뇌의 기능적 국소화를 위한 전산화 신경심리 검사의 개발 (Development of Computerized Neuropsychological Tests for Functional Localization of Brain)

  • 정신과 임상에서 편리하게 사용할 수 있는 전산화 신경심리 검사 시스템을 개발하였다. 이 시스템은 뇌의 전반적인 인지 기능과 뇌의 기능적 국재화를 평가할 수 있는 14개의 신경심리 검사로 구성되어 있다. 개발된 시스템은 멀티미디어와 객체 지향 개념에 의해 설계, 구현되었고, 마이크로 소프트 Windows에 기반을 둔 그래픽 인터페이스를 채용하여 운용이 용이하도록 하였다. 또한 마우스와 터치 스크린을 이용하여 피검사자로부터 입력을 손쉽게 받아드릴 수 있도록 하였으며, 색채 단어 검사, 범주 검사, 그리고 언어 기억 검사, 언어 지각 검사 등은 한글화하여 검사의 효율성을 향상시켰다. 개발된 시스템을 뇌손상 환자와 정신과 환자에 적용한 결과 그 인지 장애와 뇌의 기능적 국재화를 평가하는데 유용하게 사용될 수 있었다.

자웅마우스 장기내 Hg 축적에 미치는 Se과 마늘의 영향에 관한 연구 (A Study on the Effect of Selenium and Garlic on the Accumulation of Mercury in Male and Female Mouse)

  • To study on the effect of selenium and garlic on the toxicity of MMC(methylmercury chloride), the change of body, liver, kidney and brain weight and the concentration of mercury in liver, kidney, brain and blood are measured to each group between male and female. Results obtained are as follows: 1. In female, the body weight of MMC(5mg/kg) single treatment group and MMC(5mg/kg) and selenium (0.3mg/kg) combined treatment group and significantly lower than that of control group from the 4th week, but the body weight of MMC(5mg/kg), selenium (0.3mg/kg) and garlic(1g/kg) combined treatment group is similar to that of control group. 2. For the relative weight of kidney, only MMC(5mg/kg), selenium(0.3mg/kg) and garlic(1g/kg) combined treatment female group is lower than control femal group. For the relative weight of brain, MMC, selenium and garlic combined treatment male and female groups are lower than control group, respectively. 3. When male and female mice were injected with single MMC, the concentrations of mercury in kidney, brain, blood and liver are 4.721 ppm, 4.079 ppm, 1.946 ppm and 1.196. ppm for male and 7.715 ppm, 4.527 ppm, 2.657 ppm and 1.994 ppm for female, respectively, on the 5th week. 4. When male and female mice were injected with MMC, selenium and.garlic, the accumulations of mercury in kidney, brain, blood and liver are 29.04%, 33.91%, 54.47% and 12.12% for male and 25.74%, 22.64%, 24.31% and 36.21% for female lower than that of MMC single treatment group, respectively, on the 5th week. 5. When male and female mice were injected with MMC and selenium, the accumulation of mercury in brain is 16.55% about male and 11.86% about'female higher than that of MMC single treatment group, respectively, on the 5th week.

Ginsenoside Rg1 Attenuates Neuroinflammation Following Systemic Lipopolysaccharide Treatment in Mice

  • Objectives : Neuroinflammation is characterized by microglial activation and the expression of major inflammatory mediators. The present study investigated the inhibitory effect of ginsenoside Rg1 ($GRg_1$), a principle active ingredient in Panax ginseng, on pro-inflammatory cytokines and microglial activation induced by systemic lipopolysaccharide (LPS) treatment in the mouse brain tissue. Methods : Varying doses of $GRg_1$ was orally administered (10, 20, and 30 mg/kg) 1 h before the LPS injection (3 mg/kg, intraperitoneally). The mRNA expression of pro-inflammatory cytokines in the brain tissue was measured using the quantitative real-time PCR method at 4 h after the LPS injection, Microglial activation was evaluated using western blotting and immunohistochemistry against ionized calcium binding adaptor molecule 1 (Iba1) in the brain tissue. Cyclooxigenase-2 (COX-2) expressions also observed using western blotting and immunohistochemistry at 4 h after the LPS injection, In addition, double-immunofluorescent labeling of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and COX-2 with microglia and neurons was processed in the brain tissue. Results : $GRg_1$ (30 mg/kg) significantly attenuated the upregulation of TNF-${\alpha}$, interleukin (IL)-$1{\beta}$ and IL-6 mRNA in the brain tissue at 4 h after LPS injection. Morphological activation and Iba1 protein expression of microglia induced by systemic LPS injection were reduced by the $GRg_1$ (30 mg/kg) treatment. Upregulation of COX-2 protein expression in the brain tissue was also attenuated by the $GRg_1$ (30 mg/kg) treatment. Conclusion : The results suggest that $GRg_1$ is effective in the early stage of neuroinflammation which causes neurodegenerative diseases.

