Monitoring of Polycyclic Aromatic Hydrocarbons in Sediments and Organisms from Korean Coast

  • Surface sediments (0- 5 cm), mussels (Mytilus coruscus and M. edulis) and oysters (Crassostrea gigas) were sampled at 20 stations in Korean coast during a period of February to July 2000. Samples were analyzed for polycyclic aromatic hydrocarbons (PAHs) content using gas chromatography coupled to mass spectrometer detector (GC/MSD). The sediment $\sum PAH$ concentration varied from 7 to 1,214 ng/g dry weight and the level of carcinogenic PAHs of six species ranged from 1 to 563 ng/g dry weight in surface sediments. In organisms, the levels of $\sum PAH$ were in the range of 63-876 ng/g dry weight and the concentrations of carcinogenic PAHs of six species were in the range of 4-582 ng/g dry weight. The highest PAH concentrations in sediments and organisms were in samples from Station 5 in Pohang coast and Station 8 in Jinhae coast, respectively. $\sum PAH$ concentrations in sediments and organisms collected from Korean coast were slightly low or comparable to those in other countries. The contribution of ring aromatic groups to sum of 16 PAHs in sediments and organisms showed a similar pattern for most stations. In sediments, the predominant contributions were four and five ring aromatics like fluoranthene, pyrene, benzo[a]anthracene, chrysene, benzo[b]fluoranthene, benzo[k]fluoranthene and benzo[a]pyrene. In the organisms, four ring aromatics like fluoranthene, pyrene, benzo[a]anthracene and chrysene were predominant PAH species. Several molecular indices such as phenanthrene/anthracene, fluoranthene/pyrene and LMW/HMW ratio were used to identify the origin of PAH contamination in sediments. The results indicate that PAHs of Korean coast were mainly of pyro1ytic contaminants with slight petrogenic input. A high correlation within individual PAH compound in marine sediments was observed, while correlation in organisms showed a highly significant relationship for heavier molecular weight PAHs.

Monitoring of Soil Bacterial Community and Some Inoculated Bacteria After Prescribed Fire in Microcosm

  • The soil bacterial community and some inoculated bacteria were monitored to assess the microbial responses to prescribed fire in their microcosm. An acridine orange direct count of the bacteria in the unburned control soil were maintained at a relatively stable level $(2.0\~2.7\times10^9\;cells/g^{-1}{\cdot}soil)$ during the 180 day study period. The number of bacteria in the surface soil was decreased by fire, but was restored after 3 months. Inoculation of some bacteria increased the number of inoculated bacteria sev­eral times and these elevated levels lasted several months. The ratios of eubacteria detected by a flu­orescent in situ hybridization (FISH) method to direct bacterial count were in the range of $60\~80\%$ during the study period, with the exception of some lower values at the beginning, but there were no definite differences between the burned and unburned soils or the inoculated and uninoculated soils. In the unburned control soil, the ratios of $\alpha-,\beta-\;and\;\gamma-subgroups$ of the proteobacteria, Cytophaga-Fla­vobacterium and other eubacteria groups to that of the entire eubacteria were 13.7, 31.7, 17.1, 16.8 and $20.8\%,$ respectively, at time 0. The overall change on the patterns of the ratios of the 5 subgroups of eubacteria in the uninoculated burned and inoculated soils were similar to those of the unburned con­trol soil, with the exception of some minor variations during the initial period. The proportions of each group of eubacteria became similar in the different microcosms after 6 months, which may indicate the recovery of the original soil microbial community structure after fire or the inoculation of some bac­teria. The populations of Azotobacter vinelandii, Bacillus megaterium and Pseudomonas fluorescens, which had been inoculated to enhance the microbial activities, and monitored by FISH method, showed similar changes in the microcosms, and maintained high levels for several months.

