Improvement of proliferation efficiency of strawberry 'Maehyang' treated by coconut water in tissue culture ('매향' 딸기의 조직배양 시 coconut water 처리에 따른 기내 증식효율 향상)

  • This experiment was performed to determine the concentration of coconut water that is most effective in promoting the growth of 'Maehyang' strawberry in tissue culture. Cultivars in this experiment consisted of 'Maehyang' grown in the presence of 0, 20, 40, 60, 80, 100 ml·L-1 of coconut water which was added to a medium mixed with BA 0.5 mg·L-1 and IBA 0.1 mg·L-1. Morphological variation tests and SSR detection with coconut water were conducted to determine variations in proliferation. The proliferation rate of 'Maehyang' strawberry improved with the coconut water treatment compared to non-treatment. In particular, the proliferation rate with 100 ml·L-1 coconut water treatment increased by about 4 times. When 'Maeyang' was treated with 100 ml·L-1 of coconut water, two morphological variants occurred without genetic variation. Therefore, the results suggest that 100 ml·L-1 of coconut water can be used to mass-produce "Maeyang" strawberry without causing genetic variations.

Morphological Variation of Winter Buds of Quercus variabilis BL. In Korea (굴참나무 천연집단(天然集團) 동아(冬芽)의 형태적(形態的) 변이(變異))

  • For the study of morphological variation in winter buds of Q. variabilis $B_L$. natural populations in Korea, 16 populations were selected through the country in consideration of latitude, longitude, altitude, and geographical characters. Thirty trees were randomly selected from each population and 30 terminal and 30 lateral buds were sampled below 1/3 crown length of each tree. Four morphological characters including length and width of terminal and lateral buds were measured. 1. Length and width of terminal and lateral buds were in the ranges 6.9~11.3cm, 3.0~3.7cm, 6.1~8.9cm, and 2.5~3.1cm, respectively. The coefficient of variation were in about 20% in all the characters investigated. 2. All the characters were significantly different among populations as well as among individuals within populations. The degree of contribution of variance among individuals within populations was higher than that among populations. 3. Length of terminal bud showed positive correlation with length and width of lateral bud; and width of lateral bud with width of terminal bud and length of lateral bud. Also, positive correlations were observed between longitude and width of terminal bud, between latitude and length of terminal and lateral bud. 4. Cluster analysis using complete linkage method for winter bud characters showed two groups to Euclidean distance 3.4. They were group I of population 8 and group II of populations 1, 2, 3, 4, 5, 6, 7, 9, 10, 11, 12, 13, 14, 15, and 16. However, group II was divided into two at Euclidean distance 1.5 that are a group including populations 1, 2, 7, 9, and 11(groupII-1) and the other group including populations 3, 4, 5, 6, 10, 12, 13, 14, 15, and 16(groupII-2).

Morphological Variation of Foxtail Millet (Setaria italica (L.) P. Beauv.) Germplasm Collected in Korea, China and Pakistan (우리나라와 중국, 파키스탄에서 수집한 조 계통들에 대한 형태적 변이)

  • To understand the morphological differentiation of the 26 accessions of Foxtail Millet collected in Korea (15 accessions), China (7 accessions) and Pakistan (4 accessions), we analyzed 9 morphological characteristics such as plant height, panicle length, leaf number, tiller number, heading time, seed weight and panicle color etc. Most accessions of foxtail millet collected in Korea showed late heading time, tall plant height and long conical panicles. While foxtail millet accessions of Pakistan showed early heading time, short plant height and short conical panicles. In case of Chinese accessions, some accessions of them showed similar characteristics with Korean accessions, and the other showed similar characteristics to Pakistan accessions. In ANOVA analysis, most of quantitative characteristics such as plant height, leaf number, internode number and heading time showed significant differences among foxtail millet accessions collected from Korea, China and Pakistan. Principal component analyses clearly discriminate foxtail millet accessions of Korea from those of China and Pakistan. In PCA analysis, most of quantitative characters such as panicle length, leaf number and internode number greatly contributed in positive direction, whereas several quantitative characters such as tiller number, seed weight and panicle color contributed in negative direction on the first axis. Thus, these morphological characteristics could be used to classify the foxtail millet accessions collected in Korea, China and Pakistan. The present results could expand our understanding of the morphological variation in foxtail millet accessions from Korea, China and Pakistan, and also could be useful for foxtail millet germplasm preservation.

