오디의 추출 공정에 따른 성분 변화 및 분말 과립차의 관능 특성 (Sensory Characteristics of Granular Tea and the Components of Mulberry Fruit Extracts by Different Extraction Process)

  • In the present work, mulberry fruit extracts by four extraction processes, namely wet pressing extraction (WPE), hot-water extraction (HWE), enzymatic hydrolysis (EH), and lactic-acid bacteria fermentation (LBF) by Lactobacillus plantarum TO-2100, were analyzed for nutrients and functional compounds. The sugar contents of extracts by WPE, HWE, EH, and LBF were 12.0, 10.9, 14.5, and 14.3 brix, respectively, and the extraction yields by EH and LBF were 1.65 and 1.50 times higher than those by WPE. Among the organic acids, tartaric acid and malic acid contents were the highest in the extracts by WPE. Acetic acid was best extracted by LBF, and citric acid was best extracted by EH. Lactic acid was detected only in LBF. The extracts by EH showed the highest contents of all vitamins with an exception that the extracts by LBF showed the highest contents of the folic acid, vitamin B12, and vitamin C. We also noted that vitamin B group was not detected in the extracts by LBF. The extracts by EH showed the highest contents of all the amino acids, whereas LBF showed the lowest. Polyphenol contents of extracts by EH and LBF were 3.05 and 2.51 times more than those by WPE respectively. Anthocyanin contents were 7.66, 7.14 times higher for EH and LBF compare to WPE. We manufactured mulberry fruit granular teas with different compositions and tested them for their sensory characteristics. We found that 15% mulberry fruit extracts by enzymatic hydrolysis and 85% dextrin composition gave the most satisfactory result.

Comparison with various mulberry leaves' and fruit's extract in lipid accumulation inhibitory effect at adipocyte model

  • In relation to the global increase trend of obesity population, there is a demand for the development of foods having high functional activity by mass-extracting anti-obesity active substances using mulberry product such as leaf and fruit. Therefore, we evaluated the antiobesity efficacy according to varieties by using the mulberry leaves and fruit extracts. At the same time, high active varieties were selected. For this purpose, the effects of the extracts of the mulberry leaf and fruit on 3T3-L1 adipocyte differentiation were examined. As a result, in the case of mulberry leaves, the lipid accumulation inhibitory rate of 'Cheongolppong' was higher than that of the control at $500{\mu}g/ml$ treatment. And in the case of the extract of mulberry fruit, 'Daesim' showed the highest lipid accumulation inhibitory rate compared with the control at 50 times of diluted extract.

품종이 다른 뽕잎추출물의 Helicobacter Pylori에 대한 항균활성 탐색과 항산화 효과 (The Antimicrobial Activity against Helicobacter pylori and Antioxidant effect from the extracts of Mulberry leaves (Morus Alba L.))

  • 108종의 표본묘목으로부터 채취한 뽕나무잎을 이용하여 항산화효과와 Helicobacter pylort에 대한 억제효과를 측정하였다. 항산화효과 중 DPPH에 대한 전자공여능은 물 추출물에서는 양면상 등 13종의 뽕잎에서 80% 이상의 높은 전자공여능이 확인되었으며, 80% ethanol 추출물에서는 고수9 등 59종의 뽕잎에서 80% 이상의 높은 전자공여능이 확인되어, 물 추출물보다 상대적으로 높은 효과를 나타내었다. ABTS radical decolorlization을 측정한 결과 물 추출물과 80% ethanol 추출물 모두에서 대부분의 뽕잎 추출물들이 90% 이상의 높은 항산화 효과를 나타내었다. TBARS 값은 control값이 $0.493{\times}10^{-2}{\mu}M$로 나타났으며, 물 추출물에서는 청운뽕, 도내, 축무, 사천금, 수원상 1호, 불전 2호, 오노다마구와, 야마나까다까스께를 제외한 모든 품종에서 대조구보다 낮게 나타났으며, 80% ethanol 추출물에서도 대축면을 제외한 107종의 품종에서 대조구보다 낮은 TBARS 값을 나타내어 산화촉진인자를 binding하는 능력이 뛰어난 것으로 판단되었다. TBARS와 같이 지용성 물질의 항산화력을 나타내는 antioxidant protection factor(PF)에서는 많은 종류의 뽕잎 80% ethanol 추출물에서 2이상의 높은 PF를 나타내었으며, TBARS값과 PF값 모두 80% ethanol 추출물이 물 추출물보다 효과가 우수한 것으로 나타났다. 뽕잎 품종별 추출물로 Helicobacter pylori균을 저해하는 물질을 탐색한 결과 108종의 뽕잎 중에서 감락상, 다호조생, 신일뽕, 국상 27호, 개량십문자, 국부, 수중상, 백광 6호, 이상, 사천금, 영변추우, 사175, 당상 2호, 청목노상 및 수중상 등 15 품종의 뽕잎에서 저해환을 나타냈으며, 그 중 물 추출물에서는 다호조생 품종이 지름 13mm의 저해환을, 알코올 추출물에서는 청목노상 품종이 지름 15mm의 저해환으로 효과가 가장 우수하여 천연 항균제 및 생리활성 물질로의 이용가능성을 추측할 수 있었다.

