Control of the Photo-induced Discoloration of the Handmade Korean Paper with Polyethylene Glycol (Polyethylene Glycol에 의한 수록한지(手漉韓紙)의 광(光) 변색(變色) 억제(抑制))

  • Classified handmade Korean paper which has been commonly used as the material for the paper cultural assets divide into four classes. We irradiated xenon arc lamp, which has almost the same spectrum distribution as natural sunlight, to test photo-induced of discoloration according to PEG molecular weight and concentration. So, we reach the followings. 1) Unbleached handmade Korean paper with PEG showed rapid decrease of brightness for 10 hours after photo-irradiation, regardless of molecular weight of PEG, but, after 10 hours, it showed gradual increase of brightness. From this, it proved that there was an effect of preventing discoloration of photo-irradiation. 2) Chemical bleached handmade Korean paper showed increase of brightness after 20 hours. Though time was delayed in increase of brightness, there was also an effect of preventing discoloration of photo-irradiation in chemical bleached paper. 3) Handmade Korean paper made with abet-mosk showed the lowest of brightness when it was tested on 400-20% PEG. Before photo-irradiation, the brightness of paper, applied it, was the lowest, but it showed the least difference of brightness after photo-irradiation. From this, it proved that there was the greatest effect of preventing discoloration in this paper. 4) Preventing yellowish, the first phenomena by photo-irradiation ageing with applying PEG was proved that there was the decrease of value of $b^*$ +, and increase value of $L^*$, which could fairly prevent decrease of brightness.

Characteristics of PAHs Concentration in Soil Contamination Concerned Area of Gwangju (광주지역 토양오염우려지역의 PAHs 농도 특성 연구)

  • The concentration levels and distribution characteristics of 16 polycyclic aromatic hydrocarbons (PAHs) were investigated and evaluated for total 100 soil samples as a part of the survey on soil contamination in Gwangju. The results (median and range) of T-PAHs (sum of 16 PAH concentrations), C-PAHs (sum of carcinogenic PAH concentrations) and T-TEQs (sum of 16 TEQ concentrations) were 20.8 (7.6~1158.1), 2.2 (N.D~509.6), and 0.3 (N.D~424.6) ㎍/kg, respectively. There was a positive correlation between C-PAHs/T-PAHs and T-TEQs/T-PAHs except one point where the concentration of benzo(a)pyrene was high. The ratios of the C-PAHs/T-PAHs were 31.7% for low molecular weight-PAHs and 68.3% for high molecular weight-PAHs, suggesting that PAHs generation mainly arose from combustion sources. The ratio of isomers of individual PAHs, Phe/Ant, Flu/Pyr, Ant/(Ant+Phe), Flu/(Flu+Pyr), and BaA/(BaA+Chr), also confirmed the predominance of PAHs from combustion activities. Statistical tracing of the source of PAHs through principal component analysis indicated that the main sources of combustion were automobile fuel and coal. The overall results of this study suggested HMW-PAHs, T-PAHs, C-PAHs and T-TEQs should be separately evaluated to better assess the toxicity and environmental behavior of individual PAHs.

The SIMDIST (Simulated Distillation) Analysis of Distributing Engine Oil (국내 유통 엔진오일 고온모사증류시험 분석)

  • The vehicle lubricant occupies upto 35% in a total lubricant market and engine oil occupies upto 77% in the vehicle lubricant market in Korea. A suitable quality management of the circulating engine oil is necessary for driver and engine protection. But, KS and synthetic engine oil products (involved over 30% synthetic oil) are exempt to any quality management under Petroleum and Alternative Fuel Business Act. It is also known that synthetic oils such as PAO (poly alpha olefin) have excellent properties and performance like anti-wear, varnish control and oxidation stability than those of mineral oils. For this reason, PAO has been used for an engine oil, rotary screw and reciprocating compressor in addition to heavy duty and other extreme service applications. In this study, our research group analyzed the chromatogram pattern for the mineral oil, PAO and mineral oil involved a typical ratio of PAO using SIMDIST (simulated distillation). In the SIMDIST chromatogram, the mineral oil showed a broad peak, while PAO showed a sharp typical peak. Also the oil with a large viscosity grade exhibited a long retention time due to the heavy molecular weight and high boiling point. In particular, the blended mineral oil with 20% PAO sample showed a distinctly different pattern compared to that of using the conventional mineral oil. For monitoring PAO contents in distributing engine oils, we analyzed the SIMDIST for 27 kinds of engine oils which were popularly sold in Korea. The analytic results indicate that all kinds of engine oils showed that PAO contents were below 20% in engine oil products. Moreover, the PAO titled product was found to have a small amount of PAO. Thus, we conclude that the related laws for the proper quality management of synthetic oils are needed to be established.

