Myosin Heavy Chain Covalenily Modified at Its Reactive Site Sulfhydryl Residues is Preferentially Degraded by Calpain (활성화 부위의 황화기가 화학적으로 변형된 Myosin Heavy Chain의 Calpain에 의한 선택적 분해)

  • N-ethylmaleimide at low concentrations is known to interact specifically with 2 sulfhydryl residues in myosin heavy chain. Calpain, a Ca$^2$+-dependent neutral protease isolated from chick skeletal muscle, was found to preferentially degrade the alkylated protein but much less significantly the unmodified protein. Exposure of myosin to KMnO$_4$, which is also known to interact with sulthydryl groups, also caused the rapid degradation of the myosin heavy chain. Furthermore, treatment of each agent with increasing concentrations results in a greater loss of the myosin ATPase activity, indicating that the modification occurred at the reactive site sulfhydryl residues. These results suggest that the covalent modification at the reactive site salfhydryl residues in the myosin heavy chain may mark the protein for degradation by intracellular proteases such as calpain.

Identification of Amino Acid Residues Involved in Xylanase Activity from Bacillus Pumilus TX703 (Bacillus Pumilus TX703 유래 Xylanase의 활성에 관여하는 아미노산 잔기의 확인)

  • The purified xylanase from Bacillus pumilus TX703 was modified with various chemical modifiers to determine the active sites of the enzyme. Treatment of the enzyme with group-specific reagents such as carbodiimide or N-bromosuccinimide resulted in complete loss of enzyme activity. These results assumed that these reagents reacted with glutamic acid or aspartic acid and tryptophan residues located at or near the active site. In each case, inactivation was performed by pseudo first-order kinetics. Inhibition of enzyme activity by carbodiimide and W-bromosuccinimide showed non-competitive and competitive inhibition type, respectively. Addition of xylan to the enzyme solution containing N-bromosuccinimide prevented the inactivation, indicating the presence of tryptophan at the substrate binding site. Analysis of kinetics for inactivation showed that the loss of enzyme activity was due to modification of two glutamic acid or aspartic acid residues and single tryptophan residue.

Ecological Studies on the Montane Grassland of Mt. Soback in Korea (소백산 산지초원의 생태학적 연구 II. 물질생산과 염류순환)

  • Comparative study of the biomass productioin and the cyclings of nitrogen, phosphorus and potassium was carried out on the east and the northwest facing slope in the montane grassland of Mt. Soback. The maximum productin during the growing season in the east and the northwest slope were 1, 150g/m2.yr, and 755g/m2.yr. in the August, respectively. The positive correlation was appearent between biomass production and A-layer depth of the soil and the correlation coefficient (r=.964) was very significant at 1% level. In the above ground materials, the nutrients contents were high at the early of the growing season but decreased gradually. In roots, however, there are no significant trend throughout the growing season. Total amounts of minerals uptaken by plants during the growing season in the east and the northwest site were 18.20 and 10.66g/m2.yr for N, 0.06 and 0.03g/m2.yr for P, 0.23 and 0.11g/m2.yr for K, respectively. the minerals returned to the soil by litter decomposition and roots decay in the east and the northwest site were 16.19 and 9.62g/m2.yr for N, 0.046 and 0.021g/m2.yr for P, 0.211 and 0.099g/m2.yr for K, respectively. The turnover the rate (absorbed/returned) of the nutrients in the east and the northwest site were 1.13 and 1.14 for N, 1.30 and 1.43 for P, 1.09 and 1.11 for K, and the absorption rates of minerals were 0.39 and 0.29% for N, 3.16 and 1.88% for P, 0.91 and 0.57% for K, respectively.

  • Role of cl$^{[-10]}$ in photosynthetic oxygen-evolving complex was studied by light-induced Fourier transform infrared (FTIR) spectroscopy. cl$^{[-10]}$ depletion resulted in the suppression of amide I and amide II IR modes upon S$_1$ to S$_2$ transition. Br$^{[-10]}$ , 1$^{[-10]}$ and N0$_3$$^{[-10]}$ substituted FTIR difference spectra were very similar to that in cl$^{[-10]}$ reconstitution. F$^{[-10]}$ and $CH_3$COO$^{[-10]}$ substituted spectra were largely distorted. We succeeded in detecting the structural change of N0$_3$ $^{[-10]}$ in the cl$^{[-10]}$ site upon the S$_1$ to S$_2$ transition from $^{14}$ N0$_3$$^{[-10]}$ /$^{15}$ N0$_3$$^{[-10]}$ difference spectrum.

