오가피전탕액(五加皮煎湯液)이 에탄올에 의해 유발되는 TM3 mouse Leydig 세포주의 apoptosis에 미치는 영향 (Protective effects Acanthopanax senticosus extracts on ethanol-induced apoptosis in TM3 mouse Leydig cells)

  • 목적: 오가피가 mouse testis에서 유래된 Leydig 세포주에서 에탄올에 의해 유발된 아폽토시스에 미치는 영향을 조사하였다. 방법: TM3 세포주에서의 아폽토시스 변화를 관찰하기 위해서 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, 4,6-diamidino-2-phenylindole (DAPI), DNA fragmentation assay, 및 reverse transcription-polymerase chain reaction (RT-PCR) 방법을 이용하였다. 결과: MTT assay를 이용하여 분석한 결과 농도에 따른 세포독성의 효과가 에탄올 투여부터 관찰되었다. 또한 오가피로 전처치하고 에탄올을 처치하였을 때 세포독성이 크게 감소되었다. DAPI staining에서 오가피 투여군은 에탄올 투여군에 비해서 fragmentation이 억제되었다. RT-PCR의 분석에 의하여 caspase-3 mRNA 발현이 오가피 투여군은 알코올 투여군보다 유의성있게 억제됨을 보여주었다. 결론: TM3 Leydig 세포주에서 에탄올에 의해 유발된 아폽토시스는 전형적인 세포사멸 형태를 나타내었다. 반면에 오가피 투여군은 에탄올에 의해서 유발된 아폽토시스에서 세포보호 효과가 있음이 확인되었다.

Effects of Several Salt Marsh Plants on Mouse Spleen and Thymus Cell Proliferation Using MTT Assay

  • In the present study, we have tested the effects of 21 salt marsh plants on cell proliferation of mouse immune cells (spleen and thymus) using MTT assay in culture. The methanolic extracts of six salt marsh plants (Rosa rugosa, Ixeris tamagawaensis, Artemisia capillaris, Tetragonia tetragonoides, Erigeron annus, and Glehnia littoralis) showed very powerful suppressive effects of mouse immune cell death and significant activities of cell proliferation in vitro. Especially, the methanolic extract of Rosa rugosa was found to have fifteen times compared to the control treatment, demonstrating that Rosa rugosa may have a potent stimulation effect on immune cell proliferation. These results suggest that several salt marsh plants including Rosa rugosa could be useful for further study as an immunomodulating agent.

백굴채(白屈菜)가 손상된 배양척수감각신경세포에 미치는 영향 (Effects of Herbar Chelidonii on the Cultured Spinal Sensory Neurons Damaged by XO/HX)

  • Objectives and Methods : To evaluate the mechanism of oxidative damage by xanthine oxydase(XO) and hypoxanthine(HX)-induced oxygen radicals, MTT assay and NR assay were carried out after the cultured mouse spinal sensory neurons were preincubated for 4 hours with various concentrations of XO/HX. And the amount of total protein. neurofilament EIA. lipid peroxidation and LDH activity were measured, to evaluate the protective effect of Herbar Chelidonii(HC) water extract on cultured spinal sensory neurons damaged by XO/HX. after the cultured mouse spinal sensory neurons were preincubated with various concentrations of HC water extract for 3 hours prior to exposure of XO/HX. Results : XO/HX decreased significantly the survival rate of the cultured mouse sensory neurons by NR assay and MTT assay In proportion to concentration and exposed time. In proportion to concentration and exposed time on cultured spinal sensory neurons, XO/HX showed the quantitative decrease of neurofilament by EIA. the decrease of total protein amount by SRB assay and the Increase of lipid peroxidation as well as LDH. HC showed the quantitative increase of neurofilament and total protein, but showed the decrease of lipid peroxidation and LDH activity against the neurotoxicity of XO/HX. Conclusions : From the above results, it is concluded that XO/HX have a neurotoxic effect on cultured spinal sensory neurons and that the herbs extract, such as HC, prevent the toxicity of XO/HX effectively in that they decrease lipid peroxidation and LDH activity.