Response of Mulberry Brown Leaf Spot Fungus Myrothecium roridum to Different Plant Extracts

  • Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.

식용대두유에 대한 뽕잎추출물의 항산화 작용 (The Antioxidative Activities of Mullberry leaves Extracts on Edible Soybean Oil)

  • Recently, the concern about safety and functional substances of foods are increased therefore antioxidant substances and plants which had pharmacological effect have been studied actively. It has been known that the mulberry leaf is effective in curing many diseases. Nowaday, the pharmachological effects of mulberry leaves on diabetes mellitus and their usage for many aspects were confirmed. Mulberry leaves are known for effective in prevention of diabetes mellitus, because of specific amino acids and fibers. In this study, methanol, hexane, chlorform, ethyl acetate, and butanol extracts obtained from mulberry leaves were added to soybean oil and they were stored for 30 days at $60{\pm}2^{\circ}C$ and peroxide value(POV) and conjugated diene value(CDV) were measured periodically. Results of this study were obtained as follows; 1. The POV of soybean oil after the addition of each mulberry leaves powder(MLP) extracts generally enhanced as the storage time was prolonged, so the POV of all samples was reached higher than 100meq./kg.oil after 10 days storage without the addition of butanol, methanol, ethylacetate, hexane extracts at 0.1% level. Especially, the POV of soybean oils including butanol extract was 87.35meq/kg.oil after 10 days storage and antioxidant activity of butanol extract was shown to be superior to that of BHT. The pattern of the changes of the CDV of soybean oil after the addition of MLP extracts at 0.02%, 0.05% and 0.1%, respectively, were almost constant during 10 days of storage and then rapidly increased during the rest of experimental periods. During 10 days of storage in case of 0.1% adding level, the antioxidant activities of the butanol extract was superior to that of the each MLP extracts.

Physiological and Whitening Effects of Morus alba Extracts

  • Mulberry extracts can be incorporated into skin-whitening products. The compound attributed to lighten the skin is arbutin, a form of hydroquinone that inhibits melanin release by suppressing the tyrosinase enzyme. For the cosmetic applications, the physiological effects of mulberry (Morus alba) extracts were investigated. The water soluble fraction of mulberry contains higher amount of protein (16.28~4.47%) in contrast to fat (1.55~1.41%). In addition, the fraction abundantly contains succinic acid (972.4-275.8 mg/g) and phosphoric acid (1,628.4-121.9 mg/g) in different parts of mulberry. The free radical scavenging ability in water soluble fraction was found to display remarkable effects in comparison with methanol and ethyl acetate fraction. The ethyl acetate-soluble of root and leaf showed remarkable tyrosinase inhibition activity by IC 50 (${\mu}g/ml$). The anticancer activity of methanol fraction obtained from mulberry using human cancer cell lines showed growth inhibition effect (270.14 mg/ml in Calu-6 cells, 295.29 mg/ml in HCT-116, and 332.29 mg/ml in MCF-7 cells, respectively). Based on the results, Morus alba extracts include cosmetic ingredients with antioxidizing and whitening properties.

뽕잎의 유산발효에 의한 Piperidine Alkaloid 함량 증진 (Enhancement of Piperidine Alkaloid Contents by Lactic Acid Fermentation of Mulberry Leaves (Morus alba L.))