산수유(山茱萸) 메탄올 추출물이 편측 경동맥 폐색으로 유도된 생쥐의 허혈성 뇌손상에 미치는 영향 (The protective effect of methanol extract of Corni Fructus on brain injury caused by unilateral common carotid artery occlusion in mice)

  • Objectives : This study was conducted to evaluate the effects of Corni Fructus, the dried fruits of Cornus officinalis Sieb., on unilateral common carotid artery occlusion (UCCAO) in mouse model. Methods : The Corni Fructus used in the experiment was extracted with anhydrous methanol, then filtered and freeze-dried. C57BL/6 mice used in the experiments were conducted left UCCAO surgery to set up UCCAO rodent model for mice. The mice were divided into five groups for evaluate the effect of methanol extract of Corni Fructus (COM) on UCCAO induced ischemic brain injury. The expression levels of nitric oxide in cerebrum and serum, body weight change were measured. To determine the effect of UCCAO and COM administration on brain neurons, morphological changes of the cerebrum through a microscope was conducted. And western blot was performed to confirm the underlying mechanism of neuroprotective effect of COM administration. Results : COM administered UCCAO groups (CO50, CO150, and CO500) had no significant effects on nitric oxide production in ipsilateral hemisphere proteins and sera. The CO500, 500 mg/kg COM administration, attenuated UCCAO-induced p38 inflammatory signaling pathway and inflammatory mediators such as iNOS and COX-2. The CO500 group showed resilient morphological changes of hippocampus neuronal cells about brain damage caused by decreased flow of blood. These group also showed decreased inflammation and cellular stress response in neuronal cells. Conclusions : From these results, COM has a neuroprotective property via moderating inflammatory factors and cellular stress inducing factors in brain cells.

Therapeutic effects of selective p300 histone acetyl-transferase inhibitor on liver fibrosis

  • Liver fibrosis is caused by chronic liver damage and results in the aberrant accumulation of extracellular matrix during disease progression. Despite the identification of the HAT enzyme p300 as a major factor for liver fibrosis, the development of therapeutic agents targeting the regulation of p300 has not been reported. We validated a novel p300 inhibitor (A6) on the improvement of liver fibrosis using two mouse models, mice on a choline-deficient high-fat diet and thioacetamide-treated mice. We demonstrated that pathological hall-marks of liver fibrosis were significantly diminished by A6 treatment through Masson's trichrome and Sirius red staining on liver tissue and found that A6 treatment reduced the expression of matricellular protein genes. We further showed that A6 treatment improved liver fibrosis by reducing the stability of p300 protein via disruption of p300 binding to AKT. Our findings suggest that targeting p300 through the specific inhibitor A6 has potential as a major therapeutic avenue for treating liver fibrosis.

6-Shogaol, an Active Ingredient of Ginger, Improves Intestinal and Brain Abnormalities in Proteus Mirabilis-Induced Parkinson's Disease Mouse Model