Molecular Cloning and Bioinformatic Analysis of SPATA4 Gene

  • Full-length cDNA sequences of four novel SPATA4 genes in chimpanzee, cow, chicken and ascidian were identified by bioinformatic analysis using mouse or human SPATA4 cDNA fragment as electronic probe. All these genes have 6 exons and have similar protein molecular weight and do not localize in sex chromosome. The mouse SPATA4 sequence is identified as significantly changed in cryptorchidism, which shares no significant homology with any known protein in swissprot databases except for the homologous genes in various vertebrates. Our searching results showed that all SPATA4 proteins have a putative conserved domain DUF1042. The percentages of putative SPATA4 protein sequence identity ranging from 30% to 99%. The high similarity was also found in 1 kb promoter regions of human, mouse and rat SPATA4 gene. The similarities of the sequences upstream of SPATA4 promoter also have a high proportion. The results of searching SymAtlas (http://symatlas.gnf.org/SymAtlas/) showed that human SPATA4 has a high expression in testis, especially in testis interstitial, leydig cell, seminiferous tubule and germ cell. Mouse SPATA4 was observed exclusively in adult mouse testis and almost no signal was detected in other tissues. The pI values of the protein are negative, ranging from 9.44 to 10.15. The subcellular location of the protein is usually in the nucleus. And the signal peptide possibilities for SPATA4 are always zero. Using the SNPs data in NCBI, we found 33 SNPs in human SPATA4 gene genomic DNA region, with the distribution of 29 SNPs in the introns. CpG island searching gives the data about CpG island, which shows that the regions of the CpG island have a high similarity with each other, though the length of the CpG island is different from each other.This research is a fundamental work in the fields of the bioinformational analysis, and also put forward a new way for the bioinformatic analysis of other genes.

Range of physicochemical parameters for active ingredients of herbicides (제초제의 활성 성분에 대한 물리-화학 파라미터의 범위)

  • Various physicochemical parameters for the active ingredients of 245 herbicides were calculated to develope a diagnosis and estimation system for utility as herbicide. The range of physico-chemical parameters for each inhibitors of photo system II (H1), acetolactate synthase (ALS) (H2) and herbicides were confirmed. The distribution ranges of 85% dependence for each physicochemical parameters were Obs.logP :$-0.90\sim4.50$, dipol moment: $1.80\sim12.22$ (debye), molecular refractivity: $53.0\sim104.0(cm^3/mol)$, polarizability: $19.0\sim37.0(\AA^3)$, HOMO energy: $-9.98\sim-7.34$ (eV), LUMO energy:$-2.76\sim0.40$ (eV), Van der Waals molecular volumes: $558.0\sim995.0(cm^3)$, molecular weight: $202.0\sim430.0$ (amu) and surface areas (Grids): $194.0\sim356.0(\AA^2)$, hydration energy: $-10.16\sim114.7$ Kcal/mol, respectively. It is suggested that MR and polarizability constants will be able to distinguish between herbicides and medicinal drugs. Results revealed that various compounds based on the range of physicochemical parameters of herbicides could be diagnosed and estimated.

Plasma Levels of High Molecular Weight Adiponectin are Associated with Cardiometabolic Risks in Patients with Hypertension (고혈압 환자에서 혈장 고분자량 아디포넥틴 농도와 심장-대사위험인자와의 관련성 연구)

  • In the present study, we comprehensively examined the associations of plasma levels of total adiponectin and high molecular weight (HMW) adiponectin with the features of cardiometabolic risks including body fat distribution, dyslipidemia, insulin resistance and inflammatory markers in a cross-sectional study of 110 treated hypertensive patients. Blood lipid profiles, high sensitivity C-reactive protein (hsCRP) and homeostasis model assessment of insulin resistance (HOMA- IR) derived from fasting glucose and insulin concentrations were determined. Plasma levels of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-6 (IL-6) and intercellular adhesion molecule-1 (ICAM-1) were analyzed using ELISA. The results showed that plasma levels of HMW-adiponectin were negatively associated with body mass index (BMI, r = - 0.203, p < 0.05) and waist circumference (r = -0.307, p < 0.01), which was not shown in total adiponectin. Plasma levels of HMW-adiponectin were negatively associated with triglyceride (r = -0.223, p < 0.05) and positively associated with HDL-cholesterol (r = 0.228, p < 0.05). Plasma levels of adiponectin were positively associated with HDL-cholesterol (r = 0.224, p < 0.05). Plasma levels of HMW-adiponectin were negatively associated with hsCRP (r = -0.276, p < 0.01) and IL-6 (r = -0.272, p < 0.01). In addition, there were weak associations between plasma levels of HMWadiponectin and TNF-${\alpha}$ (r = -0.163, p = 0.07) and ICAM-1 (r = -0.158, p = 0.09). However, there were no significant associations of total adiponectin with inflammatory markers except hsCRP (r = -0.203, p < 0.05). Stepwise multiple linear regression analysis showed that only plasma levels of HMW-adiponectin was an independent factor influencing serum levels of hsCRP, a marker of systemic low grade inflammation, after adjusting for age, gender, BMI, waist circumference, alcohol intake, smoking status, blood lipids, total adiponectin and drug use (p < 0.01). These results suggest that HMW-adiponectin, rather than total adiponectin, is likely to be closely associated with the features of cardiometabolic risks in treated hypertensive patients and might be effective biomarker for the prediction of cardiovascular disease.