Baicalein이 Lipoteichoic acid로 자극된 RAW 264.7 mouse macrophages의 hydrogen peroxide 생성에 미치는 영향 (Effects of Baicalein on hydrogen peroxide productions in RAW 264.7 macrophages stimulated by lipoteichoic acid)

  • Objectives : The aim of this study was to investigate the effect of Baicalein (BA) on the production of hydrogen peroxide in lipoteichoic acid-stimulated RAW 264.7 mouse macrophages. Methods : Lipoteichoic acid-stressed RAW 264.7 mouse macrophages were incubated with baicalein at concentrations of 50 and 100 µM. Incubation time is 30 minutes, 2 h, 12 h, and 18 h. After incubation, The production of hydrogen peroxide in RAW 264.7 mouse macrophages was measured with dihydrorhodamine 123 assay. Streptococcus aureus lipoteichoic acid and Streptococcus pyogenes lipoteichoic acid were used as cell-stimulating lipoteichoic acid. Cell viabilities were measured with a modified MTT assay. Berberine, indomethacin, and gallic acid were incubated for the same time as the comparative materials. Results : BA at the concentration of 50 and 100 µM did not show cytotoxicity on RAW 264.7 mouse macrophages for 24 h incubation. For 30 minutes, 2 h, 12 h, and 18 h incubation, BA at the concentration of 50 and 100 µM significantly inhibited the production of hydrogen peroxide in RAW 264.7 mouse macrophages stimulated by Streptococcus aureus lipoteichoic acid (p < 0.05); also, BA at the concentration of 50 and 100 µM also inhibited the productions of hydrogen peroxide in RAW 264.7 mouse macrophages stimulated by Streptococcus pyogenes lipoteichoic acid significantly (p < 0.05). Conclusions : BA might have anti-bacterial activity related to its inhibition of hydrogen peroxide production in lipoteichoic acid-stimulated RAW 264.7 mouse macrophages.

SDK시제품(가칭)에 대한 변이원성시험 (Mutagenecity Test of SDK)

  • In order to evaluate the mutagenic potential of SDK(skin decontamination kit) produced by Agency for Defense Development(ADD), were performed Salmonella typhimurium reversion assay, chromosomal aberration test on chinese hamster ovarian cells and in vivo micronucleus assay using mouse bone marrow cells according to the established regulation of Korean Food and Drug Administration. In the reverse mutation test using Salmonella typhimurium TA98, TA100, TA1535 and TA1537 did not in-crease the number of revertant at any of the concentration tested in this study. SDK did not increase the number of cells having structural or numerical chromosome aberration in cytogenetic test. In mouse micronucleus test, no significant increase in the occurrence oj micro nucleated polychromatic erythrocytes were observed in ICR male mice intraperitoneally administered with SDK. These results indicate that SDK has no mutagenic effects under these experimental conditions.

천연감미료 스테비오사이드와 스테비올의 생체내, 시험관내 유전독성평가 (In vitro and In vivo Evaluation of Genotoxicity of Stevioside and Steviol, Natural Sweetner)

  • The standard operation procedure of mouse lymphoma L5178Y $tk^{+/-}-3.7.2C$ gene mutation assay (MOLY) has been regarded as a sensitive in vitro mammalian cell gene mutation assay that is capable of detecting clastogens as well as mutagens. Using MOLY, one of natural sweetner, stevioside (5mg/ml) and its aglycon, steviol ($340{\;}\mu\textrm{g}/ml$) were evaluated the mutagenicity. Stevioside and steviol did not induce mutagenicity in MOLY. On the other hand, stevioside (250mg/kg, B.W.) and steviol (200mg/kg, B.W.) were also evaluated their ability to induce micronuclei in regenerating hepatocytes and bone marrow cells of ddY mice. From these results, stevioside and steviol did not induce any mutagenic effect both MOLY and in vivo micronucleus test.

