한국산 꼼치과 어류의 분자계통 및 분류학적 재검토 (Molecular Phylogeny and Taxonomic Review of the Family Liparidae (Scorpaenoidei) from Korea)

  • 쏨뱅이아목 (Scorpaenoidei) 어류에 속하는 꼼치과 (Liparidae) 어류는 형태적으로 유사하고 오동정 가능성이 크며 체색과 몸의 상대적인 크기 변이가 심하여 분류학적으로도 상당히 혼란스러운 분류군이다. 따라서, 본 연구는 꼼치과 어류의 자원변동을 이해하고 자원관리를 위해 국내 보고된 3속 10종을 대상으로 분자계통 및 분류학적 재검토를 수행하였다. 미토콘드리아DNA COI 영역과 핵DNA RAG2 영역의 염기서열을 이용한 분자계통 분석 결과, 물미거지가 분홍꼼치와 가깝게 유집되는 mtCOI 계통 결과를 제외하면 3속 (분홍꼼치속, 물미거지속, 꼼치속)이 대등한 단계통성을 나타내었다. Gilbert and Burke (1912)는 L. ochotensis와 구분되는 한국산 미거지를 L. ingens로 신종 보고하였으나, 본 연구에서 한국산 미거지는 일본산 및 러시아산 L. ochotensis와 형태 및 분자에서 잘 일치하였으므로 한국산 미거지의 학명을 L. ingens에서 L. ochotensis로 변경한다.

Morphology, Molecular Phylogeny and Pathogenicity of Colletotrichum panacicola Causing Anthracnose of Korean Ginseng

  • Colletotrichum panacicola isolates were obtained from anthracnose lesions of Korean ginseng and compared with four Colletotrichum species in morphology, molecular phylogeny and pathogenicity. Based on morphological characteristics, C. panacicola was easily distinguished from Colletotrichum gloeosporioides but not from Colletotrichum higginsianum, Colletotrichum destructivum and Colletotrichum coccodes. A phylogenetic tree generated from ribosomal DNA-internal transcribed spacer sequences revealed that C. panacicola is remarkably distinguished from C. gloeosporioides and C. coccodes but not from C. higginsianum and C. destructivum. However, molecular sequence analysis of three combined genes (actin + elongation factor-$1{\alpha}$ + glutamine synthatase) provided sufficient variability to distinguish C. panacicola from other Colletotrichum species. Pathogencity tests showed that C. panacicola is pathogenic to Korean ginseng but not to other plants. These results suggest that C. panacicola is an independent taxon distin-zguishable from C. gloeosporioides and other morphologically similar Colletotrichum species.

Sinuolinea capsularis (Myxosporea: Sinuolineidae) Isolated from Urinary Bladder of Cultured Olive Flounder Paralichthys olivaceus

  • Sinuolinea capsularis Davis, 1917 is myxosporean that infect the urinary system of the host fish. Insufficient morphological and molecular data of S. capsularis exits, and it is therefore difficult to make an accurate identification of the parasite. We tried a series of morphological and molecular analysis to identify an myxosporean isolated from urinary bladder of cultured olive flounder, Paralichthys olivaceus, from Jeju island in the Republic of Korea. Some of them were observed under a light microscope and SEM, and remain samples were used molecular and phylogenetic analysis. Mature spores were subspherical, measuring $13.9{\pm}0.6{\mu}m$ in length and $13.8{\pm}0.8{\mu}m$ in width. Two spherical polar capsules on opposite sides in the middle of the spore had a diameter range of $4.3{\pm}0.4{\mu}m$. Scanning electron microscopy revealed that spores a severely twisted the suture line. By the morphological comparison and analysis, it was identified as S. capsularis. In addition, we obtained the partial 18S rDNA of S. capsularis and first registered it in NCBI. Phylogenetic analysis showed that S. capsularis clustered with Zschokkella subclade infecting the urinary system of marine fish, and it supported the infection site tropism effect on phylogeny of marine myxosporeans as well as the origin of Sinuolinea is not monophyly.

Occurrence and Molecular Identification of Microcotyle sebastis Isolated from Fish Farms of the Korean Rockfish, Sebastes schlegelii

  • Microcotyle sebastis is a gill monogenean ectoparasite that causes serious problems in the mariculture of the Korean rockfish, Sebastes schlegelii. In this study, we isolated the parasite from fish farms along the coasts of Tongyeong, South Korea in 2016, and characterized its infection, morphology and molecular phylogeny. The prevalence of M. sebastis infection during the study period ranged from 46.7% to 96.7%, and the mean intensity was 2.3 to 31.4 ind./fish, indicating that the fish was constantly exposed to parasitic infections throughout the year. Morphological observations under light and scanning electron microscopes of the M. sebastis isolates in this study showed the typical characteristics of the anterior prohaptor and posterior opisthaptor of monogenean parasites. In phylogenetic trees reconstructed using the nuclear 28S ribosomal RNA gene and the mitochondrial cytochrome c oxidase I gene (cox1), they consistently clustered together with their congeneric species, and showed the closest phylogenetic relationships to M. caudata and M. kasago in the cox1 tree.

