This study describes the ability of capillary electrochromatography (CEC) for the determination of imidacloprid and carbendazim in tomato samples. A novel liquid crystal crown ether modified hybrid silica monolithic column was synthesized, characterized and developed as separation column for the first time. Baseline separation of imidacloprid and carbendazim could be achieved using a mobile phase containing 90% (v/v) 20 mmol/L phosphate buffer (pH 7.0) and 10% (v/v) acetonitrile. The matrix matched calibration curves were linear with correlation coefficient $r^2$ > 0.9998 in the range of 0.20-10.00 mg/L. The limits of detection for imidacloprid and carbendazim were 0.061 and 0.15 mg/kg, respectively, which were below the maximum residue limits established by the European Union as well as Codex Alimentarius. Average recoveries for imidacloprid and carbendazim varied from 101.6-108.0% with relative standard deviations lower than 6.3%. This method was applied to the analysis of tomatoes collected from local markets.
The compressive strength of horizontal joints in precast concrete large panel structures depends on parameters such as grout and panel strength, detail of joint, joint moment, width of grout column etc. As the panels are only connected at the pocket, it results in the reduction of compressive strength area. The purposes of this study are to develop the suitable grout material the joint pocket and to enable us to evaluate structural capacity. The validity of the design formulas provides us more economic system in construction. Test results of 15 specimens show that the proper construction procedure and grouting material develop the sufficient compressive strength of the wall as monolithic system.
One of the shortcomings of seismic bridge design codes is the lack of clarity in defining the role of different seismic isolation systems with linear or nonlinear behavior in terms of R-factor. For example, based on AASHTO guide specifications for seismic isolation design, R-factor for all substructure elements of isolated bridges should be half of those expressed in the AASHTO standard specifications for highway bridges (i.e., R=3 for single columns and R=5 for multiple column bent) but not less than 1.50. However, no distinction is made between two commonly used types of seismic isolation devices, i.e., elastomeric rubber bearing (ERB) with linear behavior, and lead rubber bearing (LRB) with nonlinear behavior. In this paper, five existing bridges located in Iran with two types of deck-pier connection including ERB and LRB isolators, and two bridge models with monolithic deck-pier connection are developed and their R-factor values are assessed based on the Uang's method. The average R-factors for the bridges with ERB isolators are calculated as 3.89 and 4.91 in the longitudinal and transverse directions, respectively, which are not in consonance with the AASHTO guide specifications for seismic isolation design (i.e., R=3/2=1.5 for the longitudinal direction and R=5/2=2.5 for the transverse direction). This is a clear indicator that the code-prescribed R-factors are conservative for typical bridges with ERB isolators. Also for the bridges with LRB isolators, the average computed R-factors equal 1.652 and 2.232 in the longitudinal and transverse directions, respectively, which are in a good agreement with the code-specified R-factor values. Moreover, in the bridges with monolithic deck-pier connection, the average R-factor in the longitudinal direction is obtained as 2.92 which is close to the specified R-factor in the bridge design codes (i.e., 3), and in the transverse direction is obtained as 2.41 which is about half of the corresponding R-factor value in the specifications (i.e., 5).
본 연구는 플랫 플레이트의 전단보강체에 관한 연구의 일부이고, 슬래브-기둥 접합부의 수직전단력 실험에서 보강체의 종류에 따라 뚫림전단강도에 대한 영향을 확인하는 것을 목표로 한다. 제안된 4개의 보강체는 s/s bar, c/s bar, wiremesh, CFS이다. 전체 5개의 실험체를 제작하고 실험을 행하였으며, 슬래브의 제원은 $2.63{\ast}2.725.0.18m$이며, 중앙에 0.6${\ast}$0.8m의 단면을 가진 기둥을 가지고 있다. 실험결과를 요약하면 CFS 전단보강체는 콘크리트와의 부착이 충분히 되지 않아서 보강체가 보강력을 발휘하기 전에 보강체와 콘크리트가 분리되어 보강효과를 발휘하지 못하였다. CFS를 제외한 3개의 전단보강체는 슬래브-기둥 접합부의 뚫림전단 저항력을 증가시켜 높은 보강효과를 나타내었다. 전단보강이 되지 않은 실험체에 비하여 강도면에서는 16${\sim}$35%의 증가율을 보였고, 변형능력면에서는 35${\sim}$63%의 증가율을 보였다.
