Phylogenetic and Morphological Characterization of Cladosporium perangustum Associated with Flyspeck on Shine Muscat Grapes in South Korea

  • The Shine Muscat is a table grape, popular in South Korea for its unique mango-flavor taste. Flyspeck is a disease that is characterized by small, black, and circular specks on the grape cuticle was first observed in several commercial orchards in Sangju, South Korea, in August 2019. Here we identified the causal agent of flyspeck based on an advanced diagnosis approach, comprised of both morphological and molecular analyses. Morphological characteristics of the cultures isolated from grape flyspeck were identical to the fungus Cladosporium perangustum. The concatenated sequences of ITS, ACT, and EF1-α were used for molecular phylogenetic analysis, BLAST searches along with Bayesian inference-based phylogeny, confirmed that the causal agent of grape flyspeck is C. perangustum. The cultured fungal isolates also produced flyspeck symptoms on healthy fruits in pathogenicity tests. To the best of my knowledge, this is the first documented evidence of any Cladosporium sp. producing flyspeck symptoms on any plant.

Morphological and Phylogenetic Characteristics of Tuber himalayense Collected from Rhizosphere of Quercus dentata in Korea

  • We collected the ascomata of Tuber species from the rhizosphere of Quercus dentata in Danyang, Korea. We observed the morphological characteristics of ectomycorrhizal roots and ascomata, and identified the species based on the results of the phylogenetic analysis conducted using the DNA sequences of an internal transcribed spacer, a large-subunit rDNA, translation elongation factor 1-α DNA (TEF1), and MAT. Finally, we identified the fungal species as Tuber himalayense B.C. Zhang & Minter, which has not been recorded previously in Korea. We evaluated the morphological characteristics and conducted phylogenetic analysis of the ascoma and mycorrhiza (associated with Q. dentata) of T. himalayense.

A Novel Acremonium Species Isolated from Air Samples in Korea

  • The aim of this study was to characterize a new fungal species, Acremonium conglutinatum, isolated from air samples collected in Wando, South Korea. Phylogenetic analysis based on the internal transcribed spacer and large subunit regions revealed its unique position within the genus Acremonium. The isolated strain displayed distinct morphological characteristics, including ellipsoid or bent-ellipsoid conidia formed in clusters on the phialides. These features differentiate the new species from closely related species within the genus. This study describes the morphological and molecular characteristics of A. conglutinatum and emphasizes its phylogenetic relationships with other Acremonium spp. The identification of this novel species contributes to our understanding of the diversity and ecological role of Acremonium.

Studies on Morphological Variation Among Provenances of a Rare Rhododendron micranthum in Korea (희귀 식물 꼬리진달래의 형태적 변이)

  • The objectives of this study, an analysis of the morphological characteristics among six provenances of a rare Rhododendron micranthum could be used for the conservation of gene resources and could provide information on superior trees selection. The following results were obtained. Approximately Mt.worak region showed larger values at petal character. On the other hand, Bonghwa region showed smaller values at petal character. Yeonha-ri region showed larger values at leaf character. On the other hand, Bonghwa region showed smaller values at leaf character. The results of principal component analysis (PCA) for morphological characteristics showed that the first for principal components(PC's) explained 41.6% of the total variation. From th third PC explained 81.5% of the total variation. The first PC was correlated with those characteristics that were mainly related to the Petal length (PL), Leaf length (LL) width (LW), Stigma length (SL). The second PC was correlated with the Petiole length (PTW), Anther length (AL). The third PC was correlated with the Flower pedicel length (FPL), Filament length (FL). Therefore, these characteristics was important to analysis of the variation for morphological characteristics among provenances of Rhododendron micranthum. Cluster analysis using single linkage method based on morphological characteristics showed that six provenances of Rhododendron micranthum could be clustered into three groups. Group I is Jicdong-ri, Group II is Mt.worak and Yeonha-ri, and Group III is Taeback, Bonghwa, and Samcheok. These results corresponded well with that of principal component analysis.

Analysis and evaluation of morphological and molecular polymorphism in the hybridization of Elaeagnus ×maritima and E. ×submacrophylla (잡종 기원 녹보리똥나무와 큰보리장나무의 형태학적 및 분자적 다양성 분석 및 평가)

  • The taxonomic identity of Elaeagnus ×maritima and E. ×submacrophylla (Elaeagnaceae) in Korea is unclear, yet they are presumed to be hybrid taxa based on their morphology. To determine their hybrid origins, a morphological analysis (field surveys and specimen examinations) and a molecular analysis involving two nuclear ribosomal DNA (nrDNA) regions (internal transcribed spacer and 5S non-transcribed spacer) and one chloroplast DNA (cpDNA) region (matK) were conducted. The morphological analysis revealed that E. ×maritima showed certain morphological similarities to E. glabra, whereas E. ×submacrophylla showed certain morphological similarities to E. pungens. However, the molecular analysis indicated that E. ×maritima exhibited additive species-specific sites of E. glabra and E. macrophylla in the nrDNA regions. Notably, E. ×submacrophylla showed various aspects, with some individuals exhibiting additive species-specific sites of E. pungens and E. macrophylla in the nrDNA and E. macrophylla sequences in the cpDNA regions, some individuals exhibiting E. macrophylla sequences in the nrDNA and E. pungens sequences in the cpDNA regions, and some individuals displaying E. macrophylla sequences in both the nrDNA and cpDNA regions, despite an intermediate morphology between E. pungens and E. macrophylla. These results indicate that these two species are of hybrid origin and frequently cross between parental and hybrid individuals.

