Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.
We assessed the genetic relations and epidemiological links among bloodstream isolates of Candida albicans, which were obtained from a university hospital over a period of five years. The 54 bloodstream isolates from the 38 patients yielded 14 different karyotypes, 29 different patterns after digestion with SfiI (REAG-S), and 31 different patterns after digestion with BssHII (REAG-B) when analyzed using three different pulsed-field gel electrophoresis (PFGE) typing methods. In 11 patients with serial blood stream isolates, all strains from each patient had the same PFGE pattern. The dendrograms for all of the strains revealed that the distribution of similarity values ranged from 0.70 to 1.0 in the REAG-S patterns, and from 0.35 to 1.0 in the REAG-B patterns. Overall, the combination of the three different PFGE methods identified 31 distinct types, reflecting the results obtained using the REAG-B alone different. different Five PFGE types were shared among 22 isolates from 12 patients. These types of strains were more frequently associated with central venous catheter-related fungemia than the other 26 type strains $(92\%\;versus\;31\%;\;P<0.005)$. Of five PFGE types, four isolates were determined to be epidemiologically related: each of these types was primarily from two or three patients who had been hospitalized concurrently within the same intensive care unit. Our results suggest that the REAG-B constitutes perhaps the most useful PFGE method for investigating C. albicans candidemia and also shows that a relatively high proportion of C. albicans candidemia may be associated with exogenous acquisition of clonal strains.
Kunitz-type soybean trypsin inhibitor (SBTI) and chondroitin sulfate (A, and C type) were conjugated using sodium periodate method. And the physicochemical, pharmacokinetic properties and immunogenecity of the conjugates (Chon-A-SBTI or Chon-C-SBTI) were characterized. We expected the conjugation using chondroitin sulfate to reduce the immunogenecity and to improve the pharmacological effect. As the results, the mean molecular weight of the conjugate highly increased. After I.V. injection of the radiolabeled conjugates or native SBTI into mice, it was found that native SBTI showed rapid elimination from plasma, whereas Chon-A-SBTI and Chon-C-SBTI were slowly eliminated. Organ distribution of the two agents at 30 min after I.V. injection was different : Chon-A-SBTI or Chon-C-SBTI accumulated to a large extent in the liver (13% in Chon-A-SBTI and 16% in Chon-C-SBTI), whereas native SBTI was taken up more rapidly by the kidney (107% dose/g of tissue) and excreated into the urine (26%). In addition we evaluated the therapeutic value of the conjugates by using the sublethal septic shock model caused by pseudomonal elastase and tested the immunogenecity by passive cutaneous anaphylaxis shock (PCA). The conjugates were more effective than native SBTI against pseudomonal elastase induced septic shock in guinea pig. In case of the conjugates, the pharmacological and therapeutic effect lasted over 3 hours long. In immunogenecity test, both of the conjugates showed the reduction of their immunogenecity, especially Chon-A-SBTI looked most effective.
Peroxidase를 다량 함유하는 식물체를 검색할 목적으로 십자화과 식물 9종에 대하여 효소활성 분포를 조사하였다. 검색한 식물체 중 배추 뿌리에서 본 효소의 활성이 강하게 나타나 배추 뿌리로 부터 본 효소의 정제를 행하였다. 정제된 효소는 분자량 50,000의 단량체로서 pH 7.0, $50^{\circ}C$에서 최적 활성을 나타내었다. phenol 및 phenol이 유도체가 좋은 기질로 작용하였으며 pyrogallol에 대한 $H_2O_2$의 Km 값은 1.6mM이었다. 본 효소는 406nm 에서 soret 흡수대를 나타내어 다른 peroxidase와 마찬가지로 분자내 보결분자단으로 heme을 가지고 있음을 알았다. 정제된 배추뿌리 기원의 효소는 면역화학적, 전기영동적 성질이 horseradish 기원의 peroxidase와 유사하였다.
