Effect of n-6/n-3 fatty acid ratio on metabolic partitioning in hyperlipidemic rats (n-6/n-3 지방산 비율이 고지혈증 랫드의 지질대사 분할에 미치는 영향)

  • This study was investigated the mechanism of action of n-6/n-3 fatty acid ratio on the metabolic partitioning of blood glycerolipids by in vivo monitoring technique in hyperlipidemic animal model rats. The ratio of cholesteryl 14C-oleate metabolized in the liver of total glycerolipids was lower in the order of n-6/n-3 ratios of 4:1, 15:1, 30:1 and control group (p<0.05). The secretion amount of phospholipid was higher in the order of n-6/n-3 ratio 4:1, 15:1, 30:1 than the control (p<0.05). The secretion amount of triglyceride was lower in especially 4:1, in order of n-6/n-3 4:1, 15:1 and 30:1 compared with the control. The ratio of phospholipid partitioning to total glycerolipid was high in orfer of n-6/n-3 ratio 4:1, 15:1, 30:1 and control (p<0.05). The triacylglycerol partitioning (%) via liver was higher 72.97, 75.93, 78.12% in n-6/n-3 4;1, 15:1, 30:1, respectively than the control of 82.25%, according to increased n-6/n-3 (p<0.05). The phospholipid partitioning (%) was lower 25.15, 18.87, 18.15% in n-6/n-3 4;1, 15:1, 30:1, respectively, compared to control 11.04%, according to increased n-6/n-3 (p<0.05).

Fatty Acid Composition in Blood Plasma and Follicular Liquid in Cows Supplemented with Linseed or Canola Grains

  • This study was carried out to evaluate the fatty acid composition in Nellore cows supplemented with either linseed (n-3) or canola grains (n-6 and n-9). Fifteen Nellore cows, aged five years and bodyweight 550 kg${\pm}$48 kg, were randomly distributed to the following treatments: CON (control), LIN (linseed) and CAN (canola grains). The cows were fed for 80 days. The concentrations of C18:0, C18:2 n-6 and C20:3 n-6 fatty acid were higher (p<0.10) in CON blood plasma in comparison to follicular liquid. Likewise, PUFA, n-6 contents, PUFA:SFA and n-6:n-3 ratios were higher (p<0.10) in blood plasma. On the other hand, C18:1 n-9, C22:5 n-3, MUFA and n-3 contents were lower (p<0.10) in blood plasma. C18:0, C18:2 n-6, C18:3 n-3, C22:5 n-3, PUFA, n-6, n-3 contents and PUFA:SFA ratio were higher (p<0.10) in LIN blood plasma than in the follicular liquid. Nevertheless, C14:0, C16:0, C16:1 n-7, PUFA, C16:0, C18:1 n-9 and MUFA contents were lower (p<0.10) in LIN blood plasma. On treatment CAN, the C18:0 and SFA contents, and n-6:n-3 ratios were higher (p<0.10) in blood plasma. However, C20:3 n-6, C22:5 n-3, PUFA and n-3 contents were lower (p<0.10) in blood plasma. C16:0, C18:0, PUFA, SFA contents and PUFA:SFA ratio did not differ (p>0.10) among the treatments. C14:0, C16:1 n-7, C18:2 n-6 and n-6 contents were higher (p<0.10) for CON and CAN than LIN. C17:1 n-7, C20:4 n-6 and C 22:0 contents were higher (p<0.10) for CAN than CON and LIN. C18:1 n-9, C18:3 n-3, MUFA and n-3 contents were higher (p<0.10) for LIN and CAN than CON. C20:3 n-6 content and n-6:n-3 ratio were higher (p<0.10) for CON than LIN and CAN. C22:5 n-3 content were higher (p<0.10) for CON and LIN than CAN. The concentrations of fatty acids in blood plasma and follicular liquid were not correlated for any fatty acid, independent of the treatment studied. Canola grain added to the diet of Nellore cows resulted in increased concentrations of fatty acids n-6 and n-3 in follicular liquid.

