Analysis of Genetic Relationship of Entomogenous Fungi in Korea by Morphological Characteristics and RAPD (한국에서 채집한 Entomogenous fungi의 형태와 RAPD에 의한 유연관계 분석)

  • RAPD test and the observation of morphological, cultural characteristics of fourteen selected entomogenous fungi were conducted to investigate the analysis of their internal relationships. Paecilomyces tenuipes showed snow flower form attached to numerous white conidiophores, produced globular and semi-egg types on the club types of phialides. Cordyceps militaris formed globosely conidiophores, dark yellow fruiting body on pupa. The phialide as on Acremonium-type in global conidiophores. Beauveria bassiana covered with conidia was not formed fruiting body and adhered conidia on conidiophore of zigzag type. The PDA and SDAY medium were confirmed as an optimum growth of them. P. tenuipes showed to velvet and plane types in several media whereas C. militaris was belong to centrally raised and floccose in the morphological type. In contrast, B. bassiana covered with conidia on velvet shape. The size of amplified products were analyzed by RAPD using URP primer and were from 100 bp to 2.0 kb with $10{\sim}14$ geuomic DNA. Total similarities of two groups were by dendrogram of UPGMA analysis. The homology of P. tenuipes groups was 94.8 to 100%. It also showed 70.1 to 96.6% in C. militaris group and B. bassiana was higher similarity than any other. The internal change of C. militaris, produced telemorph fruiting body, was higher seperated in species than P. tenuipes and B. bassiana in the RAPD.

Current status on the development and commercialization of GM plants (국내·외 GM식물의 개발 및 산업화 현황)

  • During a last decade, the introduced traits in commercialized GM crops have been diversified from a simple trait such as herbicide resistance gene or insectresistance gene which are related to the crop production into more complicated traits such as modification of fatty acid or essential amino acid composition, modified coloring pattern of flower. In addition, it was investigated that several other GM crops bearing more refined traits expected to lead next generation are also awaiting for risk assessment (RA) or under field test for the preparation of RA in the near future. These GM crops include abiotic stress resistance including drought or cold, increased biomass, production of bioethanol or diesel, production of pharmaceuticals or functional materials for industrial. In particular, in 2008 and 2009, it was reported that the highest number of GM crops for molecular farming are under developed in laboratory or green house level in all the world. Likewise, in Korea, 171 events from 49 plant species are under developed to introduce several important traits. At present, about 10 events are under field test to select elite lines for RA application. For the first time, herbicide resistance turfgrass developed by Korean research team has been submitted for RA and currently under requested for additional data. Moreover, GM rice resistant to leaf roll (folder) disease is expected as a next event to be submitted for RA application.

Investigation of the Medicinal Plant Resources (약용식물(藥用植物)의 유전자원수집(遺傳資源蒐集)과 이용(利用)에 대(對)한 조사(調査))

  • In order to collect the plant genetic resources in Korea, the medicinal plants were surveyed from 1987 to 1989 in Kyungbuk province and at the several areas in Kyuognam and Choongbuk province. The medicinal plants of 456 species in 118 families were investigated in these regions. Among them, 249 species were belonged to 19 families, Compositae, Liliaceae, Leguminosae, Poiygonaceae, Labiatae, Rosaceae, Umbelliferae, Gramineae, Solanaceae, Cucurbitaceae, Ranunculaceae, Cruciferae, Rutaceae, Araliaceae, Araceae, Moraceae, Scrophulariaceae, Caryopyhllaceae, and Convolvulaceae, and others were belonged to 99 families. They have been used mostly for urination, gastroenteric disoder, robustness, antiphlogistic, anodyne, cough medicine, fever remedy, hemostatic, and 9 species of them have been used for anticancer medicine. The medicinal plant resources which were planted in the fields, gardens, parks, roadsides, and glass house, were 93 species for ornamental, 50 for medicine, 27 for vegetable, 14 for food, 5 for vegetable fruit, 13 for fruit, 5 for oil, 6 for tea, 9 for shade, and 14 for other purposes. Most of investigated plants were perennial, and flowered from June to August. The root parts were most frequently used as medicine, and other parts were used in the order of leaf and stem, whole plants, flower, seed, and fruit. Among the 456 species, 191 species in 87 families were investigated to be important medicinal plants.

