Muscle-specific color stability in fresh beef from grain-finished Bos indicus cattle

  • Objective: To investigate the color and oxidative stabilities of longissimus lumborum (LL) and psoas major (PM) muscles from grain-finished Bos indicus cattle in Brazil. Methods: The LL and PM muscles were obtained 24 h post-mortem from eight (n = 8) Nellore bull carcasses, fabricated into 1.5-cm steaks, aerobically packaged, and stored at $4^{\circ}C$ for nine days. Steaks were analyzed for myoglobin concentration, pH, instrumental color, metmyoglobin reducing activity (MRA) and lipid oxidation. Results: The LL steaks exhibited greater (p<0.05) redness, color stability, and MRA than their PM counterparts on days 5 and 9. The LL and PM steaks demonstrated similar (p>0.05) lightness and yellowness on days 0, 5, and 9. On the other hand, PM steaks exhibited greater (p<0.05) myoglobin concentration, pH, and lipid oxidation than their LL counterparts. Conclusion: These results indicated that muscle source influenced the color and oxidative stabilities of beef from grain-finished Bos indicus animals. These results highlighted the necessity of muscle-specific strategies to improve the color stability of beef from grain-fed Bos indicus cattle.

실리콘 기반 타원편광계식 바이오센서를 이용한 심근경색 생체표지자의 실시간 초고감도 진단 농도 측정 (Real-time Highly Sensitive Measurement of Myocardial Infarction Biomarkers Using Silicon-based Ellipsometric Biosensors)

  • $2^{\circ}$ 기울어진 산화막 코팅 실리콘 기판의 바이오칩과 프리즘으로 제작한 바이오센서와 검광자 회전 타원편광계를 이용하여 심근경색 생체표지자인 미오글로빈과 cTnI의 진단 농도를 수백 초 내에 실시간 초고감도로 측정하는데 성공하였다. 러닝 버퍼로는 순수한 phosphate buffered saline (PBS) 또는 PBS에 10% 인간 혈청을 섞은 러닝 버퍼를 사용하였다. PBS 조건에서는 미오글로빈과 cTnI가 각각 1 ng/mL와 5 pg/mL로 측정되었으며, PBS에 인간 혈청을 10% 섞은 조건에서는 미오글로빈과 cTnI는 각각 1 ng/mL과 1 pg/mL로 측정되었다. 이러한 심근경색 생체표지자의 진단 농도는 현재 제시된 세계보건기구의 심근경색 진단 기준 농도보다 미오글로빈은 1/15배 낮고, cTnI는 1/80배 낮다.

  • 미오글로빈 시안착물(MbCN) 단백질에 대한 NMR의 HMQC 연구는 수소와 결합된 상자기성 heme 탄소 시그날의 완전한 지정을 가능토록 해준다. 이러한 상자기성 MbCN에 대한 HMQC 실험의 적용은 heme시그날뿐만 아니라 상자기성 아미노산에 대해 결합된 수소와 탄소간의 coherence를 지정하여주며 자연존재량 $^{13}C$시그날의 지정이 모든 low-spin 상자기성 heme단백질에서도 가능하다. 이러한 시그날 지정 전략은 정자기성 영역에서 공명하는 수소 시그날의 지정을 위해 사용되는 NOE에만 의존하는것 보다 훨씬 명확한 시그날지정이 가능하다. 2,4-비닐기의 ${\alpha}$-탄소들과 7-프로피온기의 ${\beta}$-탄소에서 특이한 anti-Curie형태를 보이는 것은 그들이 heme평면에 존재하고 있지 않다는 증거가 된다. Proximal His에 의해 유도된 heme의 전자 및 자성의 비대칭은 heme탄소 시그날공명이 $25^{\circ}C$에서 250 ppm의 범위에 이르도록 한다. 이러한 heme 탄소 시그날 공명은 미오그로빈 heme의 전자구조를 분석하는데 있어서 수소 시그날의 공명보다 더욱 민감한 증거로 작용할수 있다.

