Studies on Submerged Culture and Mycelial Components of Naematoloma sublateritium Mycelia (개암버섯균의 액체배양과 균사체의 성분에 관한 연구)

  • This study was carried out to get the basic information for the submerged culture and analyze the biochemical components of Naematoloma sublateritium mycelia. The optimal temperature, pH, agitation speed and cultural time for the mycelial growth of Naematoloma sublateritium were $25^{\circ}C$, 5.5, 150rpm and 20 days, respectively. The proximate composition of mycelia was as follows; carbohydrate 55.8% (total sugar 48.7%), crude protein 22.4%, fat 4.1 % and ash 4.7% respectively. Among the free amino acid contents, phenylalanine, alanine and lysine were predominant component. The linoleic acid and palmitic acid were found to be the highest among the free fatty acids. The biopolymer extracts of mycelia was identified to be protein-bounded polysaccharide by color reaction and sepharose CL-4B gel chromatography.

Optimization for Production of Phellinus linteus Mycelia with Anti-Complement Activity (항보체 활성을 갖는 Phellinus linteus 균사체 생산의 최적화)

  • To produce the functional food materials from edible mushrooms, hot-water extracts from 70 kinds of mushroom mycelia were examined for anti-complementary activity and Phellinus linteus showed the highest activity through the complement fIxation test. The maximum production of Phellinus linteus mycelia with anti-complementary activity was observed in culture medium containing soluble starch 3.0%, peptone 0.3%, yeast extract 0.4%, $MgSO_4{\cdot}7H_2O$ 0.1%, $K_2HPO_4$ 0.2% and in the culture conditions controlled at initial pH 7.0, $30^{\circ}C$ and 150 rpm by the rotary shaker. In addition, the maximum production of mycelial dry weight was 15 mg/mL after 18 days under the optimal conditions, and anti-complementary activity was reached to 88% in 5 L-jar fermenter.

Extraction and Purification of Polysaccharides from Phellinus linteus Mycelia (상황버섯의 균사체로부터 다당류의 추출과 정제)

  • To maximize the obtained polysaccharides from Phellinus linteus mycelia, effects of extraction and purification conditions including dialysis time (8, 16, 24, 48 h) on an amount of the polysaccharides were investigated. As extraction temperature ($50{\sim}95^{\circ}C$), ratio of solvent volume to the dry weight of mycelia (10, 15, 20, 25 ml/g), extraction time(2, 4, 6, 8 h), final concentration of ethanol (70, 75, 80, 85%), and aging time (1, 4, 8, 16 h) increased, an amount of the polysaccharides was increased. An increase in precipitation time up to 24 h increased an amount of the polysaccharides but a further increase in precipitation time after 24 h did not changed largely an amount of the polysaccharides. Three precipitation solvents (ethanol, methanol, acetone) were tested. An amount of the polysaccharides was increased in order to acetone, ethanol, and methanol. On the optimal extraction condition, extraction temperature, ratio of water volume to the dry weight of mycelia, and extraction time were $95^{\circ}C$, 25 ml/g, and 8 h, respectively.

Anti-oxidization Effect of Extracts from Oriental Medicine and Cereal Medium Where Tricholoma matsutake Mycelia were Cultured

  • In order to explore the anti-oxidization effect of oriental medicine and cereal medium(OCM) where Tricholoma matsutake mycelia were cultured, measurement of hot water extract and UMPM(extraction method using ultra sonic waves, micro waves, micro bubble) extract, the total polyphenol content of crude polysaccharide from each extract, SOD-like activity, electron donating ability(EDA), xanthine oxidase inhibitory activity, and tyrosinase inhibitory activity was conducted. The total polyphenol content of each extract was found to be 16.36% for hot water extract(HE) group and 15.73% for UMPM extract(UE) group and the amount of crude polysaccharide precipitated into ethanol of extracts was found to be 8.79% for UMPM ethanol extract(UEE) group and 6.48% for hot water ethanol extract(HEE) group. As a result of measurement of SOD-like activity by concentration of each extract, it was found to be 96.17% for UE group, 91.23% for HE group, 91.33% for UEE group, and 87.11% for HEE group at 20 mg/mL. In the case of EDA, it was found to be 47.55% for UE group, 44.93% for HE group, 25.38% for UEE group, and 18.36% for HEE group. And in the cases of the rates of xanthine oxidase inhibitory activity and tyrosinase inhibitory activity, as the concentration of each extract increased, the inhibition rate increased accordingly. As a result of comparison between hot water extract method and UMPM extract method using extracts obtained from oriental medicine compound medium where Tricholoma matsutake mycelia were cultured, all of the extracts were judged to have a high anti-oxidization effect. In particular, UMPM extracts were found to have higher polyphenol content, SOD-like activity, EDA, xanthine oxidase inhibitory activity and tyrosinase inhibitory activity compared to hot water extract method. In this regard, extracts obtained from OCM where Tricholoma matsutake mycelia were cultured are considered to have high availability as functional material when and if they are prepared using UMPM extract method.