  • This study was carried out to investigate solid-state fermentation method using cellulolytic lactic acid bacteria Lactobacillus plantarum TO-2100 in order to increase piperidine alkaloid contents in mulberry leaves. Piperidine alkaloid, one type of which include 1-deoxynojirimycin (1-DNJ), is known to inhibit ${\alpha}$-glycosidase activities. Using this strain, the optimal solid-state fermentation conditions on mulberry leaves powder were found as the following: initial moisture content, temperature and relative humidity were 20%, $30{\sim}35^{\circ}C$ and 60 ~ 70%, respectively, and the fermentation time was 72 hrs. The piperidine alkaloid contents in the fermented mulberry leaves were 2.86% on dry powder, which is 7-fold increase from that of non-fermented mulberry leaves. The 1-deoxynojirimycin contents after applying preparative thin layer chromatography were 2.02% on dry powder, which is 8 times higher than that of non-fermented mulberry leaves. ${\alpha}$-Glycosidase activities was inhibited by 65.7 ~ 84.7% with 3 ~ 5% treatments of hot-water extracts of the fermented mulberry leaves, compared to 16.2 ~ 40.2% with 3 ~ 5% treatments of hot-water extracts of non-fermented mulberry leaves. Therefore, the results suggest that solid-state fermentation method does indeed increase of piperidine alkaloid contents on mulberry leaves.

Screening of Botanicals Against Root Knot Disease Complex in Mulberry (Morus indica L.)

  • For developing an integrated eco-friendly package against root knot disease complex of mulberry caused by the association of Meloidogyne incognita with Fusarium solani and F. oxysporum causing serious loss in terms of leaf yield and quality during cultivation, twenty botanical extracts at 5, 10 & 20% concentrations were screened under in vitro conditions. Among the extracts, Allium sativum followed by Lasownia inermis were found to be effective at 20% concentration against both the virulent fungi and nematode. Both the extracts reduced the mycelial growth of virulent fungi to an extent of 76-100%, inhibited the hatching of nematode eggs by 80-90% and 76-85% larval mortality over the control. The other extracts were found either moderately or poorly effective in reducing the growth of fungi, hatching of nematode eggs and enhancing the mortality of larvae. The two effective botanical extracts, which rated as strong inhibitors against both nematode and virulent fungi, can be utilized in developing an integrated ecofriendly technology for better management of root knot disease complex in mulberry.

Evaluation of Certain Plant Extracts and Antagonists Against Fusarium solani and Alternaria tenuissima, the Incitants of Root Rot and Die-Back Diseases of Mulberry

  • The studies have been conducted to control the soil borne fungal pathogens viz, Fusarium solani (Mart) Sacc. and Alternaria tenuissima the incitants of root rot and die-back diseases on mulberry stem cuttings planted in the mulberry nurseries and also in established mulberry gardens ten plant extracts with 10% concentration except Lantana camara (undiluted) were tested through poisoned food technique and four biofungicides were also screened by dual culture method under in vitro conditions. Plant extract of Prosopis juliflora showed the maximum inhibition on the mycelial growth (81.2% over A. tenuissima and 80.0% over F. solani) and followed by L. camara (66.7% over A. tenuissima and 68.9% over F. solani). Among the antagonists Pseudomonas fluorescens and Trichoderma viride showed maximum inhibition on the mycelial growth of both pathogenic fungi. The promising plant extracts (P. juliflora and L. camara) and antagonists (P. fluorescens and T. viride) were tested against both the pathogenic fungi under in vivo conditions along with the existing popular chemical Mancozeb. All the tested plant products and bio-fungicides showed inhibitory effect on both fungi. But the maximum survival percentage of mulberry cuttings was recorded in the treatment with T. viride (95% against F. solani and 90% against A. tenuisssima) followed by P. fluorescens (90% against both fungi) and T. harzianum (80% against F. solani and 85% against A. tenuisssima). Incase of the treatments with plant extracts and chemical fungicide the P. juliflora (60% against F. solani and 55% against A. tenuisssima) showed higher survival percentage and followed by L. camara (55% against F. solani and 50% against A. tenuisssima) and Mancozeb (55% against both fungi). In case of control only 10% of survival was recorded in F. solani inoculated cuttings and 15% survival in A. tenuissima inoculated cuttings.

Antioxidant Activities and Phenolic Compounds Composition of Extracts from Mulberry (Morus alba L.) Fruit

  • The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity
