Isolation and Identification of Fungal Species from the Insect Pest Tribolium castaneum in Rice Processing Complexes in Korea

  • The red flour beetle, Tribolium castaneum, is one of the most common and economically important pests of stored cereal products worldwide. Furthermore, these beetles can act as vectors for several fungal post-harvest diseases. In this study, we collected T. castaneum from 49 rice processing complexes (RPCs) nationwide during 2016-2017 and identified contaminating fungal species on the surface of the beetles. Five beetles from each region were placed on potato dextrose agar media or Fusarium selection media after wet processing with 100% relative humidity at $27^{\circ}C$ for one week. A total of 142 fungal isolates were thus collected. By sequence analysis of the internal transcribed spacer region, 23 fungal genera including one unidentified taxon were found to be associated with T. castaneum. The genus Aspergillus spp. (28.9%) was the most frequently present, followed by Cladosporium spp. (12.0%), Hyphopichia burtonii (9.2%), Penicillium spp. (8.5%), Mucor spp. (6.3%), Rhizopus spp. (5.6%), Cephaliophora spp. (3.5%), Alternaria alternata (2.8%) and Monascus sp. (2.8%). Less commonly identified were genera Fusarium, Nigrospora, Beauveria, Chaetomium, Coprinellus, Irpex, Lichtheimia, Trichoderma, Byssochlamys, Cochliobolus, Cunninghamella, Mortierella, Polyporales, Rhizomucor and Talaromyces. Among the isolates, two known mycotoxin-producing fungi, Aspergillus flavus and Fusarium spp. were also identified. This result is consistent with previous studies that surveyed fungal and mycotoxin contamination in rice from RPCs. Our study indicates that the storage pest, T. castaneum, would play an important role in spreading fungal contaminants and consequently increasing mycotoxin contamination in stored rice.

조선 전통 메주에서 균독소(Mycotoxin) 측정 (Detections of the Mycotoxins on the Korean Traditional Home made Mejus)

  • 우리 나라에 중요한 식품의 원료인 메주에 대한 균독소($Aflatoxin\;B_1$와 Ochratoxin A) 생성 여부를 인위적인 혹은 자연적인 환경에서 조사하였다. 채집된 메주에서 균을 분리하여 접종함으로, 인위적인 환경인 독소생성 배지와 콩을 이용한 배지를 이용하여 균독소 생성을 관찰하였다. 조선 전통메주에서 채집된 메주 균인 불완전 진균들은 몇 몇 균이 독소를 생성하였으나, 멸균된 날 콩에는 다만 $2{\sim}3$개의 분리균들이 $Aflatoxin\;B_1$와 Ochratoxin A를 분비하였다. 그러나, 언급된 두 개의 균독소가 함께 생성하는 분리균은 관찰되지 않았다. 그러나, 조선 전통메주에서는 어떤 채집된 메주에서도 위의 균독소가 $0.01{\sim}100\;ng$ 범위에서 관찰되지 않았다. 이러한 결과로 메주 제조에 관련된 메주 균에서 털곰팡이(Mucor species)가 메주 제조에 초기에 작용한 균으로 전통 메주의 발효와 균독소 생성과 관계에서 중요한 것으로 사료된다.

Influence of Temperature and Water Activity on Deleterious Fungi and Mycotoxin Production during Grain Storage

  • Cereal grains are the most important food source for humans. As the global population continues to grow exponentially, the need for the enhanced yield and minimal loss of agricultural crops, mainly cereal grains, is increasing. In general, harvested grains are stored for specific time periods to guarantee their continuous supply throughout the year. During storage, economic losses due to reduction in quality and quantity of grains can become very significant. Grain loss is usually the result of its deterioration due to fungal contamination that can occur from preharvest to postharvest stages. The deleterious fungi can be classified based on predominance at different stages of crop growth and harvest that are affected by environmental factors such as water activity ($a_w$) and eco-physiological requirements. These fungi include species such as those belonging to the genera Aspergillus and Penicillium that can produce mycotoxins harmful to animals and humans. The grain type and condition, environment, and biological factors can also influence the occurrence and predominance of mycotoxigenic fungi in stored grains. The main environmental factors influencing grain fungi and mycotoxins are temperature and $a_w$. This review discusses the effects of temperature and $a_w$ on fungal growth and mycotoxin production in stored grains. The focus is on the occurrence and optimum and minimum growth requirements for grain fungi and mycotoxin production. The environmental influence on aflatoxin production and hypothesized mechanisms of its molecular suppression in response to environmental changes are also discussed. In addition, the use of controlled or modified atmosphere as an environmentally safe alternative to harmful agricultural chemicals is discussed and recommended future research issues are highlighted.