Molecular Identification of Sagittaria trifolia and S. aginash Based on Barcode (바코드에 의한 보풀속(Genus Sagittaria L.) 보풀과 벗풀의 동정)

  • Since sulfonylurea (SU) herbicide-resistant Monochoria korsakowii in Seosan reclaimed land in 1998 was reported first, herbicide-resistant weed species and their area of occurrence have been steadily increasing. In recent years, Sagittaria trifolia resistant to SU herbicides has been reported in Gimhae, Gyeongnam province. While collecting S. trifolia for constructing barcode database of major weeds we were suspicious by the continuous variation in the leave morphology of the species. In order to identify the S. trifolia from S. aginash we barcoded the species collected from domestic using ITS and compared the sequence variation with the ITS sequences of S. aginash downloaded from NCBI. As a result, it was found that the plants collected from the domestic did not have any variation among individuals although they showed wide morphological variation. On the other hand, interspecific variation between S. trifolia and S. aginash was 4.6%. Plants that are difficult to identify using morphological characters can be identified quickly and accurately using the barcode technique. Herbicide-resistant weeds may require different management practices depending on the species even in the same genus. Domestic herbicide-resistant weeds are steadily increasing. Therefore, accurate identification of these species must be preceded for effective weed control.

Geosmin and Morphological Characteristics of Anabaena circinalis, Obtained from the Bukhan River (북한강에서 출현한 Anabaena circinalis의 형태학적 특성 및 지오스민(geosmin) 발생 양상)

  • This study was carried out in the Bukhan River in the summer of 2014 and 2015, to identify the relationship between geosmin and the morphological changes in Anabaena. Identification of Anabaena was conducted using morphological and molecular analyses. Anabaena in this study was similar to Anabaena circinalis, A. crass, and A. spiroides with regard to regular coils, vegetative cell, akinete shape, and size, hoever, it was distinguishabl from A. crass and A. spiroides because of its larger trichome coil size. Additionally, the sequences of phycocyanin (PC) gene from Anabaena showed a 99% genetic similarity with A. circinalis NIES-1647 strain. The coil diameter of trichome ranged from 106 to $899{\mu}m$, and the diameter and abundance showed an insignificant positive correlation (r=0.544, p<0.05). The result of relationship between the coil diameter and the cell number per 360-degree rotation was kept at $33.8{\pm}5.2$ cells per $100{\mu}m$ diameter despite variable diameter. The average geosmin concentrations in 2014 and 2015 were investigated to be 99 ng/L and 35 ng/L, respectively. A. circinalis cell density contributed considerably to the change in geosmin and was positively correlated with geosmin concentration (2014; r=0.599, p<0.01, 2015; r=0.559, p<0.01). Our results suggest that geosmin and coil diameter could be estimated with the help of cell density.

Morphological Examination of Zostera asiatica Miki (Zosteraceae) from Various Habitats (서식지에 따른 왕거머리말(Zostera asiatica Miki, Zosteraceae)의 형태적 특성)

  • The morphological variations and habitat environment of the Zostera asiatica Miki (Zosteraceae) were examined in the waters off the eastern coast of Korea and Akkeshi Bay of Japan. In this study, we report new habitat of Z. asiatica along the eastern coast of south Korea; Gonghunjin, Donghori, Youngdong and Chilpo. Z. asiatica is found at the depth of 8∼15 m in these areas. However, Z. asiatica grows at the depth of 3 m in Akkeshi Bay of Japan. The sediment of habitats in the eastern coast of Korea were principally composed of sand. The stems were 64.0∼75.7 cm long in mature plants with flowering shoots and the leaves were 11.0∼15.0 mm wide for Korean populations. Plants from Akkeshi Bay of Japan had stems of 187.0∼234.0 cm in length. The difference in size are considered to be due to the depth of habitats. In spite of the significant morphological difference between Korea and Japan population of Z. asiatica, there was no difference in ITS sequences.

