A Study on Screening of Oriental Medicines Against Antibiotics-resistant Staphylococcus aureus (항생제(抗生劑) 내성(耐性) 화농균(化膿菌)의 생육(生育)을 억제(抑制)하는 한약재(韓藥材) 탐색(探索)에 관(關)한 硏究(연구))

  • The various oriental herbal medicines, which have usually been used for treatment of reducing fever, purging intense heat and detoxication, were screened to determine the antibacterial activity and the minimal inhibitory concentration against antibiotics-resistant Staphylococcus aureus. The results obtained were as follows: 1. Among the 23 oriental medicines tested, the water-soluble extract of Coptis japonica showed very excellent antibacterial activity against antibiotics-sensitive and -resistant S. aureus. A little activity was found in the extracts of Scutellaria baicalensis, Eriobotrya japonica and Prunus mume against antibiotics-sensitive S. aureus and in that extracts of P. mume, Schizandra chinensis and S. baicalensis against antibiotics-resistant S. aureus. When C japonica was used, the high antibacterial activity was shown against Bacillus subtilis and other extracts showed a little or no antibacterial activity against B. subtilis and E. coli as a control. 2. The highest antibacterial activities against antibiotics-resistant S. aureus were found in the ethanol-soluble extract of C japonica. Other ethanol-soluble extracts of S, baicalensis, S. chinensis and P. mume showed a little activity against both antibiotics-sensitive and antibiotics-resistant S. aureus. 3. Among the medicinal herbs tested, the water- and ethanol extracts of C. japonica showed very excellent antibacterial activity against the pathogenic S. aureus and both Escherichia coli and Bacillus subtilis used as a control. 4. When the water-soluble extracts of C. japonica and S. baicalensis, minimal inhibitory concentrations (MICs) against antibiotics-sensitive S. aureus were $20mg/m{\ell}\;and\;30mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. scabriosaefolia were $7.5mg/m{\ell}\;and\;12mg/m{\ell}$, respectively. 5. For the MICs against antibiotics-resistant S. aureus, the water-soluble extracts of C. japonica and P. mume showed $15mg/m{\ell}\;and\;10mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. mume were $13mg/m{\ell}\;and\;20mg/m{\ell}$, respectively. As a result, the highest antibacterial activity was found in both water- and ethanol-soluble extracts of C. japonica against antibiotics-resistant S. aureus. And the water- and ethanol-soluble extracts of S. baicalensis, S. chinensis and P. mume showed comparatively high antibacterial activities against both antibiotics-sensitive and -resistant S. aureus.

Antimicrobial Activity of Prunus mume and Schizandra chinenis H-20 Extracts and Their Effects on Quality of Functional Kochujang (매실과 오미자 추출물의 항균성과 기능성 고추장의 제조)

  • Prunus mume and Schizandra chinenis H-20 were found to have antibacterial properties against B. subtilis, S. aureus, E. coli KCCM 11591, and P. aeruginosa KCTC 1750. Prunus mume and S. chinenis H-20 showed clear zones of $6{\sim}10\;mm$ for the microbes used, while jujube, Lycii fructus and pine needle showed clear zones of $0.1{\sim}1\;mm$. In the experiment on DNA action, S. chinenis H-20 completely inhibited the growth of B. subtilis, S. aureus, and E. coli KCCM 11591 in the beginning of cultivation and even when added 5 hr after initiating cultivation. Moreover, as the microbes of about $l0^8$ cfu were shown by this test to have strong antibacterial power, they will most likely have an effect in vivo. Prunus mume and S. chinenis H-20 extracts were added to kochujang and fermented at $30^{\circ}C$ for 35 days. Prunus mume kochujang showed a sensory score of 4.29 (somewhat good), suggesting that Prunus mume can he utilized as a functional food ingredient.

Analysis of Consumer Behavior toward and Preferences for Prunus mume (Maesil), the Chinese Plum (매실에 대한 소비자 선호 및 소비행태분석)

  • We sought to define the main motivational factors promoting consumption of Prunus mume products. We surveyed both specialized high-volume consumers and public consumers, and focused on consumption of and preferences for Prunus mume and Prunus mume products. We investigated how products were preferentially consumed, purchase experience, purchase location, purchase price, consumption experience, factors important in a purchase decision, preference factors, dissatisfaction factors, consumption outlook, comments on new processed foods, and activation of consumption. A total of 534 responses were received and data were analyzed using the SAS program (Version 9.1) of the Frequency test; Chi-square test was used to detect significant factors. A greater percentage of high-volume(compared with low-volume) consumers processed personal products at home. The public consumer tended to buy commercial products. However, public consumers who were older, who had higher incomes, and who were heads of households, tended to both buy commercial products and to prepare foods at home. The common purchase methods used by high-volume consumers were internet and direct marketing, but the public consumer frequented department stores and hypermarkets. High-volume consumers observed that commercial products were expensive, and such consumers seemed to be particularly cost-sensitive. The decisive factor triggering purchase decisions in high-volume consumers was the geographical origin of the fruit, whereas the public consumer was more concerned with taste. In public, positive factors were that the product was good for health and had a pleasant taste. Some of those surveyed complained of a lack of variety in Prunus mume products and that the amount of Prunus mume in certain products was low. To promote and increase consumption, convenient-to-eat products with healthy images are needed.