In order to obtain the impact of socket connection interface forms and socket gap sizes on the seismic performance of reinforced concrete (RC) socket prefabricated bridge piers, quasi-static tests for three socket prefabricated piers with different column-foundation connection interface forms and reserved socket gap sizes, as well as to the corresponding cast-in-situ reinforced concrete piers, were carried out. The influence of socket connection structure on various seismic performance indexes of socket prefabricated piers was studied by comparing and analyzing the hysteresis curve and skeleton curve obtained through the experiment. Results showed that the ultimate failure mode of the socket prefabricated pier with circumferential corrugated treatment at the connection interface was the closest to that of the monolithic pier, the maximum bearing capacity was slightly less than that of the cast-in-situ pier but larger than that of the socket pier with roughened connection interface, and the displacement ductility and accumulated energy consumption capacity were smaller than those of socket piers with roughened connection interface. The connection interface treatment form had less influence on the residual deformation of socket prefabricated bridge piers. With the increase in the reserved socket gap size between the precast pier column and the precast foundation, the bearing capacity of the prefabricated socket bridge pier component, as well as the ductility and residual displacement of the component, would be reduced and had unfavorable effect on the energy dissipation property of the bridge pier component.
This study presents code-compliant seismic details by addressing dry mechanical splices for precast concrete (PC) beam-column connections in the ACI 318-19 code. To this end, critical observations of previous test results on precast beam-column connection specimens with the proposed seismic detail are briefly reported in this study, along with a typical reinforced concrete (RC) monolithic connection. On this basis, nonlinear dynamic models were developed to verify seismic responses of the PC emulative moment-resisting frame systems. As the current design code allows only the emulative design approach, this study aims at identifying the seismic performances of PC moment frame systems depending on their emulative levels, for which two extreme cases were intentionally chosen as the non-emulative (unbonded self-centering with marginal energy dissipation) and fully-emulative connection details. Their corresponding hysteresis models were set by using commercial finite element analysis software. According to the current seismic design provisions, a typical five-story building was designed as a target PC building. Subsequently, nonlinear dynamic time history analyses were performed with seven ground motions to investigate the impact of emulation level or hysteresis models (i.e., energy dissipation performance) on system responses between the emulative and non-emulative PC moment frames. The analytical results showed that both the base shear and story drift ratio were substantially reduced in the emulative system compared to that of the non-emulative one, and it indicates the importance of the code-compliant (i.e., emulative) connection details on the seismic performance of the precast building.