Effect of n-3 Fatty Acid Deficiency on Fatty Acid Composition in Brain, Retina and Liver Using a Novel Artificial Rearing System (인공 사육 동물 모델 시스템을 이용한 n-3 지방산 결핍이 쥐의 뇌, 망막, 간의 지방산 조성에 미치는 영향)

  • Docosahexaenoic acid (22:6n-3, DHA) is highly enriched in membrane of brain and retina, and plays an important role in maintaining an optimal function of the central nervous system. We investigated the effect of n-3 fatty acid deficiency on rat brain, retina and liver fatty acyl composition at two different ages (3 wks and 15 wks) under DHA deficient condition. Rat pups born to dams fed a diet with $3.1\%$ of total fatty acids as $\alpha-linolenic$ acid (LNA) were fed using an artificial rearing system either an n-3 deficient (n-3 Def) or n-3 adequate (n-3 Adq) diet. Both diets contained $17.1\%$ linoleic acid (LA) but the n-3 Adq diet also contained $3.1\%$ LNA. Rats consuming the n-3 Def diet showed a lower brain $(50\%\;in\;13\;wks\;and\;70\%\;in\;15\;wks,\;p<0.05)$ and retinal $(50\%\;in\;13\;wks\;and\;63\%\;in\;15\;wks,\;p<0.05)$ DHA than those on the n-3 Adq diet, which was largely compensated for by an increase in docosapentaenoic acid (22:5n-6, DPAn-6). In the liver of the n-3 Def group, the percentage of DHA decreased by $97\%$ at 3 wks of age with an apparent increase in DPAn-6 relative to the n-3 Adq group (p<0.05), while there was a $65\%$ lower liver DHA in n-3 Def group at 15 wks of age than the n-3 Adq group (p<0.05). Liver arachidonic acid (20:4n-6, AA) was increased at 3 wks of age but decreased at 15 wks of age in the n-3 Def group compared with n-3 Adq group (p<0.05). In conclusion, the replacement of DHA by DPAn-6 in brain and retina fatty acid composition may be related to the suboptimal function in spatial learning, memory and visual acuity. This artificial rearing method presents a first generation model for n-3 deficiency that is similar to the case of human nutrition that commonly employed two generation model.

Effect of Different Dietary n-6 to n-3 Fatty Acid Ratios on the Performance and Fatty Acid Composition in Muscles of Broiler Chickens

  • The objective of this study was to investigate the different dietary ratios of n-6 to n-3 (n-6/n-3) fatty acid (FA) on performance and n-6/n-3 FA in muscles of broiler chickens. A total of 300 one-day-old Cobb chicks were randomly assigned to 3 treatments of 10 replicates in each (10 birds/replicate). Birds were fed on a corn-soybean meal-based diet containing 1% oil during starter (day 1 to 21) and 2% oil during finisher (day 22 to 39) phases, respectively. Treatments of high, medium and low dietary n-6/n-3 FA were formulated by replacing rice bran oil with linseed oil to achieve n-6/n-3 FA close to >20:1, 10:1 and 5:1, respectively. Average daily gain, average daily feed intake, and feed conversion ratio were similar (p>0.05) among the treatments. Serum glucose, cholesterol and triglycerides concentrations were not affected (p>0.05) by dietary treatments. In breast, concentration of C18:3n-3 was significantly greater (p = 0.001) for medium and low vs high n-6/n-3 FA, while concentrations of C20:5n-3, C22:6n-3, total n-3 FA, and n-6/n-3 FA were significantly higher for low vs medium, and medium vs high dietary n-6/n-3 FA. In contrast, concentrations of C18:2 and mono-unsaturated FA (MUFA) were lower for low vs high dietary n-6/n-3 FA. In thigh muscles, concentrations of C20:5n-3 were higher (p<0.05) for medium and low vs high dietary n-6/n-3 FA, and concentrations of C18:3n-3, C22:6, and n-3 FA were greater (p<0.05) for medium vs high, low vs medium dietary n-6/n-3 FA. However, concentrations of C18:1, MUFA, n-6/n-3 were lower (p<0.05) for low and medium vs high dietary n-6/n-3 FA. In conclusion, lowering the dietary n-6/n-3 FA did not affect the performance of chickens, but enhanced beneficial long-chain n-3 FA and decreased n-6/n-3 FA in chicken breast and thigh, which could be advantageous for obtaining healthy chicken products.

  • p·Si-전해질 접합을 전해질로 6N H2SO4, 6N H2SO4(Ti3+), 6N H2SO4(Ti4+), 6N H2SO4(Ti4+/Ti3+)을 사용하여 만들었다. 이들 전해질중 6N H2SO4(Ti4-/Ti3+)을 사용할 때 p·Si 광음극이 안정하게 동학하며 높은 광전 감도를 가지고 있었다. p·Si-electrolyte junction are prepared by using p·Si photocatode in four different electrolytes such as 6N H2SO4, 6N H2SO4(Ti3+), 6N H2SO4(Ti4+), 6N H2SO4(Ti4+/Ti3+) respectively. Among those electrolytes 6N H2SO4(Ti4-/Ti3+) shows very good results, in which p·Si photocathode is stable.