본 연구에서는 바이오매스의 탄화공정 시 발생되는 목초액을 응축성 가스물질의 전구물질로 이용하여 발생되는 응축성 가스상 오염물질을 제거하기 위하여 액체상 흡착제중 실리카 전구물질인 HMDS의 주입조건에 따른 응축성물질의 입경분포 변화특성을 명확히 파악하고자 하였다. 응축성 가스상 물질을 입자화시켜 제거하기 위하여 액체상 흡착제인 HMDS를 이용할 경우, 흡착제의 주입량, 체류시간 및 가열온도, MFC 유량 등의 적절한 조절로써 입자화 크기를 조절할 수 있었다. 특히 실리카 전구물질을 도입시 끓는점, 밀도(비중), 분자량 등의 물리화학적 특성을 고려하면, 발생되는 응축성 물질을 효과적으로 입자화시켜 제어 가능할 것으로 판단된다.
나도국수나무족(Neillieae)은 전통적으로 나도국수나무속(Neillia), 국수나무속(Stephanandra), 산국수나무속(Physocarpus)의 3속으로 구성된 장미과의 분류군으로 인식되어 왔다. 그러나, 국수나무속은 최근의 분자계통학적 연구에 근거하여 나도국수나무속에 포함되어, 나도국수나무족은 나도국수나무속과 산국수나무속의 2속으로 구성된 족으로 인식된다. 나도국수나무속은 난형 내지 피침형의 잎, 점첨두 내지 꼬리형의 엽선, 총상 또는 원추화서 및 단심피의 암술을 갖는 특징에 의해 산국수나무속과 뚜렷이 구분된다. 본 속의 식물은 히말라야에서 중국 및 한국을 거쳐 동쪽으로 일본, 남쪽으로 인도네시아에 걸쳐 분포한다. 본 연구에서는 나도국수나무족의 종속지적 연구의 일환으로 나도국수나무속의 분류를 제시하였다. 기준표본을 포함한 표본 조사 및 야외 조사를 바탕으로 형태형질 분석 결과와 현재까지 밝혀진 분자계통학적 연구 결과를 토대로 각 종에 대한 검색표, 모든 학명에 대한 자세한 명명법적인 검증 및 처리를 포함하여 나도국수나무속 및 각 종에 대한 기재, 분포도 및 조사한 표본의 목록을 제시하였다. 본 연구에서는 나도국수나무속에 12종을 인식하였으며, 이 중 3종에서 총 10 변종을 인식하였다. 또한, N. sinensis f. glanduligera 및 N. thyrsiflora의 학명에 대한 선정기준표본을 지정하였다.
For the prolonged delivery and sustained release rates of low molecular weight drugs, poly(lactic-co-glycolic acid) (PLGA) microparticles containing the drug SKL-2020 have been investigated. On increasing polyvinyl alcohol (PVA) concentration (from 0.2% to 5%), the size of microparticles decreased (from $48.02{\mu}m$ to $10.63{\mu}m$) and more uniform size distribution was noticeable due to the powerful emulsifying ability of PVA. A higher drug loading (from 5% to 20%) caused a larger concentration gradient between 2 phases at the polymer precipitation step; this resulted in decreased encapsulation efficiency (from 34.19% to 25.67%) and a greater initial burst (from 61.71% to 70.05%). SKL-2020-loaded PLGA microparticles prepared with different fabrication conditions exhibited unique release patterns of SKL-2020. High PVA concentration and high drug loading led to an initial burst effect by rapid drug diffusion through the polymer matrix. Since PLGA microparticles enabled the slow release of SKL-2020 over 1 week in vitro and in vivo, more convenient and comfortable treatment could be facilitated with less frequent administration. It is feasible to design a release profile by mixing microparticles that were prepared with different fabrication conditions. By this method, the initial burst could be repressed properly and drug release rate could decrease.