  • Parkinson's disease (PD) which has various pathological mechanisms, recently, it is attracting attention to the mechanism via microbiome-gut-brain axis. 6-Shogaol, a representative compound of ginger, have been known for improving PD phenotypes by reducing neuroinflammatory responses. In the present study, we investigated whether 6-shogaol and ginger attenuate degeneration induced by Proteus mirabilis (P. mirabilis) on the intestine and brain, simultaneously. C57BL/6J mice received P. mirabilis for 5 days. Ginger (300 mg/kg) and 6-shogaol (10 mg/kg) were treated by gavage feeding for 22 days including the period of P. mirabilis treatment. Results showed that 6-shogaol and ginger improved motor dysfunction and dopaminergic neuronal death induced by P. mirabilis treatment. In addition, they suppressed P. mirabilis-induced intestinal barrier disruption, pro-inflammatory signals such as toll-like receptor and TNF-α, and intestinal α-synuclein aggregation. Moreover, ginger and 6-shogaol significantly inhibited neuroinflammation and α-synuclein in the brain. Taken together, 6-shogaol and ginger have the potential to ameliorate PD-like motor behavior and degeneration of dopaminergic neurons induced by P. mirabilis in mice. Here, these findings are meaningful in that they provide the first experimental evidence that 6-shogaol might attenuate PD via regulating gut-brain axis.

Harpagophytum Procumbens Suppresses Lipopolysaccharide Induced Expressions of Cyclooxygenase-2 and Inducible Nitric Oxide Synthase in Mouse BV2 Microglial Cells

  • The excessive release of proinflammatory products by activated microglia causes neurotoxicity, and this has been implicated in the pathogenesis of neurodegenerative diseases. Harpagophytum procumbens (Pedaliaceae) has been widely used for the treatment of pain and arthritis in the clinical field. In this study, we investigated the effect of Harpagophytum procumbens against lipopolysaccharide-induced inflammation. From the present results, the aqueous extract of Harpagophytum procumbens was shown to suppress prostaglandin-E2 synthesis and nitric oxide production by inhibiting the lipopolysaccharide-stimulated enhancement of cyclooxygenase-2 and inducible nitric oxide synthase expressions in mouse BV2 microglial cells. These results suggest that Harpagophytum procumbens may offer a valuable means of therapy for the treatment of brain inflammatory diseases by attenuating lipopolysaccharide-induced prostaglandin-E2 synthesis and nitric oxide production.

General Pharmacology of PEG-Hemoglobin SB1

  • PEG-hemoglobin SB1 (SB1), which is a hemoglobin-based oxygen carrier, is intended to use as a safe blood substitute against brain ischemia and stroke. The general pharmacological profiles of SB1 were studied. The doses given were 0, 5, 10, 20 ml/kg and drugs were administered intravenously. The animals used for this study were mouse, rat and guinea pig. SB1 showed no effects on general behavior, motor coordination, spontaneous locomotor activity, hexobarbital sleeping time, anticonvulsant activity, analgesic activity, blood pressure and heart rate, left ventricular peak systolic pressure, left ventricular end diastolic pressure, left ventricular developing pressure, double product, heart rate, coronary flow rate, smooth muscle contraction using guinea pig ileum, gastrointestinal transport, gastric secretion, urinary volume and electrolyte excretion at all doses tested except the decrease of body temperature. These findings demonstrated that SB1 possesses no general pharmacological effects at all doses tested.

Human placental extract suppresses lipopolysaccharide-induced expressions of cyclooxygenase-2 and inducible nitric oxide synthase in mouse BV2 microglial cells

  • Human placental extract (HPE), which is prepared from the placenta of healthy pregnant females, has been widely used in clinical field. HPE is known to possess anti-inflammatory, anti-viral, anti-oxidative, anti-mutagenic, and analgesic properties. In this study, the effect of HPE against lipopolysaccharide (LPS)-induced inflammation was investigated. From the present results, HPE was shown to suppress prostaglandin E2 synthesis (PGE2) and nitric oxide (NO) production by inhibition on the LPS-stimulated enhancement of the cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expressions in mouse BV2 microglial cells. These results suggest that HPE may offer a valuable mean of therapy for the treatment of brain inflammatory diseases by attenuating LPS-induced PGE2 and NO production.

  • The purpose of this study is to investigate the biological effects in black mouse by neutron irradiation at HANARO reactor in KAERI. Neutrons readily penetrate the charged field of an atomic nucleus because they are electrically neural. And so it can fight cancer with the radiation released when an atom of the element boron absorbs a neutron. The main patient in BNCT facility is brain cancer and sometimes skin cancer in foreign countries until now. Therefore, mice were laid flat and so irradiated at the direction of the front. (omitted)
