Although Anisakis type larvae have been shown to cause various injuries directly or indirectly in humans and animals, the epidemiological studies on these larval infestations are in insufficient state. The status of larval infestation was investigated in 989 fishes of 44 species, which are inhabiting around the east-westernsouth costal area of Korea during the period from March 2007 to February 2008. The Anisakis type larvae were infected approximately 38% (377 fishes) in 989 fishes. Most of the worms were identified as Anisakis simplex type I by morphological finding and 18S ribosomal DNA sequence analysis. In the seasonal variations of infestation, most of the fishes showed higher infestation rate during spring and summer, while the fishes such as herring Clupea pallasii did during winter. From the histopathological studies of infested fishes, it has been observed that Anisakis type larvae are harbouring mainly around the intestinal viscera such as liver, pancreas, stomach, pylolic cecum, and cloaca.

Numerical design optimization techniques are implemented for the improvement of the ram accelerator performance. The design object is to find the minimum ram tube length required to accelerate projectile from initial velocity $V_o$ to target velocity $V_e$. The premixture is composed of $H_2$, $O_2$, $N_2$ and the mole numbers of these species are selected as design variables. The objective function and the constraints are linearized during the optimization process and gradient-based Simplex method and SLP(Sequential Linear Programming) have been employed. With the assumption of two dimensional inviscid flow for internal flow field, the analyses of the nonequilibrium chemical reactions for 8 steps 7 species have been performed. To determined the tube length, ram tube internal flow field is assumed to be in a quasi-steady state and the flow velocity is divided into several subregions with equal interval. Hence the thrust coefficients and accelerations for corresponding subregions are obtained and integrated for the whole velocity region. With the proposed design optimization techniques, the total ram tube length had been reduced 19% within 7 design iterations. This optimization procedure can be directly applied to the multi-stage, multi-premixture ram accelerator design optimization problems.
The infection status of anisakid larvae was examined in 290 marine fish of 25 species and in 108 cephalopods of 3 species purchased in Bayuquan region, Yingko city nearby the coast of the Bohai Sea from may to August 1992. A total of 7,327 laivae were collected from 156 fish of 19 species and 8 squids of one species. The 3rd-stage larvae of Anisokis simplex were collected from 121 fish (63..4%) of 15 species (N : 191) and from 8 squids (14.8%) of one species (N : %4), and they were total, 5,992 (81.8%). Out of remaining 1,335 larvae, 154 (2.1%) were classified as Rvnnqscnris type B from 23 fish of 4 species, 1,013 (13.8%) as avnnosccris type C from 79 fish of 13 species, 164 (2.2%) as Hvsterothylacium China type V from 20 fish of 4 species, 3 (0.04%) as Raphinoscnris from 3 fish of 2 species and one was Pseunotewonovc necipiens larva. When endoplasmic reticulum (ER) functions are perturbed, the ER induces several signaling pathways called unfolded protein response to reestablish ER homeostasis through three ER transmembrane proteins: inositol-requiring enzyme 1 (IRE1), PKR-like ER kinase (PERK), and activating transcription factor 6 (ATF6). Although it is important to measure the activity of ATF6 that can indicate the status of the ER, no specific cell-based reporter assay is currently available. Here, we report a new cell-based method for monitoring ER stress based on the cleavage of $ATF6{\alpha}$ by sequential actions of proteases at the Golgi apparatus during ER stress. A new expressing vector was constructed by using fusion gene of GAL4 DNA binding domain (GAL4DBD) and activation domain derived from herpes simplex virus VP16 protein (VP16AD) followed by a human $ATF6{\alpha}$ N-terminal deletion variant. During ER stress, the GAL4DBD-VP16AD(GV)-$hATF6{\alpha}$ deletion variant was cleaved to liberate active transcription activator encompassing GV-$hATF6{\alpha}$ fragment which could translocate into the nucleus. The translocated GV-$hATF6{\alpha}$ fragment strongly induced the expression of firefly luciferase in HeLa Luciferase Reporter cell line containing a stably integrated 5X GAL4 site-luciferase gene. The established double stable reporter cell line HLR-GV-$hATF6{\alpha}$(333) represents an innovative tool to investigate regulated intramembrane proteolysis of $ATF6{\alpha}$. It can substitute active pATF6(N) binding motif-based reporter cell lines.