Angiogenesis-inhibiting Effects of Prunus mume Butanol Fractions on Human Umbilical Vein Endothelial Cells (매실 부탄올 분획물에 의한 혈관 신생 억제 효과)

  • Prunus mume Sieb. et Zucc is distributed throughout Asia and has traditionally been used as medicine and food. P. mume is known to contain large amounts of various organic acids, minerals, and phenol components. To date, the trend of P. mume research has focused only on the effects of antioxidant, anticancer and antibacterial, with only a few studies have focused on angiogenesis. Angiogenesis is a common characteristic of metastatic cancer through which oxygen and nutrients are delivered to the cells and tissues. In the present study, angiogenesis-inhibiting activity was investigated by evaluating the total polyphenol and flavonoid contents of the P. mume butanol fraction (PBF) and their ability to inhibit VEGF-induced human umbilical vein endothelial cells (HUVECs) proliferation, migration, invasion, and capillary formation. The polyphenols (12.81 mg GAE/g) and flavonoids (28.4 mg QE/g) of the PBF exhibited high antioxidant activity. The results of this study showed that PBF did not inhibit the proliferation of HUVECs at concentrations of 25-200 ㎍/ml and did not exhibit toxicity to normal cells. However, PBF inhibited the VEGF-induced mobility, invasion, and capillary formation of HUVECs. These results show that PBF inhibits the angiogenesis of HUVECs induced by VEGF. Therefore, PBF could serve as a therapeutic agent for the inhibition of angiogenesis.

Bioactivities of feral peach (Prunus persica Batsch var. davidiana Max.) flower extracts and Prunus mume flower extracts (개복숭아꽃과 매화꽃 추출물의 생물활성)

  • In this study, we investigated the antibacterial, antioxidant, and whitening effects of Prunus persica Batsch var. davidiana Max. (Feral Peach) and Prunus mume (white and pink) flowers. The extracts of the three kinds of flowers showed antibacterial activity against Staphylococcus. aureus. The chloroform fractions of the white Prunus mume, Feral Peach, and pink Prunus mume flowers exhibited antibacterial activities of 84, 49, and 30%, respectively, against Staphylococcus. aureus at a concentration of 0.5 mg/mL. The flower extracts of the three species also exhibited antibacterial effects against Pseudomonas. aeruginosa. The chloroform fractions of the Feral Peach and pink Prunus meme flowers exhibited antibacterial activities of 36 and 30%, respectively, at a concentration of 0.5 mg/mL. These extracts did not exhibit any significant antibacterial activity against Staphylococcus. epidermidis and Escherichia. coli. The extracts of the three kinds of flowers did not significantly affect the survival of HaCaT cells. The distilled water fraction of the pink Prunus mume flower extract exhibited antioxidant effects at concentrations of both 20 and 40 ㎍/mL. The ethyl acetate fraction of the pink Prunus mume flower extract exhibited an antioxidant activity superior to glutathione at a concentration of 40 ㎍/mL. The flower extracts did not significantly affect the survival rate of B16F10 cells. The chloroform fraction of the Feral Peach flower exhibited a whitening effect of 18% at a concentration of 40 ㎍/mL. Based on these results, we conclude that the three kinds of flower extracts are raw materials exhibiting antibacterial, antioxidant, and whitening effects.

The Anthelmintic Principle of "O-Mae", the Roasted Fruits of Prunus mume, Against Clonorchis sinensis (오매의 간디스토마 살충성 물질에 관한 연구)

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

Effect of Harvesting Time and Storage Duration of Viscum album Seeds on in vitro and ex vitro Germination on the Branch of Prunus mume

  • Viscum album var. coloratum (mistletoe) is considered as one of the endangered plant species in Korea. Our objective is to restore its population and multiplication of plant by ex situ method. In this research we explored the maximum germination (in vitro) of freshly harvested and stored seeds of mistletoe collected in different time intervals. Ex vitro germination after artificial inoculation on the branches of Prunus mume in different physiological conditions was also monitored. The research revealed that the lately harvested seeds (Feb. and March 2014) were superior over early harvested seeds (Nov. 2013 and Jan. 2014) of mistletoe due to the higher percentage of germination (above 93%). According to the data, it is also revealed that the survival and germination rate of mistletoe seeds decreased with the increase in storage duration. In ex vitro germination, the fluctuated temperature of a glass house in natural condition enhanced (four fold) the rate of germination on the branches of Prunus mume than the constant temperature condition in the glass house.

Antimicrobial Activities of Maesil(Prunus mume) Extract (매실농축액의 항균성 검색)

  • To develop natural food preservatives with Maesil (prunus mume), investigation of antimicrobial activities against food-related bacteria and yeast was conducted. Maesil (prunu mume) extract exhibited growth-inhibiting activities for most of the microorganisms tested. Minimum inhibitory concentrations (MIC) of maesil extract for Staphylococcus aureus and Bacillus subtilis were as low as 0.1mg/ml. The antimicrobial activity of the maesil (prunu mume) extract was reduced by heating or alkali treatment. Moreover, growth of Staphylococcus aureus was completely inhibited within 24 hours by the addition of at least 100 ppm of maesil extract. These findings suggest that maesil extract may play a role in the development of natural food preservatives.