Near Infrared Spectroscopy for Diagnosis: Influence of Mammary Gland Inflammation on Cow´s Milk Composition Measurement

  • Nowadays, medical diagnostics is efficiently supported by clinical chemistry and near infrared spectroscopy is becoming a new dimension, which has shown high potential to provide valuable information for diagnosis. The investigation was carried out to study the influence of mammary gland inflammation, called mastitis, on cow´s milk spectra and milk composition measured by near infrared spectroscopy (NIRS). Milk somatic cell counts (SCC) in milk were used as a measure of mammary gland inflammation. Naturally occurred variations with milk composition within lactation and in the process of milking were included in the experimental design of this study. Time series of unhomogenized, raw milk spectral data were collected from 3 cow along morning and evening milking, for 5 consecutive months, within their second lactation. In the time of the trial, the investigated cows had periods with mammary gland inflammation. Transmittance spectra of 258 milk samples were obtained by NIRSystem 6500 spectrophotometer in 1100-2400 nm region. Calibration equations for the examined milk components were developed by PLS regression using 3 different sets of samples: samples with low somatic cell count (SCC), samples with high SCC and combined data set. The NIR calibration and prediction of individual cow´s milk fat, protein, and lactose were highly influenced by the presence of mil samples from animals with mammary gland inflammation in the data set. The best accuracy of prediction (i.e. the lower SEP and the higher correlation coefficient) for fat, protein and lactose was obtained for equations, developed when using only “healthy” samples, with low SCC. The standard error of prediction increased and correlation coefficient decreased significantly when equations for low SCC milk were used to predict examined components in “mastitis” samples with high SCC, and vice versa. Combined data set that included samples from healthy and mastitis animals could be used to build up regression models for screening. Further use of separate model for healthy samples improved milk composition measurement. Regression vectors for NIR mild protein measurement obtained for “healthy” and “mastitic” group were compared and revealed differences in 1390-1450 nm, 1500-1740 nm and 1900-2200 nm regions and thus illustrated post-secretory breakdown of milk proteins by hydrolytic enzymes that occurred with mastitis. For the first time it has been found that monitoring the spectral differences in water bands at 1440 nm and 1912 nm could provide valuable information for inflammation diagnosis.

Characteristics of Environmental Solar Ultraviolet Irradiance

  • Direct, continuous, and accurate measurements of solar ultraviolet irradiance (290-400 nm: UVR) have been carried out since 1990, by using both band-spectral ultraviolet-B (290-320 nm: UV-B) and ultraviolet-A (320-400 nm: UV-A) radiometers at Tokai University in Hiratsuka, Japan (35$^{\circ}$N, 139$^{\circ}$E). From our observations, the following findings are provided: 1) an increasing trend in solar UV -B from Oct. 1990 to Sept. 2000; 2) a regional comparison of solar UVR in Japan; 3) the distinct characteristics of UV-B and UV-A irradiance, such as diffuse property, daily and seasonal variation; and 4) human body protection against solar UVR. An increasing 10-year trend in global solar UV - B in Hiratsuka corresponded to a decrease in the total ozone amount measured at Tsukuba (36$^{\circ}$N, 140$^{\circ}$E), giving supportive evidence for a direct link between these two parameters. Furthermore, a strong correlation was found between solar UV-B and total ozone amount from results of UVR measurements at four Tokai University monitoring stations dispersed throughout Japan. Additional results revealed different diffuse properties in global solar UV and in global solar total (300-3000 nm: Total) irradiances. For example, in the global UVR, the diffuse component was dominant: about 80 % independent of weather, with more than 60 % of global UV-B, and more than 50% of global UV-A with even a cloudless clear sky. On the other hand, the portion of the diffuse in the global total irradiance was very low, less than 10 % on a cloudless clear day. Daily and seasonal variations of UV -B and UV -A irradiances were found to be quite different, because of the marked dependence of UV -B irradiance on the atmospheric ozone amount. Moreover, UV -B irradiance showed large daily and seasonal variations: the ratio between maximum and minimum irradiances was more than 5. In contrast, the variation in UV-A was small: the ratio between maximum and minimum was less than 2. Three important facts are proposed concerning solar UVR protection of the human body: 1) the personal minimal erythema dose (MED); 2) gender based difference in MED values; and 3) proper colors for UVR protective clothing.