쥐노래미과 어류 2종의 미토콘드리아 Cytochrome b 유전자의 분자계통 (Molecular Phylogeny of Two Species of Hexagrammidae (Greenlings) Inferred from Mitochondrial Cytochrome b Gene)

  • 본 연구는 쥐노래미과(Hexagrammidae)에 속하고 양식산업에서 중요한 위치를 차지하고 있는 노래미, Hexagrammos otakii(Jordan et Starks)와 쥐노래미, H. agrammus(Temminck et Schlegel)의 미토콘드리아 cytochrome b(cyt b)유전자의 염기서열을 서로 비교 분석하였다. 총 489 bp크기의 cyt b 염기서열에서 종간의 변이는 거의 없었다(96%의 유사성).Neighbor-joining방법에 의한 노래미와 쥐노래미에서의 0.0342인 pairwise distance는 두 종이 분자계통유전학적으로 유사함을 시사한다. 이러한 연구 결과들은 쥐노래미(Hexagrammos)속을 대상으로 한 양식, 수산유전학 및 분자계통학 분야에 적용 가능할 것이다.

Phylogenetic analysis of Neottia japonica (Orchidaceae) based on ITS and matK regions

  • To elucidate the molecular phylogeny of Neottia japonica, which is a terrestrial orchid distributed in East Asia, the internal transcribed spacer (ITS) of nuclear DNA and the matK of chloroplast DNA were used. A total 22 species of 69 accessions for ITS and 21 species of 114 accessions for matK phylogeny were analyzed with the maximum parsimony and Bayesian methods. In addition, we sought to establish a correlation between the distribution, morphology of the auricles and genetic association of N. japonica with phylogenetic data. The phylogenetic results suggest that N. japonica is monophyletic and a sister to N. suzukii in terms of the ITS phylogeny, while it is paraphyletic with N. suzukii in terms of the matK phylogeny. N. japonica and N. suzukii show similar morphologies of the lip and column, they both flower in April, and they are both distributed sympatrically in Taiwan. Therefore, it appears to be clear that N. japonica and N. suzukii are close taxa within Neottia, although there is incongruence between the nrDNA and cpDNA phylogenies of N. japonica. The incongruence between the two datasets may have various causes, meaning that further studies are needed to confirm the evolutionary process of N. japonica. The phylogenetic status of N. kiusiana, which was not included in previous studies, was as a sister to N. nidus-avis. Meanwhile, the ITS and matK phylogenies are unsuitable for identifying genetic associations with the characteristic of auricles. The phylogenetic topologies of Korean, Taiwanese and mainland Chinese individuals suggest that the populations of N. japonica in Korea originated from China's mainland and island areas. The characterization of regional gene differences could provide useful preliminary data for future studies.

tufA gene as molecular marker for freshwater Chlorophyceae

  • Green microalgae from the class Chlorophyceae represent a major biodiversity component of eukaryotic algae in continental water. Identification and classification of this group through morphology is a hard task, since it may present cryptic species and phenotypic plasticity. Despite the increasing use of molecular methods for identification of microorganisms, no single standard barcode marker is yet established for this important group of green microalgae. Some available studies present results with a limited number of chlorophycean genera or using markers that require many different primers for different groups within the class. Thus, we aimed to find a single marker easily amplified and with wide coverage within Chlorophyceae using only one pair of primers. Here, we tested the universality of primers for different genes (tufA, ITS, rbcL, and UCP4) in 22 strains, comprising 18 different species from different orders of Chlorophyceae. The ITS primers sequenced only 3 strains and the UCP primer failed to amplify any strain. We tested two pairs of primers for rbcL and the best pair provided sequences for 10 strains whereas the second one provided sequences for only 7 strains. The pair of primers for the tufA gene presented good results for Chlorophyceae, successfully sequencing 21 strains and recovering the expected phylogeny relationships within the class. Thus, the tufA marker stands out as a good choice to be used as molecular marker for the class.

Studies on Biological Diversity of Firefly in Japan

  • Taxonomic and phylogenetic studies of firefly in Japan have been reviewed. Fourty-six lampyrid species and one rhagophthalmid are distributed in the Japanese Islands including the Ryukyus. Recently, molecular phylogenetic approaches have been employed in the systematic study of firefly using mitochondrial and luciferase genes. Based on the molecular phylogenetic trees, evolutionary process of flashing patterns related strictly to mating behavior was estimated. Furthermore, genetic diversity studies revealed geographic differentiation patterns within species, and conservation measures of firefly were proposed to protect genetic resources endemic to the localities.

Molecular Identification of Two Strains of Phellinus sp. by Internal Transcribed Spacer Sequence Analysis

  • Two species of cultivated Phellinus sp. were identified as P. baumii by internal transcribed spacer (ITS) sequence analysis. The fruit bodies of the examined strains were similar to those of naturally occurring strains, having a bracket-like form, yellow-to-orange color, and poroid hymenial surfaces. The DNA sequences of ITS region of both strains showed a homology of 99% with ITS1 to ITS2 sequences of P. (Inonotus) baumii strain PB0806.
