혐기처리한 뽕잎의 혈당강하 효과 (Hypoglycemic Effect of Mulberry Leaves with Anaerobic Treatment in Alloxan-induced Diabetic Mice)

  • The hypoglycemic effect of mulberry leaves using alloxan-induced diabetic mice was investigated. The hypoglycemic effect of mulberry leaves was significantly affected with anaerobic incubation up to for 6 hrs. The body weight of mice treated with 20% anaerobic conditioned mulberry leaves was higher than that in the control group. Mulberry leaves which were treated with anaerobic condition significantly decreased blood glucose level in alloxan-induced type I diabetes mice. Furthermore, glucose tolerance was significantly recovered by feeding mulberry leaves anaerobically treated. Pathological analysis revealed that the disappearance of insulin-secreting beta-cells in the islets of Langerhans from the alloxan induced diabetic mice was strongly inhibited in the mulberry leaves feeding group. The content of 1-deoxynojirimycin increased up to 5% in detached mulberry leaves incubated under nitrogen gas for 6 hrs. Nevertheless, it is not clear whether the synthesis of 1-deoxynojirimycin is increased or the degradation of it is decreased. Overall, results obtained from this study indicate that the hypoglycemic effect was increased by the mulberry leaves with anaerobic treatment.

The Effects of Mulberry Fruit on the Antioxidative Defense Systems and Oxidative Stress in the Erythrocytes of Streptozotocin-Induced Diabetic Rats

  • The current study examined the effects of mulberry fruit on the antioxidative defense systems and oxidative stress in the erythrocytes of diabetes-induced rats. Sprague-Dawley male rats were randomly assigned to one normal and three streptozotocin (STZ)-induced diabetic groups. 1be diabetic groups were fed a mulberry fruit-free diet (DM-group), 0.3% mulberry fruit diet (DM-F group) or 0.6% mulberry fruit diet (DM-2F group). Diabetes was induced with STZ after three weeks of the experimental diets. 1be rats were sacrificed 9 days later for examination of the antioxidative defense systems and oxidative stress in the erythrocytes. Means of cy-3-Ο-glucopyranoside, cy-3-Ο-rutinoside, rutin, isoquercitrin, quercetin, morin and dehydroquercetin contents were 230.45, 131.5, 142.5, 10.3, 5.8, 1.6 and 3.83mg per l00g dry weight, respectively, in the mulberry fruit. Mulberry fruit strengthened the antioxidative defense systems through increased activity of the antioxidant enzymes, such as glutathione peroxidase (GSH-px) and catalase (CAT), in the erythrocytes of the diabetes-induced rats. Accrdingly, mulberry fruit was found to reduce the accumulation of thiobarbituric acid reactive substance (WARS). Therefore, mulberry fruit was found to be excellent for strengthening the antioxidative defense system and reducing damaging oxidative substances in the erythrocytes of the diabetes-induced rats.

  • 뽕잎 분말이 1, 10%가 되도록 사료에 첨가시켜 4주동안 SD 흰쥐에게 급여한 후 이들의 대장 내용물로부터 총균수와 주요장내미생물의 생균수를 배지를 이용하여 혐기배양한 후 측정한 결과 총균수는 뽕잎 투여군에서 대조군에 비해 감소하는 경향을 보였다. 그리고 유해균으로 분류할 수 있는 Clostridium은 대조군보다 뽕잎투여군에서 유의성있게 감소하였고, E. coli와 Staphylococcus도 감소하는 경향을 보였다. 뽕잎 메탄올 추출물과 분획물들의 미생물의 생장억제효과를 측정한 결과, 헥산분획과 물분획물이 Clostridium perfringens에 대하여 억제효과를 보였다. 장내균총의 개선 측면에서 뽕잎은 기능성 식품소재로서의 이용가능성이 있는 것으로 나타났다.

상엽 추출물을 이용한 항당뇨 음료의 개발 (I) - 상엽 추출물의 제제화 탐색 - (Development of An Anti-Diabetic Functional Drink (I) - Screening of the Manufacturing of Mulberry Leaf Extract -)

  • 본 연구는 천연 자원인 뽕나무에서 혈당강하 효능을 가진 뽕잎(상엽)의 열수 추출법을 이용하여 상엽 추출물의 공정화 확립과 Formulation 개발을 위해 상엽 추출물과 silk peptide 및 한약재를 첨가한 시제품을 동물실험을 통하여 Formulation을 최종 결정하여 제품화하여 기능성 음료를 개발하고자 하였다. 연구 결과, 상엽 추출의 최적 조건은 상엽 크기는 3mm 이하로 하고, 2/3 정도 packing된 면포자루로 포장된 상엽을 사용하며, 상엽 사용량은 50~100 g/L, 추출 온도는 9$0^{\circ}C$, 추출 시간은 2 hr으로 하였으며 이때 수율은 20~25%였다. 또한 시료 배합에 따른 혈당강하 실험을 통하여 상엽 추출물과 silk peptide 의 농도는 각각 1%, 0.1%일 때 혈당강하 작용이 가장 우수하였으며 부재료로 사용된 둥글레는 상엽추출물의 10% 첨가한 경우가 혈당강하작용이 가장 컸으며, polydextrose도 효과가 있으므로 첨가하는 것이 바람직하다고 사료된다.

