Breeding of mulberry variety "Shimgang" showing resistance to popcorn disease

  • We bred a mulberry cultivar named Shimgang, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four local places(Suwon, Chuncheon, Buan and Jinju) for five years from 2011. This variety 'Shimgang' belongs to Morus Microphylla Buckl. selected from seedlings imported from USA. Shimgang was resistant to popcorn disease and high yielding variety in fruit productivity by 111% compared to control cultivar 'Shimheung(Morus alba L.)' for three years. Although fruits of 'Shimgang' was smaller in size and lower in sugar content of mulberry fruits, it showed stronger resistance than that of 'Shimheung'. It is adaptable to every where except the places where cold and late frost damage happen frequently.

Morphological Characteristics and Physiological Effects of Mulberry Leaves and Fruits with Wild Varieties (횡성군 일대에서 채취한 야생뽕의 형태학적 특성 및 오디의 효능)

  • We conducted this study to investigate the morphological characteristics and physiological effects of mulberry leaves and fruits with wild varieties. Morphologically sampling materials were showed Morus bombycis type and Morus alba type characteristics. Water contents of mulberry fruits were higher than that of mulberry leaves. Sugar degree of sampling materials was 5.1∼22.7 Brix %. The yield of 85% MeOH extract from freezing dried Mulberry Fruits was 60.2% and Hexane, BuOH and H$_2$O fraction from MeOH extract were 2.8%, 4.6%, 92.3% respectively. In the cholesterol-induced hyperlipidemia rats administered with subfraction of mulberry fruit extract, total cholesterol and serum triglyceride were decreased in the MeOH extract group and H$_2$O soluble fraction group. Also H$_2$O soluble fraction group decreased GPT, GOT and LDH level. Therefore, the above results suggested that mulberry fruit with wild varieties can help to maintain normal liver functions and to protect hyperlipidemia.

  • Much attention has been focused on the activity of the natural antioxidants present in fruits and vegetables, because potentially these components may reduce the level of oxidative stress. Especially, mulberry leaves containing many natural components are considerable resource far natural antioxidants. The antioxidant capacity of mulberry leaves was investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The appropriate conditions for pre-treatments of mulberry leaves were 80% MeOH, 30 sec. vortex mixing. The antioxidant capacity of 16 varieties was 3303.4 nmol (ascorbic acid equivalents) at opening stage of five leaves in spring. The highest stage of antioxidant capacity (3708.0 nmol) and yield rate was just before the coloration stare with anthocyanin in fruits, whereas the lowest stage was middle of June (2231.6 nmol) and about two months growing stage after summer pruning (2064.6 nmol). But after summer pruning, the antioxidant capacity of mulberry.

Identification of Ciboria carunculoides RS103V, a Fungus Causing Popcorn Disease on Mulberry Fruits in Korea

  • The popcorn disease caused by sclerotia forming fungi reduces the productivity of mulberry fruits in world wide. In Korea, only two species (Ciboria shiraiana and Scleromitrula shiraiana) have been reported as the major causal organisms and their morphological features are also largely unknown. Hereby, we report the first identification of another species (i.e. Ciboria carunculoides) in Korea and detailed features of their anamorphic stage. Fungi dominantly associated with sclerotia were purely isolated from infected mulberry fruits under the microscope. PCR-amplified DNA encoding 5.8S rRNA displayed 100% similarity to Ciboria carunculoides. The anamorphic features exhibited the absence of true mycelia. Instead, very short, aseptated, branched conidiophores were directly emerged from sclerotia. Phialides were usually three in number from each conidiophore, ampuliform to navicular in shape, slightly curved and tapering towards the apex. Conidia were produced from phialides and mostly found as one celled, pear shaped, not hyaline with smooth to uneven surface walled. Diversely modified features in phialides formed pseudo-mycelial structures around the host tissue. Combined all, current study is the first report of C. carunculoides isolated in Korea and the foremost detailed description of its anamorph stage.

Effect of CaO Treatment on Quality Characteristics and Storage of Mulberry (Morus alba L.) Fruits in Yecheon

  • The effects of aqueous calcium oxide (CaO) treatment on the quality characteristics and shelf life of mulberry (Morus alba L.) were investigated. Mulberry fruits were immersed in 0, 0.5, 1, and $2g{\cdot}L^{-1}$ CaO solutions for 0, 1, 3, 6, and 12 min. Mulberries were then rinsed with potable tap water for 1 min and stored at $-1^{\circ}C$ for 14 days. CaO treatment was effective at promoting the retention of titratable acid, pH, and ascorbic acid as well as total flavonoid contents. CaO concentration and treatment time were significant factors affecting the sensory qualities of the fruits, including off-odor, flavor, and texture. For shelf life determinations, the total bacterial count was reduced by CaO treatment so that the samples treated with $1g{\cdot}L^{-1}$ CaO for 12 min had bacterial levels at 14 days comparable to those of the control at 4 day, and no coliform group was detected after CaO treatment. These results indicate that calcium oxide treatment is a promising approach for the preservation of mulberry fruit.

Fertility and Mulberry Fruit Characteristics of Three Korean Indigenous Mulberry Species (한반도에서 자생하는 뽕나무 3종이 자연교잡된 때의 稔性과 오디의 과실특성)

  • Three indigenous mulberry species of Morus tiliaefolia Makino, Morus mongolica C. K. Shneider and Morus bombycis Koidz. were naturally crossed and their fertilities and fruit characteristics were investigated during the years of 1996 and 1997. M. tiliaefolia Makino and M. mongolica C.K. Schneider showed low fertility, but M. bombycis Koidz. was medium in fertility. The fruits of M. tiliaefolia Makino were red-purple and midium in size and high sugar content, but those of M. mongolica C.K. Schneider were light red and small but extremely high sugar content. The total acidity of fruits was 0.73, 0.50 and 0.36-0.94% in M. tiliaefolia Makino, M. mongolica C.K. Schneider and M. bombycis Koidz. respectively. Regarding ripenening stage, M. mongolica C.K. Schneider was early and M. tiliaefolia Makino was medium, while M. bombycis Koidz was early or medium.

Identification of C3G(cyanidin-3-glucoside) from Mulberry Fruits and Quantification with Different Varieties (오디에서 C3G(cyanidin-3-glucoside)의 분리, 동정 및 계통별 함량분석)

  • This study was carried out to identify of C3G (cyanidin-3-glucoside) from mulberry fruits and quantify with different varieties. C3G of mulberry fruits was extracted with 1% HCl-MeOH and purified with open column (5${\times}$90cm) which filled with Amberlite IRC-50 ion exchange resin. The $\lambda$max ranges of the purified C3G on UV/vis spectrum were 516nm and 280nm. Also, molecular weight of C3G from mulberry fruits by LC-Mass was determined as 449. From above results, we concluded that anthocyanin pigment of mulberry fruits was C3G only. The cyanidin-3-glucoside (C3G) was separated and quantified by High Performance Liquid Chromatography (HPLC) system using a Nova-Pack C$\_$18/ column. Mean content of the 35 tested accessions was 0.89%. Also fruity characteristics as well as C3G content to select the desirable mulberry varieties for the production of fruit were researched and analyzed. We selected three suitable varieties such as 'Susungppong', 'Kangsun', and 'Jeolgokchosaeng(Chungpuk)'.

  • The morphological and chemical characteristics of leaves and fruits were observed in the three mulberry varieties, including Daeryugppong(Morus Lhou(Ser.) koidz), Kugsang 20(Morus Lhou(Ser.) koidz) and Cataneo(Morus alba L.). The leaf development in spring was earliest in Cataneo and latest in Kugsang. Content of total nitrogen and Mg in leaf was the highest in Cataneo while that of P2O5, K and Ca in Daeryukppong. Flowers started to bloom from May 8 and in full bloom around May 15. Mature fruits began to set from June 10 to 15 and lasted by July 10 in Cataneo. Average fruit weight was heaviest in Kugsang 20(3.52 g/fruit), while lowest in Daeryukppong(1.61 g/fruit). In fruits, glucose and fructose were the major sugars. Citric acid was the most abundant organic acid in three varieties with its average content from 0.8 to 0.14%. The major pigment in fruit was anthocyanin and its content varied among varieties. The stability of anthocyanin was evaluated under various pH, temperature, and sugar concentrations. Rutin was the major flavonol glycoside present in fruits and its content varied from 0.92 to 3.36 mg/gDW. Other flavonol glycosides such as isoquercitrin and quercitrin were also detected in fruit.

  • Though mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a new income crop. But, a stable production of mulberry was issued on damage by the weather disaster, so it is requires that agricultural disaster insurance import. So, in this study as a basic research for a importation of agricultural disaster insurance, the cultivation of mulberry for the production of mulberry fruits, the shape of tree, the number of a bearing fruit from each cultivar, the occurrence of mulberry fruit sclerotic disease, and a yield and the market price of mulberry fruits were investigated and that results obtained were as follows; The number of mulberry tree each 10a was abundant in Buan and Jeongeup region that cultivated a suitable cultivar on low cut shape, however, a large cultivated Gochang region was little. The number of a bearing fruit per 1m on the branch was highest in the variety of Cheongilppong at Gochang. Yield of mulberry fruits each 10a was Cheongilppong at Gochang, followed by Iksuppong at Sangju. Average sale price of the mulberry fruits was high in the order of Gwasang No. 2, Suwonppong, Iksuppong, and Cheongilppong.

  • Mulberry containing many bioactive components is very useful as functional food resource. Mulberry leaf ice cream, mulberry leaf tea, mulberry noodle, mulberry fruit jam, mulberry wine etc. have been developed in Korea. Because mulberry fruit has abundant natural pigments belonging to anthocyanins, we can expect their therapeutic and physiological effects. However, there are few reports about mulberry seed. So we studied on fatty acids of mulberry seed for the purpose of increasing the functional utilization of mulberry fruit. (omitted)