Monitoring of Selected Veterinary Antibiotics in Animal Carcass Disposal Site and Adjacent Agricultural Soil (가축매몰지 및 인근 농경지의 축산용 잔류 항생제 모니터링)

  • In Korea, over 4,700 animal carcass disposal sites were installed until 2011 due to the outbreak of foot and mouth disease. Due to the putrefaction of buried animals, the leachate containing veterinary antibiotics may release into surrounding environments. Antibiotic residues in the environment cause the formation of antibiotic resistance bacteria threatening human and ecosystem health. This study reports the concentrations of five antibiotics, including tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC), sulfamethazine (SMZ), and sulfamethoxazole (SMX), in soils from animal carcass disposal site and adjacent agricultural field. Concentrations of antibiotics at animal carcass disposal sites (TC: $144.26-350.73{\mu}g/kg$, SMZ: $17.72-44.94{\mu}g/kg$) were higher than those at agricultural field (TC: $134.16-320.73{\mu}g/kg$, SMZ: $6.48-8.85{\mu}g/kg$) whereas the concentrations of CTC, OTC, and SMX were below detection limit in both sites. Results showed that the antibiotics in animal carcass site might leach to the soil and possibly contaminating the groundwater. Future studies will focus on the transfer of antibiotics residues into food crops.

Determination of Efficient Operating Condition of UV/H2O2 Process Using the OH Radical Scavenging Factor (수산화라디칼 소모인자를 이용한 자외선/과산화수소공정의 효율적인 운전 조건도출)

  • This study investigated a method to determine an efficient operating condition for the $UV/H_2O_2$ process. The OH radical scavenging factor is the most important factor to predict the removal efficiency of the target compound and determine the operating condition of the $UV/H_2O_2$ process. To rapidly and simply measure the scavenging factor, Rhodamine B (RhB) was selected as a probe compound. Its reliability was verified by comparing it with a typical probe compound (para-chlorobenzoic acid, pCBA); the difference between RhB and pCBA was only 1.1%. In a prediction test for the removal of Ibuprofen, the RhB method also shows a high reliability with an error rate of about 5% between the experimental result and the model prediction using the measured scavenging factor. In the monitoring result, the scavenging factor in the influent water of the $UV/H_2O_2$ pilot plant was changed up to 200% for about 8 months, suggesting that the required UV dose could be increased about 1.7 times to achieve 90% caffeine removal. These results show the importance of the scavenging factor measurement in the $UV/H_2O_2$ process, and the operating condition could simply be determined from the scavenging factor, absorbance, and information pertaining to the target compound.

Optimization of Peach Wine Fermentation by Response Surface Methodology (반응표면분석에 의한 복숭아주 발효 최적화)

  • To prepare peach wine using peach juice, fermentation properties were monitored, and fermentation conditions (initial sugar concentration, temperature, and time) were optimized by a response surface methodology. Alcohol content for peach wine fermentation ranged from 3.4~9.2% [$R^2=0.9229$ (P<0.01)] and 8.54% (maximum value) at $18.73^{\circ}Brix$, $16.81^{\circ}C$, and 12.99 day. Acidity ranged from 0.30~0.74%, and 0.25% (minimum value) at $15.11^{\circ}Brix$, $17.09^{\circ}C$, and 13.61 day. Residual sugar concentration was $6.67^{\circ}Brix$ (maximum residual sugar content) at $17.79^{\circ}Brix$, $20.63^{\circ}C$, and 3.37 day. Yellow color intensity was 18.92 (maximum Hunter's color b value) at $13.19^{\circ}Brix$, $20.81^{\circ}C$, and 12.81 day. Based on the above study results, optimization conditions for peach wine fermentation were 9 days, below $20^{\circ}C$, and $19^{\circ}Brix$ peach juice.