금속 친화성 액 이상분계 시스템에서 단백질의 분배 향상 (Enhanced Partitioning of Proteins in Metal-Affinity Aqueous Two-Phase Systems)

  • In metal-affinify aqueous two-phase systems, protein partitioning is affected by a variety of parameters such as pH, the number of surface-accessible histidines, and the amount and partition coefficient of metallated polythylene glyco(PEG) ligand. To enhance partitioning of proteins with surface-accessible histidines, we have synthesized and used a (Cu(II)-ininodiacetic acid)$_2$-PEG20,000($Cu(II)_2IDA_2$-PEG20,000) as well as Cu(II)IDA-PEG5,000 as an affinity ligand. The partition coefficient of $Cu(II)_2-IDA_2$-PEG20,000 in a PEG5,000/dextran two-phase system was 30.1, which corresponded to a 3.8-fold increase over that of Cu(II)IDA-PEG5,000. The partitioning experiments were performed on four proteins, horse cytochrome c, S. cerevisiae cytochrome c, horse myoglobin, and sheep myoglobin. Partitioning of proteins which convey surface-accessible histidines was enhanced dramatically by the addition of $Cu(II)_2IDA_2$-PEG20,000 ligand. These results demonstrate that enhanced partitioning of metal-binding proteins in an aqueous two -phase system can by achieved by using an appropriate metallated PEG ligand.

Virus-associated Rhabdomyolysis in Children

  • Purpose: Virus-associated rhabdomyolysis is very rare. We report 15 patients with rhabdomyolysis caused by various viruses. Methods: Fifteen patients who were diagnosed with rhabdomyolysis and a viral infection were included in this study. Clinical, laboratory, and radiologic findings were evaluated through retrospective chart reviews. Results: Chief complaints were severe bilateral lower leg pain and leg weakness. The median age was 5.7 years. The male:female ratio was 2:5. The viral infections were caused by influenza virus B, parainfluenza virus, and rhinovirus. One patient with influenza virus B had coinfection with coronavirus. Median initial laboratory values and ranges were as follows: serum creatinine, 0.4 (0.1-0.5) mg/dL; serum aspartate transaminase, 124 (48-1,098) IU/L; serum alanine transaminase, 30 (16- 1,455) IU/L; serum creatine kinase, 2,965 (672-16,594) IU; serum lactate dehydrogenase, 400 (269-7,394) IU/L; serum myoglobin, 644 (314-3,867) ng/mL; urine myoglobin, 3 (3-10,431) ng/mL. All patients recovered without complications. Conclusion: This is the first report of the simultaneous occurrence of rhabdomyolysis caused by various viruses. This is also the first report of rhinovirus-associated rhabdomyolysis.

  • Study the variation of low frequency electrical stimulation on human metabolism which serum to test the increasement of creatine- kinase, myoglobin, lactate .and pyruvate. The results were as followed: 1) Creatine kinase increasement showed significant difference between 200 Hz stimulation group and control group, and also between 300Hz stimulation group and control group, but no significant increasement difference showed in 50 Hz stimulation group compared to control group. 2) Lactate increasement showed significant difference in 200 Hz stimulation group and 300 Hz stimulation group compared to control group, but no significant difference showed in 50 Hz stimulation group. 3) Pyruvate increasement showed significant difference in 50 Hz stimulation group and 200 Hz stimulation group compared to control group, but no significant increasement difference showed between 50 Hz stimulation group and control group. (P < 0.05) 4) Myoglobin increasement showed significant difference in 50Hz stimulation group and 200 Hz stimulation group compared to control group, and also in 200 Hz stimulation group and 300 Hz stimulation group compared to 50Hz stimulation group. Effects of Short-Term Presalting and Salt Level on the Development of Pink Color in Cooked Chicken Breasts

  • The objective of this study was to determine the effects of short-term presalting on pink color and pigment characteristics in ground chicken breasts after cooking. Four salt levels (0%, 1%, 2%, and 3%) were presalted and stored for 0 and 3 d prior to cooking. Cooking yield was increased as salt level was increased. However, no significant differences in pH values or oxidation reduction potential (ORP) of cooked chicken breasts were observed. Cooked products with more than 2% of salt level had less redder (lower CIE $a^*$ value) on day 3 than on those on day 0. As salt level was increased to 2%, myoglobin was denatured greatly. Myoglobin denaturation was leveled off when samples had 3% of salt. With increasing salt levels, residual nitrite contents were increased while nitrosyl hemochrome contents were decreased. These results demonstrate that salt addition to a level of more than 2% to ground meat may reduce the redness of cooked products and that presalting storage longer than 3 d should be employed to develop a natural pink color of ground chicken products when less than 1% salt is added to ground chicken meat.
