Inhibitory Effect of Lipid Bilayer Membrane on Protein Phosphatase 2A (Protein Phosphatase 2A의 활성화에 미치는 Lipid Bilayer Membrane의 저해 효과)

  • Protein phosphatase 2A was obtained from a cytosolic fraction of bovine brain homogenate. The phosphatase activity using phosphorylated histone Hl as substrate was suppressed in the presence of liposomes composed of dipalmitoylphosphatidylcholine(DPPC) or the mixture of phosphatidylserine and DPPC. The binding of protein phosphatase to liposome was indicated by the facts that the phosphatase activity of the supernatant of protein phosphatase/multilayer vesicle mixture was decreased with increasing amount of liposome, and that [$^{125}I$]-labeled protein phosphatase was coeluted with liposome. However, the affinity of the protein for phospholipid membrane was not so high. On the other hand, okadaic acid and liposome reduced the phosphatase activity synergistically, which means that okadaic acid binds neither to lipid membrane nor to the membrane-associated phosphatase, The inhibitory effect of liposome was, therefore, ascribed to association of the protein phosphatase 2A with the lipid bilayer membrane.

Construction of Membrane Sieves Using Stoichiometric and Stress-Reduced $Si_3N_4/SiO_2/Si_3N_4$ Multilayer Films and Their Applications in Blood Plasma Separation

  • The novelty of this study resides in the fabrication of stoichiometric and stress-reduced $Si_3N_4/SiO_2/Si_3N_4$ triple-layer membrane sieves. The membrane sieves were designed to be very flat and thin, mechanically stress-reduced, and stable in their electrical and chemical properties. All insulating materials are deposited stoichiometrically by a low-pressure chemical vapor deposition system. The membranes with a thickness of 0.4 ${\mu}m$ have pores with a diameter of about 1 ${\mu}m$. The device is fabricated on a 6" silicon wafer with the semiconductor processes. We utilized the membrane sieves for plasma separations from human whole blood. To enhance the separation ability of blood plasma, an agarose gel matrix was attached to the membrane sieves. We could separate about 1 ${\mu}L$ of blood plasma from 5 ${\mu}L$ of human whole blood. Our device can be used in the cell-based biosensors or analysis systems in analytical chemistry.

  • Ultrathin polyelectrolyte (PE) multilayer films prepared by the versatile layer-by layer (LbL) assembly method have been utilized for the preparation of light-emitting diodes, electrochromic, membrane, and drug delivery system, as well as for selective area patterning and particle surface modification because the various materials with specific properties can be inserted into the film with nano-level thickness irrespective of the size or the shape of substrate. Since the introduction of the LbL technique in 1991 by Decher and Hong, various hydrophilic materials can be inserted within LbL films through complementary interactions (i.e., electrostatic, hydrogen-bonding or covalent interaction). In this study, it is demonstrated that LbL SA multilayer films based on nucleophilic substitution reaction can allow the preparation of the highly efficient magnetic and/or optical films and nonvolatile memory devices.

  • A hollow fiber membrane containing BSA as ligand was Prepared by radiation-induced grafting GMA onto a porous polyethylene hollow fiber and subsequent reacting with DEA and TEA. The density of the DEA and TEA of the membrane were 3.4 mmol/g, 1.7r mmol/g, respectively. The DEA membrane exhibited a higher amount of than the TEA membrane. BSA was immobilized by the graft chains during the permeation of BSA solution throught the DEA and TEA membrane. The BSA was adsorbed in multilayer binding of 8 onto the DEA membrane whereas adsorption onto the TEA membrane remained constant. A two-stage stepwise BTC was observed due to independent chiral recognition for L, D-Trp solution by DEA-BSA membrane.

  • Membrane filtration processes are increasingly popular for drinking water treatment that requires high quality of water. Low pressure membrane(LPM) processes such as microfiltration(MF) and ultrafiltration(UF), however, are ineffective in the removal of dissolved organic matter and also membrane fouling is still an important issue to be resolved. High pressure membranes(HPMs) may guarantee better water quality, but at the high energy consumption. Thus, various approaches to combine LPM processes with other physicochemical methods have been recently made to achieve their efficiency to the level comparable to that of HPM processes. In this work, therefore, hybrid processes that coupled MF/UF with coagulation, adsorption, chemical reactions(e.g., chelation and oxidation) are reviewed regarding system design and performance and also membrane surface modifications conducted by grafting and polyelectrolyte multilayer formation were assessed.

  • 막분리에 의한 폐윤활유 재생공정을 개발하기 위한 기초 연구로써 폐윤활유 분리/재생용으로 적합한 복층(multilayer)세라믹 복합막의 제조와 합성막의 폐유 분리 효율등이 연구되었다. 결함이 없고 두께가 균일한 지르코니아 복합막 (기공크기 0.07 $\mu$m 이하)은 압출 성형법으로 제조한 튜브형 $\alpha$-알루미나 담체 (외경 7.8 mm, 두께 0.6 mm, 기공크기 0.7 $\mu$m)내부표면에 역침지 인상법(reverse dip-drawing technique)에 의하여 지르코니아 슬러리를 코팅 한 후 950$\circ$C에서 1시간 열처리하여 제조 되었다. 또한 지르코니아 복합막 위에 니타니아 졸-겔 코팅을 한 후 450$\circ$C에서 2시간 열처리하여 기공크기가 15 nm정도인 3층 복합막을 제조 하였다. SEM, Bubble Point Test, Mercury Porosimeter 그리고 분획 분자량 측정등에 의하여 복합막의 코팅층 두께, 결함유무 및 막의 기공크기등을 분석하였다.