The OH($X^2{\Pi},\;{\nu}$"=0, 1) internal state distributions from the reaction of electronically ground state oxygen atoms with HSi$Cl_3$ were measured using laser-induced fluorescence. The ground-state O$(^3P_J)$ atoms with kinetic energies above the reaction barrier were produced by photolysis of N$O_2$ at 355 nm. The OH product revealed strong vibrational population inversion, P(${\nu}$"=1)/P(${\nu}$"=0) = 4.0 ${\pm}$ 0.6, and rotational distributions in both vibrational states exhibit substantial rotational excitations to the limit of total available energy. However, no preferential populations in either of the two $\Lambda$ doublet states were observed from the micropopulations, which supports a mechanism involving a direct abstraction of hydrogen by the atomic oxygen. It was also found that the collision energy between O and HSi$Cl_3$ is effectively coupled into the excitation of the internal degrees of freedom of the OH product ($$ = 0.62, and $<\;f_{rot}>$ = 0.20). The dynamics appear consistent with expectations for the kinematically constrained reaction which supports the reaction type, heavy + light-heavy $\rightarrow$ heavy-light + heavy (H + LH′ $\rightarrow$ HL + H′). The dynamics of oxygen atom collision with HSi$Cl_3$ are discussed in comparison to those with Si$H_4$.

Surface sediments(0~5 cm) were sampled from 22 stations in Ulsan Bay, one of the most highly industrialized regions in Korea, in November 2000. The sediment samples were analyzed for their polycyclic aromatic hydrocarbon(PAH) content using a gas chromatography coupled to a mass spectrometer detector(GC/MSD). The total PAH concentrations in the sediments varied from 14 to 7108 ng/g dry weight with a mean value of 1052 ng/g dry weight. The level of carcinogenic PAHs ranged from 6 to 2396 ng/g dry weight with a mean value of 433 ng/g dry weight. The highest PAH concentrations in the sediments from Ulsan Bay were found at Station U8, whereas the lowest levels were observed at Stations U2 and U 17. The PAH distribution exhibited a decreasing gradient from the inner basin to the outer bay. The predominant contributors to the aromatic ring groups of the 16 PAHs were four- and five-ring groups, such as fluoranthene, pyrene, benzo[a]anthracene, chrysene, benzor[b]fluoranthene, benzor[k]fluoranthene and benzo[a]pyrene, while two- and three-ring aromatics, like naphthalene, acenapthylene, acenaphthene, fluorene, phenanthrene and anthracene, only exhibited a low concentration. The molecular indices for phenanthrene/anthracene and fluoranthen/pyrene were used to Identify the origin of the PAH contamination in the sediments. The results indicated that the PAH contamination in Ulsan Bay was mostly Pyrolytic in origin with a Petrogenic input adjacent to Ulsan and Jangsuengpo harbor.
One of the characteristics of polycrystalline graphene that determines its material properties is grain size. Mechanical properties such as Young's modulus, yield strain and tensile strength depend on the grain size and show a reverse Hall-Petch effect at small grain size limit for some properties under certain conditions. While there is agreement on the grain size effect for Young's modulus and yield strain, certain MD simulations have led to disagreement for tensile strength. Song et al. showed a decreasing behavior for tensile strength, that is, a pseudo Hall-Petch effect for the small grain size domain up to 5 nm. On the other hand, Sha et al. showed an increasing behavior, a reverse Hall-Petch effect, for grain size domain up to 10 nm. Mortazavi et al. also showed results similar to those of Sha et al. We suspect that the main difference of these two inconsistent results is due to the different modeling. The modeling of polycrystalline graphene with regular size and (hexagonal) shape shows the pseudo Hall-Petch effect, while the modeling with random size and shape shows the reverse Hall-Petch effect. Therefore, this study is conducted to confirm that different modeling is the main reason for the different behavior of tensile strength of the polycrystalline structures. We conducted MD simulations with models derived from the Voronoi tessellation for two types of grain size distributions. One type is grains of relatively similar sizes; the other is grains of random sizes. We found that the pseudo Hall-Petch effect and the reverse Hall-Petch effect of tensile strength were consistently shown for the two different models. We suspect that this result comes from the different crack paths, which are related to the grain patterns in the models.
