Photodynamic Therapy for Methicillin-resistant Staphylococcus aureus with High-level Mupirocin Resistance using 630 nm Light-emitting Diode

  • This study aims to evaluate the effect of Photodynamic Therapy (PDT) against methicillin-resistant Staphylococcus aureus with high-level mupirocin resistance (Hi-Mup MRSA). To examine the antimicrobial effect of photogem-mediated PDT against Hi-Mup MRSA, CFU quantifications, bacteria cell viability tests, and disk diffusion antimicrobial susceptibility tests were evaluated. In addition, one of PDT mechanisms was investigated by accumulating photogem ($10\;{\mu}g/ml$) in Hi-Mup MRSA. Photogem-mediated PDT properly inhibited the colony formation of Hi-Mup MRSA. Viable bacteria decreased greatly after a PDT application with photogem $10\;{\mu}g/ml$ at energy density $15\;J/cm^2$. The diameter of the inhibition zone around susceptible disks increased after PDT. In addition, we confirmed the accumulation of photogem in bacteria through fluorescent images. These results demonstrated that excellent photosensitization of Hi-Mup MRSA can be achieved using photogem with 630 nm LED irradiation. Thus, PDT may make survival Hi-Mup MRSA inactive.

Enhancing Mulberry Leaf Meal with Urea by Pelleting to Improve Rumen Fermentation in Cattle

  • Four, ruminally fistulated crossbred (Brahman${\times}$native) beef cattle with initial body weight of $420{\pm}15kg$ were randomly assigned according to a $4{\times}4$ Latin square design. The dietary treatments were mulberry leaf pellet (MUP) supplementation at 0, 200, 400 and 600 g/hd/d with rice straw fed to allow ad libitum intake. All steers were kept in individual pens and supplemented with concentrate at 5 g/kg of body weight daily. The experiment was 4 periods, and each lasted 21 d. During the first 14 d, all steers were fed their respective diets ad libitum and during the last 7 d, they were moved to metabolism crates for total urine and fecal collection. It was found that increasing MUP levels resulted in linearly increasing rice straw and total intakes (p<0.05). Ruminal temperature and pH were not significantly affected by MUP supplementation while $NH_3$-N concentration was increased (p<0.05) and maintained at a high level (18.5 mg/dl) with supplementation of MUP at 600 g/hd/d. Similarly, viable total bacteria in the rumen and cellulolytic bacteria were enriched by MUP supplementation at 600 g/hd/d. However, the rumen microbial diversity determined with a PCR-DGGE technique showed similar methanogenic diversity between treatments and sampling times and were similar at a 69% genetic relationship as determined by a UPGMA method. Based on this study, it could be concluded that supplementation of MUP at 600 g/hd/d improved DM intake, ruminal $NH_3$-N, and cellulolytic bacteria thus iimproving rumen ecology in beef cattle fed with rice straw.

Influence of Varying Levels of Dietary Undegraded Intake Protein Intake on Nutrient Intake, Body Weight Change and Reproductive Parameters in Postpartum Awassi Ewes

  • The objective of this study was to evaluate the effect of dietary undegradable protein (UP) level on body weight change, nutrient intake, milk production and postpartum reproductive performance of Awassi ewes. Twenty-seven multiparous Awassi ewes (initial body weight = 53.3${\pm}$1.6 kg) were randomly assigned to three dietary treatments (9 ewes/treatment) for 62 days using a completely randomized design. Experimental diets were isonitrogenous (15.5% CP), isocaloric, and were formulated to contain 17.9 (LUP), 27.1 (MUP), and 34.0% (HUP) of the dietary CP as UP. On day 10${\pm}$3 (day 0 = parturition) ewes were housed in individual pens for 5 weeks. Feed offered and refused was recorded daily. At the end of this period, animals were removed from their pens and combined into 3 separate groups (LUP, MUP and HUP). One fertile, harnessed ram was allowed with each group for 34 days. Rams were rotated every 2 days among the three groups. Each group was offered the corresponding experimental diet. Organic matter, CP, UP and metabolizable energy intakes were higher (p<0.05) for ewes fed the HUP diet compared with ewes fed the LUP and MUP diets. Ewes fed the HUP diet gained more (p<0.05) weight compared with ewes fed the MUP diet (7.3 vs. 2.1 kg), while ewes fed the LUP diet lost an average of 2.1 kg. Pregnancy rate of ewes fed the HUP diet was 100%, compared with 66 and 33% for ewes fed the MUP and LUP diets, respectively. Lambing rate was greater (p<0.05) for ewes fed HUP (8/9) diet compared with ewes fed the MUP (4/9) and LUP (3/9) diets. These results indicate that Awassi ewes receiving adequate dietary UP level consume more feed and are capable of returning to estrus shortly after parturition and are capable of producing two lamb crops per year.