Effects of dietary arginine in ameliorating the deleterious effects induced by mycotoxins on growth, immune system, body organs in growing pigs

  • Mycotoxin contamination in pig feeds has a negative impact on growth performance, the immune system, and major body organs. Arginine (Arg) plays an important role in animals' body biochemistry and physiology. This study aimed to determine the effect of dietary Arg supplementation on mitigating the negative effects of mycotoxins in growing pigs. A total of 72 growing pigs (Landrace × Large white) with initial mean body weight (BW) = 55 ± 2.5 kg were allotted to four treatment groups with three replicates per group of six pigs per replicate in a completely randomized design. The treatments included a non-toxin diet with 1.2% Arg (NT1.2) and mycotoxin-challenged treatments supplemented with 1.2% Arg (TX1.2), 1.3% Arg (TX1.3), and 1.4% Arg (TX1.4). Statistical analysis of data included the effects of dietary level of Arg. The results indicated a significantly higher BW (p < 0.05), average daily gain (p < 0.05), and gain-to-feed ratio (p < 0.05) in the NT1.2 group than in the TX1.2, TX1.3, and TX1.4 groups. The relative weight of the liver was higher (p < 0.05) in the TX1.2 compared to that of the NT1.2 group, although it was not different from that of TX1.3 and TX1.4. The level of tumor necrosis factor-alpha was significantly up-regulated (p < 0.05) in the liver tissue of the TX1.2 group compared to that of the other treatments. Overall, dietary Arg supplementation remedied liver injury and alleviated the compromised immune system caused by mycotoxin toxicity.

국내 조사료용 볏짚의 수확 전 작물포 및 수확 후 저장에 따른 곰팡이독소 오염 실태 연구 (The Studies on Real Condition of Mycotoxin Contamination in the Fields before Harvest and by the Storage of Rice Straw using as Roughage in Korea)