A Study of Geum Silk from Seokgatap in Bulguksa (불국사 석가탑 내 발견 금직물(錦織物) 고찰)

  • In 1966, $Seokgatap$ pagoda in $Bulguksa$ temple was damaged by the tomb robbers and was dismantled to fix the damage. In the process, many offerings to Buddha and containers for Sarira(the cremated remains) were found in $Sarigong$(specially designated space for the Sarira casket) inside the second floor of the pagoda. Many fabrics like $Geum$, $Neung$(twill), $Rha$(complex gauze), silk tabby and linen were also excavated. In this study, $Geum$ fabric from the $Seokgatap$ was closely examined. $Geum$ of $seokgatap$ is weft-faced compound weave according to the analysis of its weaving pattern which was wrongly presumed as warp-faced compound weave for some time. Technical analysis of $Geum$: Main: silk, Binding: silk, Proportion: 1 main warp to 1 binding warp, Count: 15 main warps and 15 binding warps per centimeter, Weft: polychrome silk without apparent twist, Colors: yellow, mustard yellow, deep blue, green and purple, Weave: weft-faced compound twill, 1/2 S. $Geum$ of $Seokgatap$ was made in the $8^{th}$ century, since it was weaved in weft-faced compound weave twill which was popular in the $8-9^{th}$ century. And also, the arrangement of the colors was done in the same way of gradation $Geum$ silk which was popular in the $7-8^{th}$ C in China and Japan. Third, we restored the pattern of $Geum$ of the Unified Shilla Dynasty for the first time. It was very difficult to figure out the shape and the size of pattern since the fabric was partially lost and ruined. We tried to draw the diagram of structure with the cross point of the warp and the weft to restore the pattern. By doing so, we could identify two kinds of small flower pattern, palmette and the pattern of repeating vines. Fourth, we could infer that the $Geum$ of $Seokgatap$ was used for $geumdae$(a pouch made of $geum$) by analyzing all the documents and the characteristics of the fabric.

  • In this paper, mechanisms of gene editing technologies including ZFN, TALENS and CRISPR were briefly discussed with mutual advantages and disadvantages. Classification criteria of gene edited, site-directed mutagenesis (SDN) crops for regulatory purpose were also discussed. The number of studies using CRISPR technology was high and studies conducted on Arabidopsis thaliana and rice were highest, followed by tobacco, tomato, wheat, and corn. It has been applied to a variety of plants such as other grain crops, flower crops, vegetable crops, and fruit trees. The number of studies focused on practical application or commercialization in the future were also increasing yearly, and the scope of studies also expanded to include research on metabolic engineering for mass production of useful proteins or substances, development of disease resistant crops against viruses, bacteria, and fungi, abiotic environmental stressresistant crops, and increased yields. In addition to this, it was revealed that application range is becoming more diversified, including the development of parthenocarpic tomatoes, hybrid rice lines using male sterility and increased shattering resistance Brassica napus. It was also revealed that the number of CRISPR gene edited crops permitted by the USDA(APHIS) increases yearly, to be released in the international seed market soon.

A Study on Korean Twill Fabrics in the 17th Century (17세기 한국 능(綾)직물 연구)

  • This study studies the fabrics from excavated 17th century tombs of Mrs. Min from Yeoheung family(1586~1656), Yeo-on Kim(1596~1665) and Won-rip Choi(1618~1690) and attempts to clarify the relationship between the fashion trend in fabrics of those times and the background behind it by viewing and examining the proportion of twill fabrics to the total silk fabrics and the characteristics of its weave and patterns. Looking into fabrics from the above tombs, twill fabrics accounted for 10.4%(13 pieces) 19.3%(16 pieces) and 9.2%(9 pieces) of total silk fabrics in each of the respective tombs. This forms a remarkable contrast with the fact that there was only one piece of twill silk fabrics(0.5%) and not any from the 16th century tombs of Mrs. Yoon from Papyeong family (0.5%) and Soo-ryoon Sim(0%). In particular, the percentage of hwamun-neung(patterned twill fabrics) in each of the tombs is 8.0%(10 pieces), 13.3%(11 pieces), 9.2%(9 pieces), which is much higher than that of non-patterned one. This is common to the twill fabrics from above three 17th century tombs. Patterns of hwamun-neung(patterned twill), simplified small flower patterns or geometrical figures, from the three excavated tombs are mostly arranged sporadically with blank space. It is supposed that these figurative characteristics reflected the aesthetic sense of the gentry at that time which valued simplicity and moderation for their Confucian standard. This phenomenon of increased use and production of twill fabrics in the 17th century resulted from different factors such as wars like Japanese Invasion of Korea(1592~1598), economic difficulty, government regulations against the production of high-class fabrics, development of weaving skill and its fixation, changes of fabric production environments, and changes of aesthetic sense preferring naive and moderate things to showy ones. As for the weaving characteristics of twill fabrics from the three 17th century tombs 3 leaf warp-faced twill was often used for the ground texture and 4 leaf warp-faced one was occasionally used. For pattern texture 6 leaf weft-faced twill was frequently used, 4 leaf weft-faced twill and 3 leaf weft-faced twill were used at times, and floating one was occasionally used as well.