  • The major urinary proteins(MUPs) of mice that bind hydrophobic molecules known as pheromones are regulated in part by the actions of growth hormone. The expression of the MUPs was therefore investigated in transgenic mice that express a human growth hormone-releasing factor gene from a metallothionein gene promoter(MT-GRF) and as a result have elevated growth hormone levels. MUPs were severely down-regulated in the urine of these animals compared to normal mice or to control transgenic mice expressing another gene(the inhibin a subunit) from the same metallothionein promoter(MT-Inh) and more MUPs disappeared in male mice than female ones. MUPs were also down-regulated in the urine of the UT-GRF-injected mice. In addition, it was observed that the urine of the MT-GRF mice included a high molecular weight protein that co-migrates with the major serum protein albumin, indicating an impairment in glomerular filtration within the kidney. The urinary loss of serum proteins was more severe in male MT-GRF mice than female ones. Thus the overexpression of human GRF mimics changes observed in MUP protein expression and glomerular function in other models of growth hormone hypersecretion with sex-dependent differential effects.

High-level mupirocin resistance in Gram-positive bacteria isolated from diseased companion animals

  • The purpose of this study was to investigate the high-level mupirocin resistance (HLMR) in Gram-positive bacteria isolated from companion animals. A total of 931 clinical specimens were collected from diseased pets. The detection of mupirocin-resistant bacteria and plasmid-mediated mupirocin resistance genes were evaluated by antimicrobial susceptibility tests, polymerase chain reactions, and sequencing analysis. Four-hundred and six (43.6%) bacteria were isolated and 17 (4.2%), including 14 staphylococci and 3 Corynebacterium were high-level mupirocin-resistant (MICs, ≥ 1,024 ug/mL) harboring mupA. Six staphylococci of HLMR strains had plasmid-mediated mupA-IS257 flanking regions. The results show that HLMR bacteria could spread in veterinary medicine in the near future.

Characteristics of Coagulase-negative Staphylococci Isolates from Dental Clinic Environments in Busan, Korea (부산지역 치과환경에서 분리된 coagulase-negative staphylococci의 특성)

  • Coagulase-negative staphylococci (CNS) have recently become the bacteria most frequently found in clinical infections. The aim of this study was to investigate the prevalence, antimicrobial susceptibilities, and molecular characteristics of CNS isolates from dental clinic environments in Busan, Korea. One hundred and fifty-four samples were collected from 10 dental clinics and dental hospitals in Busan from December 2014 to January 2015. Species were identified by matrix-assisted laser desorption/ionization–time-of-flight. Antimicrobial susceptibility was determined by disk diffusion methods. A polymerase chain reaction was performed to detect mecA, mupA gene, and SCCmec types. Of the 154 samples, 10(6.5%) isolates were identified as CNS (5 Staphylococcus epidermidis, 2 Staphylococcus capitis, 2 Staphylococcus, and 1 Staphylococcus haemolyticus). Among the 10 isolates, 6 were resistant to penicillin, 5 were resistant to gentamicin, 3 were resistant to tetracycline, and 2 were resistant to cefoxitin and erythromycin. However, clindamycin, ciprofloxacin, teicoplanin, and trimethoprim-sulfamethoxazole resistant isolates were not present. Genes encoding mecA were detected in 4 (2 S. warneri and 2 S. haemolyticus) isolates, and mupA in 1 (S. epidermidis) isolate. One methicillin-resistant CNS (S. warneri) isolate was determined as being of the SCCmec type I. It is concluded that CNS resistant to various antimicrobial agents was widely distributed in dental clinic environments in Korea.

  • 광역도시계획은 전략계획, 정책계획, 지침계획의 성격을 지니는 공간계획으로 청사진 계획으로 운용되어 온 기존의 광역개발계획 및 도시기본계획과 구분된다. 광역도시계획이 정하는 사항은 도시기본계획 및 관련 하위계획에서 기본적 취지를 검토하여 집행됨으로써 이들 계획에 대하여 지침 (guideline)적 성격을 가지나, 직접적으로 개별 개발행위나 토지이용행위를 구속하지는 않는다. 행정중심복합도시 광역도시계획에 대한 전략환경평가는 국토해양부 시행지침에서 제시한 절차를 준용하되, 계획수립 여건을 고려하여 평가계획 수립, 항목과 범위 설정(scoping), 환경성평가, 보고서 작성, 보고서 검토, 모니터링 및 사후평가의 6단계로 진행하였다. 전략환경평가 시 구체적인 항목과 범위, 평가방법 등은 계획의 범위와 성격, 내용 등에 대해 대안의 비교 검토가 가능하도록 4가지 사항을 중점적으로 고려하였다. 즉, 정책목표 달성방법의 환경 적 적정성, 대안의 설정, 자원과 에너지 이용의 효율성, 계획이 지구환경에 미치는 영향 등이다. 계획의 환경성 제고를 위해 설정한 항목은 인구, 토지이용계획, 교통계획, 녹지관리 등 4가지이며, 평가과정에서 반복 및 환류를 통하여 계획과 평가간의 연계성을 도모하였다. 이 과정에서 공간구조 변화는 행정중심복합도시를 정점으로 하고 주변 연결거점을 3개로 늘려 조정하였다.

