatile blood flow and hemodynamic energy without any significant energy loss.

Stage specific transcriptome analysis of liver tissue from a crossbred Korean Native Pig (KNP × Yorkshire)

  • Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP ${\times}$ Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (${\left|log2FC\right|}$ > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.

Effect of Nitrogen Fertilization on the Forage Growth, Yield and Quality of Native Reed (Phragmites communis) (갈대 생육지에서 질소 시비가 사초의 생육과 수량 및 사료가치에 미치는 영향)

  • This study was carried out to investigate the effect of nitrogen (N) fertilization on the forage growth, production and quality of native reed (Phragmites communis) grasses. Field experiments were conducted in Cheonan and in Ansan, 2012. Treatments were control (no N fertilization), 50 kg/ha and 100 kg/ha in Cheonan plots (fertilization on April $30^{th}$, and harvest on June $21^{st}$). Treatments in Ansan plots were control (no N fertilization) and 60 kg/ha (fertilization on May first, and harvest on August first). Forage growth and leaf colors were improved in N fertilized plots. However, the drymatter (DM) percentage was slightly decreased with N fertilization. Forage yields, in terms of DM, crude protein (CP) and digestible DM (DDM), were significantly increased with N fertilization in both sites. In Cheonan, DM, CP and DDM yields per ha were 4,026 kg, 235 kg and 1,850 kg, respectively, in the control plot, and were 4,658 kg, 306 kg and 2,388 kg, respectively, in the N 50 kg plot, and 5,622 kg, 446 kg and 3,143 kg, respectively, in the N 100 kg plot. In Ansan, DM, CP and DDM yields per ha were 2,802 kg, 177 kg and 1,288 kg, respectively, in the control plot, and were 3,876 kg, 294 kg and 1,853 kg, respectively, in the N 60 kg plot. Forage quality in terms of CP content, in vitro DM digestibility (IVDMD) and relative feed value (RFV) were also increased with N fertilization in both sites. In Cheonan, the CP content, IVDMD and RFV were 5.85%, 45.96% and 64.5 (grade 5), respectively, in the control plot, 6.58%, 51.27% and 72.3 (grade 5), respectively, in the N 50 kg plot, and 7.94%, 55.91% and 72.7 (grade 5), respectively, in the N 100 kg plot. In Ansan, the CP content, IVDMD and RFV were 6.30%, 45.98% and 70.2 (grade 5), respectively, in the control plot, and 7.59%, 47.80% and 78.3 (grade 4), respectively, in the N 60 kg plot. In conclusion, N fertilization of 60~100 kg/ha was desirable for greater forage production, with a higher quality of native Phragmites communis achievable. This should only be applied if the fertilization area is not located at a riverside/streamside or in riparian land where there is a high risk of water pollution by fertilization.

Physicochemical Characteristics of Native Korean Allium wakegi Araki according to Different Seed Bulb Origins (종구산지에 따른 한국산 쪽파(Allium wakegi Araki)의 이화학적 특성)

  • The aim of this study was to investigate the physicochemical characteristics of native Korean Allium wakegi Araki samples, which were grown from seven local seed bulbs(Yesan, Muan, Anmyon-do, Deokjeok-do, Jeju-do, Yecheon, and China) to produce high quality native Korean Allium wakegi Araki. For the proximate composition of samples, moisture contents were in the range of 90.69~92.43%. The crude protein content of the Jeju-do sample was highest compared to the other samples. However, there were no significant differences in total sugar contents between samples. The seed bulb origin did not affect the hardness of the stem part, but was high for the leaves of the Yesan sample compared to the other samples. The results for anti-oxidative activity were as follows: Yesan(2.30 mg/mL) > China(2.51 mg/mL) > Muan (2.56 mg/mL) > Yecheon(2.74 mg/mL) > Jeju-do(2.85 mg/mL) > Anmyon-do(2.87 mg/mL) > Deokjeok-do(3.18 mg/mL). In terms of mineral and amino acid contents, the Yesan sample showed the highest levels, respectively, compared with the other samples. Food values such as contents of total phenolics and pyruvic acid were highest in the Jeju-do sample. These results show that the physicochemical characteristics of Allium wakegi Araki were significantly different according to different seed bulb origins.

Dyeability of Ramie Fabrics Using Extract of the Native Plant of Rubia akane Nakai Grown in Korea (한국 자생 꼭두서니 추출물에 의한 모치섬유의 염색성)

  • This study was carried out to investigate into the dyeability of ramie fabrics using the extract of native plant, red-dye madder (Rubia akane Nakai) in different conditions. Surface color of ramie fabrics dyed with extracts from red-dye madder plants showed an order of descent YR. However, it was changed by the different mordants. Surface color of ramie fabrics treated with FeSO$_4$, SnCl$_4$, and tartaric acid was an order of descent Y, and that treated with the ashes of common camellia, and bean straw was an order of descent R. At higher temperatures, surface color and coloring matter concentration of ramie fabrics were as follows that values of a increased, however, values of b decreased and values of L lowered. Coloring matter concentration increased in the order of 7$0^{\circ}C$, 9$0^{\circ}C$, and 6$0^{\circ}C$, and so on. Dyeing colors on the ramie fabrics showed an order of descent Y by pH 4 and 9 in dyestuffs solutions. Coloring matter concentration in terms of K/S values, more or less, was increased at strong acids. Surface color by the concentration of dyestuffs solutions showed an order of descent YR irrelevant to the concentration of dyestuffs solutions. Coloring matter concentration was increased at higher concentrations of dyestuffs solutions. Ten minutes was sufficient for the dyeing of ramie fabrics in terms of surface color and coloring matter concentration.