  • Membranes are used in most processes of biopharmaceutical production. It is used for pretreatment of other processes, separation of impurities in the process, virus removal, control of products concentration and buffer solution exchange. Virus filters play an important role in ensuring product efficacy and stability because viral contamination of biopharmaceuticals for humans is a sensitive issue that is directly related to serious clinical outcomes. Virus filters typically have complex multilayer structures made of various polymers such as surface-modified PVDF, PES, CRC. Depending on the manufacturer, filters have different pore structures and shapes, such as symmetric or asymmetric, and is used in the form of pleated membrane, flat sheets or hollow fibers. Virus filters are exclusively supplied by few foreign companies such as Asahi Kasei, Millipore, Pall and Sartorius. Replacing virus filters can be time consuming and expensive, including approval from regulatory agencies through validation. As localization has become important due to Japan's recent export regulations, it is necessary to increase the degree of technical independence.

Islet function within a multilayer microcapsule and efficacy of angiogenic protein delivery in an omentum pouch graft

  • We have previously described a new multilayer alginate microcapsule system, and the goals of the present study were to assess the in vitro function of islets encapsulated in its inner layer, and the angiogenic ability of FGF-1 delivered from the external layer in an omentum pouch. Following isolation and culture, islets were encapsulated in the inner core of microspheres ($500-600{\mu}m$ in diameter) with a semi-permeable poly-L-ornithine (PLO) membrane separating two alginate layers, and both unencapsulated and encapsulated islet function was assessed by a dynamic glucose perifusion. For angiogenesis experiments, one group of microcapsules without FGF-1 (control) and another (test) containing FGF-1 with heparin encapsulated in the external layer were made. One hundred microcapsules of each group were transplanted in Lewis rats (n = 5/group) and were retrieved after 14 days for assessment of angiogenesis. Glucose perifusion of unencapsulated and encapsulated islets resulted in similar stimulation indices. The release of FGF-1 resulted in increased vascular density compared to controls. In conclusion, islets encapsulated in the core of multilayer alginate microcapsules maintain functionality and the microcapsule's external layer is effective in delivery of FGF-1 to enhance graft neovascularization in a retrievable omentum pouch.

Effects of Substrates on Nanofiltration Characteristics of Multilayer Polyelectrolyte Membranes (다층 고분자 전해질 막의 나노여과 특성에 미치는 지지체의 영향)

  • In a previous study, we probed the potential of poly(styrene sulfonate) (PSS)/poly(diallyldimethylammonium chloride) (PDADMAC) nanofiltration (NF) membranes for the separation of monovalent anions, with an emphasis on the selective rejection of $F^-$. Remarkably, deposition of $(PSS/PBADMAC)_4PSS$ films on porous alumina supports yielded membranes that exhibited $Cl^-/F^-$ selectivity > 3 with minimal $Cl^-$ rejection, and a solution flux of $3.5m^3/m^2$-day at 4.8 bar. When the number of PSS/PDADMAC bilayers was increased from 4.5 to 5.5, however, $F^-$ rejection decreased from 73% to 50% and $Cl^-/F^-$ selectivity dropped to 1.9. Addition of another bilayer to form $(PSS/PDADMAC)_6$ PSS films resulted in a significant increase in $Cl^-$ rejection to give essentially no $Cl^-/F^-$ selectivity. The decrease of selectivity with deposition of more than 4.5 bilayers was not expected and it was unclear whether this characteristic was substrate independent. In this study, to investigate the effect of substrates on NF performance of multilayer polyelectrolyte membranes, PSS/PDADMAC films were deposited on 50 kDa polyethersulfone (PES) ultrafiltration supports instead of porous alumina supports. The results indicate that, although fluoride rejection and the number of bilayers at which a maximum $F^-$ rejection occurs are different, the trend is similar for both types of substrates. Therefore, we can conclude that the M: characteristics of multilayer polyelectrolyte membranes may be substrate independent.

Hydrolysis of Urea by Immobilized Urease Membrane (우레아제(Urease) 고정막에 의한 요소(Urea)의 가수분해)

  • In this study, we examined the preparation and hydrolysis property of immobilized urease membrane to decompose harmful urea in the body and remove ammonia which was produced by its decomposition. Urease immobilized membrane was prepared by introducing anion-exchange group DEA into porous hollow-fiber membrane by radiation graft polymerization method, and immobilization of urease. When urease was immobilized at membrane introduced with anion-exchange group, the more increasing grafting rate, the more increasing immobilization amount. The result originates from the fact that a greater amount of protein was immobilized by forming a multilayer on the longer grafted chain. Meanwhile, the addition of the cross-linker was possible not only to suppress separation phenomenon produced during a washing process of immobilized urease membrane but also to enable the recycling of membrane. Urease Immobilized membrane with no separation phenomenon was prepared by cross-linking reaction for 5 h, and the hydrolysis rate of prepared urease immobilized membrane was over 98% and 50%, respectively, in 1 mol and 4 mol urea solutions.