A Study on Runoff Properties of Non-point Pollutant in Nakdong watershed by using SWAT model (SWAT 모형을 이용한 낙동강 하구언의 비점오염물질 유출특성 규명)

  • Non-point source pollutants in down stream of Nakdong river were simulated by SWAT. GIS was utilized to make input data of SWAT such as landuse pattern and soil. Meteorological data of 2007 and 2009 were applied for the calibration and validation of runoff in SWAT. It was difficult to calibrate and validate the runoff and nutrient results since a study area was influenced by the tidal effects. Jindong site was selected to escape from the bias of runoff simulation in the coastal area. $R^2$ values of calibration and validation were 0.8 and 0.79. However, $R^2$ values of water qualities were very low level in comparison to runoff. These resulted from the concentration scale of water qualities such as BOD, T-N and T-P. Additionally, tidal influence could effected on the measurements of nutrients. The simulated annual averages and patterns of BOD, T-N and T-P in SWAT were similar to the measurement data. 80 ~ 96 % of nonpoint source pollutants at Nakbon M site were released from April to August of 2009. The ratio of T-N and T-P from nonpoint source were above 50 % during the rainy season.

Determination of Cytoplasmic Male Sterile Factors in Onion Plants (Allium cepa L.) Using PCR-RFLP and SNP Markers

  • We have developed a polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) marker that can distinguish male-fertile (N) and male-sterile (S) cytoplasm in onions. The PCR-RFLP marker was located in a chloroplast psbA gene amplicon. Digesting the amplicons from different cytoplasm-containing varieties with the restriction enzyme MspI revealed that N-cytoplasm plants have a functional MspI site (CCGG), whereas the S-cytoplasm plants has a substitution in that site (CTGG), and thus no MspI target. The results obtained using this PCR-RFLP marker to distinguish between cytoplasmic male sterile factors in 35 onion varieties corresponded with those using a CMS-specific sequence-characterized amplified region (SCAR) marker. Moreover, the PCR-RFLP marker can identify N- ot S-cytoplasms in DNA sample mixtures in which they are in up to a 10-fold minority, indicating that use of the marker has high diagnostic precision. We also demonstrated the usefulness of the SNP detected in the psbA gene for high-throughput discrimination of CMS factors using Real-time PCR and a TaqMan probe assay.

Synthesis of Nitrogen-Doped Graphene by Thermal Annealing of Graphene Oxide with Melamine Compounds (멜라민 화합물을 이용한 산화 그래핀 도핑 및 특성 평가)

  • In this paper, nitrogen-doped reduced graphene oxide(rGO) is obtained by thermal annealing of nitrogen-containing compounds and graphene oxide (GO) manufactured by modified Hummers' method. We use melamine as a nitrogen-containing compound and treat GO thermally with melamine at over $800{\sim}1,000^{\circ}C$ and 1 ~ 3 hr under Ar atmosphere. The electrical conductivity of doped rGO is measured by 4-point probe method. As a result, nitrogen contents on rGO are found to be in the range of 2.5 to 12.5 at% depending on the doping conditions after thermal annealing. The main doping site on graphene oxide is changed from pyridinic-N and pyrrolinic N to the graphitic site as the heat treatment temperature increases. The electrical conductivity of doped rGO increases as the N doping content increases. As the thermal treatment time increases, the change of both total doping contents and doping sites is slight and the surface resistance is remarkably reduced, which is caused by healing effects of doped graphene oxide at high temperature.