Genotoxicity Study on Khal, a Halocidin Derivative, in Bacterial and Mammalian Cells

  • Khal was a synthetic congener of halocidin, a heterodimeric peptide consisting of 19 and 15 amino acid residues detected in Halocynthia aurantium. This compound was considered a candidate for the development of a novel peptide antibiotic. The genotoxicity of Khal was subjected to high throughput toxicity screening (HTTS) because they revealed strong antibacterial effects. Mouse lymphoma thymidine kinase ($tk^{+/-}$) gene assay (MOLY), single cell gel electrophoresis (Comet) assay and chromosomal aberration assay in mammalian cells and Ames reverse mutation assay in bacterial system were used as simplified, inexpensive, short-term in vitro screening tests in our laboratory. These compounds are not mutagenic in S. typhimurium TA98 and TA100 strains both in the presence and absence of metabolic activation. Before performing the comet assay, $IC_{20}$ of Khal was determined the concentration of $25.51\;{\mu}/mL\;and\;21.99\;{\mu}g/mL$ with and without S-9, respectively. In the comet assay, Khal was not induced DNA damage in mouse lymphoma cell line. Also, the mutation frequencies in the Khal-treated cultures were similar to the vehicle controls. It is suggests that Khal is non-mutagenic in MOLY assay. And no clastogenicity was observed in Khal-treated Chinese hamster lung cells. The results of this battery of assays indicate that Khal has no genotoxic potential in bacterial or mammalian cell systems. Therefore, we suggest that Khal, as the optimal candidates with both no genotoxic potential and antibacterial effects must be chosen.

소 흉선 추출물의 암세포 증식 및 림프구에 미치는 영향 (Antitumor and Immunological Effects of Bovine Thymic Extract)

  • Thymic extract showed antitumor effect to sarcoma mice with higher dose$(200{\mu}g/mouse/day$ i.p., 4weeks) but not with low dose$(5{\mu}g/mouse/day$ i.p., 6 weeks). Direct cytotoxicities were exhibited against sarcoma 180, L1210 and MOLT-4 by MTT assay. The spleen weight of mice were increased but the number of circulating lymphocytes were not increased after long-term(2 weeks) administration of thymic extract. Evaluating the mitogenesis by MTT assay. $\%$ absorbance of human lymphocytes was not increased by thymic extract. Cell cycle statistics of S phase and $G_2/M$ phase was not increased in the presence of that by PI staining. The formation of rosette was induced, irrespectively of exposure time short-term(l hour) and long-term(2 weeks). The population of mouse blood T-cell to bind Lyt2-antimonoclonal antibody and to $L_2T_4$ were increased after administration of thymic extract$(2-200{\mu}g/mouse/day)$. From the above results, it is suggested that thymic extract exerts antitumor activity by stimulating T cells to differeniate in vivo but not in vitro.

조각자(皂角刺)의 간암세포주(Hep G2)에 대한 세포독성, Apoptosis 및 NO에 대한 실험 (Effect of Gleditsiae Spina on Hep G2 cells cytotoxicity and Apoptosis and No)

  • In this study, antineoplastic activity against human hepatocellular carcinoma cell line(Hep G2) was tested in Gleditsiae Spina. Gleditsiae Spina was extracted with water, and the cytotoxic activity was tested using a calorimetric tetrazolium assay(MTT assay), the apoptosis was tested using a DNA electrophoresis and flow cytometry. The nitric oxide production from mouse peritoneal macrophage was tested using a Griess method. Gleditsiae Spina extracts against the proliferation of Hep G2 cells not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$, and Gleditsiae Spina extracts not showed the cytotoxicity of mitomycin C and the cytotoxicity of cisplatin on Hep G2 cells. Gleditsiae Spina extracts aginist the proliperation of BALB/c 3T3 cells not showed cytotoxicity, the proliperation of mouse thymocytes and splenocytes not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$. Gleditsiae Spina extracts not showed nitric oxide production from mouse peritoneal macrophage in vitro. Gleditsiae Spina was administered orally for 7 days at 300mg/kg increased nitric oxide production from mouse peritoneal macrophage.