Image Analysis of Micro Lesions According to Grid Frequency After Removal of Moire Artifact (Moire artifact 제거 후 그리드 주파수에 따른 미세병변의 영상분석)

  • Morphological information such as shape and margin of micro lesion is important information for diagnosis of disease in clinical imaging. In this study, we investigated the morphological changes of the micro lesions by comparing the contrast and area in grid suppressed DR images according to grid frequency. In the profile analysis of the image, the mass showed an average intensity variation of 8.6 ~ 72.4 after suppression, The higher the grid frequency, the more the contrast was increased. However, in the images obtained using 103 lp / inch, which is a grid frequency less than the sampling frequency, the contrast of the mass in the vertical direction decreased after suppression. In the binary image, the area change of the mass was also large. As a result, the shape, size, and margin of the mass changed. In the case of very small calcification, the higher the grid frequency is the larger the change in contrast, so that a clear image can be obtained in the post-suppression image. However, we could confirm that the margin of the lesion was blurred and the lesion was lost in some of the images using the 103 lp / inch grid. The higher the frequency of the grid, The change of the contrast of fiber occurred largely and clear boundary was confirmed. The decrease of the number of pixels was small and morphological change was small. In conclusion, when using a grid frequency that is not suitable for the sample frequency, morphological changes or lesion loss of micro lesions in the post- suppression image may give the possibility of misdiagnosis in diagnosis and differentiation of the image.

The morphological study of Galium L. (Rubiaceae) in Korea (분계분석을 이용한 한국산 갈퀴덩굴속(Galium L.) 식물의 외부형태학적 연구)

  • To clarify the morphological variations of Galium, its morphological characteristics were examined using maximum parsimony analysis. The key characteristics as well as the range of variation in each characteristic were investigated at the species level and were employed to elucidate the relationships between the taxa. This study confirmed that species belonging to genus Galium formed a monophyletic group and comprised two main clades. Sect. Cymogaliae and sect. Leptogalium are polyphyly groups, and other sections are monophyletic group. This study also suggests that the fruit hairs, the types of petal apexes, and the colors of the petals are the most valuable taxonomic characteristics for differentiating different sections. The numbers of the leaves and leaf shape provide useful taxonomic characteristics for the identification of different species.

Recent Morphological Changes off the Shoreface of Jinwoodo and Sinjado in the Nakdong River Estuary: 2007-2012 (낙동강 하구역 진우도와 신자도 전면의 최근 지형 변화: 2007년-2012년)

  • Recently, more attention has been paid to the geomorphological changes in the Nakdong River Estuary, because those changes are caused by artificial activities including weirs, reclamation and construction. In order to analyze quantitatively the recent geomorphological variability in the Nakdong River Estuary, we surveyed the depth and elevation of submarine topography near Jinwoodo and Sinjado from March 2007 to February 2012. A statistical method (based on Digital Shoreline Analysis System) and an Empirical Orthogonal Functions method were used to evaluate the morphological changes. According to the statistical variables (DCE, NDC, EPR, LRR), the highest amount and rate of accumulation were recorded around the Gadeokdo whereas the greatest amount of erosion appeared around the coast off the eastern part of Sinjado. In particular, a dynamic variation of morphology was clearly observed in the vicinity of the sub-tidal channel located between Jinwoodo and Sinjado, which seems to be attributable to channel migration. As a result of the EOF method, the first mode (48.7%) is most closely related to the pattern of morphological variability that might be associated with the westerly movement of sediment by longshore current. The spatial variability of the second mode (16.6%) was high in the shoreface of Sinjado, showing a 4-year periodicity of temporal variability. The strong correlation (coefficient 0.73) between the time coefficient and suspended sediment discharge from Nakdong River emphasizes the role of sediment discharge to deposition in this area. The spatial variability of the third mode (11.3%) was distributed mainly around the coast off the eastern part of Sinjado, which is related to the movement of the coastline of Sinjado. Based on the last 5 year's data, our results suggest that the study area is characterized on the whole by a depositional pattern, but the extent of sedimentation is different locally.

A Study on the Morphological Characteristics of Leaves and Fruit of Cudrania tricuspidata in Korea (국내 자생 꾸지뽕나무 잎과 열매의 형태적 특성에 관한 연구)

  • This study was conducted to classify the variation in leaf and fruit morphology among natural grown 16 Cudrania tricuspidata stands. As results of the analysis in C. tricuspidata stands total 134 Species were found and as results of the analysis in leaf morphology, it was grouped as Group IV. By the analysis of variance in length, width, and weight of fruits were differed in 15 C. tricuspidata stands. The length, width, and weight of fruits were higher in Jinan and Sunchang, than Gochang, Gimje, and Goheung. By the cluster analysis of 3 fruit morphology were classified as 3 Groups. According to the results of this study C. tricuspidata leaves and fruit morphological characteristics for each group were compared by analysis there were differences between the morphological characteristics and the characteristics of the fruit did not match.