  • 본 연구는 국내 생산 볏짚의 곰팡이독소 오염 실태를 파악하기 위하여 벼의 생육 단계별, 볏짚의 보관 형태별 및 곤포사일리지의 지역별 오염을 조사하였다. 수확하기 전 논에서 생장 중인 비에서 곰팡이독소 및 곰팡이 오염이 확인되었다. 벼의 생장 초기에는 deoxynivalenol($297{\mu}g/kg$) 이 발견되었고, 중기에는 ochratoxin A($1.8{\mu}g/kg$) 및 deoxynivalenol($264{\mu}g/kg$) 이 오염되어 있었다. 그리고 수확시기에는 deoxynivalenol($190{\mu}g/kg$) 및 zearalenone ($168{\mu}g/kg$) 이 오염되어 있었다. 그리고 곰팡이 오염은 생장 초기에는 $10^{3-4}$ cfu/g 그리고 생장 중기와 수확시기에는 $10^{4-5}$ cfu/g 이었다. 오염된 곰팡이로는 Fusarium sp., Fusarium proliferatum, Penicillium sp., Gibberella sp., Gibberella zeae, Mucor circinelloides, Aspergillus oryzae 등 다양한 종류가 발견되었다. 사료용 볏짚의 통 볏짚단, 사각 볏짚단 및 곤포사일지 등 저장 방법에 따른 오염원 mycotoxin을 조사하였을 때 zearalenone, deoxynivalenol 및 ochratoxins A가 검출되었다. 통 볏짚단의 곰팡이독소 오염도는 23% 이었고, 사각 볏짚단과 곤포사일지지는 각각 38%가 오염되어 있었다. 그리고 105점의 볏짚 곤포사일리지에서 곰팡이독소를 분석하였을 때 46%의 샘플에서 곰팡이독소가 발견되었고, 지역적으로는 경기, 강원, 충청, 영남 및 호남지역에서 각각 48, 33, 40, 50 및 57%의 샘플이 오염되어 있었다. 2가지의 곰팡이독소(zearalenone, deoxynivalenol)가 발견된 충청지역을 제외한 모든 지역의 볏짚 곤포사일리지에서 3가지의 곰팡이독소(zearalenone, deoxynivalenol, ochratoxins A)가 발견되었다. Ochratoxins A의 오염 수준은 전체적으로 $2.6{\mu}g/kg$으로 충청지역에서는 발견되지 않았지만 호남지역은 $3.0{\mu}g/kg$으로 다른 지역보다 높았다. Deoxynivalenol의 오염 수준은 전체적으로 $413{\mu}g/kg$으로 충청지역이 $800{\mu}g/kg$로 가장 높았고, 다른 지역은 $246{\mu}g/kg$에서 $433{\mu}g/kg$의 수준으로 발견되었다. 그리고 zearalenone은 전체적으로 $338{\mu}g/kg$으로 경기지역이 $168{\mu}g/kg$로 다른 지역보다 낮았고, 강원과 호남지역이 각각 $467{\mu}g/kg$$442{\mu}g/kg$으로 높았다. 이상의 연구 결과는 볏짚은 수확하기 전 벼의 생장 기간에도 곰팡이독소 및 곰팡이 오염에 노출되어 있으며, 볏짚의 보관 형태 및 지역에 상관없이 곰팡이독소의 오염이 발생할 수 있음을 명확히 시사한다. 따라서 안전하고 질 좋은 조사료를 확보하기 위해서는 이들 곰팡이독소 관련 더 구체적 연구와 대책관련 탐구가 지속적으로 이루어져야 할 것으로 사료된다.

Occurrence and Decontamination of Mycotoxins in Swine Feed

  • Contamination of agricultural crops by mycotoxins results in significant economic losses for grain producers and, when consumed, it can cause reduced growth and health in a wide range of animal species. Hundreds of mycotoxin producing molds exist, however each has a different frequency and pattern of occurrence, as well as differences in the severity of the diseases (mycotoxicoses) they cause. Among the mycotoxins considered to be major contaminates are aflatoxin, deoxynivalenol, fumonisin, ochratoxin, and zearalenone. Although a multitude of species can be harmed by consumption of these mycotoxins, swine appear to be the most commonly affected commodity species. The swine industry can thus experience great losses due to the presence of mycotoxin contamination in feeds. Subsequently, recognition and prevention of mycotoxicoses is extremely important and dependent on adequate grain sampling and analysis methods pre-harvest, as well as effective strategies post-harvest to reduce consumption by animals. The aim of this review is to provide an overview of the major mycotoxin contaminants in grains, to describe methods of analysis and prevention to reduce mycotoxicoses in swine and other animals, and finally to discuss how mycotoxins directly affect swine production.

Co-contamination of Aflatoxins with Ochratoxin A and Zearalenone in Thuja orientalis Semen

  • Korea is representative of a country that consumes herbal medicines; most of the herbal medicines circulating in South Korea have been imported from developing countries in Southeast Asia, such as China and Indonesia. Recently, domestic hygiene and safety are issues that have come to the forefront, because herbal medicines currently in circulation could possibly contain contaminants or residues. Furthermore, the appearance or discovery of harmful new species due to environmental and industrial developments is becoming a social problem. Therefore, it may be necessary to consider and investigate these matters on a continual basis. Recently, mycotoxin contaminations in such foods as cereals, nuts, and powdered red pepper have been reported. They have become a problematic issue; the possibility of contamination in herbal medicines has also been considered. Nevertheless, recognition of and research into mycotoxin contamination in herbal medicines has been scarce because herbal medicine is used in only a few nations. In this research, we identified contamination by aflatoxin which is known to be the most potent mutagenic, carcinogenic, and teratogenic mycotoxin in Thujae Semen, a herbal medicine. We also found co-contaminations involving other mycotoxins, including ochratoxin A and zeraleanone.