Nonoperative Management of Blunt Liver Trauma (둔상성 간 손상환자의 비수술적 치료)

  • Background: The management of hepatic injuries has changed dramatically during the past two decade after the technologic breakthroughs in radiologic imaging techniques. Recently, the non-operative management of blunt hepatic trauma has become the standard of care in hemodynamically stable patients. We reviewed our experience of the non-operative management of blunt hepatic trauma. And the purpose of this study was to examine the prognostic factors and indicators affecting the decision for treatment modality of emergent hepatic trauma. Methods: The medical records of 84 patients who were treated for blunt hepatic injury at Masan Samsung Hospital from January 2002 to December 2003. The patients were divided two groups, non-operative(Non-OP) and operative(OP), according to the treatment modality. The two groups were compares for age, sex, mechanism of injury, grade of liver injury scale, combined injury, systolic blood pressure, pulse rate, hemoglobin, hematocrit, WBC count, S-GOT, S-GPT, ALP, transfusion amount during initial 24 hours, amount of infused crystalloid fluid, length of ICU stay, length of ward care, morbidity and mortality. The grade of the liver injury were determined by using the organ injury scale(OSI). Results: Among the 84 patients, 46 cases(54.8%) were managed non-surgically, and 3 cases of Non-OP group were treated by transarterial embolization. Between the two groups, there were significant difference in age, injury grade, combined injury, hemoglobin, hematocrit, initial systolic blood pressure, amount of infused crystalloid fluid, amount of transfusion during the first 24 hours, and length of ICU care, morbidity and mortality.(p<0.05) The overall mortality rate was 8.3%, but 2.2% mortality in the non-operative group. Conclusion: Non-operative management may be considered as a first choice in hemodynamic stable patients with blunt liver trauma. The reliable indicators affecting the treatment modality of blunt hepatic trauma were systolic BP, Hb, Hct, amount of infused crystalloid fluid, amount of transfusion during the first 24 hours, liver injury grade and combined injury. Strict selection of treatment madality and aggresive monitoring with intensive care unit were more important.

Development of Wireless Measurement System for Bridge Using PDA and Fiber Optical Sensor (PDA와 광섬유 센서를 이용한 교량의 무선계측 시스템 개발)

  • This study proposes a wireless measurement system that is a new safety management system by using an FBG sensor and a PDA. The sensor part has many advantages of implementing a wireless measurement system, and the study emploies an FBG-LVDT sensor, FBG-STRAIN sensor, FBG-TEMP sensor, and FBG-ACC sensor, using FBG sensors. Also, the study show a configuration of a signal process system for operating a wireless transmission system of FBG sensors applied to the signal process system, and engrafted the cutting edge information technology industry in order to display from a remote distance using a PDA. In order to verify the applicability of the developed FBG sensors and wireless measurement monitoring system to the field, their accuracy, and usability, the study has conducted a static and dynamic test to a bridge in the field. The study made an assessment of service for the vibration of the bridge by applying dynamic data measured by an FBG-LVDT sensor and FBG-ACC sensor to Meister's curve and prepared methods for assessing the vibration of the bridge by proposing a standard of vibration limitation given the service of vibration of the bridge. As a follow up for this study, it would be necessary to set up an overall model for the standard of service assessment established in this study.

A Study on Tension for Cables of a Cable-stayed Bridge Damper is Attached (댐퍼가 부착된 사장교의 케이블 장력에 관한연구)

  • Recently, many ocean bridges that connect land to island or island to island have been constructed along with the improvement of the nation's economy. Long-span bridges can be categorized as suspension bridge, cable-stayed bridge, arch bridge and truss bridge. In this study, correction with respect to construction error can be presented on site through the monitoring of the cable tension change of real structure for four major construction stages so that construction accuracy, including the management of profiles, can be improved. A vibration method, the so-called indirect method that uses the cable's natural frequency changes from the acceleration sensor installed on the cable, is applied in measuring cable tension. In this study, the estimation formula for the effective length of cable with damper is presented by comparing and analyzing between actual measurement and analysis result for the change of the cable's effective length. By the way, it is known that the reliability of estimating cable tension by applying the former method that uses the net distance from damper to anchorage is low. Therefore, for future reference of the maintenance stage, the presented formula for estimating the effective length of cable can be used as a reference for the rational decision-making, such as the re-tensioning and replacement of cable.