Genetic Relationship of Genus Rosa Germplasm and Genetic Diversity of Rosa rugosa in Korea (장미속 유전자원의 유전적 관계와 국내 해당화의 유전적 다양성)

  • Fifty-nine accessions of 23 species in genus Rosa were collected, and 15 accessions of Rosa rugosa were collected throughout 10 regions of Korea. Their genetic relationship was investigated by using morphological analysis and RAPD marker. The morphological analysis was measured for 7 quantitative and quantified for 4 qualitative traits. RAPD analysis obtained a total of 959 polymorphic bands by using twenty primers. Morphological analysis classified most according to the rose section system except for several species. The cluster analysis of genus Rosa based on RAPD data could identify the subgenus $Platyrhodon$ and $Eurosa$. The subgenus $Eurosa$ separated five sections; $Gallicanae$, $Cinnamomeae$, $Pimpinellifoliae$, $Synstylae$ and $Caninae$. Correlation analysis between morphological and RAPD analysis showed low significance ($r$ = 0.35). The accessions of R. rugosa belonged to the section $Cinnamomeae$ clustered into three groups at genetic distance ranging from 0.28 on the base of RAPD analysis. In conclusion, the genetic relationship of the genus Rosa was consistent to the previously reported rose section system, and domestic collections of $R.$ $rugosa$ were separated from 3 groups on the base of RAPD marker.

Effect of Structural and Morphological Changes on the Conductivity of Stretched PANI-DBSA/HIPS Film

  • We studied the effect of structural and morphological changes on the conductivity of a stretched conducting polymer film. To improve the poor processability of polyaniline, we used dodecylbenzenesulfonic acid as both a surfactant and a dopant during emulsion polymerization, followed by blending with high-impact polystyrene. UV-Vis/NIR spectra were obtained to observe conformational changes, and SEM and AFM were used to investigate morphological changes. FT-IR dichroism was applied to determine the microscopic orientation, and XRD patterns were obtained for quantitative crystallinity analysis. The electrical conductivity (${\sigma}_{\parallel}/{\sigma}_{\perp}$) was measured as a function of draw ratio. We found a clear correlation between morphological changes and (${\sigma}_{\parallel}/{\sigma}_{\perp}$), especially at the stretching limit. The conductivity of the films can be modified according to the desired application by controlling their structure and morphology.

Molecular and Morphological Identification of a Muraenichthys gymnopterus (Ophichthidae: Anguilliformes) Leptocephalus Collected on Jeju island, Korea (한국 제주도에서 채집된 갯물뱀(뱀장어목: 바다뱀과) 엽상자어(Leptocephalus)의 형태 및 분자동정)

  • One leptocephalus (TL 59.9 mm) from the family Ophichthidae collected from Jeju Island, Korea, was identified using morphological and molecular methods. Our ophichthid leptocephalus was identified as belonging to the genus Muraenichthys based on morphological characters: 157 myomeres; the origin of the dorsal fin located a little in front of the anus; a distinct melanophore present on the opercle; and six gut swellings present. An analysis of 886 base pairs of the 12S rRNA mtDNA sequences showed that our leptocephalus must be Muraenichthys gymnopterus, because its sequences were concordant with those of an adult M. gymnopterus (d=0.001) and next to those of Muraenichthys sp. leptocephalus (d=0.034). Here, we are the first to describe the morphological characteristics of the M. gymnopterus leptocephalus.

Development of Electrocardiogram Identification Algorithm for a Biometric System (생체 인식 시스템을 위한 심전도 개인인식 알고리즘 개발)

  • This paper is about the personal identification algorithm using an ECG that has been studied by a few researchers recently. Previously published algorithm can be classified as two methods. One is the method that analyzes ECG features and the other is the morphological analysis of ECG. The main characteristic of proposed algorithm uses together two methods. The algorithm consists of training and testing procedures. In training procedure, the features of all recognition objects' ECG were extracted and the PCA was performed for morphological analysis of ECG. In testing procedure, 6 candidate ECG's were chosen by morphological analysis and then the analysis of features among candidate ECG's was performed for final recognition. We choose 18 ECG files from MIT-BIH Normal Sinus Rhythm Database for estimating algorithm performance. The algorithm extracts 100 heartbeats from each ECG file, and use 40 heartbeats for training and 60 heartbeats for testing. The proposed algorithm shows clearly superior performance in all ECG data, amounting to 90.96% heartbeat recognition rate and 100% ECG recognition rate.

A New Morphological Analysis for the Spoken Language Translation System (음성언어 번역 시스템을 위한 새로운 형태소 분석)

  • It is difficult to integrate the speech processing systems and machine translation system in the spoken language translation system by reason that each system uses its own data and basic processing unit. So, we need a common I/O unit which is used in the whole system. In this paper, we propose a Pscudo-Morpheme as the interface between speech processing systems and language translation system. We implement a morphological analysis system for Pseudo-morpheme. The speech processing system using this pseudo-morpheme can get better result than other systems using the phrase or the general morpheme. So, the quality of the whole spoken language translation system can be improved. The analysis-ratio of our implemented system is 98.9%. This is similar to the common morphological analysis systems.