Expression Methods of Peony Patterns in Korean Textiles (한국 직물 모란무늬의 표현방법에 관한 연구)

  • This study examines history of Korean peony patterns and characteristics of peony patterns on the fabric. It is classified according to expression methods, and it analyzes the characteristics of 71 kinds of peony woven on 66 Korean fabrics. First, it was observed that 38 of the 66 studied fabrics (57.6%) were relics from the 17th century, and from this it can be deduced that the peony patterns began to appear regularly around the latter half of the 16th century and were used habitually in the 17th century. Second, 71 kinds on 66 fabrics can be divided into Real Type, Design Type, and Abstraction Type according to expression methods. Among these types, 49 kinds of them are Real Types (69.0%), forming the greatest part and 19 kinds belonging to the Design Type (26.8%), and lastly, 3 kinds were under the Abstraction Type (4.2%). In particular, peony patterns of Design Types and Abstraction Types from the 17th century and from the 19th century to the 20th century were more prevalent, compared with those before the 16th century or the 18th century. Third, Real Types shown on the Korean fabrics are subdivided into 9 types, and the Real Type A among them, which describes to be as real as possible, is 12 kinds, the largest number of them. Therefore, real and natural pattern of peony is the favorite type in Korea, while rather emphasized pattern of peony is the more preferred pattern in China. And also Design Types are subdivided into 6 types again; There are 6 kinds of design type A, the largest part of Design Types. Patterns of Design Type A are most similar to real peony flowers, but more simplified than the Real Type A. This result also contrasted with the trend in China, where the Design Type C, expressed petals in detail was the favored pattern. Fourth, 9 kinds of unique types are found in Korean fabrics, especially Real Type M has not been shown on Chinese fabrics. Real type M, consisting of two parts, inner and outer, where two curve lines between two parts appear as antennas of a butterfly.

Characteristics of Photosynthesis of Dwarf and Street Tree Cultivars of Hibiscus syriacus L. (분화용 및 가로수형 무궁화 품종의 광합성 특성)

  • This study was conducted to find characteristics of photosynthesis for dwarf and street tree cultivars of Hibiscus syriacus L. Characteristics of growth and flowering on some cultivars were investigated. The photosynthetic capacity for the cultivars was also measured. Bulsae showed the best height increase; Soyang represented the lowest growth. Baektanshim ${\times}$ Kyungbuk1 had the largest number of branches and leaves. Sundeok and Bulsae were the best in terms of height and width of flower, Soyang showed the smallest flowers. According to the analysis of chlorophyll content (chlorophyll a, b, total) for the cultivars, there was little difference between Sundeok and Bulsae. Based on light response curves for cultivars, it was found that Baektanshim ${\times}$ Kyungbuk1 and Soyang had lower light compensation and light saturation points than Bulsae and Sundeok. Cultivars Baektanshim ${\times}$ Kyungbuk1 and Soyang were also low in dark respiration, photosynthetic capacity, and net apparent quantum yield. In conclusion, it is recommended that cultivars Baektanshim ${\times}$ Kyungbuk1 and Soyang might be used as a dwarf type due to photosynthetic capacity.

Effects of Cutting on Nodule Development and Nitrogen Fixation in Alfalfa (예취가 알팔파 근류의 발달과 질소고정활성에 미치는 영향)

  • This experiment was conducted to evaluate the effects of cutting in field and solution culture. Periodical samplings of alfalfa in cutting and uncutting plots were taken to measure nodule development and nodule activity. Regrowth of plant and nodule development after shoot cutting by different heights and nodule removal at different levels were investigated in solution culture of alfalfa plant. 1. Nodule weight in the field was reduced 30% after the first cutting and 25% after the second cutting, but during the following 30 days after second cutting, there was no significant difference between cutting and uncutting plots. 2. Specific nodule activities of cutting plots at the beginning of June and at the beginning of September were 80% and loo%, higher than those of uncutting plots respectively. Total nodule activities of cutting plots in late August and early September were 40% higher than those of uncutting plot. The decrease of nodule activity can be prevented by cutting at flowering stage. 3. The decrease of nodules in solution culture when 50% of the shoot was cut, was as much as that when shoot was not cut or flower buds were removered. But when 90% of the shoot was cut, the number of the nodules were decreased more remarkably than the above treatments. New nodules, when 90% of the shoot was cut, were reformed slowly and did not grow fully until 15 days after cutting.

Influence of Crown Gall Infection on Growth and Flowering of Rose (뿌리혹병 감염이 장미의 생육과 개화에 미치는 영향)

  • Crown gall of rose (Rosa hybrida) was observed in greenhouse during 2003-2007. The average disease incidence was up to 38.1 % and was more severe in hydroponic culture as compared to soil culture. The typical gall symptom occurred mainly on the root, crown, or both, and resulted on poor rooting, growth retardation and yield loss. The reduction rate of rooting influenced by crown gall was 57.5% as compared to healthy plants on nursery stock. The location of gall formation in the plant influenced growth vigor resulting in symptoms such as poor shooting. Healthy plants produced 19.1 flowers/$m^2$, while diseased plants produced 9.5 flowers/$m^2$ during the same cultivation period. Moreover, the number of days to flowering was longer for the diseased plants than for healthy plants - 51.2 days and 39.8 days for first harvest, and 60.6 days and 52.1 days for the second harvest, respectively. Conclusively, infection on the basal stem caused serious loss of the number of shoot formation; yield loss of cut flower was 38.7% due to crown gall infection and delay of harvesting time about 8-10 days.
