Solid Phase Extraction of Celecoxib from Drug Matrix and Biological Fluids by Grafted Poly β-cyclodextrine/allyl Amine Magnetic Nano-particles

  • Using nanotechnology, magnetic nanoparticles of iron oxide were produced via co-precipitation method and followed modification with organic compounds. In the next step, functionalized monomer was provided via coupling ${\beta}$-cyclodextrine and allylamine onto modified magnetic nanoparticles. These nanoparticles were used to establish the adsorption rate of celecoxib. Magnetic nanoparticles are modified by (3-mercaptopropyl)trimethoxysilane. Nano-adsorbent was characterized by analytical and spectroscopic methods, such as Fourier transform infrared spectroscopy, elemental analysis, thermo-gravimetric analysis, and transmission electron microscopy (TEM). Laboratory parameters, such as the kinetics of adsorption isotherms, pH, reaction temperature and capacity were optimized. Finally, by using this nano-adsorbent in the optimized condition, extraction of celecoxib from biological samples as urine, drug matrix and blood plasma was carried out by high performance liquid chromatography with sensitivity and high accuracy.

  • Ceramic layered double hydroxide (LDH) nanohybrids have attracted considerable interest in biomedical science due to their unique structural feature and characteristics in biological condition. Many studies on LDH nanoparticles have been reported in diagnosis applications including magnetic resonance imaging (MRI) contrast agents in order to not only provide better imaging performance through multimodal imaging strategy, but realize therapeutic function which treat cancers in one platform. This review highlights the recent progress in MRI T1 contrast agent, dual modal imaging system, and MRI-guided drug delivery systems ranging from synthetic method and characterization to evaluation in vitro and in vivo based on the ceramic LDH nanohybrids. Future research directions are also suggested for next-generation bio-imaging contrast agent.

  • Phase behavior of the ternary systems of water-insoluble simvastatin drug, which is well known to be effective drugs for hypercholesterolemia therapy, in solvent mixtures of dimethyl ether (DME) and supercritical carbon dioxide was investigated to present a guideline of establishing operating conditions in the particle formation of the drugs by a supercritical anti-solvent recrystallization process utilizing DME as a solvent and carbon dioxide as an anti-solvent. The solubilities of simvastatin in the mixtures of DME and carbon dioxide were determined as functions of temperature, pressure and solvent composition by measuring the cloud points of the ternary mixtures at various conditions using a high-pressure phase equilibrium apparatus equipped with a variable-volume view cell. The solubility of the drug increased as the DME composition in solution and the system pressure increases at a fixed temperature. A lower solubility of the drug was obtained at a higher temperature.

3D Bioprinted GelMA/PEGDA Hybrid Scaffold for Establishing an In Vitro Model of Melanoma

  • Due to the high incidence of malignant melanoma, the establishment of in vitro models that recapitulate the tumor microenvironment is of great biological and clinical importance for tumor treatment and drug research. In this study, 3D printing technology was used to prepare GelMA/PEGDA composite scaffolds that mimic the microenvironment of human malignant melanoma cell (A375) growth and construct in vitro melanoma micro-models. The GelMA/PEGDA hybrid scaffold was tested by the mechanical property, cell live/dead assay, cell proliferation assay, cytoskeleton staining and drug loading assay. The growth of tumor cells in two- and three-dimensional culture systems and the anti-cancer effect of luteolin were evaluated using the live/dead staining method and the Cell Counting Kit-8 (CCK-8) method. The results showed a high aggregation of tumor cells on the 3D scaffold, which was suitable for long-term culture. Cytoskeleton staining and immunofluorescent protein staining were used to evaluate the degree of differentiation of tumor cells under 2D and 3D culture systems. The results indicated that 3D bioprinted scaffolds were more suitable for tumor cell expansion and differentiation, and the tumor cells were more aggressive. In addition, luteolin was time- and dose-dependent on tumor cells, and tumor cells in the 3D culture system were more resistant to the drug.

  • Objective : This research aims to investigates the effects of CMT and HCMT on Dementia. and we also want to know the different effect of CMT, HCMT by drug types. Methods : The research is progressed by two types of experiment. one experiment is BV2 microglial cell line treated by LPS in vitro and another experiment is memory deficit mice induced by scopolamine in vivo. Results : The CMT and HCMT is effective in BV2 microglial cell line treated by LPS in vitro and in the serum of the memory deficit mice induced by scopolamine in vivo. But, there is no significant difference between CMT and HCMT extract&nano powder in experimental conclusion. Conclusions : These results suggest that the two drug types of CMT and HCMT may be effective for the prevention and treatment of Dementia. Investigation into the further study two drug types of the CMT and HCMT for Dementia is suggested for future research.

  • While there have been great demands on improving domestic water pollution issues, the necessity for real time monitoring of particular drug residues in water resources has been raised since drug residues including antibiotics could provoke new trains of drug-resistant bacteria in water environments. Among many different types of drugs used for pharmaceutical treatment, antibiotics are considered to be one of the most hazardous to our ecosystem since they can rapidly promote the spreading of drug-resistant bacteria in water environments. In this mini-review, we will highlight recent developments made on creating in-situ sensing platforms for the fast monitoring of antibiotic residues in aquatic environmental samples focusing on optical and electrochemical techniques. Related recent technology developments and the resulting economy effects will also be discussed.

  • Colloid and surface chemistry have been focused on surface area and surface energy. Local surface properties such as surface density, interaction, molecular orientation and reactivity have been one of interesting subjects. Systems of such surface energy being important would be listed as association colloid, emulsion, particle dispersion, foam, and 2-D surface and film. Such nanoparticle systems would be applied to drug delivery systems and functional cosmetics with biocompatible and degradable materials, while nanoparticles having its size of several nm to micron, and wide surface area, have been accepted as a possible drug carrier because their preparation, characteristics and drug loading have been inves-tigated. The biocompatible carriers were also used for the solubilization of insoluble drugs, the enhancement of skin absorption, the block out of UV radiation, the chemical stabilization and controlled release. Nano/micro emulstion system is classified into nano/microsphere, nano/microcapsule, nano/microemulsion, polymeric micelle, liposome according to its prep-aration method and size. Specially, the preparation method and industrial applications have been introduced for polymeric micelles self-assembled in aqueous solution, nano/microapsules controlling the concentration and activity of high concen-tration and activity materials, and monolayer or multilayer liposomes carrying bioactive ingredients.

  • Zaltoprofen, a propionic acid derivative non-steroidal anti-inflammatory drug, was known to have powerful inhibitory effects on acute, subacute and chronic inflammation. For initial release and sustained release, the microspheres were prepared using an emulsion-solvent evaporation method like an O/W emulsion method with varying the ratio of zaltoprofen-loaded polyoxalate (POX)/PLGA micropheres. The morphology of the microspheres was confirmed by scanning electron microscopy. The crystallinity of microspheres was analyzed by X-ray diffraction and differential scanning calorimeter. Fourier transform infrared spectroscopy was used to analyze the chemical structure of microspheres. The increased ratio of POX microspheres affected the initial drug release, and the sustained release of drug was influenced by ratio of PLGA microspheres. In this study, the initial release behavior of zaltoprofen can be controlled by the ratio of POX/PLGA microspheres.

  • Studies on drug delivery using nano-size particles of magnetic fluid and hyperthermia have been performed by some researchers [1] because interests in human health increased according to industry development. However, there are few studies on systems which can accurately control delivery of the magnetic fluids to a diseased part of body [2]. In this study, Cu-added magnetic ferrofluid was prepared and the external magnetic field system was designed for drug localization.
