Fabrication of Au Nanoparticle for Au-conjugate Immuno Chemistry Probe (Au-conjugate 면역화학 진단용 금 나노입자 제조)

  • Current nanogold cluster synthesized by chemical routine with 11 or 55 atoms of gold has been widely used for immuno chemistry probe as a form of nanocluster conjugated with biomolecules. It would be an undeveloped region that the 1 nm size of nanogold could be made by materials engineering processing. Therefore, objective of this study is to minimize the size of gold nanocluster as a function of operating temperature and chamber pressure in inert gas condensation (IGC) processing. Evaporation temperature was controlled by input current from 50 A to 65 A. Chamber pressure was controlled by argon gas with a range of 0.05 to 2 torr. The gold nanocluster by IGC was evaluated by X-ray diffraction (XRD) and transmission electron microscopy (TEM). The gold nanocluster for TEM analysis was directly sampled with special in-situ method during the processing. Atomic force microscopy (AFM) was used to observe 3-D nanogold layer surfaces on a slide glass for the following biomolecule conjugation step. The size of gold nanoclusters had a close relationship with the processing condition such as evaporation temperature and chamber pressure. The approximately 1 nm size of nanogold was obtained at the processing condition for 1 torr at $1124 ^{\circ}C$.

Preparation and Characterization of Thiolate-Protected Gold Nanoparticles Using Modified One-Phase Method (개선된 단일상 합성법을 이용한 티올화 나노 금의 합성 및 확인)

  • One-phase method to prevent the initial formation of Ag(I) thiolate layered materal from the mixture of $AgNO_3$ and thiols was previously developed to generate TP (Thiolate-Protected)-nanosilver. In this modified method, $AgNO_3$ is added to the mixtures of $NaBH_4$ and thiols in ethanol. This method was so successful that it was applied to synthesize TP-nanogold and nanoplatinum. The synthesis and characterization of these nanoparticles by ultraviolet-visible (UV-vis) absorption spectra, transmission electron microscopy (TEM) pictures, X-ray powder differaction (XRD) patterns and infrared(IR) spectra are described. The results show that colloidal nanoparticles are spherical or oval shape and the mean sizes for TP-nanogold and nanoplatinum are about 3~7 nm and below 2 nm, respectively. The conformation of polymethylene [$-(CH_2)_7-$] sequence in octanethiolate attached to nanogold was elucidated as trans.

Study on Ophthalmic Materials Possessing UV-Blocking/Antimicrobial Functions (자외선 차단 기능을 가진 항균성 안 의료용 소재에 관한 연구)

  • We manufactured functional contact lens materials using 2-hydroxy-4-methoxy-benzophenone, titanium(VI) isopropoxide and nanogold, nanoplatinum with UV-blocking and antimicrobial effects. Contact lens was manufactured by cast mould method. The resulting mixture was copolymerized by heating at $70^{\circ}C$ for about 40 min, at $80^{\circ}C$ for about 40 min, and finally at $100^{\circ}C$ for about 40 min. The refractive index of 1.434 ~ 1.436, water content of 35.24 ~ 36.32%, and visible transmittance of 88.3 ~ 90.8% were obtained for the contact lens materials. The polymer materials satisfied the physical properties required to make the material suitable to be applied as a functional material for ophthalmological purposes.

Microfluidic Immuno-Sensor Chip using Electrical Detection System (전기 검출 시스템을 이용한 Microfluidic Immuno-Sensor Chip)

  • This study presents the characterization of an integrated portable microfluidic electrical detection system for fast and low volume immunoassay using polystyrene microbead, which are used as immobilization surfaces. In our chip, a filtration method using the microbead was adopted for sample immobilization and immunogold silver staining(IGSS) was used to increase the electrical signal. The chip is composed of an inexpensive and biocompatible Polydimethylsiloxane(PDMS) layer and Pyrex glass substrate. Platinum microelectrodes for electric signal detection were fabricated on the substrate and microchannel and pillar-type microfilters were formed in the PDMS layer. With a fabricated chip, we reacted antigen and antibody according to the procedures. Then, silver enhancer was injected to increase the size of nanogold particles tagged with the second antibody. As a result, microbeads were connected to each other and formed an electrical bridge between microelectrodes. Resistance measured through the electrodes showed a difference of two orders of magnitude between specific and nonspecific immuno-reactions. The detection limit was 10 ng/ml. The developed immunoassay chip reduced the total analysis time from 3 hours to 50 min. Fast and low-volume biochemical analysis has been successfully achieved with the developed microfilter and immuno-sensor chip, which is integrated to the microfluidic system.

Nanogold-modified Carbon Paste Electrode for the Determination of Atenolol in Pharmaceutical Formulations and Urine by Voltammetric Methods

  • A gold nanoparticles modified carbon paste electrode (GN-CPE) has been used for the determination of atenolol (ATN) in drug formulations by cyclic voltammetry (CV), differential pulse voltammetry (DPV) and chronocoulometric methods. The results revealed that the modified electrode shows an electrocatalytic activity toward the anodic oxidation of atenolol by a marked enhancement in the current response in buffered solution at pH 10.0. The anodic peak potential shifts by -80.0 mV when compared with the potential using bare carbon paste electrde. A linear analytical curve was observed in the range of $1.96\;{\times}\;10^{-6}$ to $9.09\;{\times}\;10^{-4}\;mol\;L^{-1}$. The detection limit for this method is $7.3\;{\times}\;10^{-8}\;mol\;L^{-1}$. The method was then successfully applied to the determination of atenolol in tablets and human urine. The percent recoveries in urine ranged from 92.0 to 110.0%.

Highly catalysis Zinc MOF-loaded nanogold coupled with aptamer to assay trace carbendazim by SERS

  • Zinc metal organic framework (MOFZn)-loaded goad nanoparticles (AuNPs) sol (Au@MOFZn), which was characterized by TEM, Mapping, FTIR, XRD, and molecular spectrum, was prepared conveniently by solvothermal method. The results indicated that Au@MOFZn had a very strong catalytic effect with the nanoreaction of AuNPs formation between sodium oxalate (SO) and HAuCl4. AuNPs in the new indicator reaction had a strong resonance Rayleigh scattering (RRS) signal at 370 nm. The indicator AuNPs generated by this reaction, which had the most intense surface enhanced Raman scattering (SERS) peak at 1621 cm -1. The new SERS/RRS indicator reaction in combination with specific aptamer (Apt) to fabricate a sensitive and selective Au@MOFZn catalytic amplification-aptamer SERS/RRS assay platform for carbendazim (CBZ), with SERS/RRS linear range of 0.025-0.5 ng/mL. The detection limit was 0.02 ng/mL. Similarly, this assay platform has been also utilized to detect oxytetracycline (OTC) and profenofos (PF).
