Sensitization Effects of Hyperthermia on Bleomycininduced DNA Strand Breaks and Replication Inhibition in CHO-$K_1$ Cells in Vitro

  • Effects of hyperthermia on the induction of DNA single strand breaks and replication inhibition were studied in bleomycin-treated CHO-K$_1$ cells by alkaline elution and alkaline sucrose gradient sedimentation. Bleomycin-induced DNA single strand breaks of DNA were dose-and time-dependently increased, and these strand breaks of DNA were gradually rejoined as post-incubation time passed. Treatment with hyperthermia alone did not affect the induction of DNA single strand breaks. However, pre-exposure of cells to hyperthermia followed by bleomycin treatment greatly increased the single strand breaks, and also reduced the rejoining processes of bleomycin-induced DNA single strand breaks. Bleomycin selectively inhibited the replicon initiation. The combined treatment with hyperthermia and bleomycin markedly potentiated the nonspecific inhibition of replication.

  • Deficiencies in wear and frost resistance as well as mechanical strength constitute the main causes of equipment failure under the harsh climatic conditions of the Earth's polar regions. To improve the properties of the materials used in this equipment, nanoparticle composites have been prepared from clays such as kaolinite, hectorite, and montmorillonite in combination with polytetrafluoroethylene (PTFE) or ultrahigh molecular weight polyethylene (UHMWPE). A number of techniques have been proposed to disperse silicate particles in PTFE or UHMWPE polymer matrices, and several successful processes have even been widely applied. Polymer nanocomposites that exhibit enhanced mechanical and thermal properties are promising materials for replacing metals and glass in the equipment intended for Arctic use. In this article, we will review PTFE- and UHMWPE-based layered silicate nanocomposites.

From Cytosol to Mitochondria: The Bax Translocation Story

  • The balance between life and death of a cell regulates essential developmental processes in multicellular organisms. Apoptotic cell death is a complex, stepwise program involving multiple protein components that trigger and execute the demise of the cell. Of the many triggers of apoptosis, most are not well understood, but some key components have been identified, such as those of the Bcl-2 family, which function as anti-apoptotic or proapoptotic factors. Bax, a pro-apoptotic member of this family, has been shown to serve as a component of many apoptotic triggering cascades and its mechanism of action is the focus of intense study. Herein we discuss current, differing ideas on the function of Bax and its structure, and suggest novel mechanisms for how this death protein targets mitochondria, triggering apoptosis.

  • Recent progress in the development of non-invasive imaging technologies continues to strengthen the role of biomedical research. These tools have been validated recently in variety of research models, and have born shown to provide continuous quantitative monitoring of the location(s), magnitude, and time-variation of gene delivery and/or expression. This article reviews the use of PET technologies as they have been used in imaging biological processes for molecular imaging applications. The studies published to date demonstrate that noninvasive imaging tools will help to accelerate pre-clinical model validation as well as allow for clinical monitoring of human diseases.

Single Molecule Method for Molecular Biology

  • In order to understand biological phenomena accurately, single molecule techniques using a physical research approach to molecular interactions have been developed, and are now widely being used to study complex biological processes. In this review, we discuss some of the single molecule methods which are composed of two major parts: single molecule spectroscopy and manipulation. In particular, we explain how these techniques work and introduce the current research which uses them. Finally, we present the oral biology research using the single molecule methods.

  • $Ca^{2+}$ is a universal signalling molecule that affects a variety of cellular processes including cardiac development. The majority of intracellular $Ca^{2+}$ is stored in the endoplasmic and sarcoplasmic reticulum of muscle and non-muscle cells. Calreticulin is a well studied $Ca^{2+}$-buffering protein in the endoplasmic reticulum, and calreticulin deficiency is embryonic lethal due to impaired cardiac development. Despite calsequestrin being the most abundant $Ca^{2+}$-buffering protein in the sarcoplasmic reticulum, viability is maintained in embryos without calsequestrin and normal $Ca^{2+}$ release and contractile function is observed. The $Ca^{2+}$ homeostasis regulated by the endoplasmic and sarcoplasmic reticulum is critical for the development and proper function of the heart.

  • It has recently been shown that the instability of thin film of a nanoscale can be used in the processes of building nano-size structures, which have potential practical importance in nanotechnology. Molecular dynamics simulation is conducted to probe the thin fluid film of a nano-size and its dynamic behavior during destabilization and structure formation. Non-continuum characteristics are shown in the properties like pressure tensor, viscosity, and thermal conductivity. The thermocapillary force induces a slow growth of long waves in the scale considered. A long-range interaction with the solid wall induces vertical structures, whose formation time and space between neighbors are proportional to the strength of the interaction.

  • Application of the Rheocatch Hotwire Monitoring System for food biotechnology process was evaluated. The growth of microogranism, E coli (JM 83 and Sigma) and Corynesccfertun glutamicum, were monitored. in the fermentor. The cell growth could not be detected the temperature differences between the hotwire and samples($\Delta$T) as indicated by the monitoring system during the fermentation processes. The cell concentration of less than 2g/dl was not sufficient to generate the measurable temperature difference in the fermentor. In order to calibrate the Rheocatch Monitoring System, the temperature difference as a function of solute concentration (microbial cells, sodium cholide, sucrose and dextran) was studied. The relationship between $\Delta$T and the concentration of microbial cells, sucrose and dextran can be expressed in a power series. Further studied with dextran indicated that viscosity and/or kinematic viscosity increase exponentially with an increase in $\Delta$T This is regardless of the concentration and molecular weight of dextran. $\Delta$T linearly increases with the logarithm of molecular weight, while the logarithm of viscosity and the logarithm of kinematic viscosity increase with the logarithm of molecular weight.

  • Recently, high vacuum pumps are widely used in the semi-conduction and liquid-crystal display ( LCD ) process. The composite-type high vacuum pumps are widely used in the various processes. In this study, the pumping performance of composite-type molecular pumps has been investigated experimentally. The experimented pumps are a compound molecular pump ( CMP ) and hybrid molecular pump ( HMP ). The CMP consists with helical-type drag pump, at lower part, and with turbomolecular pump ( TMP ), at upper part. The HMP consists with disk-type drag pump, at lower part, and with TMP, at upper part. The experiments are performed in the outlet pressure of $0.2\;{\sim}\;533\;Pa$. We have measured the ultimate pressure, compression ratio, and pumping speed

Alternative Polyadenylation of mRNAs: 3'-Untranslated Region Matters in Gene Expression

  • Almost all of eukaryotic mRNAs are subjected to polyadenylation during mRNA processing. Recent discoveries showed that many of these mRNAs contain more than one polyadenylation sites in their 3' untranslated regions (UTR) and that alternative polyadenylation (APA) is prevalent among these genes. Many biological processes such as differentiation, proliferation, and tumorigenesis have been correlated to global APA events in the 3' UTR of mRNAs, suggesting that these APA events are tightly regulated and may play important physiological roles. In this review, recent discoveries in the physiological roles of APA events, as well as the known and proposed mechanisms are summarized. Perspective for future directions is also discussed.