  • This study was carried out to investigate the proximate composition , free sugars,amino acids, organic acid, minerals and extracted color of the fruit of paper mulberry (Broussonetia Kazinoki Siebold) . the proximate compositins were 6.1% of moisture, 15.9% of crude protein, 28.5% of crude fat, 8.9% of crude ash and 40.6% of carbohydrates in the fruit of paper mulberry. Free sugars showed low content as 0.1% of glucose , 0.1% of sucrose, 0.1% of fructose and 0.01% of sorbitol, respectively. In amino acid composition of the fruit of paper mulberry, glycine was the highest in the content, followed by methionine, aspartic acid and alanine, The ratio of essential /total mino acids was 0.43 . The richestmineral contained in the fruit of paper mulberry was Ca and followed by K and Mg. Optical density (490-500nm) of the extracted color with 50% ehtanol and 80 % ethanol were 0.75 and 0.30 , respectively.

Biological Control of Mulberry Root Knot Nematode Meloidogyne incognita by Trichoderma harzianum

  • Trichoderma harzianum-THN1 parasitising the egg masses of root knot nematode Meloidogyne incognita was isolated from galled mulberry roots and evaluated for its potential to control root knot disease. In pot experiments root galling was reduced and leaf yield increased significantly following soil treatment with T. harzianum-THN1. The extracts obtained from the soils inoculated with T. harzianum-THN1 drastically inhibited the hatching of nematode eggs and the effect was irreversible even after the eggs were transferred to fresh water. The fungus was equally effective in controlling the disease in nematode infested mulberry garden under field conditions which was significant over the most commonly used egg parasitic fungus Paecilomyces lilacinus. The disease reduction recorded with T. harzianum was on par with the plants treated with the nematicide Carbofuran. The results suggest that T. harzianum- THN1 could be used as a potent ecofriendly biocontrol agent against M. incognita in mulberry without any residual toxicity to silkworms. T. harzianum- THN1 can form an important component of integrated disease management package in mulberry cultivation.

뽕잎차의 제조와 그 품질 특성 (Preparation of Mulberry Leaves Tea and Its Quality Characteristics)

  • An optimal manufacturing process was established for the mass-production of mulberry leaves tea. The produced tea contained abundant dietary fibers, $\beta$-carotenes, and calcium. Also the polyphenol content and antioxidant activity of mulberry leaves tea were higher than those of simply dried mulberry leaves. These results imply that this tea manufacturing process is effective to utilize the useful components of mulberry leaves.

지각과 상엽 혼합추출물이 지질 개선에 미치는 영향 (The Improvement of Mixed Extracts of Fructus Aurantii and Mulberry Leaves on Lipid Metabolism)

  • Background : Diabetes is a chronic disease characterized by deficiency of insulin. It has been reported that Fructus aurantii and mulberry leaves have the effect of anti-diabetes. Methods : To investigate the improvement of mixed extracts of Fructus aurantii and mulberry leaves on lipid metabolism, we performed double-blinded, randomized and placebo-controlled multi-center clinical trial. Results : Mixed extracts of Fructus aurantii and mulberry leaves significantly reduced total cholesterol. Conclusion : These results suggest that mixed extracts of Fructus aurantii and mulberry leaves improve lipid metabolism.

건조 및 초저온 처리에 의한 뽕나무 종자의 장기 보존 (Cryopreservation of Mulberry(Morus) Seeds in Liquid Nitrogen(L$N_2$))

  • To investigate the possibility of crypreservation of mulberry seeds in liquid nitrogen(LN$_2$), characteristics of the seeds were examined after picking mulberry syncarps and drying-heat treatment. Storage in LN$_2$has the potential of providing indifinite preservation of valuable seed germplasm. Determining the tolerance of seeds among given cultivars to LN$_2$cooling and subsequent rewarming is the first step to establishing the feasibility of LN$_2$storage. Seeds of 4 mulberry varities were treated to LN$_2$(-196$\^{C}$) for 24 hours after drying heat treatment. Seed moisture content of Daeryukppong was the highest. As moisture content of mulberry seed was below 1%, storage in LN$_2$was safe. And drying heat treatment for 60 minutes was suitable to prevent decreased germination rate and germination vigor of seeds. The seeds of Cheongilppong were unsuibable to cryopreserve in LN$_2$for longterm storage.

뽕잎추출물이 Zucker Rat의 체지방 축적에 미치는 효과 (The Effect of Mulberry-Leaf Extract on the Body Fat Accumulation in Obese fa/fa Male Zucker Rats)

  • This study was conducted to investigate whether the obesity of obese fa/fa male Zucker rats can be ameliorated by mulberry leaves. There was no significant difference between mulberry treatment group and control group in body weight gain and food efficiency ratio. It also showed decreases on plasma triglyceride and total-cholesterol concentration compared to the control group. But there was no change in plasma leptin concentration between the control and mulberry treatment groups. These results suggest that mulberry leaves reduce serum lipid levels, but do not effect the plasma leptin concentration.