최근 일련의 연구로부터, 중공이 있는 프리캐스트 콘크리트(hollewed precast concrete, HPC) 기둥를 사용할 경우, 일반적인 PC기둥에 비하여 접합부 콘크리트를 충실하게 채울수 있으며 이에 따라, 접합부의 고정도는 일체식 철근콘크리트(RC)와 같이 향상될 수 있음이 확인되었다. 그러나 충전콘크리트로 중공부가 채워진 후, HPC부재와 충전콘크리트의 강도가 서로 다르고 이 두 부재 사이에는 접촉면이 있음에 따라 합성구조와 같은 거동을 보이게 되며 이는 합성된 기둥의 구조적 거동과 강도에 영향을 미치게 된다. 이 논문에서는 HPC 기둥에서 중공의 크기와 중공부분 콘크리트의 채움 유무에 따른 기둥의 압축강도 시험을 실시하였다. 중공의 비율은 35, 50 그리고 59%이며 실험체의 양단을 단순지지 형태로서 지지한 뒤 중심압축력을 기둥의 상단에 작용시켰다. 또한 압축에 대한 HPC 기둥의 파괴거동을 묘사하기 위하여 유한요소 해석을 실시하였다. 실험 결과, 중공내부가 충전콘크리트로 채워진 기둥에서, PC와 충전콘크리트의 강도차이와 상관없이 중공률은 기둥의 초기강성에 영향을 미치지 않는 것으로 나타났다. 그러나 기둥의 압축강도는 중공률에 반비례하는 것으로 나타났다. 충전콘크리트가 채워지지 않은 HPC기둥의 구조성능은 중공의 직경에 밀접하게 관련이 있는 것으로 나타났으며, 특히, HPC의 두께가 지나치게 얇은 경우에는 국부적인 파괴가 전체 파괴를 지배하는 것으로 나타났다. 이러한 영향을 고려하여 HPC기둥의 압축내력을 산정할 수 있는 식을 제시하였다. 유한요소해석에서는, HPC와 충전콘크리트 사이의 접촉면을 고려할 경우, 해석 결과가 실험 결과와 좋은 대응을 보이는 것으로 나타났다.
Modem drug discovery requires rapid pharmacokinetic evaluation of chemically diverse compounds for early candidate selection. This demands the development of analytical methods that offer high-throughput of samples. Naturally, liquid chromatography / tandem mass spectrometry (LC-MS/MS) is choice of the analytical method because of its superior sensitivity and selectivity. As a result of the short analysis time(typically 3-5min) by LC-MS/MS, sample preparation has become the rate- determining step in the whole analytical cycle. Consequently tremendous efforts are being made to speed up and automate this step. In a typical automated 96-well SPE(solid-phase extraction) procedure, plasma samples are transferred to the 96-well SPE plate, internal standard and aqueous buffer solutions are added and then vacuum is applied using the robotic liquid handling system. It takes only 20-90 min to process 96 samples by automated SPE and the analyst is physically occupied for only approximately 10 min. Recently, the ultra-high flow rate liquid chromatography (turbulent-flow chromatography)has sparked a huge interest for rapid and direct quantitation of drugs in plasma. There is no sample preparation except for sample aliquotting, internal standard addition and centrifugation. This type of analysis is achieved by using a small diameter column with a large particle size(30-5O ${\mu}$m) and a high flow rate, typically between 3-5 ml/min. Silica-based monolithic HPLC columns contain a novel chromatographic support in which the traditional particulate packing has been replaced with a single, continuous network (monolith) of pcrous silica. The main advantage of such a network is decreased backpressure due to macropores (2 ${\mu}$m) throughout the network. This allows high flow rates, and hence fast analyses that are unattainable with traditional particulate columns. The reduction of particle diameter in HPLC results in increased column efficiency. use of small particles (<2 urn), however, requires p.essu.es beyond the traditional 6,000 psi of conventional pumping devices. Instrumental development in recent years has resulted in pumping devices capable of handling the requirements of columns packed with small particles. The staggered parallel HPLC system consists of four fully independent binary HPLC pumps, a modified auto sampler, and a series of switching and selector valves all controlled by a single computer program. The system improves sample throughput without sacrificing chromatographic separation or data quality. Sample throughput can be increased nearly four-fold without requiring significant changes in current analytical procedures. The process of Bioanalytical Method Validation is required by the FDA to assess and verify the performance of a chronlatographic method prior to its application in sample analysis. The validation should address the selectivity, linearity, accuracy, precision and stability of the method. This presentation will provide all overview of the work required to accomplish a full validation and show how a chromatographic method is suitable for toxirokinetic sample analysis. A liquid chromatography/tandem mass spectrometry (LC-MS/MS) method developed to quantitate drug levels in dog plasma will be used as an example of tile process.