In radiotherapy treatment planning, it is critical to deliver the radiation dose to tumor and protect surrounding normal tissue. Recent developments in functional imaging and radiotherapy treatment technology have been raising chances to control tumor saving normal tissues. A brain phantom which could be used for image registration technique of CT-MR and CT-SPECT images using surface matching was developed. The brain phantom was specially designed to obtain imaging dataset of CT, MR, and SPECT. The phantom had an external frame with 4 N-shaped pipes filled with acryl rods, Pb rods for CT, MR, and SPECT imaging, respectively. 8 acrylic pipes were inserted into the empty space of the brain phantom to be imaged for geometric evaluation of the matching. For an optimization algorithm of image registration, we used Downhill simplex algorithm suggested as a fast surface matching algorithm. Accuracy of image fusion was assessed by the comparison between the center points of the section of N-shaped bars in the external frame and the inserted pipes of the phantom and minimized cost functions of the optimization algorithm. Technique with partially transparent, mixed images using color on gray was used for visual assessment of the image registration process. The errors of image registration of CT-MR and CT-SPECT were within 2mm and 4mm, respectively. Since these errors were considered within a reasonable margin from the phantom study, the phantom is expected to be used for conventional image registration between multimodal image datasets..
Purpose: Hydrodynamic-based procedure is a simple and effective gene delivery method to lead a high gene expression in liver tissue. Non-invasive imaging reporter gene system has been used widely with herpes simplex virus type 1 thymidine kinase (HSV1-tk) and its various substrates. In the present study, we investigated to image the expression of HSV1-tk gene with 5-(2-iodovinyD-2'-deoxyuridine (IVDU) in mouse liver by the hydrodynamicbased procedure. Materials and Methods: HSV1-tk or enhanced green fluorescence protein (EGFP) encoded plasmid DNA was transferred into the mouse liver by hydrodynaminc injection. At 24 h post-injection, RT-PCR, biodistribution, fluorescence imaging, nuclear imaging and digital wholebody autoradiography (DWBA) were performed to confirm transferred gene expression. Results: In RT-PCR assay using mRNA from the mouse liver, specific bands of HSV1-tk and EGFP gene were observed in HSV1-tk and EGFP expressing plasmid injected mouse, respectively. Higher uptake of radiolabeled IVDU was exhibited in liver of HSV1-tk gene transferred mouse by biodistribution study. In fluorescence imaging, the liver showed specific fluorescence signal in EGFP gene transferred mouse. Gamma-camera image and DWBA results showed that radiolabeled IVDU was accumulated in the liver of HSV1-tk gene transferred mouse. Conclusion: In this study, hydrodynamic-based procedure was effective in liver-specific gene delivery and it could be quantified with molecular imaging methods. Therefore, co-expression of HSV1-tk reporter gene and target gene by hydrodynamic-based procedure is expected to be a useful method for the evaluation of the target gene expression level with radiolabeled IVDU.

The purpose in this study has to development the system on increasing operation of old-typed transceiver for solve the question that limited allocation frequencies and continuesly icreasment of traffic. Therefore, we are desigened the remote control system that has the function for variable channels, PTT and monitoring of transmission power and frequencies. Exchange of control data is to hold in common the twist two-wire or telephone line for the voice transmission. The H/W is consist of FSK and MCS-51 processor which are up-down control of channel, n control and monitoring display by serial data transmission. According to the simplex traffic operation is designed the algorithm of serial data transmission by sequential transmission sequence and protocol. The S/W of sequential transmission sequence is designed to usefully the intergrated communications system which is able to connection between the multi-transceiver and multi-terminal by master processor.