Nutrient Dynamics in Litterfall and Decomposing Leaf Litter at the Kwangneung Deciduous Broad-Leaved Natural Forest (광릉 천연활엽수림의 낙엽낙지와 낙엽분해에 따른 양분동태)

  • Litterfall and litter decomposition represent a major contribution to the carbon and nutrient inputs in a forest ecosystem. We measured litterfall quantity and nutrient dynamics in decomposing litter for two years at the Kwangneung broadleaf natural forest (DK site) in Korea. Litterfall was collected in circular littertraps (collecting area : 0.25 $m^2$) and mass loss rates and nutrient release patterns in decomposing litter were estimated using the litterbag technique employing 30 cm ${\times}$ 30 cm nylon bags with 1.5 mm mesh size. Total annual litterfall was 5,627 kg/ha/yr and leaf litter accounted for 61 % of the litterfall. The leaf litter quantity was highest in Quercus serrata, fallowed by Carpinus laxiflora and C. cordata, etc., which are dominant tree species in the site. Mass loss from decomposing leaf litter was more rapid in C. laxiflora and C. cordata than in Q. serrata litter. About 77% of C. laxiflora and 84% of C. cordata litter disappeared, while about 48% in Q. serrata litter lost over two years. Lower mass loss rates of Q. serrata litter may be attributed to the difference of substrate quality such as lower nutrient concentrations compared with the other litter types. Nutrient concentrations (N, P, Mg) of three litter types except for potassium (K) increased compared with initial nutrient concentrations of litter over the study period. Compared with Q. serrata litter, nutrients (N, P, K, Ca, Mg) in C. laxiflora and C. cordata litter were released rapidly. The results suggest that litter mass loss and nutrient dynamic processes among tree species vary considerably in the same site conditions.

The Composting Techniques for On-site Recycling of Wood Waste (임목폐기물의 현장 재활용을 위한 퇴비화기술)

  • The main purpose of this study is as follows : Finding a solution for fresh wood chips to be used as an alternative growing-media through a study of the formation method of a compost base that is applicable on both construction site and composting factory to ferment fresh wood chips produced from construction site as well as a study on adjuvant or secondary materials. The result from the experiment plot using wood chips sized 50mm or less, manure and fertilizer mixed manure as a source of nitrogen to compost fresh wood chips has shown that the temperatures 9days and 3days after the beginning of the experiment reached to $49.0^{\circ}C$ and $40.4^{\circ}C$ respectively, the heating duration was 7days and 4days respectively, and the C/N ratio was 26.5 and 25.3, each satisfying the standard for composted manure (25.0~40.0). Also, the other result from another experiment plot using mixture of sandy soil and microorganism as an inoculation source of microorganism has shown that the temperature 10days after the beginning of the experiment reached to $67.6^{\circ}C$ and lasted 16days. The experiment plot using sandy soil has shown the highest figure of $5.3{\sim}108$ CFU/g in terms of number of microorganism. The result from the experiment plots for on-site composting of fresh wood chips have shown that the experiment plot that used sandy soil with fertilizer due to supply restriction of manure as a nitrogen source in construction site reached the high temperature of $54.7^{\circ}C$ after 3days of experiment beginning and maintained heating state for 17days and reached 30.6 of C/N ratio, satisfying the standard for composted manure.

Immediate regraft of the remnant skin on the donor site in split-thickness skin grafting

  • Background: Skin defects of head and neck need reconstruction using various local flaps. In some cases, surgeons should consider skin graft for large skin defect. It is important to heal skin graft and donor sites. The authors investigated wound healing mechanisms at the donor sites with split-thick-ness skin graft (STSG). In this study, the authors compared two types of immediate regraft including sheets and islands for the donor site after facial skin graft using remnant skin. Methods: The author reviewed 10 patients who underwent STSG, from March 2015 to May 2017, for skin defects in the craniofacial area. The donor site was immediately covered with the two types using remnant skin after harvesting skin onto the recipient site. Depending on the size of the remnant skin, we conducted regraft with the single sheet (n= 5) and island types (n= 5). Results: On postoperative day 1 and 3 months, the scar formation was evaluated using the Patient and Observer Scar Assessment Scale (POSAS) and Vancouver Scar Scale (VSS). Total POSAS and VSS scores for the island type were lower than in single sheet group after 3 months postoperatively. There was significant difference in specific categories of POSAS and VSS. Conclusion: This study showed a reduction in scar formation following immediate regrafting of the remnant skin at the donor site after STSG surgery. Particularly, the island type is useful for clinical application to facilitate healing of donor sites with STSG.