Reduction of the Antigenicity of Whey Protein by Enzymatic Hydrolysis (효소가수분해에 의한 유청단백질의 항원성 저하)

  • As a preliminary study about the reduction of the antigenicity of whey protein isolate(WPI) by treatment of chymotrypsin, trypsin, pancreatin, and protease from Aspergillus oryzae, the properties and antigenicities of whey protein hydrolysates(WPH) were investigated. When degrees of hydrolysis (DH) were measured by use of trinitrobenzensulfonic acid(TNBS), the DH of the WPH treated by pancreatin or protease from Aspergillus oryzae$(5.05{\sim}11.47)$ were much higher than those of the tryptic or chymotryptic WPH$(15.67{\sim}20.20)$. And the pretreatments of heat$(75^{\circ}C)$, 20 min and/or pepsin resulted in higher DH of WPH, generally. When the molecular distributions of the WPH were determined by high performance size exclusion chromatography(HPSEC), the ratios of polypeptides with molecular weight more than 10kDa ranged from 12% to 36%, and the average molecular weights and the average peptide lengths of the WPH were $4,252{\sim}9,132$ dalton and $38{\sim}83$ amino acids, respectively. And there was no bitter taste in all of the WPH. Results of SDS-PAGE showed that most of intact native proteins were eliminated by the enzymatic hydrolysis but there were a few bands of peptides larger than 14.2 kDa in some WPH. When antigenicity was assayed by competitive inhibition enzyme-linked immunosorbent assay(cELISA), monovalent antigenicity of WPH to rabbit anti-WPI antiserum were lowered to $10^{-1.7}-10^{-4.9}$ times and less by the enzymatic hydrolysis. And the pretreatments of heat and pepsin resulted in the lowest antigenicicy within a group of enzymatic hydrolysis, especially in case of the pancreatic hydrolysate(PDP) whose antigenicity was found almost to be removed.

Effect of High Dose ${\gamma}-irradiation$ on the Physicochemical Properties of Shell Eggs during Storage (고선량 감마선 조사가 신선란의 저장 중 이화학적 특성에 미치는 영향)

  • To determine the quality change of the irradiated eggs during storage, fresh shell eggs were irradiated using $^{60}Co$ at 0, 1, 5, 10, 30 kGy and stored for 30 days. The york index, color, pH, viscosity, egg weight, and SDS-PAGE profile of the irradiated eggs were examined. During storage, york index values of the irradiated eggs and the control were decreased and the increase of dose decreased yolk index. However, the yolk index values were increased temporarily at 10 kGy and 30 kGy. The yolk color had a bright yellow with increases in dose level and there was no significant change during storage. The albumen viscosities were decreased with increases in dosage and were decreased during storage. Also, the albumen pH values of the irradiated eggs were higher than that of the control and were increased during storage. The weight losses of eggs were increased during storage and there were no significant changes by dose level. SDS-PAGE profile of the egg white proteins of the shell eggs showed the change in molecular weight distribution and had aggregation pattern as well as degradation. CD and fluorescence spectroscopy study showed changes in the secondary and tertiary structure of egg white proteins by ${\gamma}-irradiation$. Therefore, this study clearly indicates that irradiation dose of eggs should be appropriate to prevent the loss of egg qualities.

The Effects of Teaching Methods on Conceptual Change of Atmospheric Pressure in Middle School Students (수업방안이 중학생들의 대기압 개념 변화에 미치는 영향)

  • The purpose of this study is to inquire into the effects of teaching methods in the class on the conceptual change of atmospheric pressure for middle school students. After analyzing the concept of atmospheric pressure in the middle school science textbooks on the present 7th Curriculum, classes were performed adopting classified Method A and Method 3. For Method A, the textbook is used to explain the concept in the view of weight. For Method B, the textbook is used to approach the concept in the views of molecular movement as well as of weight. This study consists of four classes in the third grade students of middle school in Busan, where they were divided into the Method A group and the Method B group. These study was carried out with pre-post on each of these classes on the learning achievement and on the conceptual change of atmospheric pressure. The results of this study were as follows: First, the effect on the learning achievement was displayed the average score of the Method B was showing a meaningful difference comparing to the Method A. Second, the effect on the conceptual change measured by verifying the score for the difference among the averages for each sub-scale three out of four conceptual factors,'the direction of atmospheric influence and the reason','the principle of atmospheric action' and 'the atmospheric changes by the temperature rise on the surface of the earth and the reason', showed meaningful improvement. But, the one left factor,'the distribution of atmospheric pressure by altitudes and the reason', displayed no meaningful difference. Third, The concept of atmospheric pressure is better defined as the pressure created by the movement of air particles, in the view of kinetic theory of gas, rather than explained by the notion of the weight of air.