A Kinematical Analysis of Mae-ukemi (forward breakfall) in Judo (유도전방낙법의 운동학적 특성 분석[I])

  • The purpose of this study was to analyze kinematic variables of a Mae-ukemi (forward breakfall :MU) in Judo through 3-D image analysis with five Judoka with over 2 years' career (2nd dan, 3rd dan). The kinematic variables include posture, and distance variables; the mean values and the standard deviations for each variable were obtained to analyze kinematic variables of a MU in Judo. From the data analysis and discussion, the following conclusions were drawn : 1) Posture variables : The angles of wrists during right before Mae-ukemi (RBU) were extended from the just MU position(MUP) and flexed from in the right after MU(RAU), and there was a difference in the displacement between the angles of the right and left wrists. The angles of elbows were flexed in the ready position from in the RAU. The angles of ankles were flexed from the ready position to that down position, extended from the down position to that transition, and flexed from the transition positiont to that RAU. 2) Distance variables : The distance height between the mat and shoulders in the RBU(32.2cm) to in the MUP(18.3cm) and in the RAU(20.5cm). during that height between the mat and hips in the RBU(48.0cm) to in the MUP(23.4cm), and in the RAU(30.6cm).

A Cell-Based Assay System for Monitoring NF-$\kappa$B Activity in Human Epidermal Keratinocytes: A Screening Tool of the Antioxidants and Anti-inflammatories for Dermatological Purpose

  • A cell-based assay system for monitoring NF-$textsc{k}$B activity was developed to determine the influence of activated NF-$textsc{k}$B in human HaCaT cells. The pNF-$textsc{k}$B-SEAP-NPT plasmid that permits expression of the secreted alkaline phosphatase (SEAP) reported gene in response to the NF-$textsc{k}$B activity and contains neomycin phosphotransferase (NPT) gene for the geneticin resistance in host cells was constructed and transfected into human keratinocyte cell line HaCaT. Human HaCaT transfectant cells secreted the SEAP enzyme into the culture medium in a time-dependent manner until 72h. NF-$textsc{k}$B activities were measured in the SEAP reporter gene assay using a fluorescent detection method. The treatment of HaCaT cell transfectants with known antioxidants [e.g., N-acetyl-L-cysteine and vitamin C] showed inhibition of NF-$textsc{k}$B activity in a time-and concentration-dependent manner. The phorbol 12-myristate 13-acetate (PMA) known as a stimulator of NF-$textsc{k}$B expression demonstrated that it increased NF-$textsc{k}$B activity in a time- and concentration-dependent manner. This assay system could be used to determine the quantitative measurement of NF-$textsc{k}$B activity in the human skin and allow the screening of anti-inflammatory agents from various synthetic chemicals and natural products for dermatological purpose. Genomic Characterization and Safety Assessment of Bifidobacterium breve BS2-PB3 as Functional Food

  • Our group had isolated Bifidobacterium breve strain BS2-PB3 from human breast milk. In this study, we sequenced the whole genome of B. breve BS2-PB3, and with a focus on its safety profile, various probiotic characteristics (presence of antibiotic resistance genes, virulence factors, and mobile elements) were then determined through bioinformatic analyses. The antibiotic resistance profile of B. breve BS2-PB3 was also evaluated. The whole genome of B. breve BS2-PB3 consisted of 2,268,931 base pairs with a G-C content of 58.89% and 2,108 coding regions. The average nucleotide identity and whole-genome phylogenetic analyses supported the classification of B. breve BS2-PB3. According to our in silico assessment, B. breve BS2-PB3 possesses antioxidant and immunomodulation properties in addition to various genes related to the probiotic properties of heat, cold, and acid stress, bile tolerance, and adhesion. Antibiotic susceptibility was evaluated using the Kirby-Bauer disk-diffusion test, in which the minimum inhibitory concentrations for selected antibiotics were subsequently tested using the Epsilometer test. B. breve BS2-PB3 only exhibited selected resistance phenotypes, i.e., to mupirocin (minimum inhibitory concentration/MIC >1,024 ㎍/ml), sulfamethoxazole (MIC>1,024 ㎍/ml), and oxacillin (MIC >3 ㎍/ml). The resistance genes against those antibiotics, i.e., ileS, mupB, sul4, mecC and ramA, were detected within its genome as well. While no virulence factor was detected, four insertion sequences were identified within the genome but were located away from the identified antibiotic resistance genes. In conclusion, B. breve BS2-PB3 demonstrated a sufficient safety profile, making it a promising candidate for further development as a potential functional food.