Effects of Dietary n-3/n-6 Fatty Acid Ratio on In Vitro Fermentation Characteristics and Fatty Acid Profiles

  • This study was conducted to examine the effect of dietary n-3/n-6 fatty acid (FA) ratio on in vitro dry matter digestibility (IVDMD), fermentation indices and FA profile. Rice bran was mixed with oil sources (cotton seed oil and linseed oil) to make the diets at 0.02, 0.29 and 0.61 of dietary n-3/n-6 FA ratio. These diets (0.5g) were placed into the incubation bottles with 40 ml of anaerobic culture medium, which contained rumen fluid and Van Soest medium at 1:2 ratio. Five replicates of each diet and two blanks were incubated at $39^{\circ}C$ for 48 hours. After incubation, the incubated contents were centrifuged. The residues were freeze-dried for DMD and FA analyses. The supernatant was used for pH, $NH_3-N$ and volatile fatty acid analyses. The concentrations of lactate (p<0.001) and iso-valerate (p<0.001) decreased linearly with increasing dietary n-3/n-6 FA ratio, but acetate concentration (p=0.056) and the ratio of acetate to propionate (p=0.005) was increased linearly. The concentrations of n-3, n-6 FA and the ratio of n-3/n-6 FA in residues increased (p<0.001) linearly with increasing dietary n-3/n-6 FA ratio, but C18:1n-9 FA concentration was decreased (p<0.001) linearly. With these results, it could affect fermentation characteristics and FA profile of rumen content by dietary n-3/n-6 FA ratio.

Effects of Diet and Time on Feed on Fatty Acid Composition in Muscle of Charolais Steers (사료급원과 급여기간이 Charolais 거세우 근내 지방산 조성에 미치는 영향)

  • This study investigated the effects of feeding Charolais steers on diets rich in either n-6 or n-3 polyunsaturated fatty acids (PUFA) and time on feed (TOF) on muscle fatty acid composition and content. Twenty eight steers were fed on ad libitum forage and one of two concentrates varying in the source of fat; soya (high in C18:2 n-6) or whole linseed (high in C18:3 n-3) for either 60 or 90 days in IGER (Institute of Grassland and Environmental Research, UK). The concentrates were fed at approximately 0.73 of total DM intake. TOF influenced carcass weight, conformation and fatness scores, which were higher at 90 v. 60 days (P<0.05). Diet did not affect total fatty acid content of neutral lipid in m. longissimus thoracis but feeding linseed increased total phospholipid fatty acid by approx- imately 15%(P<0.05). Linseed increased the amount and proportion of C18:3 n-3 (P<0.001) and the proportion of CLA (cis-9, trans-11 conjugated linoleic acid), while soya increased the content (P<0.05) and proportion (P<0.001) of C18:2 n-6 in muscle neutral lipid. In muscle phospholipid, linseed significantly increased the amount of CLA, C18:3 n-3 and its longer chain derivatives as well as C14:0, C16:0, C18:0. C18:1 trans and C18:2 n-6. The amount and proportion of C18:2 n-6 and its longer chain C20 derivatives were higher on feeding soya. TOF (90 v. 60 day) increased the content of C14:0, C16:0, C16:1, CLA, C18:1 n-9, C18:2 n-6 and C18:3 n-3 in muscle neutral lipid. The P:S was not affected by diet or TOF. The ratio of C18:2 n-6 : C18:3 n-3 and sum of n-6 : n-3 fatty acids were higher in muscle from animals fed on linseed v. soya (P<0.001). The study indicates that the PUFA composition of beef muscle may be significantly modified by feeding contrasting dietary lipids, soya vs. linseed. Feeding linseed produced a better balance of muscle fatty acids, more in line with current nutritional recommendations with a lower C18:2 n-6:C18:3 n-3 ratio associated with higher muscle content of C18:3 n-3 and C20:5 n-3 and CLA and lower C20:4 n-6.