  • The industrial procedure for the preparation of dietary fiber material from Lentinus edodes by drying and grinding process was developed. The chemical composition of dietary fiber of mycelia was as follows; crude protein: 16.16%, crude fat: 2.96%, crude ash: 3.25% and carbohydrates: 77.63%. The mycelia contained total dietary fiber of 54.5%. The 40 mesh pass of dietary fiber exhibited water-holding capacity 7.39g water/g and oil-holding capacity 2.03g oil/g, while $40{\sim}80$ mesh of dietary fiber had water-holding capacity 7.80g water/g and oil-holding capacity 2.77g oil/g.

  • Anti-diabetic activities of cultured mycelia from Ganoderma applanatum are being evaluated in this study. The OGTT and 4-weeks of repeated oral efficacy tests are conducted in mice at the doses of 0 (vehicle treatment), 25, 75 and 225 mg/kg/day, respectively. In human study, the test article was administered orally every day for 8-week at a dose of 1,500 mg/kg, tid and placebo group. The blood glucose levels (BGL) at 0.5 hour after treatment are significant decreased in all treatment groups of OGTT test. In the 4-week test, BGL of 75 and 225 mg/kg/day group is continuously decreased during all treatment periods and the BGL of 25 mg/kg/day group show decreasing trends at the final week, the pancreas weight of all treatment groups are being increased, and the Langerhans-islet numbers were increased at all treatment groups with a dose-response manner. There are no test article-related abnormal signs and the fasted blood glucose (FBG), postprandial blood glucose (PPG) and HbA1c are decreased significantly after 8-week treatments. These results that the cultured mycelia from Ganoderma applanatum could decrease BGL by protecting the degeneration of Langerhans islets.

A STUDY ON THE MYCELIA CUL TURE AND GENETIC CHARACTERISTICS IN Lepista nuda (민자주방망이버섯(Lepista nuda)의 균사배양 및 유전적 특성에 관한 연구)

  • This study was carried out to investigate the cultural characteristics and the genetic characteristics in Lepists nuda. The mycelia cultural characteristics which include the specificity for pH, carbon source, nitrogen source and medium were studied using petridish culture and liquid media culture respectively. The genetic characteristics were also investigated using random amplified polymorphic DNA (RAPD) analysis. To find out the mycelial growth rate by various medium, mycelia was cultured for 25 days in PDA, YM, MEA, GPB and Yamanaka. The Yamanaka was superior to the other media in supporting the mycelial growth. pH6 produced the best result in the test of an optimal pH. In experiment for optimal carbon source, starch showa 46.8$\pm$1.7mm of diameter of mycelial colony as the best and for nitrogen source, yeast extract showed a good effect as well. In investigation for genetic characteristics, Lepists nuda was amplified by 7 primers among 10 primers and Pleurotus ostreatus was also amplified by 7 primers. From the RAPD analysis between species, the band patterns were different.

  • Aspergillus awamori, B-2 which has a high ability to decolorize melanoidin was selected among various fungi. Aspergillus awamori, B-2 showed the highest decolorization activity when it was cultivated in a melanoidin medium containing 3.0% glucose, 0.5% yeast extract, 0.1 % $KH_2PO_4\;and\;0.05%\;MgSO_4{\cdot}7H_2O$ at an initial pH 7.0 at $37^{\circ}C$ for 5 days. Mycelia immobilized system with. Ca-alginate was more effective on melanoidin decolorization activity showed approximately 70% in 10 days under the optimal conditions. Continuous decolorization of melanoidin using, reuse of immobilized mycelia showed an almost constant decolorization of abort 60-70% for 15 days.

  • Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. Throughout the culture, sporulation was occured. Highly phosphorylated nucleotides in sporulating mycelia were detected to assure whether the eucaryotic Aspergillus niger produce these substances or not during the differentiation. Phosphorylated nucleotides were extracted from the conidiophore, bearing mycelia and spore forming body, these nucleotides were identified by TLC with P.E.I. cellulose. Guanosine tetraphosphate was found in both phialide forming mycelia and spore forming body. The contents of free amino acids were assayed and its level was found to increase at early stage of sporulation. The effects of 8-azaguanine examined, it was found to prevent spore formation and to made abnormal structure. The effects of inosinic monophosphate and guanosine monophosphate on spore formation were examined, spore formation was enhanced by these nucleotides.

Mycelial Properties of Tremella fuciformis and Hypoxylon sp. (흰목이와 Hypoxylon sp.균의 균사적 특성)

  • Two strains, each of Tremella fuciformis and Hypoxylon sp. were isolated and their mycelial properties were investigated. T. fuciformis produced yeast-like conidia and dikaryotic mycelia from white to yellow color. The diameter of the hyphae was $1.5\;to\;3.0\;{\mu}m$, and septa with clamp connections were present. Secondary mycelia with clamp connections could produce fruitbodies on sawdust medium when the environmental conditions were suitable for fructification. The symbiotic fungus, Hypoxylon sp. produced white feather-like mycelia. But the color of old mycelia was changed to light yellow or light brown and pigmented the culture medium from light brown to brown or very dark green. Generally it did not produce conidia, but in a special case it produced conidia of which color was yellow green to grass green and of which shape was sub-elliptical with the size of approximately $3\;to\;5\;{\mu}m$.