Background: The nematode species belonging to genus Anisakis occur at their third larval stage in numerous marine teleost fish species worldwide and known to cause accidental human infection through the ingestion of raw or undercooked fish or squids. They may also draw the attention of consumers because of the visual impact of both alive and dead worms. Therefore, the information on their geographical distribution and clear species identification is important for epidemiological survey and further prevention of human infection. Results: For identification of anisakid nematodes species isolated from largehead hairtail (Trichiurus japonicus), polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of internal transcribed spacers of ribosomal DNA were conducted. Mitochondrial cytochrome c oxidase subunit 2 gene was also sequenced, and phylogenetic analysis was conducted. From the largehead hairtail (n = 9), 1259 nematodes were isolated in total. Most of the nematodes were found encapsulated throughout the viscera (56.2 %, 708/1259) or moving freely in the body cavity (41.5 %, 523/1259), and only 0.3 % (4/1259) was found in the muscles. By PCR-RFLP, three different nematode species were identified. Anisakis pegreffii was the most dominantly found (98.7 %, 1243/1259) from the largehead hairtail, occupying 98.7 % (699/708) of the nematodes in the mesenteries and 98.1 % (513/523) in the body cavity. Hybrid genotype (Anisakis simplex ${\times}$ A. pegreffii) occupied 0.5 %, and Hysterothylacium sp. occupied 0.2 % of the nematodes isolated in this study. Conclusions: The largehead hairtail may not significantly contribute accidental human infection of anisakid nematode third stage larvae because most of the nematodes were found from the viscera or body cavity, which are not consumed raw. But, a high prevalence of anisakid nematode larvae in the largehead hairtail is still in concern because they may raise food safety problems to consumers. Immediate evisceration or freezing of fish after catch will be necessary before consumption.
Gene therapy with nonviral vectors using the suicide gene/prodrug activating system of herpes simplex virus type-1 thymidine kinase (HSV1-TK)/ganciclovir (GCV) is inefficient in killing malignant tumor cells due to two major factors: (a) an unsatisfactory bystander effect; (b) short-lived expression of the protein. To study the capacity of the protein transduction domain (PTD) of HIV-1 TAT protein to enhance HSV1-TK/GCV cancer gene therapy, we constructed three fusion proteins TAT-TK, TK-TAT and TK. TAT-TK retained as much enzyme activity as TK, whereas that of TK-TAT was much lower. TAT-TK can enter HepG2 cells and much of it is translocated to the nucleus. The transduced HepG2 cells are killed by exogenously added GCV and have bystander effects on untransduced HepG2 cells. Most importantly, the introduced recombinant protein is stable and remains functional for several days at least, probably because nuclear localization protects it from the cytoplasmic degradation machinery and provides access to the nuclear transcription machinery. Our results indicate that TAT fusion proteins traffic intercellularly and have enhanced stability and prodrug cell killing activity. We conclude that TAT has potential for enhancing enzyme prodrug treatment of liver cancers.

Using the design of experiment (DOE), factors affecting the particle size of hydrated liquid crystalline vesicles containing a high content of ceramide were analyzed and the mixture composition was optimized. Manufacturing temperature, amount of ethanol, and ultrasonic time were selected as the main variables affecting the droplet size of the vesicles, and the effect of these variables on the droplet size was examined through the signal to noise (S/N) ratios of Taguchi method and ANOVA analysis. In addition, mixture composition experiments of three lipid components constituting the vesicle membrane, hydrogenated phosphatidyl choline (HPC), cholesterol (Chol), and ceramide (Cer), were performed according to the simplex central design matrix of the mixture. Regression analysis was conducted with the experimental data to obtain a model equation, and the optimal mixing composition of the three lipid components to minimize the vesicle droplet size was determined as HPC (0.6), Chol (0.1), and Cer (0.3).