  • Two new nickel vanadium borophosphate cluster compounds, $(NH_4)_{10}[Ni(H_2O)_5]_4[V_2P_2BO_{12}]_6{\cdot}nH_2O$ (1) and $(NH_4)_{3.5}(C_3H_{12}N_2)_{3.5}[Ni(H_2O)_6]_{1.25}{[Ni(H_2O)_5]_2[V_2P_2BO_{12}]_6{\cdot}nH_2O$ (2) have been synthesized and structurally characterized. Inter-diffusion methods were employed to prepare the compounds. The cluster anion $[(NH_4)\;{\supset}\;V_2P_2BO_{12}]_6$ is used as a building unit in the synthesis of new compounds containing $Ni(H_2O){^{2+}_5}$ in the presence of pyrazine and 1,3-diaminopropane. Compounds contain isolated cluster anions with general composition ${[Ni(H_2O)_5]_n[(NH_4)\;{\supset}\;V_2P_2BO_{12}]_6}^{-(17-2n)}$ (n = 2, 4). Effects of dietary n-6:n-3 polyunsaturated fatty acid ratio on growth performance, blood indexes, tissue fatty acid composition and the expression of peroxisome proliferator-activated receptor gamma signaling related genes in finishing pigs

  • Objective: This study investigated the effects of dietary n-6:n-3 polyunsaturated fatty acid (PUFA) ratio on growth performance, blood indexes, tissue fatty acid composition and the gene expression in finishing pigs. Methods: Seventy-two crossbred ([Duroc×Landrace]×Yorkshire) barrows (68.5±1.8 kg) were fed one of four isoenergetic and isonitrogenous diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1, and 8:1. Results: Average daily gain, average daily feed intake and gain-to-feed ratio had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The concentrations of serum triglyceride, total cholesterol and interleukin 6 were linearly increased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio, while that of high-density lipoprotein cholesterol tended to decrease (p = 0.062), and high-density lipoprotein cholesterol:low-density lipoprotein cholesterol ratio and leptin concentration were linearly decreased (p<0.05). The concentration of serum adiponectin had a quadratic response but the measurement was decreased and then increased (quadratic, p<0.05). The proportion of C18:3n-3 was linearly decreased (p<0.05) in the longissimus thoracis (LT) and subcutaneous adipose tissue (SCAT) as dietary n-6:n-3 PUFA ratio increasing, while the proportion of C18:2n-6 and n-6:n-3 PUFA ratio were linearly increased (p<0.05). In addition, the expression levels of peroxisome proliferator-activated receptor gamma (PPARγ) and lipoprotein lipase in the LT and SCAT, and adipocyte fatty acid binding protein and hormone-sensitive lipase (HSL) in the SCAT had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The expression of HSL in the LT was linearly decreased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio. Conclusion: Dietary n-6:n-3 PUFA ratio could regulate lipid and fatty acid metabolism in blood and tissue. Reducing dietary n-6:n-3 PUFA ratio (3:1) could appropriately suppress expression of related genes in PPARγ signaling, and result in improved growth performance and n-3 PUFA deposition in muscle and adipose tissue in finishing pigs.

The Age-Related Effect of n-6/n-3 Ratio of Dietary Fats on Lipid Levels and Prostaglandin Production in Rats (식이 다가 불포화 지방산의 n-6/n-3 비율에 의한 흰쥐의 지질수준 및 Prostaglandin 생성에 대한 연령별 비교)

  • The effects of various n-6/n-3 ratios(about 2, 4, 6, 8) of dietary fatty acids on various lipid levels and prostaglandin production were studied at the constant P/S ratio (1.5-1.6) in young (5 weeks old) and adult(8 months old) Sprague-Dawley rats using palm oil, safflower oil and sardine oil. The concentration of serum cholesterol tended to increase with the increasing n-6/n-3 ratio. The tendency of HDL-cholesterol levels was similar to serum cholesterol levels. These were not apparent differences between young and adults rats. Serum triglyceride levels increased according to increasing n-6/n-3 ratio in young rats. These were generally high in the adult rats compared with the young rats. Though liver cholesterol level tended to increase according to the increasing n-6/n-3 ratio in the young rats. The liver triglyceride level did not change according to the n-6/n-3 ratio. However, these levels were apparently higher in the adult than in the young rats. The fatty acid compositions of phosphatidylcholine(PC) were similar in serum and liver. The arachidonate/linoleate ratios in tissue PC were influenced by the n-6/n-3 ratio. They tended to be lower in the adult rats compared with the young rats. It was suggested that the activity of $\Delta$6-desturase was decreased by aging. Production of platelet thromboxane A2(TXA2)and aortic prostacyclin(PGI2) was not apparently influenced with n-6/n-3 ratio. Whereas the ratio of TXA2/PGI2 was the lowest value at 3.8 of n-6/n-3 ratio, expecially in the young rats. Thus this ratio seemed to be a desirable level to protect atherosclerosis. These results indicate that the lipid level and prostaglandin production were influenced not only by n-6/n-3 ratio(under constant P/S ratio) but by aging, particulary triglycerde level and arachidonic/linoleic acid ratio.
